    Saving fun for a boring future

    We discuss how experiences that fill a future waiting period, such as focusing on fun or boring future activities, affect intertemporal choices. We propose that savoring, the positive utility derived from anticipating future pleasant outcomes, is more likely to have an impact on intertemporal choices when the future seems boring than when it seems fun. We provide empirical evidence that people who foresee a busy future full of boring activities are more likely to prefer to delay rewards than people who foresee a future not so busy with boring activities

    WOS: 000188054500005Two subspecies of mistletoe (Viscum album L. ssp. album and ssp. abietis) growing on lime and pine trees, respectively, were investigated for the monosaccharides and polyols by GC-MS spectrometry. Arabinose, xylose, glucose, galactose, mannose, xylitol and inositol were determined in methanol extracts following the acidic hydrolysis. Sugar contents of the leaves were expressed as percentage on dry weight. Xylose content was the same (1.5%) in each species, whereas the other saccharides varied. V. album ssp. abietis (collected from pine trees) were found containing significantly higher percentage of glucose (29.0%) and galactose (44.0%) than V. album ssp. album (collected from lime trees) (9.0% and 17.0%, respectively). In contrast, mannose, arabinose and sugar alcohol percentages were higher in ssp. album. Mannose content was 3.5% for ssp. album whereas 1.0% for ssp. abietis. 3.0% arabinose were determined in the former and 2.0% in the latter. Xylitol and inositol percentages were 8.0% and 58.0% for ssp. album and 1.5% and 21.0% for ssp. abietis, respectively. These results indicate that saccharide composition of mistletoes depends upon the subspecies of the plant and the host tree

    Bolstering Stochastic Gradient Descent with Model Building

    Stochastic gradient descent method and its variants constitute the core optimization algorithms that achieve good convergence rates for solving machine learning problems. These rates are obtained especially when these algorithms are fine-tuned for the application at hand. Although this tuning process can require large computational costs, recent work has shown that these costs can be reduced by line search methods that iteratively adjust the stepsize. We propose an alternative approach to stochastic line search by using a new algorithm based on forward step model building. This model building step incorporates second-order information that allows adjusting not only the stepsize but also the search direction. Noting that deep learning model parameters come in groups (layers of tensors), our method builds its model and calculates a new step for each parameter group. This novel diagonalization approach makes the selected step lengths adaptive. We provide convergence rate analysis, and experimentally show that the proposed algorithm achieves faster convergence and better generalization in well-known test problems. More precisely, SMB requires less tuning, and shows comparable performance to other adaptive methods

    A CONCEPTUAL FRAMEWORK AND MATURITY MODEL TO APPRAISE COMPANIES FOR SMART MANUFACTURING

    During the past decade, a fourth revolution has dominated the manufacturing industry within the production facilities as well as among the supply chain by utilizing today’s technologies. This new era introduced the smart manufacturing term to the world. As a result, fostering the conversion from traditional to smart manufacturing started. To survive during this transformation, companies need to understand its concept, requirements, and benefits. For this reason, this study aims to provide a comprehensive understanding of fundamental features and components of smart manufacturing, and a model that assesses companies' current status, capabilities, i.e. their maturity in this era. However, this transformation also brought fear to employees since it creates an image of unmanned production fields. Thus, a human transformation to “adapted people” is also required and assessed during smart manufacturing conversion. To identify components that build a framework for smart manufacturing and dimensions, levels, and criteria for realizing the maturity of a company in the smart manufacturing era, the last five years' literature was reviewed. As a result, this research presents a conceptual smart manufacturing framework and a maturity appraisal model for assisting companies before and during the smart manufacturing transformation within three dimensions at six maturity levels. On this basis, there are eight core technologies that bring intelligence to manufacturing operations and facilities. Additionally, humans are still one of the essential resources in manufacturing, however, they need to embrace and adapt to this new era

    WOS: 000332942700005PubMed ID: 24778561In this study, we assessed the feasibility of fetal RhD genotyping by analysis of cell-free fetal DNA(cffDNA) extracted from plasma samples of Rhesus (Rh) D-negative pregnant women by using real-time polymerase chain reaction (PCR). Fetal genotyping was performed on 30 RhD-negative women between 9 and 39 weeks of gestation who were referred to us for invasive testing [amniocentesis/chorionic villi sampling (CVS)]. The fetal RHD genotype was determined based on real-time PCR method. Exons 7 and 10 of the RHD and SRY genes were targeted. Among the pregnant women, 12 were carrying male and 17 were carrying female fetuses. Out of 29 pregnant women, 21 had RhD-positive and nine had RhD-negative fetuses. One sample) case 12, whose blood group was found to be AB Rh [+] (was excluded due to controversial results from repeated serological analyses. All prenatal results were in concordance with postnatal RhD status and fetal sex without false-positive or -negative results. Performing real-time PCR on cffDNA showed accurate, efficient and reliable results, allowing rapid and high throughput non invasive determination of fetal sex and RhD status in clinical samples

    IBD genetic risk profile in healthy first-degree relatives of Crohn's disease patients

    BACKGROUND: Family history provides important information on risk of developing inflammatory bowel disease [IBD], and genetic profiling of first-degree relatives [FDR] of Crohn's disease [CD]- affected individuals might provide additional information. We aimed to delineate the genetic contribution to the increased IBD susceptibility observed in FDR. METHODS: N = 976 Caucasian, healthy, non-related FDR; n = 4997 independent CD; and n = 5000 healthy controls [HC]; were studied. Genotyping for 158 IBD-associated single nucleotide polymorphisms [SNPs] was performed using the Illumina Immunochip. Risk allele frequency [RAF] differences between FDR and HC cohorts were correlated with those between CD and HC cohorts. CD and IBD genetic risk scores [GRS] were calculated and compared between HC, FDR, and CD cohorts. RESULTS: IBD-associated SNP RAF differences in FDR and HC cohorts were strongly correlated with those in CD and HC cohorts, correlation coefficient 0.63 (95% confidence interval [CI] 0.53 - 0.72), p = 9.90 x 10(-19). There was a significant increase in CD-GRS [mean] comparing HC, FDR, and CD cohorts: 0.0244, 0.0250, and 0.0257 respectively [p < 1.00 x 10(-7) for each comparison]. There was no significant difference in the IBD-GRS between HC and FDR cohorts [p = 0.81]; however, IBD-GRS was significantly higher in CD compared with FDR and HC cohorts [p < 1.00 x 10(-10) for each comparison]. CONCLUSION: FDR of CD-affected individuals are enriched with IBD risk alleles compared with HC. Cumulative CD-specific genetic risk is increased in FDR compared with HC. Prospective studies are required to determine if genotyping would facilitate better risk stratification of FDR

    Identifying hypothetical genetic influences on complex disease phenotypes

    <p>Abstract</p> <p>Background</p> <p>Statistical interactions between disease-associated loci of complex genetic diseases suggest that genes from these regions are involved in a common mechanism impacting, or impacted by, the disease. The computational problem we address is to discover relationships among genes from these interacting regions that may explain the observed statistical interaction and the role of these genes in the disease phenotype.</p> <p>Results</p> <p>We describe a heuristic algorithm for generating hypothetical gene relationships from loci associated with a complex disease phenotype. This approach, called Prioritizing Disease Genes by Analysis of Common Elements (PDG-ACE), mines biomedical keywords from text descriptions of genes and uses them to relate genes close to disease-associated loci. A keyword common to, and significantly over-represented in, a pair of gene descriptions may represent a preliminary hypothesis about the biological relationship between the genes, and suggest the role the genes play in the disease phenotype.</p> <p>Conclusion</p> <p>Our experimentation shows that the approach finds previously published relationships, while failing to find relationships that don't exist. The results also indicate that the approach is robust to differences in keyword vocabulary. We outline a brief case study in which results from a recently published Type 2 Diabetes association study are used to identify potential hypotheses.</p

    Clinical spectrum of early onset “Mediterranean” (homozygous p.P131L mutation) mitochondrial neurogastrointestinal encephalomyopathy

    Mitochondrial neurogastrointestinal encephalomyopathy (MNGIE) is an autosomal recessive mitochondrial disorder characterized by cumulative and progressive gastrointestinal and neurological findings. This retrospective observational study, aimed to explore the time of presentation, diagnosis and clinical follow-up of 13 patients with a confirmed MNGIE disease of Mediterranean origin. The mean age of symptom onset was 7 years (6 months−21 years) and the average diagnosis age was 15.4 years ±8.4. Four of 13 patients (30%) died before 30 years at the mean age of 19.7 years ±6.8. Cachexia and gastrointestinal symptoms were observed in all patients (100%). The mean body mass index standard deviation score at diagnosis was 4.8 ± 2.8. At least three subocclusive episodes were presented in patients who died in last year of their life. The main neurological symptom found in most patients was peripheral neuropathy (92%). Ten patients (77%) had leukoencephalopathy and the remaining three patients without were under 10 years of age. The new homozygous “Mediterranean” TYMP mutation, p.P131L (c.392 C > T) was associated with an early presentation and poor prognosis in nine patients (69%) from five separates families. Based on the observations from this Mediterranean MNGIE cohort, we propose that the unexplained abdominal pain combined with cachexia is an indicator of MNGIE. High-platelet counts and nerve conduction studies may be supportive laboratory findings and the frequent subocclusive episodes could be a negative prognostic factor for mortality. Finally, the homozygous p.P131L (c.392 C > T) mutation could be associated with rapid progressive disease with poor prognosis

    Combined effect of CCND1 and COMT polymorphisms and increased breast cancer risk

    <p>Abstract</p> <p>Background</p> <p>Estrogens are crucial tumorigenic hormones, which impact the cell growth and proliferation during breast cancer development. Estrogens are metabolized by a series of enzymes including COMT, which converts catechol estrogens into biologically non-hazardous methoxyestrogens. Several studies have also shown the relationship between estrogen and cell cycle progression through activation of CCND1 transcription.</p> <p>Methods</p> <p>In this study, we have investigated the independent and the combined effects of commonly occurring CCND1 (Pro241Pro, A870G) and COMT (Met108/158Val) polymorphisms to breast cancer risk in two independent Caucasian populations from Ontario (1228 breast cancer cases and 719 population controls) and Finland (728 breast cancer cases and 687 population controls). Both COMT and CCND1 polymorphisms have been previously shown to impact on the enzymatic activity of the coded proteins.</p> <p>Results</p> <p>Here, we have shown that the high enzymatic activity genotype of CCND1<sup>High </sup>(AA) was associated with increased breast cancer risk in both the Ontario [OR: 1.3, 95%CI (1.0–1.69)] and the Finland sample [OR: 1.4, 95%CI (1.01–1.84)]. The heterozygous COMT<sup>Medium </sup>(MetVal) and the high enzymatic activity of COMT<sup>High </sup>(ValVal) genotype was also associated with breast cancer risk in Ontario cases, [OR: 1.3, 95%CI (1.07–1.68)] and [OR: 1.4, 95%CI (1.07–1.81)], respectively. However, there was neither a statistically significant association nor increased trend of breast cancer risk with COMT<sup>High </sup>(ValVal) genotypes in the Finland cases [OR: 1.0, 95%CI (0.73–1.39)]. In the combined analysis, the higher activity alleles of the COMT and CCND1 is associated with increased breast cancer risk in both Ontario [OR: <b>2.22</b>, 95%CI (1.49–3.28)] and Finland [OR: <b>1.73</b>, 95%CI (1.08–2.78)] populations studied. The trend test was statistically significant in both the Ontario and Finland populations across the genotypes associated with increasing enzymatic activity.</p> <p>Conclusion</p> <p>Using two independent Caucasian populations, we have shown a stronger combined effect of the two commonly occurring CCND1 and COMT genotypes in the context of breast cancer predisposition.</p
