An analysis of consumer panel data
In terms of collecting comprehensive panel expenditure data, there are trade-offs to be made in terms of the demands imposed on respondents and the level of detail and spending coverage collected. Existing comprehensive spending data tends to be cross-sectional whilst panel studies include only limited expenditure questions that record spending only as broad aggregates. More recently, economists have begun to use spending information collected by market research companies that records very detailed spending down to the barcode level from a panel of households, usually recorded by in-home barcode scanners, which may provide considerable advantages over existing data more commonly used in social sciences. However, there has not been a comprehensive assessment of the strengths and weaknesses of this kind of data collection method and the potential implications of survey mode on the recorded data. This paper seeks to address this, by an in-depth examination of scanner data from one company, Taylor Nelson Sofres (TNS), on grocery purchases over a five-year period. We assess how far the ongoing demands of participation inherent in this kind of survey lead to 'fatigue' in respondents' recording of their spending and compare the demographic representativeness of the data to the well-established Expenditure and Food Survey (EFS), constructing weights for the TNS that account for observed demographic differences. We also look at demographic transitions, comparing the panel aspect of the TNS to the British Household Panel Study (BHPS). We examine in detail the expenditure data in the TNS and EFS surveys and discuss the implications of this method of data collection for survey attrition. Broadly, we suggest that problems of fatigue and attrition may not be so severe as may be expected, though there are some differences in expenditure levels (and to some extent patterns of spending) that cannot be attributed to demographic or time differences in the two surveys alone and may be suggestive of survey mode effects. Bryophytes and their distribution in the Blue Mountains region of New South Wales
The bryophytes (mosses, liverworts and hornworts) that occur in the Blue Mountains region of New South Wales (latitude 33˚–34˚ S, longitude 151˚–151˚40’ E) are listed and information is provided on their distribution in the region. Species lists are based on herbarium specimens and field collections. 348 bryophyte taxa have been recorded from 70 families, including 225 moss taxa (in 108 genera from 45 families), 120 liverwort taxa (in 51 genera from 24 families) and 3 hornwort taxa (in 3 genera from one family). The moss families with most taxa are the Pottiaceae (with 23 taxa in 13 genera), Bryaceae (with 15 taxa in 3 genera) and Fissidentaceae (with 13 taxa). The largest genera are Fissidens (13 taxa), Campylopus (9) and Macromitrium (8). The liverwort family with the most taxa is Lepidoziaceae, with 29 taxa in 10 genera. The largest liverwort genera are Frullania (11 taxa) and Riccardia (8). The species lists include collections from both bushland and urban areas. Natural features of the Blue Mountains, including topography, altitude, climate and vegetation appear to be important factors influencing the number of bryophyte species recorded from each location. The number of collections from particular locations has been considerably influenced by ease of access, particularly proximity to roads, public transport and railway stations. The species lists include many records from areas that were not accessible to the early collectors of the late 19th and early 20th centuries such as Wollemi National Park, Gardens of Stone National Park, Newnes Plateau and Kanangra-Boyd National Park
Non-bisphosphonate inhibitors of isoprenoid biosynthesis identified via computer-aided drug design.
The relaxed complex scheme, a virtual-screening methodology that accounts for protein receptor flexibility, was used to identify a low-micromolar, non-bisphosphonate inhibitor of farnesyl diphosphate synthase. Serendipitously, we also found that several predicted farnesyl diphosphate synthase inhibitors were low-micromolar inhibitors of undecaprenyl diphosphate synthase. These results are of interest because farnesyl diphosphate synthase inhibitors are being pursued as both anti-infective and anticancer agents, and undecaprenyl diphosphate synthase inhibitors are antibacterial drug leads
After the fire: an ecological, phenomenological exploration of resilience‐building following the Fuego volcanic eruption in Guatemala
Combining ecological resilience theory with a phenomenological epistemology, we explored experiential, social, and cultural factors mediating resilience-building with participants from a village destroyed by the 2018 Fuego volcanic eruption in Guatemala. The purpose of the study is to find out what strategies displaced families and communities employ for living through the aftermath of a volcano eruption and for building psychological resilience. We conducted semistructured interviews with nine survivors of the Fuego eruption, now relocated and coping with the loss of community and family members killed in the disaster. Interpretive phenomenological analysis was used to analyze transcripts. The analysis produced four main themes: (i) individual and collective challenges, (ii) social support and protection, (iii) faith and culturally endorsed belief, and (iv) looking to the future. As well as learning more about how a community faced challenges presented by a volcano eruption, the current study has a degree of transferability, with implications for understanding how other communities experience and cope with such events
Anti-obesity effect of the CB2 receptor agonist JWH-015 in diet-induced obese mice
The cannabinoid receptor 2 (CB2) is well known for its immune modulatory role. However, recent localisation of CB2 receptors in metabolically active tissue suggests that the CB2 receptor plays a significant role in energy homeostasis. This study was designed to investigate the impact of chronic CB2 receptor stimulation on food intake, body weight and mood. Lean male C57BL/6 mice were injected i.p. with the selective CB2 receptor agonist, JWH-015 (0.0, 1.0, 5.0 and 10.0 mg kg-1) to establish dose response parameters. Mice made obese following exposure to a diet consisting of 19.4 MJ/kg (4641 Kcal/kg) of energy (19.0% protein, 21.0% total fat, 4.7% crude fiber, and 4.7% AD fiber were given either vehicle or 10 mg/kg JWH-015. Impact on mood, food intake, body weight, plasma metabolites, expression of key metabolic proteins in the brown adipose tissue (BAT) and white adipose tissue (WAT), and markers of inflammation were measured. High dose (10 mg/kg) JWH-015 reduced food intake after 1, 2, 4, and 24 h in lean mice. When given to diet induced obese (DIO) mice, a 10 mg/kg dose of JWH-015 significantly reduced body weight compared to vehicle. This dose led to a shift in markers of lipid metabolism and inflammation in WAT consistent with lipolysis and improved immune response. Furthermore, JWH-015 (10 mg/kg) produced a transient reduction in food intake and significant reduction in fat mass and adipocyte cell size. Importantly, JWH-015 produced an anxiolytic response in the elevated plus maze while having no effect on immobility time in the forced swim test. It should be noted that though the 10 mg/kg dose produced positive effects on the obese state, the possibility that these effects are mediated via non-CB2 receptor mechanisms cannot be ruled out. These results demonstrate a role for CB2 receptors in modulating energy homeostasis and obesity associated metabolic pathologies in the absence of any adverse impact on mood
Image and word on the street: a reflexive, phased approach to combining participatory visual methods and qualitative interviews to explore resilience with street connected young people in Guatemala City
We present an argument for reflexively combining participatory visual methods with each other, alongside qualitative ethnographic interviewing, in exploring resilience with street-connected young people who attend two voluntary educational outreach projects in Guatemala City. We present primary research in the form of three case studies selected from a wider, mixed-methods study. The case studies adopt an approach that is rooted in phenomenology, incorporating participatory photography, participatory drawing, and image-elicited interviewing. We discuss the advantages of combining visual methods with qualitative interviewing in ways that respond to the developing phases of research and to the sensory and technological preferences of participants. We also outline the potential benefits of this approach for psychological research into resilience, place, and community, and for benefitting voluntary outreach organisations with whom we collaborated
Histone marks regulate the epithelial-to-mesenchymal transition via alternative splicing
International audienceHistone modifications impact final splicing decisions. However, there is little evidence of the driving role of these marks in inducing cell-specific splicing changes. Using CRISPR epigenome editing tools, we show in an epithelial-to-mesenchymal cell reprogramming system (epithelial-to-mesenchymal transition [EMT]) that a single change in H3K27ac or H3K27me3 levels right at the alternatively spliced exon is necessary and sufficient to induce a splicing change capable of recapitulating important aspects of EMT, such as cell motility and invasiveness. This histone-mark-dependent splicing effect is highly dynamic and mediated by direct recruitment of the splicing regulator PTB to its RNA binding sites. These results support a role for H3K27 marks in inducing a change in the cell's phenotype via regulation of alternative splicing. We propose the dynamic nature of chromatin as a rapid and reversible mechanism to coordinate the splicing response to cell-extrinsic cues, such as induction of EMT
Source-to-sink magnetic properties of NE Saharan dust in Eastern Mediterranean marine sediments: review and paleoenvironmental implications
We present a review of the magnetic properties of NE Saharan dust that was conducted, following a source-to-sink approach, to unravel the paleoclimatic significance of environmental magnetic records from Eastern Mediterranean marine sediments. Our synthesis indicates that pedogenic hematite, which formed during past wetter Green Sahara periods (GSPs), is the most common magnetic mineral in Eastern Mediterranean marine sediments as a result of its eolian transportation, along with smaller amounts of lithogenic hematite, from the NE Sahara. Coupled with the limited impact of reductive sedimentary diagenesis on hematite abundances in Eastern Mediterranean Sea sediments, this indicates that hematite concentrations provide reliable quantitative estimates of NE Saharan dust supply. Our results indicate that variations in NE Saharan dust supply record an on-off mechanism in which a key control on eolian input is provided by the monsoon-driven spread and retreat of savannah vegetation through the Sahara. Magnetite/maghemite is also a common magnetic mineral in NE Saharan dust, and also formed mainly pedogenically during GSPs but in much smaller amounts compared to hematite. Limited production of magnetite/maghemite in the source area during GSPs, along with the later imprint of diagenetic processes, indicates that magnetite/maghemite abundances cannot be used to estimate dust inputs from the NE Sahara. Goethite cannot be used either to estimate variations in NE Saharan dust supply, because its occurrence in Eastern Mediterranean marine sediments also appears to be linked to fluvial inputs.
