    Hepatoxicity of aqueous extract and fractionated methanol extract of Phytolacca americana by isolated rat liver perfusion system

    Recombinant retroviral vector containing human tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) gene was constructed and investigation of the in vitro invasion and metastasis of gastric cancer cells transfected with TIMP-2 was carried out. Human TlMP-2 was isolated from recombinant vector Bluescript 1/TIMP-2(+), and then inserted into the retroviral vector pL-MT. Correct orientation was verified by restriction endonuclease digestion. Human full length TIMP-2 gene was ligated into a plasmid, which was then transfected into PA317 cell line. G418-resistant individual clones were selected to transfect human SGC-7901 cell line. Cell proliferation, cell electrophoresis, soft agar colony formation and in vitro invasion were detected to analyze the bio-behavioral changes of cancer cells. The results from restriction endonuclease digestion were as theoretically expected. The cell electrophoresis rate, colony number and invasion ability in SGC-7901 cells and MFC cells transfected with TIMP-2 gene were significantly decreased when compared with control group. However, no significant changes were noted in the proliferation of cancer cells. We successfully construct a recombinant retroviral vector containing human TIMP-2. TIMP-2 transfection could markedly alter the membrane charge of cancer cells, resulting in decreased electrophoresis capacity, cell migration and invasion. However, cell growth was not affected by TIMP-2. These results suggested TIMP-2 transfection might exert effects on the malignant phenotype of cancer cells through affecting extracellular environment, which provided a new way to investigate gene regulation of in vitro collagen metabolism

    Possible spin-orbit driven spin-liquid ground state in the double perovskite phase of Ba3YIr2O9

    We report the structural transformation of hexagonal Ba3YIr2O9 to a cubic double perovskite form (stable in ambient conditions) under an applied pressure of 8GPa at 1273K. While the ambient pressure (AP) synthesized sample undergoes long-range magnetic ordering at 4K, the high pressure(HP) synthesized sample does not order down to 2K as evidenced from our susceptibility, heat capacity and nuclear magnetic resonance (NMR) measurements. Further, for the HP sample, our heat capacity data have the form gamma*T+beta*T3 in the temperature (T) range of 2-10K with the Sommerfeld coefficient gamma=10mJ/mol-Ir K2. The 89Y NMR shift has no T-dependence in the range of 4-120K and its spin-lattice relaxation rate varies linearly with T in the range of 8-45K (above which it is T-independent). Resistance measurements of both the samples confirm that they are semiconducting. Our data provide evidence for the formation of a 5d based, gapless, quantum spin-liquid (QSL) in the cubic (HP) phase of Ba3YIr2O9. In this picture, the T term in the heat capacity and the linear variation of 89Y 1/T1 arises from excitations out of a spinon Fermi surface. Our findings lend credence to the theoretical suggestion [G. Chen, R. Pereira, and L. Balents, Phys. Rev. B 82, 174440 (2010)] that strong spin-orbit coupling can enhance quantum fluctuations and lead to a QSL state in the double perovskite lattice.Comment: 6 pages 5 figure

    Coronary artery fistula is an uncommon but hemodynamically significant anomaly of the coronary arteries, occurring as an incidental finding in 0.1% to 0.2% of coronary angiograms. Although half of the patients with a coronary artery fistula remain asymptomatic, the other half develops CHF, infective endocarditis, myocardial ischemia, or rupture of an aneurysm.     Drug resistance patterns of bacteria isolated from patients with nosocomial pneumonia at Tehran hospitals during 2009-2011

    INTRODUCTION: Nosocomial pneumonia remains an important cause of mortality and morbidity worldwide. Surveillance programs play an important role in the identification of common etiologic agents and local patterns of antimicrobial resistance. METHODOLOGY: In this study we determined the frequency and antimicrobial susceptibility of pathogens isolated from patients with nosocomial pneumonia during 2009 to 2011. RESULTS: A total of 642 bacteria were isolated from 516 suspected samples. Acinetobacter baumannii (21.1%, n = 136), was the commonest isolated pathogen followed by Pseudomonas aeruginosa (17.4%, n = 112), Staphylococcus aureus (15.8%, n = 102) and enterococci (8.4% n = 54). The most effective therapeutic agents against A. baumannii were polymyxin B (95.5% susceptible), ceftriaxone/tazobactam (72% susceptible) and levofloxacin (52.9% susceptible). Polymixin B (89.2% susceptible), ceftriaxone/tazobactam (89.2% susceptible) and piperacillin-tazobactam (80.3% susceptible) were found to be the most active agents against P. aeruginosa. Extended-spectrum beta-lactamases were detected among isolates of K. pneumoniae (45.4%) and E. coli (20.3%). Overall, the prevalence of methicillin-resistant S. aureus and vancomycin resistant enterococci were 80.4% and 40.7% respectively. Linezolid was found to be the most active antibiotic against these pathogens. The etiology of 50% of the nosocomial infection cases was polymicrobial. CONCLUSIONS: The combination of ceftriaxone/tazobactam seems to be beneficial agent against multidrug-resistant Gram-negative bacilli isolated form respiratory tract infections. The results of our study can be used for guiding appropriate empiric therapy in this geographic region

    Facteurs liés au diagnostic tardif du cancer du sein: expérience du CHU Mohammed VI Marrakech

    Le cancer du sein est le premier cancer féminin en termes d'incidence et de mortalité. Au Maroc, il vient au premier rang des cancers de la femme avant celui du col utérin Il constitue un problème de santé publique. Son pronostic est étroitement lié au stade auquel le diagnostic est posé. Il s'agit d'une pathologie dont les moyens diagnostiques sont de nos jours développés, allant de la détection précoce à la mise en évidence de lésions infra-cliniques, ce qui a nettement amélioré le pronostic dans les pays développés. Ce travail que nous présentons a pour objectif d'identifier dans notre pratique quotidienne, les facteurs qui amènent les patientes à consulter à des stades tardifs. Une étude rétrospective a été menée de janvier 2012 à janvier 2013 portant sur 130 patientes porteuses d'un cancer du sein au sein du service d'onco-radiotherapie CHU Mohammed VI Marrakech. Un questionnaire a été élaboré et dument renseigné en ayant recours aux dossiers des malades. Ainsi 63,07% des patientes consultaient au-delà de six mois avec un délai moyen de consultation de 8,47 mois avec comme motif de consultation des lésions classées T4 dans 27,69%, et des tumeurs d'emblée métastatiques dans 13,84%. Les facteurs retrouvés à l'interrogatoire étaient le manque de moyens financiers 40%, l'éloignement des structures sanitaires dans 23%, les habitudes socioculturelles avec les traitements traditionnels en première intention 20%, et l'insuffisance de prise en charge thérapeutique 7%. Cependant, pris individuellement, aucune concordance significative n'était retrouvée entre ces facteurs et le long délai diagnostique. Dans notre pratique, c'est la conjonction de la triade ignorance, indigence et habitudes socioculturelles qui constituent le facteur essentiel du diagnostic tardif des cancers du sein

    Treatment of Iron-deficiency Anemia in Patients with Subclinical Hypothyroidism

    Subclinical hypothyroidism is a health state that is associated with hypercholesterolemia, infertility, iron-deficiency anemia, and poor obstetric outcome. This article summarizes the results of a prospective clinical investigation of whether treatment of subclinical hypothyroidism and iron-deficiency anemia with a combination of levothyroxine plus iron salt would be superior to each treatment alone. METHODS: In a randomized, double-blind, active-controlled trial, 60 patients with subclinical hypothyroidism and iron-deficiency anemia received iron salt+placebo (20 patients), levothyroxine+placebo (20 patients), or levothyroxine+iron salt (20 patients) for 3 months. Change from baseline (before) to end of study (after) in hemoglobin, ferritin, and thyroid-stimulating hormone levels were compared among groups. RESULTS: The increase from baseline in hemoglobin and ferritin in the levothyroxine+iron group was superior to the other groups, in which a decrease in thyroid-stimulating hormone in the 2 groups that received levothyroxine was superior to the group treated with iron salt. CONCLUSION: Subclinical hypothyroidism was investigated in iron-deficient patients with no acceptable response to iron salt alone. A combination of levothyroxine and iron salt is better than each one alone

    The effect of intra-wound vancomycin powder application in reducing surgical site infections after total hip arthroplasty

    Post-operative total hip arthroplasty surgical wound infections are associated with substantial morbidity, mortality, and economic burden. Intra-wound vancomycin powder application may prevent infections after hip surgery. The objectives of this study is to answer the question as to whether the application of 2 gram intra-wound vancomycin powder In patients undergoing total hip arthroplasty, lead to reduced rates of postoperative surgical site infections or not? From April 2013, 125 patients had consent prior to their hip surgery. These 125 patients had total hip arthtoplasty by three different surgeons at Imam Hossein hospital. All patients received either 2 grams of IV Cefazolin one hour prior to the incision or 1 gram of IV Vancomycin one hour prior to the incision in patient with a Penicillin allergy. Near the end of the procedure, the wound was thoroughly irrigated with normal saline. for All patients befor surgical wound closure 2 gram of Vancomycin powder was poured into the wound. Postoperatively, patients continued on intravenous antibiotics for twenty-four hours. All patients underwent routine postoperative surgical care including physical therapy, routine radiographs, and routine managment of pain medications. Laboratory tests include Hb , BUN, Cr and electrolits were done before surgery and postoperatively after 48 hours and 7 days. All patients were discharged within the first 3 days after surgery and follow-up at weeks 2 and 4 and one year after surgery was performed. During the course of follow up that has ranged from one months to one year, sixteen of patients experienced surgical site infection. 10 patients were lost to follow up. No allergic reactions or adverse outcomes were reported from the use of 2 gram of Vancomycin powder prior to closure. Although intra-wound application of Vancomycin is known to minimize infections in spinal surgery little previous studies reported if Vancomycin application will reduce surgical site infection in hip surgeries. This study demonstrates that intra-wound application of Vancomycin reduces surgical site infection in total hip arthroplasty without any adverse complications

    Impact of hybrid plasmonic nanoparticles on the charge carrier mobility of P3HT:PCBM polymer solar cells

    The solution processable polymer solar cells have shown a great promise as a cost-effective photovoltaic technology. Here, the effect of carrier mobility changes has been comprehensively investigated on the performance of P3HT:PCBM polymer solar cells using electro-optical coupled simulation regimes, which may result from the embedding of SiO2@Ag@SiO2 plasmonic nanoparticles (NPs) in the active layer. Firstly, the active layer thickness, stemmed from the low mobility of the charge carriers, is optimized. The device with 80 nm thick active layer provided maximum power conversion efficiency (PCE) of 3.47%. Subsequently, the PCE has increased to 6.75% and 6.5%, respectively, along with the benefit of light scattering, near-fields and interparticle hotspots produced by embedded spherical and cubic nanoparticles. The PCE of the devices with incorporated plasmonic nanoparticles are remarkably enhanced up to 7.61% (for spherical NPs) and 7.35% (for cubic NPs) owing to the increase of the electron and hole mobilities to [Formula: see text] and [Formula: see text], respectively (in the optimum case). Furthermore, SiO2@Ag@SiO2 NPs have been successfully synthesized by introducing and utilizing a simple and eco-friendly approach based on electroless pre-treatment deposition and Stober methods. Our findings represent a new facile approach in the fabrication of novel plasmonic NPs for efficient polymer solar cells
