    Case-control study of apoE gene polymorphism in young CHD patients and controls in the Serbian population

    Apolipoprotein E displays polymorphism with three common alleles, e2, e3, and e4. The aim of this research was to determine apoE gene polymorphism in a group of healthy patients and a group of patients with CHD, and to reveal the relation between anthropometric and biochemical parameters and the apoE genotype. In CHD group significantly higher values of blood pressure, waist circumference, BMI and fat %, triglycerides, insulin (HOMA IR) and CRP were found. A statistically significant higher presence of the e3e4 genotype and e4 allele was detected in the CHD group. Statistically significant differences between waist circumference, BMI, insulin and HOMA IR were found between subjects with e3e3 and e3e4 genotypes

    Polyglutamine-encoding microsatellite contributes to LMW GS diversity in Triticum monococcum

    Glu-A3 variability of fourteen accessions of diploid wheat species T. monococcum was analyzed by means of SDS-PAGE and PCR. Seven tentative alleles were scored based on LMW GS SDS-PAGE patterns. PCR-amplified DNA sequences corresponding to C-terminus of Glu-3 genes showed size variation that was in accordance with the B-LMW-GS variation as seen on SDS-PAGE for most of the accessions. The sequences of the amplified fragments showed that the amplicon size variation was caused solely by the expansion of the compound (CAG)(CAA) microsatellite within this region. In this paper we have provided evidence for the first time of a correlation between the polyglutamine microsatellite and the B-LMW GS electrophoretic phenotypes

    Potential uses of microsatellites in marker-assisted selection for improved bread-making quality in wheat

    Implementation of marker assisted selection (MAS) in conventional breeding programs could allow assessment of the genetic potential of specific genotypes prior to their phenotypic evaluation. Furthermore, it could identify important trait alleles or marker-trait associations for further determination of a precise position for the loci of interest. Potential uses of microsatellite markers in molecular evaluation of bread-making quality was tested in a sample of 69 wheat genotypes that were genotyped with 3 microsatellites linked to previously mapped QTLs for loaf volume and Hagberg falling number on chromosome 3A. A total of 19 alleles were found, with an average of 6.33 alleles per loci, and average PIC value of 0.40. Specific SSR alleles were tested for association with bread-making related parameters. The association study approach, which uses statistical analysis of marker and phenotypic data, showed significant association of a specific allele at the GWM674 locus with Hagberg falling number in wheat

    The determination of genetic background in metabolic syndrome (MetS) represents one of the necessary steps to prevent the disorder, thus reducing the cost of medical treatments and helping to design targeted therapy. The study explores the association between individual alleles of the LRP1 gene and the diagnosis of MetS to find correlation between the low-density lipoprotein receptor-related (LRP1) gene polymorphism and each individual anthropometric and biochemical parameter. The study included 93 males and females, aged from 19 to 65, divided into two groups. The genotype of each person was determined from the restriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR) profile. Results indicated the association of the T allele form of exon 3 LRP1 gene with development and progression of MetS that further pointed out its negative impact on tested anthropometric and biochemical parameters. The presence of the T allele in patients multiplies the chance of occurrence of deviations from the reference values of body mass index (BMI), (4.24-fold) and low-density lipoprotein (LDL) (20.26-fold) compared to C allele carriers. The results showed that T allele presence multiplies the chance (4.76 fold) for the occurrence of MetS in comparison to C allele carriers. Correlation found that the T allele of the LRP1 gene with MetS determinants is not negligible, therefore, the T allele may be considered as a risk factor for MetS development

    Last Interglacial decadal sea surface temperature variability in the eastern Mediterranean

    The Last Interglacial (~129,000ā€“116,000 years ago) is the most recent geologic period with a warmer-than-present climate. Proxy-based temperature reconstructions from this interval can help contextualize natural climate variability in our currently warming world, especially if they can define changes on decadal timescales. Here, we established a ~4.800-year-long record of sea surface temperature (SST) variability from the eastern Mediterranean Sea at 1ā€“4-year resolution by applying mass spectrometry imaging of long-chain alkenones to a finely laminated organic-matter-rich sapropel deposited during the Last Interglacial. We observe the highest amplitude of decadal variability in the early stage of sapropel deposition, plausibly due to reduced vertical mixing of the highly stratified water column. With the subsequent reorganization of oceanographic conditions in the later stage of sapropel deposition, when SST forcing resembled the modern situation, we observe that the maximum amplitude of reconstructed decadal variability did not exceed the range of the recent period of warming climate. The more gradual, centennial SST trends reveal that the maximal centennial scale SST increase in our Last Interglacial record is below the projected temperature warming in the twenty-first century
