    "A New Composite Index of Coincident Economic Indicators in Japan: How can we improve the forecast performance? "

    The purpose of this paper is to construct a new composite index of coincident economic indicators in Japan and to demonstrate their usefulness in forecasting recent short-run economic fluctuations. The method of construction is based on the single-index dynamic factor model. Our two types of indexes are highly correlated with the traditional composite index compiled by the EPA over business-cycle horizons. However, standard leading indicators, which failed to forecast the traditional composite index, make a satisfactory performance in forecasting our indexes in the 1990s. In addition, lagged values of our indexes help to improve the leading indicatorsf performance in forecasting the traditional composite index in the 1990s. The result is noteworthy because a large number of research institutes made serious errors in forecasting recent recessions in Japan.

    Evaluation of nitrogen loading in the last 80 years in an urbanized Asian coastal catchment through the reconstruction of severe contamination period

    Most semi-enclosed seas have experienced severe eutrophication owing to high nutrient loading from rivers during rapid population growth periods. In Japan, the coastal areas of some megacities (e.g. Tokyo and Osaka) experienced considerable economic growth during the 1960s-1970s. Therefore, determining the amount of nutrient loading during this period is essential to undertake measures for the conservation of coastal environments. However, determining the nutrient loading that occurred several decades ago is generally difficult owing to lacking water quality records. In this study, the nitrogen loading in the Yamato River catchment, an urbanized coastal catchment in Asia, for 80 years from the 1940s to the 2010s is reconstructed using the Soil and Water Assessment Tool. We considered factors such as population growth, wastewater treatment plant (WWTP) construction, and changes in land and fertilizer usage in different urbanization stages. Results show that the total nitrogen loading in the catchment peaked in the 1970s at 6616 tons yr(-1) owing to untreated wastewater discharge and rapid increase in population growth. By reducing 57% of the nitrogen loading in the 2010s from the catchment, WWTPs have been instrumental in improving the water environment. The decrease in and integration of agricultural land has reduced nitrogen loading attributed to nonpoint sources; however, this reduction was not obvious because of the high fertilizer usage before the 2000s. Overall, the findings of this study provide a comprehensive understanding of the impact of rapid urbanization in an Asian coastal catchment on nitrogen loading during the high economic growth period in the past.     A comparison of the signal pathways between the TNF alpha- and oridonin-induced murine L929 fibrosarcoma cell death.

    Oridonin, an active component isolated from Rabdosia rubescences, has been reported to have antitumor effects. In this study, we compared the signal transduction pathways between TNFalpha-and oridonin-induced L929 cell death. Oridonin and TNFalpha initiated apoptotic morphologic changes, but DNA fragmentation was found in TNFalpha-treated L929 cells but not in oridonin-treated ones. The pan-caspase inhibitor (z-VAD-fmk), caspase-8 inhibitor (z-IETD-fmk) and caspase-3 inhibitor (z-DEVD-fmk) augmented oridonin-and TNFalpha-induced cell death. However, the caspase-9 inhibitor (z-LEHD-fmk) only increased oridonin-induced L929 cell death. Moreover, poly (ADPribose) polymerase (PARP) was cleaved in oridonin-treated L929 cells but not in the TNFalpha-treated groups, and the caspase-3 inhibitor (z-DEVD-fmk) failed to inhibit PARP cleavage. These results showed that only oridonin-induced L929 cell death required PARP degradation in a caspase-3 independent manner. In addition, oridonin increased the ratio of Bax/Bcl-2 protein expression, but TNFalpha did not. TNFalpha induced p38 and ERK activation, whereas oridonin triggered only ERK activation. We also investigated the effect of oridonin on intracellular TNFalpha expression, and found that oridonin augmented endogenous pro-TNFalpha expression and its upstream protein IkB phosphorylation. These results indicated that although oridonin promoted endogenous pro-TNFalpha expression, a great difference existed between the signal pathways through which TNFalpha-and oridonin-induced cell death.</p

    The augmentation of TNFalpha-induced cell death in murine L929 fibrosarcoma by the pan-caspase inhibitor Z-VAD-fmk through pre-mitochondrial and MAPK-dependent pathways.

    We investigated the mechanism of the pan-caspase inhibitor z-VAD-fmk's augmentation of TNFalpha-induced L929 cell death and found this mechanism differs from that of TNFalpha-induced L929 cell death. In the presence of 20 ng/ml TNFalpha, z-VAD-fmk initiated apoptosis and necrosis in the majority of L929 cells as measured by an agarose gel electrophoresis and lactate dehydrogenase(LDH)activity based assay. Mitochondrial permeability transition (MPT) inhibitor (cyclosporine A) effectively inhibited z-VAD-fmk-augmented cell death. In addition, z-VAD-fmk plus TNFalpha increased Bax expression without affecting Bcl-2 and cytochrome expression. Western-blot analysis showed that z-VAD-fmk plus TNFalpha caused persistent JNK activation and ERK inactivation. Poly(ADP-ribose) polymerase (PARP) inhibitor (DPQ) effectively reversed the cell death which was augmented by z-VAD-fmk, and z-VAD-fmk plus TNFalpha also caused PARP cleavage to an 85 KDa fragment. These results indicate that in the presence of TNFalpha, z-VAD-fmk further augments cell death which requires the mitochondrial permeability transition and the JNK activation. However, we did not detect the changes in cytochrome c expression and the participation of caspase-9 in this process, suggesting that there might exist an unknown signal pathway(s) from the mitochondria to the downstream protein PARP, which is cleaved in a caspase-independent manner.</p

    Verification of piezometric method for the observation of dissolved component and pressure head dynamics in a tidal flat groundwater

    We carried out the verification of the piezometric method to observe concerning groundwater flow and solute transport under the unsteady state in the tidal flat. Especially, we considered whether solute moves and reacts instantaneously as pressure head changes in the tidal flat. The results of verification are summarized as follows, (1)The electric conductivity(EC) of water collected from the piezometer changed with the tidal fluctuation, and the variation was similar to that by buried EC sendor. Moreover, the variation accuracy increased by pulling out water in the piezometer just before water sampling. (2)The piezometric head (water table in the pipe) changed with the tidal fluctuation. Especially, it decreased with delaying during falling period of the tide level. This showed that pressure remained under the ground. (3)Our study corresponded to previous studies concerning between the groundwater and solute flux by the piezometric and catchment mass balance methods.Based on these results, we would apply this method to groundwater flow and solute dynamics in the tidal flat

    NO_3^--N Flux of Streams in the Setouchi Region : Effects of Fruit-Farmland Area, Water Reservoir, and Alluvial Fan

    To confirm the effects of fruit-farmland area, water reservoir, and alluvial fan on nitrate load in short streams, we measured runoff and collected water samples at five or eight sites in each of four streams, Seto Inland Sea catchment. Nitrate load of the streams increased with increasing ratio of fruit-farmland area. At a downstream site of water reservoir, nitrate concentration showed a slight decrease. On the area widely domi- nated by alluvial fan, it assumed that groundwater pollution by nitrate-nitrogen is accelerated with groundwater recharge of stream water. It is necessary for conservation of water resources to consider function of these effects as well as river-groundwater mixing

    In Vivo Imaging of Particle-Induced Inflammation and Osteolysis in the Calvariae of NFκB/Luciferase Transgenic Mice

    Wear debris causes biological response which can result in periprosthetic osteolysis after total joint replacement surgery. Nuclear factor-kappa B (NFκB), a representative transcription factor involved in inflammation, is believed to play an important role in this event by regulating the production of proinflammatory mediators and osteoclastogenesis. In this study, we sought to determine whether activation of NFκB in response to stimulation by particles could be visualized by in vivo imaging. We loaded polyethylene (PE) particles onto the calvaria of NFκB/luciferase transgenic mouse, and detected luminescence generated by activation of NFκB. On day 7 after loading, the level of luminescence was maximal. Levels of luminescence were significantly correlated with the levels of luciferase activity, proinflammatory mediator mRNAs, and bone resorption parameters. This system, which enabled us to evaluate particle-induced inflammation and osteolysis without sacrificing mice, constitutes a useful tool for evaluating the efficacy of prophylaxis or treatments for particle-induced osteolysis

    Effect of forestation of hinoki and larch on soil chemistry in mountainous water source area for water supply to Tokyo

    Effects of forestry on soil properties, stream water chemistry and mass balance in watersheds had been confieremed by many previous studies. However, they have not clarified in detail variation processes of soil chemistry and soil physics after the hinoki and larch forestation. To clarify the variation processes, it is important to confirm the difference of soil chemistry and soil physics on the artifitial and natural forest. In this research, we conducted the soil physical and chemical investigations on seven slopes covered by hinoki (chamecyparis obtusa) and larch (Larix leptolepis) artificial and beech (Fagus crenata) natural forest in a highland area, the western side of Tokyo. The water repellency of A_0 horizon was stronger on the artificial forest slope than on the natural forest. In addition, the permeability was low on the artificial forest due to the strong water repellency. On the artificial forest, both of the exchangeable base cation content and soil pH were low and Al concentration was high, as compared with those on the natural forest. These results suggest that the soil acidification is progressed on the artificial forest. The decline of soil pH by the forestation was controlled by the increase of H^+ supply at the A_0 horizon due to the property of litter decomposition and the decrease of base cation supply at the A-horizon. In general, the cation supply rate such as weathering rate is controlled by the infiltration rate and temperature. These soil physical and chemical properties suggest that infitration rate at the A-horizon. declines after the forestation, weathering rate declines and consequently soil is acidified

    TRANSPORT AND TRANSFORMATION OF CHEMICAL COMPONENTS IN THE GROUNDWATER FLOW SYSTEM OF JAKARTA METROPOLITAN AREA

    ABSTRACT The aim of this study is to examine the transport and transformation of chemical components within the groundwater flow in Jakarta area, and to evaluate the effects of accelerated urbanization on it. The collected data showed that the current hydraulic potential in the Jakarta metropolitan area is below sea level because of prior excess abstraction of groundwater. The distribution of Cl- and Mn2- concentration in groundwater suggests that the decline in hydraulic potential has caused the intrusion of seawater to shallow groundwater and the movement of shallow groundwater into deep groundwater. It implies an accumulation of contaminants in deep aquifers. On the other hands, the presentation of NO3--N in groundwater is suggested to be attenuated by the processes of denitrification and dilution in the coastal area

    Norcantharidin Induces Human Melanoma A375-S2 Cell Apoptosis through Mitochondrial and Caspase Pathways

    Norcantharidin (NCTD) is the demethylated form of cantharidin, which is the active substance of mylabris. To examine the pathway of NCTD-induced A375-S2 cell death, 3-(4, 5-dimethylthiazol-2-yl)-2, 5-dipheyltetrazolium bromide (MTT) assay, photomicroscopical observation, DNA agarose gel electrophoresis, caspase activity assay and Western blot analysis were carried out. A375-S2 cells treated with NCTD exhibited several typical characteristics of apoptosis. The inhibitory effect of NCTD on human melanoma, A375-S2 cells, was partially reversed by the inhibitors of pan-caspase, caspase-3 and caspase-9. The activities of caspase-3 and -9 were significantly increased after treatment with NCTD at different time. The expression of inhibitor of caspase-activated DNase was decreased in a time-dependent manner, simultaneously, the ratio of Bcl-2/Bax or Bcl-xL/Bax was decreased and the expression ratio of proteins could be reversed by caspase-3 inhibitor. The expression of cytochrome c in cytosol was increased after NCTD treatment and caspase-3 inhibitor had no significant effect on the up-regulation of cytochrom c. These results suggest that NCTD induced A375-S2 cell apoptosis and the activation of caspase and mitochondrial pathway were involved in the process of NCTD-induced A375-S2 cell apoptosis
