    Nucleotide-Binding Oligomerization Domain-1 and -2 Play No Role in Controlling Brucella abortus Infection in Mice

    Nucleotide-binding oligomerization domain proteins (NODs) are modular cytoplasmic proteins implicated in the recognition of peptidoglycan-derived molecules. Further, several in vivo studies have demonstrated a role for Nod1 and Nod2 in host defense against bacterial pathogens. Here, we demonstrated that macrophages from NOD1-, NOD2-, and Rip2-deficient mice produced lower levels of TNF-α following infection with live Brucella abortus compared to wild-type mice. Similar reduction on cytokine synthesis was not observed for IL-12 and IL-6. However, NOD1, NOD2, and Rip2 knockout mice were no more susceptible to infection with virulent B. abortus than wild-type mice. Additionally, spleen cells from NOD1-, NOD2-, and Rip2-deficient mice showed unaltered production of IFN-γ compared to C57BL/6 mice. Taken together, this study demonstrates that NOD1, NOD2 and Rip2 are dispensable for the control of B. abortus during in vivo infection

    Histologic and Histomorphometric Analysis of Posterior Region of the Human Temporomandibular Disc

    Objective The aim of this study was to analyze histologic and histomorphometric features of the articular disc in groups with and without disc displacement. Study design A sample of 39 temporomandibular joints TMJs (31 case specimens, 8 control specimens) from 28 patients (mean age 31.2 years) were recruited for this study. The patients were considered to be affected and treated surgically with disc repositioning when presenting painful clinical signs of disc displacement after unsuccessful nonsurgical treatment for at least 6 months. Of the control patients, 4 presented condyle fracture which required opening to be reduced for treatment, and 4 displayed active condyle hyperplasia. The posterior region of the disc was removed and sent for histologic and histomorphometric analysis. Histologic (hematoxylin-eosin) and histomorphometric (picro-Sirius red) analyses were performed. Statistically significant differences between the analyzed groups were accessed through the chi-squared test (P ≤ .05). The Mann-Whitney U test was used to observe the differences between mean values when variables did not present normal distribution [Kolmogorov-Smirnov(a) test]. Results There were no significant differences between the groups in relation to the parameters studied by histologic and histomorphometric analysis (using or not using polarization). Conclusions To the limits of this study, there were no significant histologic and histomorphometric differences in the articular disc between groups with and without TMJ dysfunction

    Human T Cell and Antibody-Mediated Responses to the Mycobacterium tuberculosis Recombinant 85A, 85B, and ESAT-6 Antigens

    Tuberculosis remains a major health problem throughout the world causing large number of deaths. Effective disease control and eradication programs require the identification of major antigens recognized by the protective responses against M. tuberculosis. In this study, we have investigated humoral and cellular immune responses to M. tuberculosis-specific Ag85A, Ag85B, and ESAT-6 antigens in Brazilian patients with pulmonary (P, n = 13) or extrapulmonary (EP, n = 12) tuberculosis, patients undergoing chemotherapy (PT, n = 23), and noninfected healthy individuals (NI, n = 7). Compared to NI, we observed increased levels of IgG1 responses to Ag85B and ESAT-6 in P and PT groups. Regarding cellular immunity, Ag85A and ESAT-6 were able to discriminate P, PT, and EP patients from healthy individuals by IFN-γ production and P and PT groups from EP individuals by production of TNF-α. In summary, these findings demonstrate the ability of Ag85A, Ag85B, and ESAT-6 to differentiate TB patients from controls by IgG1, IFN-γ and TNF-α production

    Megameter propagation and correlation of T-waves from Kermadec Trench and Islands

    On 18 June 2020 and 4 March 2021, very energetic low-frequency underwater T-wave signals (2 to 25 Hz) were recorded at the Comprehensive Nuclear-Test-Ban Treaty (CTBT) International Monitoring System (IMS) hydrophone stations in the Pacific Ocean (Stations HA11 and HA03) and the South Atlantic Ocean (Station HA10). This work investigates the long-range (megameters) propagation of these T-waves. Their sources were three powerful submarine earthquakes in the Kermadec Trench and Islands, located at approximately 6000, 8800, and 15100 km from Stations HA11, HA03, and HA10, respectively. Arrival time and back azimuth of the recorded T-waves were estimated using the Progressive Multi-Channel Correlation algorithm installed on the CTBT Organization (CTBTO) virtual Data Exploitation Centre (vDEC). Different arrivals within the duration of the earthquake signals were identified, and their correlations were also analyzed. The data analysis at HA03 and HA10 revealed intriguing T-wave propagation paths reflecting, refracting, or even transmitting through continents, as well as T-wave excitation along a chain of seamounts. The analysis also showed much higher transmission loss (TL) in the propagation paths to HA11 than to HA03 and HA10. Moreover, strong discrepancies between expected and measured back azimuths were observed for HA11, and a three-dimensional (3D) parabolic equation model was utilized to identify the cause of these differences. Numerical results revealed the importance of 3D effects induced by the Kermadec Ridge, Fiji archipelago, and Marshall Islands on T-wave propagation to HA11.     Type of diet modulates the metabolic response to sleep deprivation in rats

    <p>Abstract</p> <p>Background</p> <p>Evidence suggests that sleep loss is associated with an increased risk of obesity and diabetes; however, animal models have failed to produce weight gain under sleep deprivation (SD). Previous studies have suggested that this discrepancy could be due to more extreme SD conditions in experimental animals, their higher resting metabolic rate than that of humans, and the decreased opportunity for animals to ingest high-calorie foods. Thus, our objective was to determine whether diets with different textures/compositions could modify feeding behavior and affect the metabolic repercussions in SD in rats.</p> <p>Methods</p> <p>Three groups of male rats were used: one was designated as control, one was sleep deprived for 96 h by the platform technique (SD-96h) and one was SD-96h followed by a 24-h recovery (rebound). In the first experiment, the animals were fed chow pellets (CPs); in the second, they received high-fat diet and in the third, they were fed a liquid diet (LD).</p> <p>Results</p> <p>We observed that SD induces energy deficits that were related to changes in feeding behavior and affected by the type of diet consumed. Regardless of the diet consumed, SD consistently increased animals' glucagon levels and decreased their leptin and triacylglycerol levels and liver glycogen stores. However, such changes were mostly avoided in the rats on the liquid diet. SD induces a wide range of metabolic and hormonal changes that are strongly linked to the severity of weight loss.</p> <p>Conclusions</p> <p>The LD, but not the CP or high-fat diets, favored energy intake, consequently lessening the energy deficit induced by SD.</p

    Host Susceptibility to Brucella abortus Infection Is More Pronounced in IFN-γ knockout than IL-12/β2-Microglobulin Double-Deficient Mice

    Brucella abortus is a facultative intracellular bacterial pathogen that causes abortion in domestic animals and undulant fever in humans. IFN-γ, IL-12, and CD8+ T lymphocytes are important components of host immune responses against B. abortus. Herein, IFN-γ and IL-12/β2-microglobulin (β2-m) knockout mice were used to determine whether CD8+ T cells and IL-12-dependent IFN-γ deficiency would be more critical to control B. abortus infection compared to the lack of endogenous IFN-γ. At 1 week after infection, IFN-γ KO and IL-12/β2-m KO mice showed increased numbers of bacterial load in spleens; however, at 3 weeks postinfection (p.i.), only IFN-γ KO succumbed to Brucella. All IFN-γ KO had died at 16 days p.i. whereas death within the IL-12/β2-m KO group was delayed and occurred at 32 days until 47 days postinfection. Susceptibility of IL-12/β2-m KO animals to Brucella was associated to undetectable levels of IFN-γ in mouse splenocytes and inability of these cells to lyse Brucella-infected macrophages. However, the lack of endogenous IFN-γ was found to be more important to control brucellosis than CD8+ T cells and IL-12-dependent IFN-γ deficiencies

    Evolutionary processes underlying latitudinal differences in reef fish biodiversity

    Aim: To examine the dynamics among the processes of speciation, extinction and dispersal in marine environments using phylogenies to reveal the evolutionary mechanisms that promote latitudinal differences in biodiversity. Using phylogenetic comparative methods we assess whether tropical reef fish lineages show higher diversification rates and whether the majority of extratropical reef fish lineages have originated from tropical areas. Location: Shallow water tropical and extratropical reefs globally. Methods: Using fossil-calibrated phylogenies for four reef-associated fish families (Chaetodontidae, Labridae, Pomacentridae and Sparidae) we apply evolutionary models (GeoSSE and HiSSE) that allow the estimation of speciation, extinction and dispersal rates associated with geographical ranges and explore potential biases from unsampled characters. Results: We found that tropical lineages show higher rates of speciation and tended to have lower extinction rates. Overall, we identify higher net diversification rates for tropical lineages compared with those in extratropical regions in all four families. Rates of dispersal tended to be higher for lineages with tropical origins expanding into extratropical regions. Within the family Labridae, two tropical lineages were found to exhibit higher net diversification rates, above that expected from latitudinal differences. Main conclusions : Our results offer support for the predictions of the out of the tropics' and evolutionary speed' models of evolution, both of which highlight the marine tropics as an important evolutionary engine promoting latitudinal differences in reef fish biodiversity. Moreover, we find that two tropical labrid lineages are undergoing exceptional diversification associated with additional traits, possibly linked with the extreme sexual dichromatism observed in both clades
