    Decrease of T-cells exhaustion markers programmed cell death-1 and T-cell immunoglobulin and mucin domain-containing protein 3 and plasma IL-10 levels after successful treatment of chronic hepatitis C

    During chronic hepatitis C virus (HCV) infection, both CD4+^{+} and CD8+^{+} T-cells become functionally exhausted, which is reflected by increased expression of programmed cell death-1 (PD-1) and T-cell immunoglobulin and mucin domain-containing protein 3 (Tim-3), and elevated anti-inflammatory interleukin 10 (IL-10) plasma levels. We studied 76 DAA-treated HCV-positive patients and 18 non-infected controls. Flow cytometry measured pretreatment frequencies of CD4+^{+}PD-1+^{+}, CD4+^{+}PD-1+^{+}Tim-3+^{+} and CD8+^{+}PD-1+^{+}Tim-3+^{+} T-cells and IL-10 levels measured by ELISA were significantly higher and CD4+^{+}PD-1^{-}Tim-3^{-} and CD8+^{+}PD-1^{-}Tim-3^{-} T-cells were significantly lower in patients than in controls. Treatment resulted in significant decrease of CD4+^{+}Tim-3+^{+}, CD8+^{+}Tim-3+^{+}, CD4+^{+}PD-1+^{+}Tim-3+^{+} and CD8+^{+}PD-1+^{+}Tim-3+^{+} T-cell frequencies as well as IL-10 levels and increase in CD4+^{+}PD-1^{-}Tim-3^{-} and CD8+^{+}PD-1^{-}Tim-3^{-} T-cells. There were no significant changes in the frequencies of CD4+^{+}PD-1+^{+} T-cells, while CD8+^{+}PD-1+^{+} T-cells increased. Patients with advanced liver fibrosis had higher PD-1 and lower Tim-3 expression on CD4+^{+}T-cells and treatment had little or no effect on the exhaustion markers. HCV-specific CD8+^{+}T-cells frequency has declined significantly after treatment, but their PD-1 and Tim-3 expression did not change. Successful treatment of chronic hepatitis C with DAA is associated with reversal of immune exhaustion phenotype, but this effect is absent in patients with advanced liver fibrosis

    Experimental investigation of mid-infrared laser action from DY3+ doped fluorozirconate fiber

    Efficient continuous-wave laser operation at 2.982 μm is achieved with a Dy3:fluoride fiber pumped using an inhouse-built 1.1 μm ytterbium (III) fiber laser. The laser output power reached is 554 mW, with a maximum slope efficiency of 18% with respect to the launched pump power. Additionally, the measured spontaneous luminescence within the visible wavelength range, under 1.1 μm pumping, is presented and attributed to excited state absorption (ESA). The influence of the ESA on the laser performance is discussed. The results confirm that high output powers from Dy: fluoride fiber laser pumped at 1.1 μm are possible

    Climate indices of environmental change in the High Arctic: Study from Hornsund, SW Spitsbergen, 1979–2019

    An analysis of a suite of climatological indices was undertaken on the basis of long-term (1979–2019) climatological data from the Polish Polar Station in Hornsund, SW Spitsbergen. It was followed by an attempt to assess the scale of their impact on the local environment. The temperature and precipitation indices were based on percentiles of the variables calculated for a population of daily values from the climate normals for 1981–2010. A greater share of both cyclonic and anticyclonic circulations from the S and SW sectors, forcing the advection of warm air masses from the south, was decisive for the trends of change in comparison with the long-term mean. Both extreme precipitation and drought events depend on the 500 hPa geopotential height and precipitable water anomalies, determined by the baric field over the North Atlantic. Climate changes impact on the dynamics of local geoecosystems by causing faster glacier ablation and retreat, permafrost degradation, intensification of the hydrological cycle in glaciated and unglaciated catchments, and changes in the condition and growth of tundra vegetation

    Nuclear survivin expression is a positive prognostic factor in taxane-platinum-treated ovarian cancer patients

    <p>Abstract</p> <p>Background</p> <p>Survivin is an inhibitor of apoptosis and a regulator of mitotic progression. TP53 protein is a negative transcriptional regulator of survivin. The aim of our study was to evaluate the clinical significance of survivin expression in advanced stages ovarian cancer with respect to the TP53 status.</p> <p>Methods</p> <p>Survivin and TP53 expression was evaluated immunohistochemically in 435 archival samples of ovarian carcinomas (244 patients were treated with platinum/cyclophosphamide-PC/PAC; 191-with taxane-platinum (TP) agents). Univariate and multivariate statistical analyses were performed in patients groups divided according to the administered chemotherapeutic regimen, and in subgroups with and without TP53 accumulation (TP53+ and TP53-, respectively).</p> <p>Results</p> <p>Nuclear and cytoplasmic survivin expression was observed in 92% and 74% of the carcinomas, respectively. In patients treated with TP, high nuclear survivin expression decreased the risk of disease recurrence and death, and increased the probability of high platinum sensitivity (p < 0.01), but only in the TP53(+) group, and not in the TP53(-) group.</p> <p>Conclusions</p> <p>It appears that TP53 status determines the clinical importance of nuclear survivin expression in taxane-platinum treated ovarian cancer patients.</p
