    Hipomielinogênese associada à intoxicação transplacentária por Sida carpinifolia (Malvaceae) em fetos e neonatos bovinos

    Sida carpinifolia is a plant responsible for poisoning several species of animals. This paper describes Hypomyelinogenesis in fetuses and neonates of cattle that consumed S. carpinifolia. Neonates manifested ataxia and muscle tremors. Two bovine newborns and four fetuses were necropsied and showed no significant gross changes. Histopathologic findings included vacuolation of pancreatic acinar cells, thyroid follicular cells, hepatocytes, cells of renal tubules and neurons of the fetus and the white matter of the telencephalic frontal lobe of the neonates and also revealed axonal spheroids in the brain of the fetuses and neonates. The lectin-histochemical evaluation shoved staining for the lectins Con-A, WGA and s-WGA. The Luxol Fast Blue staining revealed a marked decrease of myelin in the brain of all the fetuses and a moderate decrease in the neonates. Histologic and lectin-histochemic findings indicate that the consumption of S. carpinifolia by pregnant bovine females can cause hypomyelinogenesis in fetuses and neonates.Sida carpinifolia é uma planta responsável por intoxicar várias espécies animais. Este artigo descreve hipomielinogênese em fetos e neonatos de bovinos que consumiram S. carpinifolia. Os neonatos manifestaram ataxia e tremores musculares. Dois neonatos e quatro fetos bovinos foram necropsiados e não havia alterações macroscópicas significativas. Os achados histopatológicos incluíram vacuolização de células acinares do pâncreas, células foliculares da tireoide, hepatócitos, células renais tubulares e neurônios nos fetos. Nos neonatos havia vacuolização na substância branca do lobo frontal telencefálico, além de esferoides axonais no encéfalo dos fetos e dos recém-nascidos. A avaliação lectino-histoquímica demonstrou marcação para as lectinas Con-A, WGA e s-WGA. A coloração de Luxol Fast Blue revelou diminuição acentuada da mielina no telencéfalo de todos os fetos e diminuição moderada nos neonatos. Os achados histológicos e lectina-histoquímicos indicam que o consumo de S. carpinifolia por fêmeas bovinas gestantes pode causar hipomielinogênese em fetos e neonatos

    High yield expression of leptospirosis vaccine candidates LigA and LipL32 in the methylotrophic yeast Pichia pastoris

    <p>Abstract</p> <p>Background</p> <p>Leptospirosis, a zoonosis caused by <it>Leptospira </it>spp., is recognized as an emergent infectious disease. Due to the lack of adequate diagnostic tools, vaccines are an attractive intervention strategy. Recombinant proteins produced in <it>Escherichia coli </it>have demonstrated promising results, albeit with variable efficacy. <it>Pichia pastoris </it>is an alternative host with several advantages for the production of recombinant proteins.</p> <p>Results</p> <p>The vaccine candidates LigANI and LipL32 were cloned and expressed in <it>P. pastoris </it>as secreted proteins. Large-scale expression resulted in a yield of 276 mg/L for LigANI and 285 mg/L for LipL32. The recombinant proteins were glycosylated and were recognized by antibodies present in the sera of patients with severe leptospirosis.</p> <p>Conclusions</p> <p>The expression of LigANI and LipL32 in <it>P. pastoris </it>resulted in a significant increase in yield compared to expression in <it>E. coli</it>. In addition, the proteins were secreted, allowing for easy purification, and retained the antigenic characteristics of the native proteins, demonstrating their potential application as subunit vaccine candidates.</p

    Evaluation of the Potential of Brazilian Propolis against UV-Induced Oxidative Stress

    This study investigated the potential use of topically and orally administered propolis extracts to prevent UV irradiation-induced oxidative stress in skin. The results illustrated that green propolis extract (GPE) contained greater amounts of polyphenols, coumaric acid, drupanin, baccharin and artepillin C than did brown propolis extract (BPE). GPE showed higher antioxidant activity than BPE when the IC50 (concentration that caused 50% inhibition) values were compared. Interesting, the oral treatment of hairless mice demonstrated a recovery of 30.0% for GPE and 22.8% for BPE with respect to UV irradiation-induced GSH depletion. The topical pretreatment of animals with both propolis extract solutions recovered around 14.0% of the depleted GSH. However, the employed treatments did not inhibit the increase of cutaneous proteinase secretion/activity caused by irradiation. These findings indicate that despite differences in composition and antioxidant properties, GPE and BPE both successfully prevent UV-induced GSH depletion in vivo and are both promising antioxidant systems against oxidative stress in skin. Based on these findings, complementary studies should be performed to enhance our understanding of the protective effects of propolis extracts in skin

    Double shunt technique for hybrid palliation of hypoplastic left heart syndrome: a case report

    We report a technique to palliate hypoplastic left heart syndrome, with no PDA stenting, but with double polytetrafluoroethylene shunt from pulmonary artery to ascending and descending aorta by combined thoracotomies. A 30-day-old female was operated with this technique. Five months after first operation, the child was submitted to Norwood/Glenn operation. Good hemodinamic recovery and initial clinical evolution was observed. The child was extubated in 8th post operatory day and reentubated in the next day due to pulmonary infection. Despite antibiotic treatment, the child died after systemic infectious complications

    Depression as a determinant of frailty in late life

    Objectives Accumulating evidence shows depression as a risk factor for frailty, but studies are mainly population-based and widely differ in their assessment of either depression or frailty. We investigated the association between depression and frailty among geriatric outpatients using different assessment instruments for both conditions. Method Among 315 geriatric outpatients (mean age 72.1 years, 68.3% female sex) participating the MiMiCS-FRAIL cohort study, major and subthreshold depression were measured with psychiatric diagnostic interview according to DSM-5 criteria (SCID-5) as well as with instruments to screen and measure severity of depressive symptoms (GDS-15 and PHQ-9). Frailty was assessed according to a screening instrument (FRAIL-BR) and a multidimensional Frailty Index (FI-36 items). Multiple logistic and linear regression were performed to assess the association between depression (independent variable) and frailty (dependent variable) adjusted for confounders. Results Frailty prevalence in patients with no, subthreshold or major depressive disorder increases from either 14.5%, 46.5% to 65.1% when using the FRAIL-BR questionnaire, and from 10.2%, 20.9%, to 30.2% when using the FI-36 index. These association remain nearly the same when adjusted for covariates. Both the FRAIL-BR and the FI-36 were strongly associated with major depressive disorder, subthreshold depression, and depressive symptoms by PHQ-9 and GDS-15. Conclusion Late life depression and frailty are associated in a dose-dependent manner, irrespective of the used definitions. Nonetheless, to avoid residual confounding, future research on underlying biological mechanisms should preferably be based on formal psychiatric diagnoses and objectively assessment frailty status

    Radiographic interpretation using high-resolution Cbct to diagnose degenerative temporomandibular joint disease

    The objective of this study was to use high-resolution cone-beam computed images (hr-CBCT) to diagnose degenerative joint disease in asymptomatic and symptomatic subjects using the Diagnostic Criteria for Temporomandibular Disorders DC/TMD imaging criteria. This observational study comprised of 92 subjects age-sex matched and divided into two groups: clinical degenerative joint disease (c-DJD, n = 46) and asymptomatic control group (n = 46). Clinical assessment of the DJD and high-resolution CBCT images (isotropic voxel size of 0.08mm) of the temporomandibular joints were performed for each participant. An American Board of Oral and Maxillofacial Radiology certified radiologist and a maxillofacial radiologist used the DC/TMD imaging criteria to evaluate the radiographic findings, followed by a consensus of the radiographic evaluation. The two radiologists presented a high agreement (Cohen’s Kappa ranging from 0.80 to 0.87) for all radiographic findings (osteophyte, erosion, cysts, flattening, and sclerosis). Five patients from the c- DJD group did not present radiographic findings, being then classified as arthralgia. In the asymptomatic control group, 82.6% of the patients presented radiographic findings determinant of DJD and were then classified as osteoarthrosis or overdiagnosis. In conclusion, our results showed a high number of radiographic findings in the asymptomatic control group, and for this reason, we suggest that there is a need for additional imaging criteria to classify DJD properly in hr-CBCT images

O projeto foi realizado no Instituto Federal Catarinense Campus Araquari, na unidade de ensino e aprendizagem (UEA) bovino cultura. A meta foi coletar dados e planilhar Posteriormente foi realizar comparações com outros períodos sobre a produtividade do setor, fazendo com que o manejo seja mais eficaz e produtivo. Com tudo observamos que seria necessário direcionar alguns ani maispara descarte, diminuindo gastos dentro da propriedade. O projeto nos mostrou também algumas orientações incorretas feitas dentro do instituto, que proporcionou dados zootécnicos favoráveis para a tomada de decisões
