Ensiling Alfalfa (Medicago sativa L.) and Orchard Grass (Dactylis glomerata L.) Forage Harvested at 08:00 or 14:00, without Wilting or 1 or 2 h Wilting and with or without Use of Bacterial Inoculant
Alfalfa forage is difficult to ensile due to low water-soluble carbohydrate content and high buffering capacity. The objective was to assess at Chapingo, Mexico, during the rainy season effects of combinations of harvest hours (08:00, 14:00), wilting time (0, 1, 2 h) and bacterial inoculants on the quality of silage made of alfalfa and orchard grass forage, made in 200-L containers. The experiment was conducted in three phases with two replicates per phase. Variables measured in freshly cut forage and silages were dry matter content (DM), buffer capacity, pH, and alcohol soluble carbohydrates (ASC). Silos remained sealed during 60 d, and additional variables measured in silage were aerobic stability, NH3 -N and in vitro disappearance of DM. In forage harvested at 14:00 h, DM and ASC contents were higher; pH and buffering capacity were not affected by harvest hour; in silages made of that forage, NH3-N levels were lower, while ASC contents and in vitro disappearance of MS were unaffected by harvest hour. Treatments with inoculants were less aerobic stable for 5 days when made of forage harvested at 08:00 h but more stable when made of forage harvested at 14:00 h. Harvesting at 14:00 h was advantageous as silage presented higher DM and ASC contents
Association between Household Air Pollution Exposure and Chronic Obstructive Pulmonary Disease Outcomes in 13 Low- and Middle-Income Country Settings.
RATIONALE: Forty percent of households worldwide burn biomass fuels for energy, which may be the most important contributor to household air pollution. OBJECTIVES: To examine the association between household air pollution exposure and chronic obstructive pulmonary disease (COPD) outcomes in 13 resource-poor settings. METHODS: We analyzed data from 12,396 adult participants living in 13 resource-poor, population-based settings. Household air pollution exposure was defined as using biomass materials as the primary fuel source in the home. We used multivariable regressions to assess the relationship between household air pollution exposure and COPD outcomes, evaluated for interactions, and conducted sensitivity analyses to test the robustness of our findings. MEASUREMENTS AND MAIN RESULTS: Average age was 54.9 years (44.2-59.6 yr across settings), 48.5% were women (38.3-54.5%), prevalence of household air pollution exposure was 38% (0.5-99.6%), and 8.8% (1.7-15.5%) had COPD. Participants with household air pollution exposure were 41% more likely to have COPD (adjusted odds ratio, 1.41; 95% confidence interval, 1.18-1.68) than those without the exposure, and 13.5% (6.4-20.6%) of COPD prevalence may be caused by household air pollution exposure, compared with 12.4% caused by cigarette smoking. The association between household air pollution exposure and COPD was stronger in women (1.70; 1.24-2.32) than in men (1.21; 0.92-1.58). CONCLUSIONS: Household air pollution exposure was associated with a higher prevalence of COPD, particularly among women, and it is likely a leading population-attributable risk factor for COPD in resource-poor settings
The Relationship Between Socioeconomic Status and CV Risk Factors: The CRONICAS Cohort Study of Peruvian Adults.
BACKGROUND: Variations in the distribution of cardiovascular disease and risk factors by socioeconomic status (SES) have been described in affluent societies, yet a better understanding of these patterns is needed for most low- and middle-income countries. OBJECTIVE: This study sought to describe the relationship between cardiovascular risk factors and SES using monthly family income, educational attainment, and assets index, in 4 Peruvian sites. METHODS: Baseline data from an age- and sex-stratified random sample of participants, ages ≥35 years, from 4 Peruvian sites (CRONICAS Cohort Study, 2010) were used. The SES indicators considered were monthly family income (n = 3,220), educational attainment (n = 3,598), and assets index (n = 3,601). Behavioral risk factors included current tobacco use, alcohol drinking, physical activity, daily intake of fruits and vegetables, and no control of salt intake. Cardiometabolic risk factors included obesity, elevated waist circumference, hypertension, insulin resistance, diabetes mellitus, low high-density lipoprotein cholesterol, and high triglyceride levels. RESULTS: In the overall population, 41.6% reported a monthly family income <US$198, and 45.6% had none or primary education. Important differences were noted between the socioeconomic indicators: for example, higher income and higher scores on an asset index were associated with greater risk of obesity, whereas higher levels of education were associated with lower risk of obesity. In contrast, higher SES according to all 3 indicators was associated with higher levels of triglycerides. CONCLUSIONS: The association between SES and cardiometabolic risk factors varies depending on the SES indicator used. These results highlight the need to contextualize risk factors by socioeconomic groups in Latin American settings
The drivers of change for the contribution of small farms to regional food security in Europe
The capacity of the food system to respond to the economic, demographic and environmental challenges ahead has become a topic of increasing interest, with particular attention to the roles and responsibilities of the different actors to ensure more sustainable food systems that can guarantee food and nutrition security for all. In this paper we approach the need to better understand the factors that can condition the potential contribution of small farms to regional food and nutrition security in Europe, acknowledging the role that small farms play in Europe at present. The analysis is based on a survey to 94 experts from 17 regions (NUTS3 level) in 11 different European countries, which identified the drivers of change according to the regional experts. These drivers were then categorized and their relative relevance assessed. The results indicate that some relevant drivers in the European context are linked to the capacity to adopt technologies and practices allowing adaptation to climate change, and the capacity to connect to food markets, with emphasis in the need for cooperation and collective action. The weight of other more European-specific drivers such as ‘consumer values and habits’ reveal that the future role of small farms will be very dependent on a societal change, with equity becoming a relevant component of consumers’ choice
Dietary Intervention Modulates the Expression of Splicing Machinery in Cardiovascular Patients at High Risk of Type 2 Diabetes Development: From the CORDIOPREV Study
Type-2 diabetes mellitus (T2DM) has become a major health problem worldwide. T2DM risk can be reduced with healthy dietary interventions, but the precise molecular underpinnings behind this association are still incompletely understood. We recently discovered that the expression profile of the splicing machinery is associated with the risk of T2DM development. Thus, the aim of this work was to evaluate the influence of 3-year dietary intervention in the expression pattern of the splicing machinery components in peripheral blood mononuclear cells (PBMCs) from patients within the CORDIOPREV study. Expression of splicing machinery components was determined in PBMCs, at baseline and after 3 years of follow-up, from all patients who developed T2DM (Incident-T2DM, n = 107) and 108 randomly selected non-T2DM subjects, who were randomly enrolled in two healthy dietary patterns (Mediterranean or low-fat diets). Dietary intervention modulated the expression of key splicing machinery components (i.e., up-regulation of SPFQ/RMB45/RNU6, etc., down-regulation of RNU2/SRSF6) after three years, independently of the type of healthy diet. Some of these changes (SPFQ/RMB45/SRSF6) were associated with key clinical features and were differentially induced in Incident-T2DM patients and non-T2DM subjects. This study reveals that splicing machinery can be modulated by long-term dietary intervention, and could become a valuable tool to screen the progression of T2DM
The Latin American Consortium of Studies in Obesity (LASO)
Current, high-quality data are needed to evaluate the health impact of the epidemic of obesity in Latin America. The Latin American Consortium of Studies of Obesity (LASO) has been established, with the objectives of (i) Accurately estimating the prevalence of obesity and its distribution by sociodemographic characteristics; (ii) Identifying ethnic, socioeconomic and behavioural determinants of obesity; (iii) Estimating the association between various anthropometric indicators or obesity and major cardiovascular risk factors and (iv) Quantifying the validity of standard definitions of the various indexes of obesity in Latin American population. To achieve these objectives, LASO makes use of individual data from existing studies. To date, the LASO consortium includes data from 11 studies from eight countries (Argentina, Chile, Colombia, Costa Rica, Dominican Republic, Peru, Puerto Rico and Venezuela), including a total of 32 462 subjects. This article describes the overall organization of LASO, the individual studies involved and the overall strategy for data analysis. LASO will foster the development of collaborative obesity research among Latin American investigators. More important, results from LASO will be instrumental to inform health policies aiming to curtail the epidemic of obesity in the region
Clinical Utility of Ghrelin-O-Acyltransferase (GOAT) Enzyme as a Diagnostic Tool and Potential Therapeutic Target in Prostate Cancer
Recent data suggested that plasma Ghrelin O-Acyl Transferase enzyme (GOAT) levels could represent a new diagnostic biomarker for prostate cancer (PCa). In this study, we aimed to explore the diagnostic and prognostic/aggressiveness capacity of GOAT in urine, as well as to interrogate its putative pathophysiological role in PCa. We analysed urine/plasma levels of GOAT in a cohort of 993 patients. In vitro (i.e., cell-proliferation) and in vivo (tumor-growth in a xenograft-model) approaches were performed in response to the modulation of GOAT expression/activity in PCa cells. Our results demonstrate that plasma and urine GOAT levels were significantly elevated in PCa patients compared to controls. Remarkably, GOAT significantly outperformed PSA in the diagnosis of PCa and significant PCa in patients with PSA levels ranging from 3 to 10 ng/mL (the so-called PSA grey-zone). Additionally, urine GOAT levels were associated to clinical (e.g., Gleason-score, PSA levels) and molecular (e.g., CDK2/CDK6/CDKN2A expression) aggressiveness parameters. Indeed, GOAT overexpression increased, while its silencing/blockade decreased cell-proliferation in PCa cells. Moreover, xenograft tumors derived from GOAT-overexpressing PCa (DU145) cells were significantly higher than those derived from the mock-overexpressing cells. Altogether, our results demonstrate that GOAT could be used as a diagnostic and aggressiveness marker in urine and a therapeutic target in PCa
Global Analysis of the post-SNO Solar Neutrino Data for Standard and Non-Standard Oscillation Mechanisms
What can we learn from solar neutrino observations? Is there any solution to
the solar neutrino anomaly which is favored by the present experimental
panorama? After SNO results, is it possible to affirm that neutrinos have mass?
In order to answer such questions we analyze the current available data from
the solar neutrino experiments, including the recent SNO result, in view of
many acceptable solutions to the solar neutrino problem based on different
conversion mechanisms, for the first time, using the same statistical
procedure. This allows us to do a direct comparison of the goodness of the fit
among different solutions, from which we can discuss and conclude on the
current status of each proposed dynamical mechanism. These solutions are based
on different assumptions: (a) neutrino mass and mixing, (b) non-vanishing
neutrino magnetic moment, (c) the existence of non-standard flavor-changing and
non-universal neutrino interactions and (d) the tiny violation of the
equivalence principle. We investigate the quality of the fit provided by each
one of these solutions not only to the total rate measured by all the solar
neutrino experiments but also to the recoil electron energy spectrum measured
at different zenith angles by the Super-Kamiokande collaboration. We conclude
that several non-standard neutrino flavor conversion mechanisms provide a very
good fit to the experimental data which is comparable with (or even slightly
better than) the most famous solution to the solar neutrino anomaly based on

Contribution of microscopy for understanding the mechanism of action against trypanosomatids
Transmission electron microscopy (TEM) has proved to be a useful tool to study the ultrastructural alterations and the target organelles of new antitrypanosomatid drugs. Thus, it has been observed that sesquiterpene lactones induce diverse ultrastructural alterations in both T. cruzi and Leishmania spp., such as cytoplasmic vacuolization, appearance of multilamellar structures, condensation of nuclear DNA, and, in some cases, an important accumulation of lipid vacuoles. This accumulation could be related to apoptotic events. Some of the sesquiterpene lactones (e.g., psilostachyin) have also been demonstrated to cause an intense mitochondrial swelling accompanied by a visible kinetoplast deformation as well as the appearance of multivesicular bodies. This mitochondrial swelling could be related to the generation of oxidative stress and associated to alterations in the ergosterol metabolism. The appearance of multilamellar structures and multiple kinetoplasts and flagella induced by the sesquiterpene lactone psilostachyin C indicates that this compound would act at the parasite cell cycle level, in an intermediate stage between kinetoplast segregation and nuclear division. In turn, the diterpene lactone icetexane has proved to induce the external membrane budding on T. cruzi together with an apparent disorganization of the pericellar cytoskeleton. Thus, ultrastructural TEM studies allow elucidating the possible mechanisms and the subsequent identification of molecular targets for the action of natural compounds on trypanosomatids.Fil: Lozano, Esteban Sebastián.
