    Effects of exercise performed during the reproductive cicle: analysis of studies using animal models

    O ciclo reprodutivo impõe várias adaptações no metabolismo lipídico, garantindo modificações específicas quanto ao direcionamento de substratos circulantes, na gestação para o crescimento fetal, durante a lactação para a produção de leite, e após o desmame para a reposição de tecido adiposo. Por outro lado, durante o exercício há aumento na captação de substratos pelo músculo. Desta forma, através desta revisão objetiva-se responder à seguinte questão: o exercício realizado durante o ciclo reprodutivo pode contribuir para o retomo ao estado pré-gestacional sem causar efeito deletério para o crescimento fetal e da cria?.     L'adsorption sur charbon actif est une technique relativement répandue de post-traitement des effluents aqueux. Cependant il ne s'agit pas d'un traitement ultime, puisque les polluants restent concentrés sur la surface solide. Des techniques de régénération doivent donc être mises en œuvre. C'est dans ce contexte qu'ont été étudiés ici les procédés d'oxydation avancée Fenton et photo-Fenton, dont l'efficacité est reconnue pour éliminer la pollution organique. Plus précisément, l'objet de cette thèse est l'étude sur plusieurs cycles consécutifs de l'adsorption du phénol sur charbon actif et de la régénération in-situ de l'adsorbant par oxydation (photo)Fenton. Deux charbons actifs différents ont été étudiés : le premier à la fois micro et mésoporeux (PICA L27) et le second essentiellement microporeux (PICA S23). Deux séries d'expériences ont été ainsi réalisées : 1) d'abord en réacteur agité (adsorption et oxydation en mode batch), 2) puis dans des conditions plus proches du procédé réel, avec une adsorption continue en lit fixe, suivie de l'oxydation batch par recirculation du réactif Fenton au travers du lit saturé. Dans le premier cas, les effets de la concentration en Fe2+ et en H2O2 ont été analysés, montrant que les conditions optimales pour l'oxydation homogène du phénol (sans charbon) ne sont pas les meilleures pour la régénération du charbon saturé par le polluant : une réduction continue de la capacité d'adsorption du L27, de 100% à 23%, est observée après 3 oxydations, en raison de la consommation du charbon et de la diminution de sa surface spécifique. Par contre, une concentration plus élevée de Fe2+ et plus faible de H2O2 (2 fois la stœchiométrie) permettent de retrouver 50% de la capacité initiale d'adsorption pendant au moins 4 cycles consécutifs. Comme il a été vu dans des études précédentes utilisant l'oxydation à l'air (sous température et pression), l'efficacité de régénération est aussi bien plus faible pour le S23 (autour de 20%). Des résultats similaires ont été obtenus sur le réacteur à lit fixe avec recirculation. Durant l'oxydation, le taux de conversion du Carbone Organique Total en phase liquide a atteint à chaque fois une valeur limite, probablement du fait de la formation de complexes entre le fer et les acides carboxyliques produits. L'utilisation de l'irradiation UV, qui est connue pour décomposer ces complexes, a conduit à une minéralisation quasi-totale et a amélioré l'efficacité de régénération du charbon, jusqu'à 56% de la capacité initiale après 2 cycles (contre 40% pour l'oxydation Fenton simple)Adsorption on activated carbon (AC) is a technique extensively applied for wastewater treatment. However adsorption alone is not an ultimate solution, since the pollutants are just immobilized on the carbon surface. There is thus a need for efficient regeneration techniques. In this context, Fenton and photo-Fenton oxidations, which are promising technologies to destroy organic pollution, have been tested to regenerate the AC. The purposes of this study are the adsorption of phenol on activated carbons and the consecutive in-situ regeneration of carbon by (photo-) Fenton oxidation. Two different operations have been carried out: 1) batch procedure in order to investigate the influence of Fe2+ and H2O2 concentrations; 2) continuous fixed bed adsorption, followed by a batch circulation of the Fenton s reagent through the saturated AC bed, to examine the efficiency of the real process. Two different activated carbons have been also studied: a both micro- and mesoporous AC (PICA L27) and an only microporous one (PICA S23). In the batch reactor the best conditions found for pollutant mineralization in the homogeneous Fenton system are not the best for AC regeneration: a continuous reduction of adsorption capacity of L27 is observed after 3 oxidations, due to the decrease of both AC weight and surface area. Higher concentration of Fe2+ and lower concentration of H2O2 (2 times the stoechiometry) lead to a 50% recovery of the initial adsorption capacity during at least 4 consecutive cycles for L27, while about 20% for S23. In the consecutive continuous adsorption/batch Fenton oxidation process, the regeneration efficiency reaches 30% to 40% for L27 after two cycles whatever the feed concentration and less than 10% for S23.     Assessment of Sugarcane-Based Ethanol Production

    This chapter aims to explain how bio-ethanol has been drawn to become a successful alternative to partially replace petroleum as a source of liquid fuels in Brazil. A brief historical analysis about the production of bio-ethanol from sugarcane is presented. The motivation to start the production of the ethanol as biofuel in the 1970s and how the governmental policies have contributed to the ups and downs, successes, and failures of the sugarcane industry is shown. Then, the efficiency of the sector is addressed; firstly, the increasing efficiency of the agricultural sector is discussed, showing how the productivity per hectare has increased in the last decades and which improvements are further expected in a near future. Finally, the industrial process is discussed: the current efficiency in processing sugarcane to produce ethanol and the emerging technologies, not only to process sugarcane juice, but also to harness bagasse, vinasse, and sugarcane straw

    A Palatable Hyperlipidic Diet Causes Obesity and Affects Brain Glucose Metabolism in Rats

    Background We have previously shown that either the continuous intake of a palatable hyperlipidic diet (H) or the alternation of chow (C) and an H diet (CH regimen) induced obesity in rats. Here, we investigated whether the time of the start and duration of these feeding regimens are relevant and whether they affect brain glucose metabolism. Methods Male Wistar rats received C, H, or CH diets during various periods of their life spans: days 30-60, days 30-90, or days 60-90. Experiments were performed the 60th or the 90th day of life. Rats were killed by decapitation. The glucose, insulin, leptin plasma concentration, and lipid content of the carcasses were determined. The brain was sliced and incubated with or without insulin for the analysis of glucose uptake, oxidation, and the conversion of [1-14C]-glucose to lipids. Results The relative carcass lipid content increased in all of the H and CH groups, and the H30-60 and H30-90 groups had the highest levels. Groups H30-60, H30-90, CH30-60, and CH30-90 exhibited a higher serum glucose level. Serum leptin increased in all H groups and in the CH60-90 and CH30-90 groups. Serum insulin was elevated in the H30-60, H60-90, CH60-90, CH30-90 groups. Basal brain glucose consumption and hypothalamic insulin receptor density were lower only in the CH30-60 group. The rate of brain lipogenesis was increased in the H30-90 and CH30-90 groups. Conclusion These findings indicate that both H and CH diet regimens increased body adiposity independent treatment and the age at which treatment was started, whereas these diets caused hyperglycemia and affected brain metabolism when started at an early age

    Early exposure to distinct sources of lipids affects differently the development and hepatic inflammatory profiles of 21-day-old rat offspring

    Introduction: Maternal diet composition of fatty acids during pregnancy and lactation seems to modify the fetal programming, epigenetic pattern and offspring phenotype. Aim: Herein, we investigated the effects of maternal consumption of normal-fat diets with distinct lipid sources during pregnancy and lactation on the somatic development and proinflammatory status of 21-day-old rat offspring. Materials and Methods: On the first day of pregnancy, female Wistar rats were divided into four groups as follows: soybean oil (M-SO), lard (M-L), hydrogenated vegetable fat (M-HVF) and fish oil (M-FO). Diets were maintained during pregnancy and lactation. Male offspring constituted the SO, L, HVF and FO groups. Pups were weighed and measured weekly. Lipopolysaccharide serum concentration was determined. Tumor necrosis factor alpha, interleukin (IL)-6 and IL-10 in the liver were evaluated by enzyme-linked immunosorbent assay. Liver gene expressions were determined by real-time polymerase chain reaction. Protein expressions in the liver were analyzed by Western blotting. Results: We observed an increase in body weight and adiposity in L and HVF groups. Moreover, HVF group showed an increase in the toll-like receptor 4 mRNA levels, IL10R alpha and phosphorylated form of I kappa B kinase (IKKp-IKK alpha+beta) protein expression. The FO group presented a decrease in body weight, relative weight of retroperitoneal adipose tissue, ADIPOR2 gene expression, lipopolysaccharide and p-IKK alpha+beta and phosphorylated form of nuclear transcription factor kappa B (NF kappa B) p50 (p-NF kappa B p50) protein expression. Conclusion: Summarily, whereas maternal intake of normal-fat diets based on L and HVF appear to affect the somatic development negatively, only early exposure to HVF impairs the pups' proinflammatory status.     Intracellular biosynthesis and removal of copper nanoparticles by dead biomass of yeast isolated from the wastewater of a mine in the brazilian Amazonia

    In this study was developed a natural process using a biological system for the biosynthesis of nanoparticles (NPs) and possible removal of copper from wastewater by dead biomass of the yeast Rhodotorula mucilaginosa. Dead and live biomass of Rhodotorula mucilaginosa was used to analyze the equilibrium and kinetics of copper biosorption by the yeast in function of the initial metal concentration, contact time, pH, temperature, agitation and inoculum volume. Dead biomass exhibited the highest biosorption capacity of copper, 26.2 mg g(-1), which was achieved within 60 min of contact, at pH 5.0, temperature of 30°C, and agitation speed of 150 rpm. The equilibrium data were best described by the Langmuir isotherm and Kinetic analysis indicated a pseudo-second-order model. The average size, morphology and location of NPs biosynthesized by the yeast were determined by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and transmission electron microscopy (TEM). The shape of the intracellularly synthesized NPs was mainly spherical, with an average size of 10.5 nm. The X-ray photoelectron spectroscopy (XPS) analysis of the copper NPs confirmed the formation of metallic copper.     Strategies for reducing body fat mass: effects of liposuction and exercise on cardiovascular risk factors and adiposity

    Liposuction is the most popular aesthetic surgery performed in Brazil and worldwide. Evidence showing that adipose tissue is a metabolically active tissue has led to the suggestion that liposuction could be a viable method for improving metabolic profile through the immediate loss of adipose tissue. However, the immediate liposuction-induced increase in the proportion of visceral to subcutaneous adipose tissue could be detrimental to metabolism, because a high proportion of visceral to subcutaneous adipose tissue is associated with risk factors for cardiovascular disease. The results of studies investigating the effects of liposuction on the metabolic profile are inconsistent, however, with most studies reporting either no change or improvements in one or more cardiovascular risk factors. In addition, animal studies have demonstrated a compensatory growth of intact adipose tissue in response to lipectomy, although studies with humans have reported inconsistent results. Exercise training improves insulin sensitivity, inflammatory balance, lipid oxidation, and adipose tissue distribution; increases or preserves the fat-free mass; and increases total energy expenditure. Thus, liposuction and exercise appear to directly affect metabolism in similar ways, which suggests a possible interaction between these two strategies. To our knowledge, no studies have reported the associated effects of liposuction and exercise in humans. Nonetheless, one could suggest that exercise training associated with liposuction could attenuate or even block the possible compensatory fat deposition in intact depots or regrowth of the fat mass and exert an additive or even a synergistic effect to liposuction on improving insulin sensitivity and the inflammatory balance, resulting in an improvement of cardiovascular risk factors. Consequently, one could suggest that liposuction and exercise appear to be safe and effective strategies for either the treatment of metabolic disorders or aesthetic purposes

    Supplementing alpha-tocopherol (vitamin E) and vitamin D3 in high fat diet decrease IL-6 production in murine epididymal adipose tissue and 3T3-L1 adipocytes following LPS stimulation

    <p>Abstract</p> <p>Background</p> <p>It is well known that high fat diets (HFDs) induce obesity and an increase in proinflammatory adipokines. Interleukin-6 (IL-6) is considered the major inflammatory mediator in obesity. Obesity is associated with a vitamin deficiency, especially of vitamins E and D3. We examined the effects of vitamin D3 and vitamin E supplementation on levels of IL-6 and IL-10 (as a marker of anti-inflammatory cytokines since, a balance between pro- and anti-inflammatory cytokines is maintained) protein expression in adipose tissue of mice provided with an HFD. Additionally, we measured the effects of vitamin E and vitamin D3 treatment on LPS-stimulated 3T3-L1 adipocytes IL-6 and IL-10 secretion.</p> <p>Results</p> <p>IL-6 protein levels and the IL-6/IL-10 ratio were decreased in epididymal white adipose tissue in groups receiving vitamins E and D3 supplementation compared to the HFD group. A 24-hour treatment of vitamin D3 and vitamin E significantly reduced the IL-6 levels in the adipocytes culture medium without affecting IL-10 levels.</p> <p>Conclusions</p> <p>Vitamin D3 and vitamin E supplementation in an HFD had an anti-inflammatory effect by decreasing IL-6 production in epididymal adipose tissue in mice and in 3T3-L1 adipocytes stimulated with LPS. Our results suggest that vitamin E and D3 supplementation can be used as an adjunctive therapy to reduce the proinflammatory cytokines present in obese patients.</p

    Hydrogenated fat diet intake during pregnancy and lactation modifies the PAI-1 gene expression in white adipose tissue of offspring in adult life

    We examine whether feeding pregnant and lactating rats hydrogenated fats rich in trans fatty acids modifies the plasma lipid profiles and the expression of adipokines involved with insulin resistance and cardiovascular disease in their 90-day-old offspring. Pregnant and lactating Wistar rats were fed with either a control diet (C group) or one enriched with hydrogenated vegetable fat (T group). Upon weaning, the male pups were sorted into four groups: CC, mothers were receiving C and pups were kept on C; CT, mothers were receiving C and pups were fed with T; TT, mothers were receiving T and pups were kept on T; TC, mothers were receiving T and pups were fed with C. Pups' food intake and body weight were quantified weekly and the pups were killed at day 90 of life by decapitation. Blood and carcass as well as retroperitoneal, epididymal, and subcutaneous white adipose tissues were collected. Food intake and body weight were lower in TC and TT, and metabolic efficiency was reduced in TT. Offspring of TT and TC rats had increased white adipose tissue PAI-1 gene expression. Insulin receptor was higher in TT than other groups. Ingestion of hydrogenated vegetable fat by the mother during gestation and lactation could promote deleterious consequences, even after the withdrawal of the causal factor

    Recuperação nutricional com farelo de arroz não modificou o balanço energético e os níveis de leptina e insulina

    OBJECTIVE: To investigate the effect of nutritional recovery with rice bran on energy balance, leptin and insulin levels. METHODS: Weaned Wistar rats were fed on a 17% (Control - C) or 0.5% (Aproteic - A) protein diet for 12d. After this, rats were kept on a C diet (C) or recovered with control (Recovered Control - RC) or control plus recovered rice bran diet (Recovered Rice Bran - RRB). RESULTS: Despite the increased food intake, group A exhibited lower carcass fat associated to low serum leptin. RRB and RC groups showed lower carcass weight and energy intake and expenditure. Energy expenditure was positively associated with food intake and carcass weight. Negative correlations between HOMA-IR and energy expenditure and energy intake were observed. CONCLUSION: Nutritional recovery with rice bran did not modify energy balance, leptin and insulin levels.OBJETIVO: Investigar o efeito da recuperação nutricional com farelo de arroz sobre o balanço energético e níveis de leptina e insulina. MÉTODOS: Ratos Wistar recém-desmamados foram alimentados com 17% (Controle - C) ou 0,5% (Aproteico - A) de proteína (caseína) durante 12 dias. Em seguida, ratos permaneceram com dieta controle (C) ou foram recuperados com controle (Recuperados Controle - RC) ou controle mais 5% de farelo de arroz (Recuperados com Farelo de Arroz - RFA) durante 21 dias. RESULTADOS: Apesar de a ingestão alimentar ter sido maior em A, a gordura na carcaça foi reduzida, sendo associada com menor nível de leptina. Os grupos RFA e RC tiveram redução no peso da carcaça, no gasto e ingestão de energia. O gasto energético foi correlacionado com a ingestão de alimentos e o peso da carcaça fresco. Foi observada correlação negativa entre HOMA-IR com gasto energético e com ingestão de energia.
