    KINEMATIC VARIABLES AND BLOOD ACID-BASE STATUS IN THE ANALYSIS OF COLLEGIATE SWIMMERS’ ANAEROBIC CAPACITY

    Short duration repeated maximal efforts are often used in swimming training to improve lactate tolerance, which gives swimmers the ability to maintain a high work rate for a longer period of time. The aim of the study was to examine the kinematics of swimming and its relation to the changes in blood acid-base status and potassium level. Seven collegiate swimmers, with at least 6 years of training experience, volunteered to participate in the study. The test consisted of 8 x 25 m front crawl performed with maximum effort. The rest period between repetitions was set to five seconds. Blood samples were taken from the fingertip at rest, after warm-up and in the 3rd minute after completion of the test. The swimming was recorded with a video recorder, for later analysis of time, velocity and technique (stroke index). Based on the swimming velocity results, the obtained curve can be divided into rapid decrease of velocity and relatively stable velocities. The breaking point of repetition in swimming velocity was assumed as the swimming velocity threshold and it was highly correlated with the decrease of the blood acid-base status (pH r=0.82, BE r=0.87, HCO3- r=0.76; p<0.05 in all cases). There was no correlation between stroke index or fatigue index and blood acid-base status. Analysis of the swimming speed in the 8 x 25 m test seems to be helpful in evaluation of lactate tolerance (anaerobic capacity) in collegiate swimmers

    Characterizing the Pore Structure of Carbonated Natural Wollastonite

    This paper focuses on examining the pore structure of a cementitious paste made with a calcium silicate (wollastonite) that reacts with carbon dioxide and water to form a hardened solid. The pore structure of the hardened solid has been characterized using vapor sorption and desorption, low-temperature differential scanning calorimetry (LT-DSC), and scanning electron microscopy (SEM). The total porosity was also measured using mass measurement in oven-dry and vacuum-saturated conditions. Evidence exists that support the hypothesis that the solid has two main pore sizes: large macropores (\u3e10 nm) appear to form between the initial calcium silicate particles and small micropores (\u3c10 \u3enm) were found in the reacted silica gel. The bimodal nature of the pore structure was evident from the desorption and LT-DSC responses. The extent of reaction was also investigated and was found to be the result of the function of the raw material particle size: only particles with radius \u3c10 \u3eμm were found to have entirely reacted even in highly reacted systems. Moreover, the degree of reaction influenced the uniformity of reaction across the sample. Only the highly reacted system showed a uniform microstructure with continuous reaction products path and low porosity

    Changes on fecal microbiota in rats exposed to permethrin during postnatal development

    Alteration of the gut microbiota through diet and environmental contaminants may disturb the mammalian digestive system, leading to various diseases. Because most exposure to environmentally pyrethroid pesticides such as permethrin (PERM) occurs through the diet, the commensal gut microbiota is likely to be exposed to PERM. The study aimed at evaluating the effect of low-dose exposure to PERM in early life on the composition of fecal microbiota in rats. Over a 4-month follow-up period, fecal microbiota and short-chain fatty acids were measured in order to identify possible differences between PERM-treated rats and controls. Further in vitro antimicrobial experiments were conducted to establish the antibacterial activity of PERM against different strains to obtain Minimal Inhibitory Concentrations. The main finding focused on the reduced abundance of Bacteroides-Prevotella-Porphyromonas species, increased Enterobacteriaceae and Lactobacillus in PERM-treated rats compared to controls. Changes of acetic and propionic acid levels were registered in PERM-treated group. From in vitro studies, PERM showed higher antibacterial activity against beneficial bacteria such as Bifidobacterium and Lactobacillus paracasei, while to inhibit potential pathogens as Staphylococcus aureus and Escherichia coli PERM concentration needed to be increased. In summary, exposure to PERM could affect the fecal microbiota and could be a crucial factor contributing to the development of diseases

    Plasma Trimethylamine-N-oxide following Cessation of L-carnitine Supplementation in Healthy Aged Women.

    L-carnitine supplementation elevates plasma trimethylamine-N-oxide (TMAO), which may participate in atherosclerosis development by affecting cholesterol metabolism. The aim of the current study was to determine the effect of increased plasma TMAO on biochemical markers in the blood following cessation of L-carnitine supplementation. The follow-up measurements were performed on subjects who completed 24 weeks of L-carnitine or placebo supplementation protocol. Blood samples were taken after finishing the supplementation and then 4 and 12 months following the supplementation withdrawal. Four months after cessation of L-carnitine supplementation, plasma TMAO concentration reached a normal level which was stable for the following eight months. During this period, no modifications in serum lipid profile and circulating leukocyte count were noted. TMAO implications in health and disease is widely discussed. The results of this study demonstrate no adverse effects of elevated plasma TMAO, induced by L-carnitine, on the measured parameters at 4 and 12 months after withdrawal of supplementation

    Objective The aim of our study was to investigate&nbsp; the adaptive effect of six sessions of high intensity interval training (HIIT) on changes in the amount and activity of mitochondrial enzymes. Methods Twenty seven students (age 21.2±0.9) were assigned to HIIT (n=10) and control (CON, n=17) group and performed six training sessions for 14 days:&nbsp; 6 × 90 s intervals at 80% maximal aerobic power (MAP) output separated by 180 s rest. Pre and post interventions anthropometric measurements, maximal activity of citrate synthase (CS) and 3-HydroxyacylCoA (HADH) was determined in muscle samples. The effect of HIIT on proteins involved in oxidative phosphorylation (OXPHOS) in the skeletal muscle was used via proteomic analysis’s. We took into consideration 89 identified subunits from the mitochondrial respiratory chain complexes and the ATP synthase complex. For these proteomic tests a muscle biopsy samples from the three representative participants HIIT and three CON before and after training were collected.&nbsp; Results Training induced the moderate and large effects in maximal enzymes activities CS and HADH. The HIIT caused the increase of level proteins involved in oxidative phosphorylation. Conclusions HIIT can be an optimal strategy for the prevention of certain civilization diseases or for the rehabilitation of diseases, especially cardiovascular disease

    Risk factors for Epstein–Barr virus reactivation after renal transplantation: Results of a large, multi‐centre study

    Epstein-Barr virus (EBV) reactivation is a very common and potentially lethal complication of renal transplantation. However, its risk factors and effects on transplant outcome are not well known. Here, we have analysed a large, multi-centre cohort (N = 512) in which 18.4% of the patients experienced EBV reactivation during the first post-transplant year. The patients were characterized pre-transplant and two weeks post-transplant by a multi-level biomarker panel. EBV reactivation was episodic for most patients, only 12 patients showed prolonged viraemia for over four months. Pre-transplant EBV shedding and male sex were associated with significantly increased incidence of post-transplant EBV reactivation. Importantly, we also identified a significant association of post-transplant EBV with acute rejection and with decreased haemoglobin levels. No further severe complications associated with EBV, either episodic or chronic, could be detected. Our data suggest that despite relatively frequent EBV reactivation, it had no association with serious complications during the first post-transplantation year. EBV shedding prior to transplantation could be employed as biomarkers for personalized immunosuppressive therapy. In summary, our results support the employed immunosuppressive regimes as relatively safe with regard to EBV. However, long-term studies are paramount to support these conclusions

    DNA methylation clock DNAmFitAge shows regular exercise is associated with slower aging and systemic adaptation

    DNAmPhenoAge, DNAmGrimAge, and the newly developed DNAmFitAge are DNA methylation (DNAm)-based biomarkers that reflect the individual aging process. Here, we examine the relationship between physical fitness and DNAm-based biomarkers in adults aged 33–88 with a wide range of physical fitness (including athletes with long-term training history). Higher levels of VO 2 max ( ρ = 0.2, p = 6.4E − 4, r = 0.19, p = 1.2E − 3), Jumpmax ( p = 0.11, p = 5.5E − 2, r = 0.13, p = 2.8E − 2), Gripmax ( ρ = 0.17, p = 3.5E − 3, r = 0.16, p = 5.6E − 3), and HDL levels ( ρ = 0.18, p = 1.95E − 3, r = 0.19, p = 1.1E − 3) are associated with better verbal short-term memory. In addition, verbal short-term memory is associated with decelerated aging assessed with the new DNAm biomarker FitAgeAcceleration ( ρ : − 0.18, p = 0.0017). DNAmFitAge can distinguish high-fitness individuals from low/medium-fitness individuals better than existing DNAm biomarkers and estimates a younger biological age in the high-fit males and females (1.5 and 2.0 years younger, respectively). Our research shows that regular physical exercise contributes to observable physiological and methylation differences which are beneficial to the aging process. DNAmFitAge has now emerged as a new biological marker of quality of life
