Streamlining skin regeneration: a ready-to-use silk bilayer wound dressing
Silk proteins have been highlighted in the past decade for tissue engineering (TE) and skin regeneration due to their biocompatibility, biodegradability, and exceptional mechanical properties. While silk fibroin (SF) has high structural and mechanical stability with high potential as an external protective layer, traditionally discarded sericin (SS) has shown great potential as a natural-based hydrogel, promoting cell–cell interactions, making it an ideal material for direct wound contact. In this context, the present study proposes a new wound dressing approach by developing an SS/SF bilayer construct for full-thickness exudative wounds. The processing methodology implemented included an innovation element and the cryopreservation of the SS intrinsic secondary structure, followed by rehydration to produce a hydrogel layer, which was integrated with a salt-leached SF scaffold to produce a bilayer structure. In addition, a sterilization protocol was developed using supercritical technology (sCO2) to allow an industrial scale-up. The resulting bilayer material presented high porosity (>85%) and interconnectivity while promoting cell adhesion, proliferation, and infiltration of human dermal fibroblasts (HDFs). SS and SF exhibit distinct secondary structures, pore sizes, and swelling properties, opening new possibilities for dual-phased systems that accommodate the different needs of a wound during the healing process. Microbiological evaluation of food manipulator's hands and nasal cavity in a food unit of a university hospital
Este trabalho teve como objetivo identificar os microorganismos presentes nas mãos e fossas nasais de manipuladores de alimentos da cozinha de um hospital universitário. Foram realizadas coletas de amostras de 24 manipuladores de alimentos, usando swab embebido em solução salina. As amostras foram semeadas em meio de cultivo específico para isolamento e identificação dos microorganismos presentes. Dos 24 manipuladores analisados, todos albergavam Staphylococcus aureus , sendo que 41,67% albergavam esta bactéria nas fossas nasais, 41,67% apenas na palma das mãos, e 16,67% albergavam tanto nas mãos quanto nas fossas nasais.Escherichia coli esteve presente em 29,17% dos manipuladores, sendo 28,57% nas fossas nasais e 71,43% nas mãos dos manipuladores. Em 12,5% dos manipuladores foram encontradas ambas as bactérias, S. aureus e E. coli , nas mãos. Ademonstração de S. aureus e E. coli nas amostras isoladas dos manipuladores de alimento apontam para a possibilidade da contaminação de alimentos, a partir da colonização das mãos e fossas nasais de manipuladores. A prevenção desta contaminação pode ser feita pelo meio de medidas educativas e disponibilização de produtos que facilitem a higienização adequada das mãos no ambiente de trabalho. This work had as objectives identify microorganisms present in the hands and nasal cavity of food handlers that work in a kitchen of a University Hospital. It was collected samples of 24 individuals usingswab Staphylococcus aureus , with 41.67% harbouring this bacterium in nasal cavity, 41.67% in the palm of the hand, and 16.67% in both hands and nasal cavity. Another microorganism detected was Escherichia coli , present in 29.17% of handlers, being 28.57% in nasal cavity and 71.43% in the hands of manipulators. In 12.5% of food handlers were found both S. aureus and E. coli in their hands. The demonstrations of S. aureus e E. coli in the food handlers´ isolated samples indicate food contamination; arise from contaminated hands and nasal cavity of them. The prevention for this contamination can be applied adopting an orientation procedure and available of products that facilitate the right sanitation of the hands in the work environment. s soaked in saline solution. The music industry has undergone many changes in the last few decades, notably since vinyl, cassettes and compact discs faded away as streaming platforms took the world by storm. This Digital evolution has made huge volumes of data about music consumption available. Based on such data, we perform cross-era comparisons between Physical and Digital media within the music market in Brazil. First, we build artists' success time series to detect and characterize hot streak periods, defined as high-impact bursts that occur in sequence, in both eras. Then, we identify groups of artists with distinct success levels by applying a cluster analysis based on hot streaks' features. We find the same clusters for both Physical and Digital eras: Spike Hit Artists, Big Hit Artists, and Top Hit Artists. Our results reveal significant changes in the music industry dynamics over the years by identifying the core of each era

Randomized in situ evaluation of surface polishing protocols on the caries-protective effect of resin Infiltrant.
The aim of this placebo-controlled randomized in situ study was to evaluate the effect of different surface polishing protocols on enamel roughness, bacterial adhesion and caries-protective effect of a resin infiltrant. Seventy-five bovine enamel samples having artificial caries lesions were treated with a resinous infiltrant and afterwards randomly dividided into five polishing protocols: aluminum oxide flexible disks (Al2O3-Disks), silicon carbide tips (SIC-Tips), silicon carbide brush (SIC-Brush), silicon carbide polyester strips (SIC-Strips) or no polishing [negative control (NC)]. Average surface roughness (Ra) was assessed by profilometry. Samples were mounted in palatal appliances under a mesh for biofilm accumulation. Fifteen volunteers wore the intraoral appliances (14-days) and cariogenic challenge was triggered by sucrose solutions. Biofilm formed was collected for microbiological analysis of caries-related bacteria (Streptococcus mutans, Lactobacillus acidophilus) and demineralization was assessed by cross-sectional microhardness. Mean Knoop hardness numbers (Kg/mm2) were plotted over lesion depth (µm) and area under the lesion curve was subtracted from sound enamel to determine demineralization (ΔS, Kg/mm2xµm). Data were analyzed by ANOVA and post-hoc comparisons (α = 0.05). NC resulted in significantly higher Ra means than Al2O3-Disks and SIC-Strips. Bacterial counts were not significantly different between the groups (p > 0.05). Regards ΔS means, however none of the groups were significantly different to NC (6983.3 kg/mm2xµm /CI 4246.1-9720.5, p > 0.05). Conclusions: Polishing protocols (Al2O3-Disks, SIC-Strips) significantly decreseased roughness of infiltrated-enamel, however none of the polishing protocols could signicantly decrease bacterial counts nor resulted in significant less demineralization
Magnetoliposomes containing multicore nanoparticles and a new antitumor thienopyridine compound with potential application in chemo/thermotherapy
Multicore magnetic nanoparticles of manganese ferrite were prepared using carboxymethyl dextran as an agglutinating compound or by an innovative method using melamine as a cross-coupling agent. The nanoparticles prepared using melamine exhibited a flower-shape structure, a saturation magnetization of 6.16 emu/g and good capabilities for magnetic hyperthermia, with a specific absorption rate (SAR) of 0.14 W/g. Magnetoliposome-like structures containing the multicore nanoparticles were prepared, and their bilayer structure was confirmed by FRET (Förster Resonance Energy Transfer) assays. The nanosystems exhibited sizes in the range of 250–400 nm and a low polydispersity index. A new antitumor thienopyridine derivative, 7-[4-(pyridin-2-yl)-1H-1,2,3-triazol-1-yl]thieno[3,2-b]pyridine, active against HeLa (cervical carcinoma), MCF-7 (breast adenocarcinoma), NCI-H460 (non-small-cell lung carcino-ma) and HepG2 (hepatocellular carcinoma) cell lines, was loaded in these nanocarriers, obtaining a high encapsulation efficiency of 98% ± 2.6%. Development of thermo-and pH-sensitive liposomal magnetic carriers for new potential antitumor thienopyridine derivatives
The development of stimuli-sensitive drug delivery systems is a very attractive area of current research in cancer therapy. The deep knowledge on the microenvironment of tumors has supported the progress of nanosystems’ ability for controlled and local fusion as well as drug release. Temperature and pH are two of the most promising triggers in the development of sensitive formulations to improve the efficacy of anticancer agents. Herein, magnetic liposomes with fusogenic sensitivity to pH and temperature were developed aiming at dual cancer therapy (by chemotherapy and magnetic hyperthermia). Magnetic nanoparticles of mixed calcium/manganese ferrite were synthesized by co-precipitation with citrate and by sol–gel method, and characterized by X-ray diffraction (XRD), scanning electron microscopy in transmission mode (STEM), and superconducting quantum interference device (SQUID). The citrate-stabilized nanoparticles showed a small-sized population (around 8 nm, determined by XRD) and suitable magnetic properties, with a low coercivity and high saturation magnetization (~54 emu/g). The nanoparticles were incorporated into liposomes of dipalmitoylphosphatidylcholine/cholesteryl hemisuccinate (DPPC:CHEMS) and of the same components with a PEGylated lipid (DPPC:CHEMS:DSPE-PEG), resulting in magnetoliposomes with sizes around 100 nm. Dynamic light scattering (DLS) and electrophoretic light scattering (ELS) measurements were performed to investigate the pH-sensitivity of the magnetoliposomes’ fusogenic ability. Habitat determinants of golden‐headed lion tamarin (Leontopithecus chrysomelas) occupancy of cacao agroforests: Gloomy conservation prospects for management intensification
Organismal distributions in human‐modified landscapes largely depend on the capacity of any given species to adapt to changes in habitat structure and quality. The golden‐headed lion tamarin (GHLT; Leontopithecus chrysomelas) is an Endangered primate from the Brazilian Atlantic Forest whose remaining populations occupy heterogeneous landscapes consisting primarily of shade cacao (Theobroma cacao) agroforestry, locally known as cabrucas. This cash crop can coexist with high densities of native tree species and holds a significant proportion of the native fauna, but its widely extolled wildlife‐friendly status is increasingly threatened by management intensification. Although this potentially threatens to reduce the distribution of GHLTs, the main determinants of tamarin's occupancy of cabrucas remain unknown, thereby limiting our ability to design and implement appropriate conservation practices. We surveyed 16 cabruca patches in southern Bahia, Brazil, and used occupancy modeling to identify the best predictors of GHLT patch occupancy. Key explanatory variables included vegetation structure, critical resources, landscape context, human disturbance, and predation pressure. We found a negative relationship between GHLT occupancy and the prevalence of jackfruit trees (Artocarpus heterophylus), which is likely associated with the low representation of other key food species for GHLTs. Conversely, cabrucas retaining large‐diameter canopy trees have a higher probability of GHLT occupancy, likely because these trees provide preferred sleeping sites. Thus, key large tree resources (food and shelter) are currently the main drivers of GHLT occupancy within cabruca agroecosystems. Since both factors can be directly affected by crop management practices, intensification of cabrucas may induce significant habitat impacts on GHLT populations over much of their remaining range‐wide distribution
