    Analysis of mRNA expression for steroidogenic enzymes in the remaining adrenal cortices attached to adrenocortical adenomas.

    DESIGN AND METHODS: We have recently demonstrated that the adrenal cortices attached to aldosterone-producing adenoma (APA) contained microscopic subcapsular micronodules suggestive of active aldosterone production. In this study, we used in situ hybridization to investigate the mRNA expression of steroidogenic enzymes in the adrenal cortices attached to cortisol-producing adenoma (CPA) and clinically silent adenoma (non-functioning adenoma; NFA), in addition to APA. RESULTS: Microscopic subcapsular micronodules, which were several hundreds of micrometers in size and spheroid in shape, were observed in the cortices attached to CPA and NFA, as well as APA, at high frequency. Most of the cortical nodules in zona fasciculata to zona reticularis showed a suppressed steroidogenesis in the cortices attached to adenoma, but some expressed intensely all necessary steroidogenic enzyme mRNAs for cortisol synthesis. CONCLUSIONS: It is thus necessary to keep in mind, on the occasion of subtotal adrenalectomy, that lesions with the potential to later develop into functional adrenocortical nodules may be present in other parts of the ipsilateral or contralateral adrenal cortices

    In this study, we clarify the classification of museum specimens of the family Viviparidae, which is composed of six species/subspecies in Japan, including three endangered species. We examined Viviparus sclateri specimens from the Tomotaro Iwakawa collection (1855-1933) in the National Museum of Nature and Science, Tokyo. The Iwakawa's collection was catalogued in 1919 and Viviparus sclateri, labelled with Naga-tanishi, the current Japanese name for H. longispira, which was, at the time, equivalent to Viviparus sclateri, was listed in this catalogue. The catalogue noted localities of Viviparus sclateri (Naga-tanishi) from outside Lake Biwa, including occurrences in Lake Kasumigaura and Lake Suwa. However, Heterogen longispira (Naga-tanishi) is currently considered to be endemic to Lake Biwa drainage. The actual status of Viviparus sclateri in Iwakawa (1919) has not been clarified until now.Our examination revealed that Viviparus sclateri from Iwakawa’s catalogue included H. japonica, H. longispira and Sinotaia quadrata histrica, based on current taxonomy. Specimens assigned to H. longispira occurred only in Lake Biwa drainage. Heterogen japonica was confirmed to be present in all lots and some H. japonica from Lake Suwa had a distinctive morphology. Sinotaia quadrata histrica was only confirmed to occur in Lake Suwa. Furthermore, some specimens from southern Lake Biwa and the Seta River had intermediate characteristics between H. japonica and H. longispira and their populations are currently almost extinct

    Indoleamine 2,3-dioxygenase (IDO) is the rate-limiting enzyme of the kynurenine pathway of tryptophan metabolism, ultimately leading to production of the excitotoxin quinolinic acid (QUIN) by monocytic cells. In the Tg2576 mouse model of Alzheimer's disease, systemic inflammation induced by lipopolysaccharide leads to an increase in IDO expression and QUIN production in microglia surrounding amyloid plaques. We examined whether the IDO over-expression in microglia could be mediated by brain proinflammatory cytokines induced during the peripheral inflammation using THP-1 cells and peripheral blood mononuclear cells (PBMC) as models for microglia. THP-1 cells pre-treated with 5-25 μM amyloid β peptide (Aβ) (1-42) but not with Aβ (1-40) or Aβ (25-35) became an activated state as indicated by their morphological changes and enhanced adhesiveness. IDO expression was only slightly increased in the reactive cells but strongly enhanced following treatment with proinflammatory cytokine interferon-γ (IFN-γ) but not with interleukin-1β, tumor necrosis factor-α, or interleukin-6 at 100 U/mL. The concomitant addition of Aβ (1-42) with IFN-γ was totally ineffective, indicating that Aβ pre-treatment is prerequisite for a high IDO expression. The priming effect of Aβ (1-42) for the IDO induction was also observed for PBMC. These findings suggest that IFN-γ induces IDO over-expression in the primed microglia surrounding amyloid plaques.10 page(s

    Construction of a novel expression vector in Pseudonocardia autotrophica and its application to efficient biotransformation of compactin to pravastatin, a specific HMG-CoA reductase inhibitor

    The novel plasmid vector (pTAOR4-Rev) suitable for gene expression in actinomycete strains of Pseudonocardia autotrophica was constructed from 2 P. autotrophica genetic elements, the novel replication origin and the acetone-inducible promoter. The replication origin was isolated from the endogenous plasmid of strain DSM 43082 and the acetone-inducible promoter was determined by analysis of the upstream region of an acetaldehyde dehydrogenase gene homologue in strain NBRC 12743. P. autotrophica strains transformed with pTAOR4-P450, carrying a gene for cytochrome P450 monooxygenase, expressed P450 from the acetone-inducible promoter, as verified by SDS-PAGE and spectral analysis. The biotransformation test of acetone-induced resting cells prepared from a strain of P. autotrophica carrying pTAOR4 that harbors a compactin (CP)-hydroxylating P450 gene revealed 3.3-fold increased production of pravastatin (PV), a drug for hypercholesterolemia. Biotransformation of CP by the same strain in batch culture yielded PV accumulation of 14.3 g/l after 100 hr. The expression vector pTAOR4-Rev and its function-enhancing derivatives provide a versatile approach to industrial biotransformation by Pseudonocardia strains, which can be good hosts for P450 monooxygenase expression

    Amelioration of Combustion of Hydrogen Rotary Engine

    Hydrogen rotary engines are promising power sources for the future hydrogen society, and improving their thermal efficiency helps propel tomorrow's car farther. This research looked into the combustion process to reduce fuel consumption. Primary studies of combustion duration and location showed that increasing the local mass burning rate in the L-side of the combustion chamber by generating turbulence was beneficial. Experiments with optical and real rotary engines proved that turbulence effectively increased mass burning rate in L-side and consequently engine's thermal efficiency and its lean limit
