    Mechanisms of flame spread and burnout in large enclosure fires

    Knowledge of the first principles defining fire behaviour in large enclosures remains limited despite their common use in modern tall buildings. The evolution of a fire in large enclosures can be defined by the relationship between the flame front and burnout velocities (VS/VBO). This relationship can be classified into three distinct fire spread modes being Mode 1 (VS/VBO → ∞), Mode 2 (VS/VBO > 1), and Mode 3 (VS/VBO ≈ 1). The mechanisms governing flame spread and burnout are investigated using four full-scale enclosure fire experiments with high porosity wood cribs with similar enclosure geometries. Flame and burnout front positions and velocities are estimated using video data. Velocities are affected by the heat feedback from the enclosure and smoke layer to the fuel. The spread velocity shows two regimes, a minimum external heat flux above which there is surface spread (q''s,min) and a heat flux that defines the onset of very rapid flame spread ((q''rs,crit)). A phenomenological model is developed to help identify the underlying mechanisms controlling the transition between the different spread modes. Both the model and data show that for wood cribs, the dependence of the burnout front velocity to the external radiation is weak, whereas the dependence of the flame spread velocity to the external and flame heat flux is strong. A transition from Mode 3 (VS/VBO ≈ 1) to Mode 2 (VS/VBO > 1) occurs with increasing external heat fluxes above q''s,min. The transition to Mode 1 (VS/VBO → ∞) is further defined once (q''rs,crit) is attained due to a sudden increase in the flame heat flux by changing the ventilation condition, or by significant increases in the external heat flux from the enclosure

    Uncertainty-based decision-making in fire safety: Analyzing the alternatives

    Large accidents throughout the 20th century marked the development of safety fields in engineering, devoted to better identify hazards, understand risks and properly manage them. As these fields evolved rather quickly and moved from a compliance to a risk-based approach, a significant delay in this transition was experienced in fire safety engineering (FSE). Devastating fires well into the 21st century and the restrictive nature of prescriptive codes signaled the need to transition towards a performance-based one. A performance-based approach provides flexibility and capitalizes on learning from accidental events and engineering disciplines such as process safety and FSE. This work provides an overview of the main alternatives to account for uncertainty in safety studies within the context of FSE, including traditional probabilistic analyses and emerging approaches such as strength of knowledge. A simple example is used to illustrate the impact of the uncertainty analysis on the results of a simple fire safety assessment. A structured evaluation is performed on each alternative to assess its ease of implementation and communication. The outcome is a compendium of advantages and disadvantages of the alternatives that constitute a toolbox for fire safety engineers to configure and use within their fire risk assessments. Process safety engineers are expected to gain an understanding of the similar and important challenges of FSE, being it directly relevant for process risk management and fire risk management in administrative buildings

    Overcoming risk assessment limitations for potential fires in a multi-occupancy building

    Decision-making under risk has been a key issue in systems with a potential for major losses such as chemical process industries (Bhopal - 1984, Toulouse - 2001) or high occupancy buildings (World Trade Center - 2001, Grenfell Tower - 2017). For the past decades, engineering disciplines have supported risk management decision-making through the implementation of risk assessments using quantitative approaches. The popularity of this approach relates to the quantitative definition of risk given by Kaplan in 1981, who decomposed risk into a set of scenarios, probability of occurrence and consequences. Recently, research on quantitative risk assessments (QRA) has reported key limitations on identifying the set of scenarios and estimating their probability of occurrence. These limitations may lead to uncertainties of up to three orders of magnitude that affect the QRA’s ability of delivering reliable information to stakeholders. This research uses an alternative definition of risk and applies it to a case study of a multi-occupancy building in the event of a fire. The proposed approach quantifies the maximum damage potential (MDP) of the system when all the active safety measures are allowed to fail, even those with low failure frequencies. The system’s MDP is compared to its maximum allowable damage (MAD), which is previously defined by the stakeholders. This approach allows defining design modifications and operational rules aiding the development of the building’s fire safety strategy. Finally, a comparison between the obtained results and a typical QRA is used to comment on the suitability of the proposed approach when evaluating risk in complex systems

    Towards a better understanding of fire performance assessment of façade systems: Current situation and a proposed new assessment framework

    This manuscript presents tools and data that serve to enable an evaluation of the risk associated with vertical fire spread on buildings. A highly detailed context to cladding fires is described to unveil the complexity and magnitude of the problem and to identify gaps of information. An engineering framework is then developed which delivers required information that fills some of those gaps and that needs to be used towards achieving quantified fire performance. The data itself has been published as a publicly available database, entitled the Cladding Materials Library (www.claddingmaterialslibrary.com.au). This data can be used to support building fire risk assessments or as the basis for more in-depth research into façade fires. This paper presents the context of the data together with the competency framework necessary for upskilling building professionals to have the capacity to implement the engineering framework

    Sentinel network for monitoring in vitro susceptibility of Plasmodium falciparum to antimalarial drugs in Colombia: a proof of concept

    Drug resistance is one of the principal obstacles blocking worldwide malaria control. In Colombia, malaria remains a major public health concern and drug-resistant parasites have been reported. In vitro drug susceptibility assays are a useful tool for monitoring the emergence and spread of drug-resistant Plasmodium falciparum. The present study was conducted as a proof of concept for an antimalarial drug resistance surveillance network based on in vitro susceptibility testing in Colombia. Sentinel laboratories were set up in three malaria endemic areas. The enzyme linked immunosorbent assay-histidine rich protein 2 and schizont maturation methods were used to assess the susceptibility of fresh P. falciparum isolates to six antimalarial drugs. This study demonstrates that an antimalarial drug resistance surveillance network based on in vitro methods is feasible in the field with the participation of a research institute, local health institutions and universities. It could also serve as a model for a regional surveillance network. Preliminary susceptibility results showed widespread chloroquine resistance, which was consistent with previous reports for the Pacific region. However, high susceptibility to dihydroartemisinin and lumefantrine compounds, currently used for treatment in the country, was also reported.     Magnetoresistance through a single molecule

    The use of single molecules to design electronic devices is an extremely challenging and fundamentally different approach to further downsizing electronic circuits. Two-terminal molecular devices such as diodes were first predicted [1] and, more recently, measured experimentally [2]. The addition of a gate then enabled the study of molecular transistors [3-5]. In general terms, in order to increase data processing capabilities, one may not only consider the electron's charge but also its spin [6,7]. This concept has been pioneered in giant magnetoresistance (GMR) junctions that consist of thin metallic films [8,9]. Spin transport across molecules, i.e. Molecular Spintronics remains, however, a challenging endeavor. As an important first step in this field, we have performed an experimental and theoretical study on spin transport across a molecular GMR junction consisting of two ferromagnetic electrodes bridged by a single hydrogen phthalocyanine (H2Pc) molecule. We observe that even though H2Pc in itself is nonmagnetic, incorporating it into a molecular junction can enhance the magnetoresistance by one order of magnitude to 52%.Comment: To appear in Nature Nanotechnology.     Attenuated Leishmania induce pro-inflammatory mediators and influence leishmanicidal activity by p38 MAPK dependent phagosome maturation in Leishmania donovani co-infected macrophages

    Promastigote form of Leishmania, an intracellular pathogen, delays phagosome maturation and resides inside macrophages. But till date limited study has been done to manipulate the phagosomal machinery of macrophages to restrict Leishmania growth. Attenuated Leishmania strain exposed RAW 264.7 cells showed a respiratory burst and enhanced production of pro-inflammatory mediators. The augmentation of pro-inflammatory activity is mostly attributed to p38 MAPK and p44/42 MAPK. In our study, these activated macrophages are found to induce phagosome maturation when infected with pathogenic Leishmania donovani. Increased co-localization of carboxyfluorescein succinimidyl ester labeled pathogenic L. donovani with Lysosome was found. Moreover, increased co-localization was observed between pathogenic L. donovani and late phagosomal markers viz. Rab7, Lysosomal Associated Membrane Protein 1, Cathepsin D, Rab9, and V-ATPase which indicate phagosome maturation. It was also observed that inhibition of V-type ATPase caused significant hindrance in attenuated Leishmania induced phagosome maturation. Finally, it was confirmed that p38 MAPK is the key player in acidification and maturation of phagosome in attenuated Leishmania strain preexposed macrophages.     Gaze following is modulated by expectations regarding others’ action goals

    Humans attend to social cues in order to understand and predict others' behavior. Facial expressions and gaze direction provide valuable information to infer others' mental states and intentions. The present study examined the mechanism of gaze following in the context of participants' expectations about successive action steps of an observed actor. We embedded a gaze-cueing manipulation within an action scenario consisting of a sequence of naturalistic photographs. Gaze-induced orienting of attention (gaze following) was analyzed with respect to whether the gaze behavior of the observed actor was in line or not with the action-related expectations of participants (i.e., whether the actor gazed at an object that was congruent or incongruent with an overarching action goal). In Experiment 1, participants followed the gaze of the observed agent, though the gaze-cueing effect was larger when the actor looked at an action-congruent object relative to an incongruent object. Experiment 2 examined whether the pattern of effects observed in Experiment 1 was due to covert, rather than overt, attentional orienting, by requiring participants to maintain eye fixation throughout the sequence of critical photographs (corroborated by monitoring eye movements). The essential pattern of results of Experiment 1 was replicated, with the gaze-cueing effect being completely eliminated when the observed agent gazed at an action-incongruent object. Thus, our findings show that covert gaze following can be modulated by expectations that humans hold regarding successive steps of the action performed by an observed agent

    Leaf colour as a signal of chemical defence to insect herbivores in wild cabbage (Brassica Oleracea)

    Leaf colour has been proposed to signal levels of host defence to insect herbivores, but we lack data on herbivory, leaf colour and levels of defence for wild host populations necessary to test this hypothesis. Such a test requires measurements of leaf spectra as they would be sensed by herbivore visual systems, as well as simultaneous measurements of chemical defences and herbivore responses to leaf colour in natural host-herbivore populations. In a large-scale field survey of wild cabbage (Brassica oleracea) populations, we show that variation in leaf colour and brightness, measured according to herbivore spectral sensitivities, predicts both levels of chemical defences (glucosinolates) and abundance of specialist lepidopteran (Pieris rapae) and hemipteran (Brevicoryne brassicae) herbivores. In subsequent experiments, P. rapae larvae achieved faster growth and greater pupal mass when feeding on plants with bluer leaves, which contained lower levels of aliphatic glucosinolates. Glucosinolate-mediated effects on larval performance may thus contribute to the association between P. rapae herbivory and leaf colour observed in the field. However, preference tests found no evidence that adult butterflies selected host plants based on leaf coloration. In the field, B. brassicae abundance varied with leaf brightness but greenhouse experiments were unable to identify any effects of brightness on aphid preference or performance. Our findings suggest that although leaf colour reflects both levels of host defences and herbivore abundance in the field, the ability of herbivores to respond to colour signals may be limited, even in species where performance is correlated with leaf colour

    The Role of MMP7 and Its Cross-Talk with the FAS/FASL System during the Acquisition of Chemoresistance to Oxaliplatin

    Background: The efficacy of oxaliplatin in cancer chemotherapy is limited by the development of drug resistance. MMP7 has been related to the loss of tumor cell response to cytotoxic agents although the exact mechanism is not fully understood. Moreover, MMP7 is an independent prognosis factor for survival in patients with colorectal cancer. The aim of the present study was to analyze the role of MMP7 and its cross-talk with the Fas/FasL system during the acquisition of oxaliplatin resistance in colon cancer cells. Principal Findings: For this purpose we have developed three different oxaliplatin-resistant cell lines (RHT29, RHCT116 p53+/+, RHCT116 p53−/−) from the parental HT29, HCT116 p53+/+ and HCT116 p53−/− colon cancer cells. MMP7 basal expression was higher in the resistant compared to the parental cell lines. MMP7 was also upregulated by oxaliplatin in both HT29 (p53 mutant) and RHCT116 p53−/− but not in the RHCT116 p53+/+. Inhibition of MMP by 1,10-phenantroline monohydrate or siRNA of MMP7 restores cell sensitivity to oxaliplatin-induced apoptosis in both HT29 and RHCT116 p53−/− but not in the RHCT116 p53+/+. Some of these effects are caused by alterations in Fas receptor. Fas is upregulated by oxaliplatin in colon cancer cells, however the RHT29 cells treated with oxaliplatin showed a 3.8-fold lower Fas expression at the cell surface than the HT29 cells. Decrease of Fas at the plasma membrane seems to be caused by MMP7 since its inhibition restores Fas levels. Moreover, functional analysis of Fas demonstrates that this receptor was less potent in inducing apoptosis in RHT29 cells and that its activation induces MAPK signaling in resistant cells. Conclusions: Taking together, these results suggest that MMP7 is related to the acquisition of oxaliplatin-resistance and that its inhibition restores drug sensitivity by increasing Fas receptor. Furthermore, Fas undergoes a change in its functionality in oxaliplatin-resistant cells inducing survival pathways instead of apoptotic signals
