    Semi-field assessment of the BG-Malaria trap for monitoring the African malaria vector, Anopheles arabiensis

    Odour-baited technologies are increasingly considered for effective monitoring of mosquito populations and for the evaluation of vector control interventions. The BG-Malaria trap (BGM), which is an upside-down variant of the widely used BG-Sentinel trap (BGS), has been demonstrated to be effective to sample the Brazilian malaria vector, Anopheles darlingi. We evaluated the BGM as an improved method for sampling the African malaria vectors, Anopheles arabiensis. Experiments were conducted inside a large semi-field cage to compare trapping efficiencies of BGM and BGS traps, both baited with the synthetic attractant, Ifakara blend, supplemented with CO2. We then compared BGMs baited with either of four synthetic mosquito lures, Ifakara blend, Mbita blend, BG-lure or CO2, and an unbaited BGM. Lastly, we compared BGMs baited with the Ifakara blend dispensed via either nylon strips, BG cartridges (attractant-infused microcapsules encased in cylindrical plastic cartridge) or BG sachets (attractant-infused microcapsules encased in plastic sachets). All tests were conducted between 6P.M. and 7A.M., with 200–600 laboratory-reared An. arabiensis released nightly in the test chamber. The median number of An. arabiensis caught by the BGM per night was 83, IQR:(73.5–97.75), demonstrating clear superiority over BGS (median catch = 32.5 (25.25–37.5)). Compared to unbaited controls, BGMs baited with Mbita blend caught most mosquitoes (45 (29.5–70.25)), followed by BGMs baited with CO2 (42.5 (27.5–64)), Ifakara blend (31 (9.25–41.25)) and BG lure (16 (4–22)). BGM caught 51 (29.5–72.25) mosquitoes/night, when the attractants were dispensed using BG-Cartridges, compared to BG-Sachet (29.5 (24.75–40.5)), and nylon strips (27 (19.25–38.25)), in all cases being significantly superior to unbaited controls (p < 000.1). The findings demonstrate potential of the BGM as a sampling tool for African malaria vectors over the standard BGS trap. Its efficacy can be optimized by selecting appropriate odour baits and odour-dispensing systems

    Comparison of alternative evidence summary and presentation formats in clinical guideline development: a mixed-method study.

    BACKGROUND: Best formats for summarising and presenting evidence for use in clinical guideline development remain less well defined. We aimed to assess the effectiveness of different evidence summary formats to address this gap. METHODS: Healthcare professionals attending a one-week Kenyan, national guideline development workshop were randomly allocated to receive evidence packaged in three different formats: systematic reviews (SRs) alone, systematic reviews with summary-of-findings tables, and 'graded-entry' formats (a 'front-end' summary and a contextually framed narrative report plus the SR). The influence of format on the proportion of correct responses to key clinical questions, the primary outcome, was assessed using a written test. The secondary outcome was a composite endpoint, measured on a 5-point scale, of the clarity of presentation and ease of locating the quality of evidence for critical neonatal outcomes. Interviews conducted within two months following completion of trial data collection explored panel members' views on the evidence summary formats and experiences with appraisal and use of research information. RESULTS: 65 (93%) of 70 participants completed questions on the prespecified outcome measures. There were no differences between groups in the odds of correct responses to key clinical questions. 'Graded-entry' formats were associated with a higher mean composite score for clarity and accessibility of information about the quality of evidence for critical neonatal outcomes compared to systematic reviews alone (adjusted mean difference 0.52, 95% CI 0.06 to 0.99). There was no difference in the mean composite score between SR with SoF tables and SR alone. Findings from interviews with 16 panelists indicated that short narrative evidence reports were preferred for the improved clarity of information presentation and ease of use. CONCLUSIONS: Our findings suggest that 'graded-entry' evidence summary formats may improve clarity and accessibility of research evidence in clinical guideline development.     Household modifications after the indoor residual spraying (IRS) campaign in Mozambique reduce the actual spray coverage and efficacy

    Indoor residual spraying of insecticides (IRS) is a key malaria vector control strategy. Whilst human attitude towards IRS is monitored before or shortly after implementation, human activities leading to the modification of insecticide-treated walls post-IRS are not. This could inadvertently reduce the protective effects of IRS. We monitored the extent of modifications to the sprayed indoor wall surfaces by household owners for six months post-IRS campaigns in two districts targeted for malaria elimination in southern Mozambique. In parallel, we assessed building of any additional rooms onto compounds, and mosquito net use. We quantified the contribution of wall modifications, added rooms, prolonged spray campaigns, and product residual efficacies on actual IRS coverage and relative mosquito bite reduction, using a mechanistic approach. Household owners continually modified insecticide-treated walls and added rooms onto compounds. Household surveys in southern Mozambique showed frequent modification of indoor walls (0–17.2% of households modified rooms monthly) and/or added rooms (0–16.2% of households added rooms monthly). Actual IRS coverage reduced from an assumed 97% to just 39% in Matutuine, but only from 96% to 91% in Boane, translating to 43% and 5.8% estimated increases in relative daily mosquito bites per person. Integrating post-IRS knowledge, attitude, and practice (KAP) surveys into programmatic evaluations to capture these modification and construction trends can help improve IRS program efficiency and product assessment

    A Rapid Assessment of the Quality of Neonatal Healthcare in Kilimanjaro Region, Northeast Tanzania.

    While child mortality is declining in Africa there has been no evidence of a comparable reduction in neonatal mortality. The quality of inpatient neonatal care is likely a contributing factor but data from resource limited settings are few. The objective of this study was to assess the quality of neonatal care in the district hospitals of the Kilimanjaro region of Tanzania. Clinical records were reviewed for ill or premature neonates admitted to 13 inpatient health facilities in the Kilimanjaro region; staffing and equipment levels were also assessed. Among the 82 neonates reviewed, key health information was missing from a substantial proportion of records: on maternal antenatal cards, blood group was recorded for 52 (63.4%) mothers, Rhesus (Rh) factor for 39 (47.6%), VDRL for 59 (71.9%) and HIV status for 77 (93.1%). From neonatal clinical records, heart rate was recorded for3 (3.7%) neonates, respiratory rate in 14, (17.1%) and temperature in 33 (40.2%). None of 13 facilities had a functioning premature unit despite calculated gestational age <36 weeks in 45.6% of evaluated neonates. Intravenous fluids and oxygen were available in 9 out of 13 of facilities, while antibiotics and essential basic equipment were available in more than two thirds. Medication dosing errors were common; under-dosage for ampicillin, gentamicin and cloxacillin was found in 44.0%, 37.9% and 50% of cases, respectively, while over-dosage was found in 20.0%, 24.2% and 19.9%, respectively. Physician or assistant physician staffing levels by the WHO indicator levels (WISN) were generally low. Key aspects of neonatal care were found to be poorly documented or incorrectly implemented in this appraisal of neonatal care in Kilimanjaro. Efforts towards quality assurance and enhanced motivation of staff may improve outcomes for this vulnerable group

    Growing smaller fish for inclusive markets? Increasing stocking density and shortening the production cycle of Nile tilapia in cages on Lake Victoria

    Fish farmers aim to maximise fish weight relative to the feed inputs needed to turn a profit. Yet, many farmers in Africa lack the cash flow to grow large fish and many consumers prefer, or are limited to purchasing, small fish. This study aimed to intentionally produce small tilapia in cages by assessing the effects of higher stocking densities and shorter growth cycles on production and financial efficiency. An experiment with 3 treatments and 6 replicates took place on Lake Victoria. The first treatment (T1) used a stocking density of 2.9 ± 0.3 kg per m−3 and aimed to produce fish to an average body weight (ABW) of 400 g (final ABW = 500.33 ± 31.01 g after 138 days). Treatment two (T2) did the same but with double the stocking density (5.9 ± 0.3 kg per m−3), resulting in a final ABW of 439.22 ± 22.22 g over 138 days. The third treatment (T3) partially harvested 50% of the cage (after 76 days) once reaching an ABW of 230.92 ± 22.55 g. The remaining fish in T3 were on-grown for a total of 138 days (final ABW = 499.86 ± 15.95 g). A fourth production scenario (M1) based on data from T3, modelled a 100% harvest after 76 days of culture. There were no significant differences in mortality between treatments. There were no statistical differences in the feed conversion ratio (FCR) between T1 (1.51 ± 0.03) and T2 (1.49 ± 0.02), though T3 was statistically lower (1.46 ± 0.02; p = 0.03). Cages in T1 had a higher proportion of fish between 400 and 599 g while fish in T2 were mostly between 300 and 499 g. T3 had a bimodal distribution with most fish either in 200–299 g or 400–499 g. There was little effect on average price per kg for T1 (3.0 ± 0.01 USD) and T2 (2.98 ± 0.01 USD), though T3 (2.89 ± 0.04 USD) was significantly lower (p = 0.001). Overall, T2 had significantly higher gross margins (17% ± 2.08) than T1 (13% ± 2.3, p = 0.021) and T3 (7.2% ± 2.43, p = 0.001), while M1 had the lowest gross margins (−11.8% ± 5.5). The results suggest that farmers can increase stocking densities. Some farmers can use partial harvesting strategies or shorter cycles to produce small tilapia and achieve faster cash flows, though the economic margins are lower. Such approaches can provide opportunities for poorer farmers and consumers

    Indigenous Grasses for Rehabilitating Degraded African Drylands

    Drylands provide an important livelihood stream to its inhabitants across the globe through a range of products and ecosystem services. However, these fragile ecosystems are threatened and believed to experience various degrees of land degradation. Estimates of the landmass affected by land degradation in the global drylands range from 10% to 20%, a percentage that is increasing at an annual global rate of 12 million ha of soil lost from desertification and drought. African drylands are especially highly susceptible to severe degradation because of their poor soil structure aggravated by scarce vegetation cover. Causes of degradation in these environments are both natural and anthropogenic in nature. Change in vegetation cover, decline in soil fertility, biodiversity loss and soil erosion demonstrate degradation in African drylands. Grass reseeding using indigenous species is one of the promising sustainable land management strategies to combat degradation in the drylands. Reseeding programmes are aimed at improving vegetation cover and biomass, and they conserve the soil to an extent not possible by grazing and land management alone. Indigenous drought-tolerant grasses notably African foxtail grass (Cenchrus ciliaris), bush rye grass (Enteropogon macrostachyus) and Maasai lovegrass (Eragrostis superba) have produced promising rehabilitation outcomes. Previous studies in African drylands have demonstrated the potential of such indigenous forage grasses in improving both vegetation cover (plant frequency and densities, basal cover) and soil hydrological properties (increased infiltration capacity, reduced runoff and sediment production) as indicators of rehabilitation success. Despite their comparative and widespread success, natural and anthropogenic challenges persist. This makes reseeding programmes a risky and often expensive venture, especially for the resource-poor pastoral communities in African drylands. Despite the risks, grass reseeding using indigenous pastures remains a viable sustainable land management option to combat degradation in African drylands. However, to ensure its continued success in the long term, multifaceted approaches and strategies that will integrate land and water management and seed systems suitable for African drylands need to be developed, strengthened and promoted.Peer reviewe

    The effects of zooprophylaxis and other mosquito control measures against malaria in Nouna, Burkina Faso

    <p>Abstract</p> <p>Background</p> <p>In the absence of large scale, organized vector control programmes, individual protective measures against mosquitoes are essential for reducing the transmission of diseases like malaria. Knowledge of the types and effectiveness of mosquito control methods used by households can aid in the development and promotion of preventive measures.</p> <p>Methods</p> <p>A matched, population-based case control study was carried out in the semi-urban region of Nouna, Burkina Faso. Surveys and mosquito captures were conducted for each participating household. Data were analysed using conditional logistic regression and Pearson's product-moment correlations.</p> <p>Results</p> <p>In Nouna, Burkina Faso, the main types of reported mosquito control measures used included sleeping under bed nets (insecticide-treated and untreated) and burning mosquito coils. Most of the study households kept animals within the compound or house at night. Insecticide house sprays, donkeys, rabbits and pigs were significantly associated with a reduced risk of malaria only in univariate analyses.</p> <p>Conclusion</p> <p>Given the conflicting results of the effects of zooprophylaxis from previous studies, other community-based preventive measures, such as bed nets, coils and insecticide house-spraying, may be of more benefit.</p

    Bacillus proteolyticus OSUB18 triggers induced systemic resistance against bacterial and fungal pathogens in Arabidopsis

    Pseudomonas syringae and Botrytis cinerea cause destructive bacterial speck and grey mold diseases in many plant species, leading to substantial economic losses in agricultural production. Our study discovered that the application of Bacillus proteolyticus strain OSUB18 as a root-drench enhanced the resistance of Arabidopsis plants against P. syringae and B. cinerea through activating Induced Systemic Resistance (ISR). The underlying mechanisms by which OSUB18 activates ISR were studied. Our results revealed that the Arabidopsis plants with OSUB18 root-drench showed the enhanced callose deposition and ROS production when inoculated with Pseudomonas syringae and Botrytis cinerea pathogens, respectively. Also, the increased salicylic acid (SA) levels were detected in the OSUB18 root-drenched plants compared with the water root-drenched plants after the P. syringae infection. In contrast, the OSUB18 root-drenched plants produced significantly higher levels of jasmonyl isoleucine (JA-Ile) than the water root-drenched control after the B. cinerea infection. The qRT-PCR analyses indicated that the ISR-responsive gene MYC2 and the ROS-responsive gene RBOHD were significantly upregulated in OSUB18 root-drenched plants upon both pathogen infections compared with the controls. Also, twenty-four hours after the bacterial or fungal inoculation, the OSUB18 root-drenched plants showed the upregulated expression levels of SA-related genes (PR1, PR2, PR5, EDS5, and SID2) or JA-related genes (PDF1.2, LOX3, JAR1 and COI1), respectively, which were consistent with the related hormone levels upon these two different pathogen infections. Moreover, OSUB18 can trigger ISR in jar1 or sid2 mutants but not in myc2 or npr1 mutants, depending on the pathogen’s lifestyles. In addition, OSUB18 prompted the production of acetoin, which was reported as a novel rhizobacterial ISR elicitor. In summary, our studies discover that OSUB18 is a novel ISR inducer that primes plants’ resistance against bacterial and fungal pathogens by enhancing the callose deposition and ROS accumulation, increasing the production of specific phytohormones and other metabolites involved in plant defense, and elevating the expression levels of multiple defense genes
