    Integrative analysis of the human cis-antisense gene pairs, miRNAs and their transcription regulation patterns

    Cis-antisense gene pairs (CASGPs) can transcribe mRNAs from an opposite strand of a given locus. To classify and understand diverse CASGP phenomena in the human we compiled a genome-wide catalog of CASGPs and integrated these sequences with microarray, SAGE and miRNA data. Using the concept of overlapping regions and clustering of SA transcripts by chromosome coordinates, we identified up to 9000 overlapping antisense loci. Four thousand three hundred and seventy-four of these CASGPs form 1759 complex gene architectures. We found that ∼35% (6347/18160) of RefSeq genes are overlapped with the antisense transcripts. About 30% of Affymetrix U133 microarray initial sequences map transcripts of ∼35% CASGPs and reveal mostly concordant expression in CASGPs. We found strong significant overrepresentation of human miRNA genes in loci of CASGPs. We developed a data-driven model of cross-talk between co-expressed CASGPs and DICER1-mediated miRNA pathway in normal spermatogenesis and in severe teratozoospermia. Specifically, we revealed complex SA structural–functional gene module composing the protein-coding genes, WDR6, DALRD3, NDUFAF3 and ncRNA precursors, mir-425 and mir-191, which could provide downregulation of ncRNA pathway via direct targeting DICER1 and basonuclin 2 transcripts by mir-425 and mir-191 in normal spermatogenesis, but this mechanism is switched off in severe teratozoospermia.     Computational Study of the Dynamics of the Taylor Bubble

    We perform high-resolution numerical simulations of three-dimensional dynamics of an elongated bubble in a microchannel at moderate Reynolds numbers up to 1800. For this purpose, we use the coupled Brinkman penalization and volume of fluid methods implemented in the open-source framework Basilisk. The new results are validated with available experimental data and compared with previous numerical and theoretical predictions. We extend existing results to regimes with significant inertia, which are characterized by intense deformations of the bubble, including cases with azimuthal symmetry breaking. Various dynamical features are analyzed in terms of their spatiotemporal characteristics, such as frequencies and wavelengths of the bubble surface undulations and vortical structures in the flow

    Modification of Mechanical Properties in Directed Energy Deposition by a Static Magnetic Field: Experimental and Theoretical Analysis

    The superimposed magnetic field affects the microstructure and mechanical properties of additively manufactured metal parts. In this work, the samples were fabricated from Inconel 718 superalloy by directed energy deposition under a 0.2 T static field. The magnetohydrodynamic 1D model is proposed for the estimation of a fluid flow inside a molten pool. According to the theoretical predictions, the fluid flow is slightly decreased by an applied field. The estimated thermoelectric magnetic convection in the mushy zone is shown to be negligible to change in subgrain size, but enough to reduce the hard-to-dissolve Nb-rich phase, thereby improving the average ultimate elongation from 23% to 27%. The obtained results confirm that an external static magnetic field can modify and enhance the mechanical properties of additively manufactured materials

    Hardening of Additive Manufactured 316L Stainless Steel by Using Bimodal Powder Containing Nanoscale Fraction

    The particle size distribution significantly affects the material properties of the additively manufactured parts. In this work, the influence of bimodal powder containing nano- and micro-scale particles on microstructure and materials properties is studied. Moreover, to study the effect of the protective atmosphere, the test samples were additively manufactured from 316L stainless steel powder in argon and nitrogen. The samples fabricated from the bimodal powder demonstrate a finer subgrain structure, regardless of protective atmospheres and an increase in the Vickers microhardness, which is in accordance with the Hall-Petch relation. The porosity analysis revealed the deterioration in the quality of as-built parts due to the poor powder flowability. The surface roughness of fabricated samples was the same regardless of the powder feedstock materials used and protective atmospheres. The results suggest that the improvement of mechanical properties is achieved by adding a nano-dispersed fraction, which dramatically increases the total surface area, thereby contributing to the nitrogen absorption by the material
