    Plant productivity : a predictor of animal species and community characteristics : ecological patterns from local to global scale

    This thesis focus on the effects of plant productivity on various aspects of geographic range size, resource use, body size and abundance of animal species from different taxa, from a local to a global scale. The aim was to identify putative scales to determine whether it is possible to find functional links between plant productivity and species richness by considering how productivity affects species characteristics for different assemblages, e.g. mammals, insects and birds. Terrestrial endemic mammal genera in the world appeared to be most common in zoo-geographic regions of intermediate plant productivity (Paper I). I could not find evidence that Australian herbivorous mammals either combine food resources in more ways or are more selective in areas of high plant productivity (Paper II). Although, these Australian herbivorous mammals were larger and more similar in size in biomes of high plant productivity than in low (Paper HI). I further, found that for two families of ground dwelling Coleoptera plant productivity does not have any effect on the relationship between number of individuals and number of species within body size classes in local forest sites (Paper IV). There was a trend that higher productivity sites could hold more individuals of large sized species, but not more species. The reverse trend was observed for small sized species. Breeding forest birds in Sweden had highest average densities in areas of both low and high plant productivity (Paper V). Also, species with a lower than expected from their range size were mainly found in areas of high plant productivity while the reverse was true for species with higher than expected abundance. Finding functional links between observed patterns of animal characteristics and between species richness and productivity are vital to our understanding of how the transfer of energy from plants to animals affects the distribution of species. The spatial scale at which to study the mechanisms does influence how these proposed functional links will be affected by productivity. In this thesis I show that changes in productivity are likely to induce complex interactive changes in all key attributes, rather than clear linear responses. I therefore suggest that future studies should consider all these attributes in conjunction

    Gender perspective on older people’s exercise preferences and motivators in the context of falls prevention: a qualitative study

    Background: Several factors have previously been identified to positively influence the uptake and adherence for fall prevention exercise programmes. There is, however, a lack of studies investigating if men and women differ in their views and preferences for fall prevention exercises. Aim: To explore exercise preferences and motivators of older community-dwelling women and men in the context of falls prevention from a gender perspective. Methods: Workshops including multistage focus group discussions were conducted with 18 older community-dwelling people with and without history of falls. Participants were purposively selected and divided into two groups. Each group met on six occasions over a period of five months. Participatory and Appreciative Action and Reflection methodology was used to guide the discussions. A qualitative content analysis approach was used in the analysis. Results: Older participants had many diverse preferences and confirmed that individually tailored exercise, in terms of mode, intensity, challenge, and social context, is important. Moreover, important factors for exercise adherence and maintenance included the experience of individual confirmation; different spirit lifters to increase enjoyment; and personal tricks to maintain exercise routines. The individual differences within genders were more diverse than the differences between women and men. Conclusion: Exercise interventions to prevent falls should be individually tailored, based on the specific needs and preferences of the older participant, and do not appear to require gender specific approaches. To increase adherence, intrinsic motivation for exercise may be encouraged by competence enhancing confirmations, energizing spirit lifters, and practical tips for exercise maintenance. The study provides an awareness about women's and men's preferences for fall prevention exercises, and this information could be used as guidance in designing inclusive exercise interventions

    Behaviour of reindeer as an indicator of an adaptation to feeding

    Abstract: We assessed behaviour of reindeer affected by nutritional deprivation and how they adapted to various feeding strategies. The activity pattern of 61 penned eight month old female reindeer calves was observed during 20 of a total of 42 experimental days in winter 1997. The dominant activities were lying, ruminating, intake of feed and water, and standing. Few recordings of agonistic behaviour or snow intake occured. Restricted feed intake, half the ad lib. ration of a lichen-based diet, affected the eating behaviour of the reindeer, and more animals were standing and fewer lying compared to reindeer fed ad lib. Lack of energy in the diet correlated with animals lying curled up (lying with the muzzle close to the hind legs). This behaviour could be a useful complement to other measurements and registrations when studying adaptations to various feeding regimens.Abstract in Swedish/Sammanfattning:Syftet med studien var att undersöka om, och i så fall hur renars beteende påverkades av otillräckligt näringsintag och vid anpassning till olika utfodringsstrategier. Aktivitetsmönstret hos 61 inhägnade åtta månader gamla honrenkalvar studerades under 20 av totalt 42 försöksdagar. De vanligaste beteendekategorierna genom hela försöket var ligga, idissla, intag av foder och vatten samt stå passivt. Endast ett fåtal observationer av aggressivt beteende och snöätande registrerades. En begränsad giva dvs. halva mängden av fodergivan vid fri tillgång av en lavbaserad diet påverkade djuren ätbeteende. Dessutom observerades fler djur stå passivt medan färre låg jämfört med kontrollgruppen. Under första fasen av utfodring efter restriktionsperioden låg fler djur låg ihoprullade (med mulen tätt intill bakbenet) jämfört med kontrollgruppen, vilket tolkades som ett tecken på energibrist. Beteendestudierna visade sig vara ett värdefullt komplement till övriga mätningar och provtagningar vid studier av renars anpassning till olika utfodringsregimer

    The human epidermal growth factor receptor 2 gene (HER2) is amplified in approximately 15%–20% of all breast cancers. This results in overexpression of the HER2 protein, which is associated with worse clinical outcomes in breast cancer patients. Several studies have shown that trastuzumab, a monoclonal antibody that interferes with the HER2/neu receptor, can improve overall survival in patients with HER2-positive breast cancer. Immunohistochemistry (IHC), combined with different methods for in situ hybridization, is currently used for routine assessment of HER2 status. The aim of the present study was to determine whether real-time polymerase chain reaction (PCR) can serve as a supplementary method for evaluation of HER2 status in primary breast cancer. For this purpose, 145 formalin-fixed paraffin-embedded primary breast cancer samples were tested by real-time PCR amplification of HER2, using amyloid precursor protein as a reference. The results were compared with HER2 status determined by fluorescence in situ hybridization (FISH) and IHC. The specificity, sensitivity, and reproducibility of real-time PCR were evaluated, and a comparison of formalin-fixed and fresh-frozen samples was performed. This showed concordance of 93% between real-time PCR and FISH, and 86% between real-time PCR and IHC. Therefore, we suggest that real-time PCR can be a useful supplementary method for assessment of HER2 status

    Molecular cloning and characterization of a nuclear androgen receptor activated by 11-ketotestosterone

    Although 11-ketotestosterone is a potent androgen and induces male secondary sex characteristics in many teleosts, androgen receptors with high binding affinity for 11-ketotestosterone or preferential activation by 11-ketotestosterone have not been identified. So, the mechanism by which 11-ketotestosterone exhibits such high potency remains unclear. Recently we cloned the cDNA of an 11-ketotestosterone regulated protein, spiggin, from three-spined stickleback renal tissue. As spiggin is the only identified gene product regulated by 11-ketotestosterone, the stickleback kidney is ideal for determination of the mechanism of 11-ketotestosterone gene regulation. A single androgen receptor gene with two splicing variants, belonging to the androgen receptor-β subfamily was cloned from stickleback kidney. A high affinity, saturable, single class of androgen specific binding sites, with the characteristics of an androgen receptor, was identified in renal cytosolic and nuclear fractions. Measurement of ligand binding moieties in the cytosolic and nuclear fractions as well as to the recombinant receptor revealed lower affinity for 11-ketotestosterone than for dihydrotestosterone. Treatment with different androgens did not up-regulate androgen receptor mRNA level or increase receptor abundance, suggesting that auto-regulation is not involved in differential ligand activation. However, comparison of the trans-activation potential of the stickleback androgen receptor with the human androgen receptor, in both human HepG2 cells and zebrafish ZFL cells, revealed preferential activation by 11-ketotestosterone of the stickleback receptor, but not of the human receptor. These findings demonstrate the presence of a receptor preferentially activated by 11-ketotestosterone in the three-spined stickleback, so far the only one known in any animal

    Carbon and nitrogen pools and mineralization rates in boreal forest soil after stump harvesting

    The use of forest-derived biomass has steadily increased in Finland and Sweden during the past decades leading to more intensive forest management practices in the region, such as whole-tree harvesting, both above- and belowground. Stump harvesting results in a direct removal of stump and coarse-root carbon (C) from the stand and can cause extensive soil disturbance, which has been suggested to increase C mineralization. In this study, the effects of stump harvesting on soil C and nitrogen (N) mineralization, and soil surface disturbance were studied in two different clear-felled Norway spruce (Picea abies) sites in Central Finland. The treatments were whole-tree harvesting (WTH, removal of stems and logging residues), and WTH and stump harvesting (WTH + S). Both sites, Honkola (2 stands) and Haukilahti (6 stands) were mounded. In both treatments, soil samples were taken from different soil layers down to a total depth of 20 cm in the mineral soil from (i) mounds, (ii) undisturbed soil and (iii) pits. The sampling was performed 11-12 years after treatments. Soil C and N mineralization rates were determined in laboratory incubation experiments. In addition, total C and N pools (g m(2)) were estimated for each disturbance class and soil layer. Soil C and N pools had a tendency to be lower following stump harvesting, but no statistically significant treatment effect was detected. Stump harvesting increased soil mixing as indicated by a significant decrease in C concentration in the mound disturbance class. There was no significant effect of stump harvesting on soil C mineralization rates. A combination of mineralization rates and soil pool data showed that field C mineralization (g CO2-C m(-2) yr(-1)) did not significantly differ between stands where stumps were removed or were retained. Further, stump harvesting did not seem to have any stimulating effect on soil CO2 efflux 11-12 years after treatment. (C) 2016 Elsevier B.V. All rights reserved.Peer reviewe
