Matrix architecture plays a pivotal role in 3D osteoblast migration: The effect of interstitial fluid flow
Osteoblast migration is a crucial process in bone regeneration, which is strongly regulated by interstitial fluid flow. However, the exact role that such flow exerts on osteoblast migration is still unclear. To deepen the understanding of this phenomenon, we cultured human osteoblasts on 3D microfluidic devices under different fluid flow regimes. Our results show that a slow fluid flow rate by itself is not able to alter the 3D migratory patterns of osteoblasts in collagen-based gels but that at higher fluid flow rates (increased flow velocity) may indirectly influence cell movement by altering the collagen microstructure. In fact, we observed that high fluid flow rates (1 µl/min) are able to alter the collagen matrix architecture and to indirectly modulate the migration pattern. However, when these collagen scaffolds were crosslinked with a chemical crosslinker, specifically, transglutaminase II, we did not find significant alterations in the scaffold architecture or in osteoblast movement. Therefore, our data suggest that high interstitial fluid flow rates can regulate osteoblast migration by means of modifying the orientation of collagen fibers. Together, these results highlight the crucial role of the matrix architecture in 3D osteoblast migration. In addition, we show that interstitial fluid flow in conjunction with the matrix architecture regulates the osteoblast morphology in 3D
Mahogunin Ring Finger 1 Is Required for Genomic Stability and Modulates the Malignant Phenotype of Melanoma Cells.
The mouse mahoganoid mutation abrogating Mahogunin Ring Finger-1 (MGRN1) E3 ubiquitin ligase expression causes hyperpigmentation, congenital heart defects and neurodegeneration. To study the pathophysiology of MGRN1 loss, we compared Mgrn1-knockout melanocytes with genetically matched controls and melan-md1 (mahoganoid) melanocytes. MGRN1 knockout induced a more differentiated and adherent phenotype, decreased motility, increased the percentage of cells in the S phase of the cell cycle and promoted genomic instability, as shown by stronger γH2AX labelling, increased burden of DNA breaks and higher abundance of aneuploid cells. Lack of MGRN1 expression decreased the ability of melanocytes to cope with DNA breaks generated by oxidizing agents or hydroxyurea-induced replicative stress, suggesting a contribution of genomic instability to the mahoganoid phenotype. MGRN1 knockout in B16-F10 melanoma cells also augmented pigmentation, increased cell adhesion to collagen, impaired 2D and 3D motility and caused genomic instability. Tumors formed by Mgrn1-KO B16-F10 cells had lower mitotic indices, fewer Ki67-positive cells and showed a trend towards smaller size. In short-term lung colonization assays Mgrn1-KO cells showed impaired colonization potential. Moreover, lower expression of MGRN1 is significantly associated with better survival of human melanoma patients. Therefore, MGRN1 might be an important phenotypic determinant of melanoma cells
Procedimiento para la selección de la estrategia de regulación más adecuada en estaciones de bombeo
[ES] La selección de la mejor estrategia de operación en una estación de bombeo que impulsa
caudales directamente a red es un problema complejo de
problema se puede plantear como la búsqueda de la mejor estrategia en una instalación ya
existente (la mejora de un sistema que está en funcionamiento con posibilidades de instalar,
por ejemplo, variadores de frecuencia en los equipos de la estación de bombeo y diseñar la
estrategia de regulación más adecuada) o bien, directamente, como un problema de
selección tanto de los equipos de bombeo como del sistema de regulación para una estación
de bombeo a diseñar.
Todo ello implica que en cada caso es necesario manejar un campo de soluciones
excesivamente amplio, no conociendo la existencia de herramientas que, filtrando y
acotando las soluciones posibles, aborden dicho problema teniendo en cuenta las variables
más significativas: modelo de las bombas a instalar con sus curvas características,
bombas de velocidad fija (BVF), número de bombas de velocidad variable (BVV),
rendimiento mínimo aceptable, velocidad mínima y máxima de giro de las bombas, tarifas
eléctricas, caudales a impulsar, etc.
Debido a esto, el técnico se ve obligado a
técnica que acote el número de soluciones. Al actuar de esta forma es posible que en
muchos casos se desestimen soluciones atractivas desde el punto de vista energético y

Influence of the ectopic location on the antigen expression and functional characteristics of endometrioma stromal cells
Research question:
Are the alterations observed in the endometriotic cells, such as progesterone resistance, already present in the eutopic endometrium or acquired in the ectopic location?
Design:
The response to decidualization with progesterone and cyclic AMP for up to 28 days was compared in different endometrial stromal cell (EnSC) lines established from samples of endometriomas (eEnSC), eutopic endometrium from women with endometriosis (eBEnSC), endometrial tissue from healthy women (BEnSC) and menstrual blood from healthy donors (mEnSC).
Results:
Usual features of decidualized cells, such as changes in cell morphology and expression of prolactin, were similarly observed in the three types of eutopic EnSC studied, but not in the ectopic cells upon decidualization. Among the phenotypic markers analysed, CD105 was down-regulated under decidualization in all cell types (mEnSC, P = 0.005; BEnSC, P = 0.029; eBEnSC, P = 0.022) except eEnSC. mEnSC and BEnSC underwent apoptosis during decidualization, whereas eBEnSC and eEnSC were resistant to the induction of cell death. Lastly, migration studies revealed that mEnSC secreted undetermined factors during decidualization that inhibited cell motility, whereas eEnSC showed a significantly lower ability to produce those migration-regulating factors (P < 0.0001, P < 0.001 and P = 0.0013 for the migration of mEnSC at 24, 48 and 72 h, respectively; P < 0.0001 for the migration of eEnSC at all times studied).
Conclusions:

Variabilidad en cepas de Xylella fastidiosa procedentes de olivo (Olea europaea L.) de Argentina [resumen]
Xylella fastidiosa, es considerada una bacteria polífaga, capaz de vivir y multiplicarse en un amplio rango de hospedantes, más de 500 especies de plantas, abarcando tanto cultivos de interés agrícola como especies ornamentales y malezas. En el año 2013, se dieron las primeras detecciones de la bacteria en fincas tradicionales de olivo de La Rioja, observando un marcado declinamiento en las plantas, hasta causar la muerte de ejemplares de más de 50 años de edad. Posteriormente, fue reportada en plantaciones de las provincias de Córdoba y Catamarca. X. fastidiosa es una bacteria genéticamente diversa que actualmente comprende las siguientes subespecies: fastidiosa, pauca, multiplex, sandyi, morus y tashke. Estilos de aprendizaje y evaluación formativa: Estudio con universitarios de Educación Física chilenos
Assessment contributes to students learning. Formative assessment is a useful strategy of the initial teacher training. Perceptions about the assessment of Chilean university Physical Education students (n = 95), depending on their learning style, gender and academic course were analyzed. The Learning Styles Questionnaire and the Spanish version of the Assessment Experience Questionnaire were used. The results showed negative perceptions about the assessment process. Significant differences were observed in the perceptions about the use of feedback depending on the learning style of the students (F = 1.062, p =. 004). Significant differences were observed in the perception about the amount of effort (F = 5.888, p =. 000), the suitability of the evaluation (F = 3.282, p =. 015) and the shallow approach to the learning (F = 5.472, p =. 001), depending on the academic course. Gender was not a determinant of perceptions. The findings suggest the use of traditional procedures of assessment (summative assessment) with the students. On the other hand, findings show the influence of learning stylesand the academic course on student perceptionsand learning styles and feedback. This information contributes to a deeper understanding of the learning mode of the students according to their characteristics and context.La evaluación contribuye a la formación del aprendizaje, facilitando el avance del estudiante. La evaluación formativa se presenta como una estrategia eficaz dentro de la formación inicial docente. Se analizaron las percepciones sobre la evaluación de universitarios de Educación Física chilenos (n = 95), en función de su estilo de aprendizaje, género y curso académico. Se utilizaron el Cuestionario de Estilos de Aprendizaje y la versión en castellano del Assessment Experience Questionnaire. Los resultados mostraron percepciones negativas sobre el proceso evaluativo. Se observaron diferencias significativas en las percepciones sobre el uso del feedback en función del estilo de aprendizaje (F= 1.062, p = .004). Se observaron diferencias significativas en la percepción de la cantidad de esfuerzo (F= 5.888, p = .000), la adecuación de la evaluación (F= 3.282, p = .015)y la aproximación superficial al aprendizaje (F= 5.472, p = .001), en función del curso académico. El género no fue determinante de las percepciones. Los hallazgos sugieren la utilización de procedimientos de evaluación tradicional con los encuestados. Se encontraron relaciones entre el estilo de enseñanza y el feedback, y entre el curso académico y las percepciones de evaluación. Esta información contribuye a un entendimiento más profundo del modo de aprender de los estudiantes en función de sus características y contexto

Mahogunin Ring Finger 1 regulates pigmentation by controlling the pH of melanosomes in melanocytes and melanoma cells.
Mahogunin Ring Finger 1 (MGRN1) is an E3-ubiquitin ligase absent in dark-furred mahoganoid mice. We investigated the mechanisms of hyperpigmentation in Mgrn1-null melan-md1 melanocytes, Mgrn1-KO cells obtained by CRISPR-Cas9-mediated knockdown of Mgrn1 in melan-a6 melanocytes, and melan-a6 cells depleted of MGRN1 by siRNA treatment. Mgrn1-deficient melanocytes showed higher melanin content associated with increased melanosome abundance and higher fraction of melanosomes in highly melanized maturation stages III-IV. Expression, post-translational processing and enzymatic activity of the rate-limiting melanogenic enzyme tyrosinase measured in cell-free extracts were comparable in control and MGRN1-depleted cells. However, tyrosinase activity measured in situ in live cells and expression of genes associated with regulation of pH increased upon MGRN1 repression. Using pH-sensitive fluorescent probes, we found that downregulation of MGRN1 expression in melanocytes and melanoma cells increased the pH of acidic organelles, including melanosomes, strongly suggesting a previously unknown role of MGRN1 in the regulation of melanosomal pH. Among the pH regulatory genes upregulated by Mgrn1 knockdown, we identified those encoding several subunits of the vacuolar adenosine triphosphatase V-ATPase (mostly Atp6v0d2) and a calcium channel of the transient receptor potential channel family, Mucolipin 3 (Mcoln3). Manipulation of expression of the Mcoln3 gene showed that overexpression of Mcoln3 played a significant role in neutralization of the pH of acidic organelles and activation of tyrosinase in MGRN1-depleted cells. Therefore, lack of MGRN1 led to cell-autonomous stimulation of pigment production in melanocytes mostly by increasing tyrosinase specific activity through neutralization of the melanosomal pH in a MCOLN3-dependent manner
