Identification of human intestinal parasites affecting an asymptomatic peri-urban Argentinian population using multi-parallel quantitative real-time polymerase chain reaction
Background: In resource-limited countries, stool microscopy is the diagnostic test of choice for intestinal parasites (soil-transmitted helminths and/or intestinal protozoa). However, sensitivity and specificity is low. Improved diagnosis of intestinal parasites is especially important for accurate measurements of prevalence and intensity of infections in endemic areas. Methods: The study was carried out in Orán, Argentina. A total of 99 stool samples from a local surveillance campaign were analyzed by concentration microscopy and McMaster egg counting technique compared to the analysis by multi-parallel quantitative real-time polymerase chain reaction (qPCR). This study compared the performance of qPCR assay and stool microscopy for 8 common intestinal parasites that infect humans including the helminths Ascaris lumbricoides, Ancylostoma duodenale, Necator americanus, Strongyloides stercoralis, Trichuris trichiura, and the protozoa Giardia lamblia, Cryptosporidium parvum/hominis, and Entamoeba histolytica, and investigated the prevalence of polyparasitism in an endemic area. Results: qPCR showed higher detection rates for all parasites as compared to stool microscopy except T. trichiura. Species-specific primers and probes were able to distinguish between A. duodenale (19.1 %) and N. americanus (36.4 %) infections. There were 48.6 % of subjects co-infected with both hookworms, and a significant increase in hookworm DNA for A. duodenale versus N. americanus (119.6 fg/μL: 0.63 fg/μL, P∈<∈0.001) respectively. qPCR outperformed microscopy by the largest margin in G. lamblia infections (63.6 % versus 8.1 %, P∈<∈0.05). Polyparasitism was detected more often by qPCR compared to microscopy (64.7 % versus 24.2 %, P∈<∈0.05). Conclusions: Multi-parallel qPCR is a quantitative molecular diagnostic method for common intestinal parasites in an endemic area that has improved diagnostic accuracy compared to stool microscopy. This first time use of multi-parallel qPCR in Argentina has demonstrated the high prevalence of intestinal parasites in a peri-urban area. These results will contribute to more accurate epidemiological survey, refined treatment strategies on a public scale, and better health outcomes in endemic settings.Fil: Cimino, Rubén Oscar. Validación de una metodología analítica para la cuantificación de polifenoles totales, en procesos de extracción asistida por microondas sobre frutos de la especie colombiana Vaccinium meridionale
En este trabajo se muestra el desarrollo y la validación de un método analítico para la cuantificación de polifenoles totales con el reactivo de Folin-Ciocalteu (F–C),en procesos de extracción asistida por microondas (MAE), sobre frutos de la especie colombiana Vaccinium meridionale. Los resultados obtenidos en los parámetros selectividad, linealidad, repetibilidad y exactitud muestran que la metodología propuesta es confiable para evaluar el efecto de las condiciones de extracción sobre la cantidad de polifenoles removidos.The in vitro pharmacological model of P. falciparum culture is crucial in the initial screening for substances or plant extracts with possible antiplasmodial activity. The parasite density can be determined by varied methods, however have been described numerous advantages and disadvantages associated with each of them.The incubation time required for staining and the use of synchronous or asynchronous cultures were assessed for optimal settings; showing optimal time of 2 h, and lower limits of detection and quantification in asynchronous cultures. Employing the FCB2 and FCR3 strains, was evidenced a background noise of 12% and 38% respectively; linearity showed a good correlation, r2 of 0.9644 (FCR3) and 0.9841 (FCB2) and a slope of 1761.8 and 852.4, respectively. It was evidenced agreement between the methods, fluorometric with SYBR Green I (SYBRG I) and microscopic with Giemsa, the mean difference was 0.00002% and 0.09109% respectively for FCR3 and FCB2, The limits of detection and quantification were 0.5% and 1.5% of parasitaemia. The Z factor was 0.376 with FCB2, whereas with FCR3 reached 0.702. The inhibitory concentration 50 (IC50) against P. falciparum FCR3, generated by chloroquine (CQ), was 0.37 mcg/mL by microscopy and 0.35 mcg/mL by fluorometry.Our findings suggest that the SYBRG I fluorescence based assay, by using fluorometers commonly available in many laboratories, is precise, robust, fast and accurate; for the in vitro evaluation of substances or extracts with possible antiplasmodial activity
Leveraging Automated Image Analysis Tools to Transform Our Capacity to Assess Status and Trends of Coral Reefs
Digital photography is widely used by coral reef monitoring programs to assess benthic status and trends. In addition to creating a permanent archive, photographic surveys can be rapidly conducted, which is important in environments where bottom-time is frequently limiting. However, substantial effort is required to manually analyze benthic images; which is expensive and leads to lags before data are available. Using previously analyzed imagery from NOAA’s Pacific Reef Assessment and Monitoring Program, we assessed the capacity of a trained and widely used machine-learning image analysis tool – CoralNet coralnet.ucsd.edu – to generate fully-automated benthic cover estimates for the main Hawaiian Islands (MHI) and American Samoa. CoralNet was able to generate estimates of site-level coral cover for both regions that were highly comparable to those generated by human analysts (Pearson’s r > 0.97, and with bias of 1% or less). CoralNet was generally effective at estimating cover of common coral genera (Pearson’s r > 0.92 and with bias of 2% or less in 6 of 7 cases), but performance was mixed for other groups including algal categories, although generally better for American Samoa than MHI. CoralNet performance was improved by simplifying the classification scheme from genus to functional group and by training within habitat types, i.e., separately for coral-rich, pavement, boulder, or “other” habitats. The close match between human-generated and CoralNet-generated estimates of coral cover pooled to the scale of island and year demonstrates that CoralNet is capable of generating data suitable for assessing spatial and temporal patterns. The imagery we used was gathered from sites randomly located in <30 m hard-bottom at multiple islands and habitat-types per region, suggesting our results are likely to be widely applicable. As image acquisition is relatively straightforward, the capacity of fully-automated image analysis tools to minimize the need for resource intensive human analysts opens possibilities for enormous increases in the quantity and consistency of coral reef benthic data that could become available to researchers and managers
Efecto antiinflamatorio de la izalpinina derivada de Chromolaena leivensis: edema de la pata inducido por λ-carragenina y modelo in silico
El flavonoide izalpinina se aisló de las partes aéreas de Chromolaena leivensis. Su determinación estructural se llevó a cabo mediante técnicas espectroscópicas de EM y RMN (1H, 13C). Se evaluó el efecto antiinflamatorio de este compuesto en un modelo de edema plantar inducido por carragenina en ratas. La inflamación de la pata se midió a intervalos de una hora durante siete horas tras la administración de -carragenano. Se evaluaron los niveles séricos de creatina quinasa (CK), obteniéndose resultados estadísticamente significativos con los tratamientos a dosis de 10 mg/kg (* p < 0,01) y 20 mg/kg (** p < 0,005). El efecto antiinflamatorio del compuesto se evaluó mediante pletismografía, y los resultados mostraron diferencias significativas en las tres concentraciones (10 mg/kg, 20 mg/kg, 40 mg/kg) en la primera y tercera hora tras el tratamiento. * p < 0,05; ** p < 0,001; **** p < 0,0001 frente al grupo de control negativo tratado con vehículo (DMSO). Por último, los análisis de acoplamiento molecular revelan que la izalpinina tiene una fuerte afinidad de unión con cinco proteínas diana implicadas en el proceso inflamatorio. El análisis mediante dinámica molecular permitió demostrar que los complejos ligando-proteína presentan una estabilidad aceptable, con valores de RMSD dentro del rango permitido.The flavonoid izalpinin was isolated from the aerial parts of Chromolaena leivensis. Its structural determination was carried out using MS and NMR spectroscopic techniques (1H, 13C). This compound was evaluated for its anti-inflammatory effect in a rat model on -carrageenan-induced plantar edema. Paw inflammation was measured at one-hour intervals for seven hours following the administration of -carrageenan. Serum creatine kinase (CK) levels were evaluated, obtaining statistically significant results with the treatments at doses of 10 mg/kg (* p < 0.01) and 20 mg/kg (** p < 0.005). The anti-inflammatory effect of the compound was evaluated by using plethysmography, and the results showed significant differences at the three concentrations (10 mg/kg, 20 mg/kg, 40 mg/kg) in the first and third hours after treatment. * p < 0.05; ** p < 0.001; **** p < 0.0001 vs. the negative control group treated with vehicle (DMSO). Lastly, molecular docking analyses reveal that izalpinin has a strong binding affinity with five target proteins involved in the inflammatory process. The analysis using molecular dynamics allowed demonstrating that the ligand–protein complexes present acceptable stability, with RMSD values within the allowed range
Estrategias pedagógicas utilizadas para la enseñanza de las ciencias del movimiento humano y la percepción estudiantil acerca de su impacto en el aprendizaje
The purpose of this research was to analyze the pedagogical strategies used for the teaching of human movement sciences and their impact on the learning of higher education students. A qualitative study was designed, with focus group, participant observation and teacher evaluation questionnaire. Participants were 32 students from 2 public and 2 private universities; 12 teachers from the four universities also participated. The conclusion is that most of the teachers were perceived by the students as clearly transmitters of information with class environments superficial and not significant. On the other hand, it was shown that those teachers, who showed interest, respect and concern for their students, the class was considered significant to learn.El propósito de esta investigación fue analizar las estrategias pedagógicas utilizadas para la enseñanza de las ciencias del movimiento humano y su impacto en el aprendizaje de estudiantes de educación superior. Se diseñó un estudio de corte cualitativo, con un grupo focal, observación participante y un cuestionario de evaluación docente. Participaron 32 estudiantes de 2 universidades públicas y 2 privadas; también participaron 12 docentes entre las cuatro universidades. La conclusión es que la mayoría de los profesores fueron percibidos como netos transmisores de información; el ambiente de clase fue percibido como superficial y poco significativo. Por otro lado, se demostró que en el caso de aquellos docentes que mostraron interés, respeto y preocupación por sus alumnos, su clase fue considerada significativa para aprender

Irrigation and Fertigation with Drip and Alternative Micro Irrigation Systems in Northern Highbush Blueberry
The use of conventional drip and alternative micro irrigation systems were evaluated for 3 years in six newly planted cultivars (Earliblue, Duke, Draper, Bluecrop, Elliott, and Aurora) of northern highbush blueberry (Vaccinium corymbosum L.). The drip system included two lines of tubing on each side of the row with in-line drip emitters at every 0.45 m. The alternative systems included geotextile tape and microsprinklers. The geotextile tape was placed alongside the plants and dispersed water and nutrients over the entire length. Microsprinklers were installed between every other plant at a height of 1.2 m. Nitrogen was applied by fertigation at annual rates of 100 and 200 kg·ha⁻¹ N by drip, 200 kg·ha⁻¹1 N by geotextile tape, and 280 kg·ha⁻¹ N by microsprinklers. By the end of the first season, plant size, in terms of canopy cover, was greatest with geotextile tape, on average, and lowest with microsprinklers or drip at the lower N rate. The following year, canopy cover was similar with geotextile tape and drip at the higher N rate in each cultivar, and was lowest with microsprinklers in all but ‘Draper’. In most of the cultivars, geotextile tape and drip at the higher N rate resulted in greater leaf N concentrations than microsprinklers or drip at the lower N rate, particularly during the first year after planting. By the third year, yield averaged 3.1–9.1 t·ha⁻¹ among the cultivars, but was similar with geotextile tape and drip at either N rate, and was only lower with microsprinklers. Overall, drip was more cost effective than geotextile tape, and fertigation with 100 kg·ha⁻¹ N by drip was sufficient to maximize early fruit production in each cultivar. Efficient NiII2LnIII2 electrocyclization catalysts for the synthesis of trans-4,5-diaminocyclopent-2-enones from 2-furaldehyde and primary or secondary amines
A series of heterometallic coordination clusters (CCs) [NiII2LnIII2(L1)4Cl2(CH3CN)2] 2CH3CN [Ln = Y (1Y), Sm (1Sm), Eu (1Eu), Gd (1Gd), or Tb (1Tb)] were synthesized by the reaction of (E)-2-(2-hydroxy-3-methoxybenzylidene-amino)phenol) (H2L1) with NiCl2·6(H2O) and LnCl3·x(H2O) in the presence of Et3N at room temperature. These air-stable CCs can be obtained in very high yields from commercially available materials and are efficient catalysts for the room-temperature domino ring-opening electrocyclization synthesis of trans-4,5-diaminocyclopent-2-enones from 2-furaldehyde and primary or secondary amines under a non-inert atmosphere. Structural modification of the catalyst to achieve immobilization or photosensitivity is possible without deterioration in catalytic activity
Impact of intestinal parasites on microbiota and cobalamin gene sequences: A pilot study
Background: Approximately 30% of children worldwide are infected with gastrointestinal parasites. Depending on the species, parasites can disrupt intestinal bacterial microbiota affecting essential vitamin biosynthesis. Methods: Stool samples were collected from 37 asymptomatic children from a previous cross-sectional Argentinian study. A multi-parallel real-time quantitative PCR was implemented for Ascaris lumbricoides, Ancylostoma duodenale, Necator americanus, Strongyloides stercoralis, Trichuris trichiura, Cryptosporidium spp., Entamoeba histolytica and Giardia duodenalis. In addition, whole-genome sequencing analysis was conducted for bacterial microbiota on all samples and analyzed using Livermore Metagenomic Analysis Toolkit and DIAMOND software. Separate analyses were carried out for uninfected, Giardia-only, Giardia + helminth co-infections, and helminth-only groups. Results: For Giardia-only infected children compared to uninfected children, DNA sequencing data showed a decrease in microbiota biodiversity that correlated with increasing Giardia burden and was statistically significant using Shannon's alpha diversity (Giardia-only > 1 fg/μl 2.346; non-infected group 3.253, P = 0.0317). An increase in diversity was observed for helminth-only infections with a decrease in diversity for Giardia + helminth co-infections (P = 0.00178). In Giardia-only infections, microbiome taxonomy changed from Firmicutes towards increasing proportions of Prevotella, with the degree of change related to the intensity of infection compared to uninfected (P = 0.0317). The abundance of Prevotella bacteria was decreased in the helminths-only group but increased for Giardia + helminth co-infections (P = 0.0262). Metagenomic analysis determined cobalamin synthesis was decreased in the Giardia > 1 fg/μl group compared to both the Giardia < 1 fg/μl and the uninfected group (P = 0.0369). Giardia + helminth group also had a decrease in cobalamin CbiM genes from helminth-only infections (P = 0.000754). Conclusion: The study results may provide evidence for an effect of parasitic infections enabling the permissive growth of anaerobic bacteria such as Prevotella, suggesting an altered capacity of vitamin B12 (cobalamin) biosynthesis and potential impact on growth and development in children.Fil: Mejia, Rojelio. Recomendaciones dirigidas a los familiares responsables del cuidado domiciliario de un paciente diagnosticado con Covid-19
The crisis that is causing the disease called COVID-19 is severely affecting the health system of many countries, which require alternative measures to care this large number of patients who are not able to be treated at medical centers. This is the reason why home care is considered an alternative, for which it must be very rigorous with the care of the sick person, since if it is not done in a proper way, the other members of the family home are under risk of contamination, which would increase the saturation of the health system. Indications such as the stay of the patient in one place, the use of protective barriers when entering this area, or the constant sanitization of hands, among other measures, can lead to the success of this type of actions, favoring the prompt recovery of the patient and the preservation of the health of the other members at home.La crisis que está ocasionando la enfermedad denominada COVID-19 está afectando severamente al sistema sanitario de diversos países, lo que obliga a tomar medidas alternativas para poder atender a una gran cantidad de pacientes que no pueden acceder a los centros de atención médica. Es por esta razón que se considera como una alternativa la atención domiciliaria para lo cual se debe ser muy riguroso en el cuidado de la persona afectada con COVID 19, ya que, si esto no se realiza de forma adecuada, los demás integrantes del hogar corren el riesgo de contaminarse, lo que incrementaría la saturación de los establecimientos de salud. Indicaciones como la permanencia del enfermo en un solo lugar, el uso de barreras de protección cuando se ingresa a esta área o la constante práctica de la higiene de manos, entre otras medidas, puede encaminar al éxito de este tipo de atención favoreciendo la pronta recuperación del paciente y la preservación de la salud de los demás habitantes del hogar.A atual crise que está causando a doença denominada COVID-19 está afetando gravemente o sistema de saúde de vários países, o que requer outras medidas a serem tomadas para poder atender este grande número de pacientes fora dos centros hospitalares. É por isso que a assistência domiciliar é considerada uma alternativa, para a qual deve ser muito rigorosa com o cuidado do doente, pois se não for realizada de forma adequada os demais membros do domicílio correm o risco de contaminar o que aumentaria a saturação das unidades de saúde.
Indicações como a permanência do paciente em um único local, o uso de barreiras protetoras na entrada nesta área ou a prática constante de higienização das mãos, entre outras medidas, podem levar ao sucesso desse tipo de atendimento, favorecendo uma recuperação rápida do paciente e a preservação da saúde dos demais habitantes.
 
Mortality and Advanced Support Requirement for Patients With Cancer With COVID-19 : A Mathematical Dynamic Model for Latin America
PURPOSE: In the midst of a global pandemic, evidence suggests that similar to other severe respiratory viral infections, patients with cancer are at higher risk of becoming infected by COVID-19 and have a poorer prognosis. METHODS: We have modeled the mortality and the intensive care unit (ICU) requirement for the care of patients with cancer infected with COVID-19 in Latin America. A dynamic multistate Markov model was constructed. Transition probabilities were estimated on the basis of published reports for cumulative probability of complications. Basic reproductive number (R0) values were modeled with R using the EpiEstim package. Estimations of days of ICU requirement and absolute mortality were calculated by imputing number of cumulative cases in the Markov model. RESULTS: Estimated median time of ICU requirement was 12.7 days, median time to mortality was 16.3 days after infection, and median time to severe event was 8.1 days. Peak ICU occupancy for patients with cancer was calculated at 16 days after infection. Deterministic sensitivity analysis revealed an interval for mortality between 18.5% and 30.4%. With the actual incidence tendency, Latin America would be expected to lose approximately 111,725 patients with cancer to SARS-CoV-2 (range, 87,116-143,154 patients) by the 60th day since the start of the outbreak. Losses calculated vary between < 1% to 17.6% of all patients with cancer in the region. CONCLUSION: Cancer-related cases and deaths attributable to SARS-CoV-2 will put a great strain on health care systems in Latin America. Early implementation of interventions on the basis of data given by disease modeling could mitigate both infections and deaths among patients with cancer
