    Effects of Malting and Fermentation on the amount of reducing Sugars and Soluble Proteins and free Amino Acids in White Macia and Red Tannin Sorghum flour

    The purpose of this study was to investigate effects of malting and fermentation methods on the amounts of sugars and proteins in food grade Macia and Red tannin sorghum flours. Measurements to determine the amounts of reducing sugars, soluble protein, and free amino acids, in sorghum flour samples were performed. Grain sorghum kernels (5.2kg) from each sorghum variety were cleaned and kernel hardness tested using Tangential Abrasive Decortication Device (TADD).Half of the kernels (2.6kg) were milled to get regular (Rg) flour samples and the other half (2.6kg) were malted and milled to get malted (mal).flour samples. About 300g of both regular (Rg) and malted (mal) flour samples from both sorghum cultivars (in duplicate) were fermented, dried and milled again to get regular and fermented (Rgfe) and malted and fermented (malfe) flour samples. Extracts were prepared for all (8) flour samples by weighing about 0.5g of flour into centrifuge tubes, then 10 ml of water was added, vortexes 3 times, boiled for 15min and then centrifuged at 5250 RCF for 10 min. The reducing sugars in the extracted material of the regular, fermented, malted and malted/fermented flours from both food grade white Macia and the red tannin sorghum samples were determined by the (DNS) colorimetric methods with glucose as the standard and the absorbance values were read at 540 mm (Miller, 1959).The quantitative measurement of free amino acids of the supernatant material from all flour samples from both the food grade white macia and red tannin sorghum samples were performed using the ninhydrin reaction (Plummer, 1978). The amounts of soluble proteins in the supernatant material were measured using the Lowry method (Lowry et al, 1951). Results showed an increase in reducing sugars, soluble proteins and free amino acids in the malted and fermented flour samples compared to the control or regular flour samples, respectively. Malting and fermentation resulted in increased amounts of reducing sugars, soluble proteins and free amino acids in both food grade Macia and Red tannins sorghum flour. Statistical analysis results showed a significant (p < 0.05) difference between malting and fermentation but no significant (p >0.05) difference between the two sorghum varieties (Macia and Red sorghum). Malting and fermentation may be used to improve the nutritional quality of sorghum food products

    Combined microbiological test to assess changes in an organic matrix used to avoid agricultural soil contamination, exposed to an insecticide

    Combined microbiological test (Biolog Ecoplate, denaturing gradient gel electrophoresis (DGGE) and Real Time PCR (qPCR)) were developed to evaluate the impact of repeated diazinon (DZN) applications at high concentration (40 mg kg-1) on microbial communities in a microcosm simulating the organic matrix (straw (50%): peat (25%): soil (25%) vv-1) of an pesticide biopurification system (PBS). Moreover, pesticide dissipation was also evaluated. After three successive exposition of DZN, dissipation efficiency was high; achieved 87%, 93% and 96% after each application, respectively showing a clear accelerated dissipation of this pesticide in the organic matrix. The results obtained with Biolog Ecoplate showed that community level physiological profiles were no affected by the addition of DZN. On the other hand, molecular assays (DGGE and QPCR) demonstrated that the microbial structure (bacteria and fungi) remained relatively stable over time with high DZN doses compared to control. Therefore, the results of the present study, clearly, demonstrate the high dissipation capacity of this biomixture and highlight the microbiological robustness of this biological system.Fil: Tortella, G.     LITpro: a model fitting software for optical interferometry

    9 pagesInternational audienceLITpro is a software for fitting models on data obtained from various stellar optical interferometers, like the VLTI. As a baseline, for modeling the object, it provides a set of elementary geometrical and center-to-limb darkening functions, all combinable together. But it is also designed to make very easy the implementation of more specific models with their own parameters, to be able to use models closer to astrophysical considerations. So LITpro only requires the modeling functions to compute the Fourier transform of the object at given spatial frequencies, and wavelengths and time if needed. From this, LITpro computes all the necessary quantities as needed (e.g. visibilities, spectral energy distribution, partial derivatives of the model, map of the object model). The fitting engine, especially designed for this kind of optimization, is based on a modified Levenberg-Marquardt algorithm and has been successfully tested on real data in a prototype version. It includes a Trust Region Method, minimizing a heterogeneous non-linear and non-convex criterion and allows the user to set boundaries on free parameters. From a robust local minimization algorithm and a starting points strategy, a global optimization solution is effectively achieved. Tools have been developped to help users to find the global minimum. LITpro is also designed for performing fitting on heterogeneous data. It will be shown, on an example, how it fits simultaneously interferometric data and spectral energy distribution, with some benefits on the reliability of the solution and a better estimation of errors and correlations on the parameters. That is indeed necessary since present interferometric data are generally multi-wavelengths

    Noncovalent Interactions by QMC: Speedup by One-Particle Basis-Set Size Reduction

    While it is empirically accepted that the fixed-node diffusion Monte-Carlo (FN-DMC) depends only weakly on the size of the one-particle basis sets used to expand its guiding functions, limits of this observation are not settled yet. Our recent work indicates that under the FN error cancellation conditions, augmented triple zeta basis sets are sufficient to achieve a benchmark level of 0.1 kcal/mol in a number of small noncovalent complexes. Here we report on a possibility of truncation of the one-particle basis sets used in FN-DMC guiding functions that has no visible effect on the accuracy of the production FN-DMC energy differences. The proposed scheme leads to no significant increase in the local energy variance, indicating that the total CPU cost of large-scale benchmark noncovalent interaction energy FN-DMC calculations may be reduced.Comment: ACS book chapter, accepte

    Promising 2,6,9-Trisubstituted Purine Derivatives for Anticancer Compounds: Synthesis, 3D-QSAR, and Preliminary Biological Assays

    We designed, synthesized, and evaluated novel 2,6,9-trisubstituted purine derivatives for their prospective role as antitumor compounds. Using simple and efficient methodologies, 31 compounds were obtained. We tested these compounds in vitro to draw conclusions about their cell toxicity on seven cancer cells lines and one non-neoplastic cell line. Structural requirements for antitumor activity on two different cancer cell lines were analyzed with SAR and 3D-QSAR. The 3D-QSAR models showed that steric properties could better explain the cytotoxicity of compounds than electronic properties (70% and 30% of contribution, respectively). From this analysis, we concluded that an arylpiperazinyl system connected at position 6 of the purine ring is beneficial for cytotoxic activity, while the use of bulky systems at position C-2 of the purine is not favorable. Compound 7h was found to be an effective potential agent when compared with a currently marketed drug, cisplatin, in four out of the seven cancer cell lines tested. Compound 7h showed the highest potency, unprecedented selectivity, and complied with all the Lipinski rules. Finally, it was demonstrated that 7h induced apoptosis and caused cell cycle arrest at the S-phase on HL-60 cells.     Interacting Kasner-type cosmologies

    It is well known that Kasner-type cosmologies provide a useful framework for analyzing the three-dimensional anisotropic expansion because of the simplification of the anisotropic dynamics. In this paper relativistic multi-fluid Kasner-type scenarios are studied. We first consider the general case of a superposition of two ideal cosmic fluids, as well as the particular cases of non-interacting and interacting ones, by introducing a phenomenological coupling function q(t)q(t). For two-fluid cosmological scenarios there exist only cosmological scaling solutions, while for three-fluid configurations there exist not only cosmological scaling ones, but also more general solutions. In the case of triply interacting cosmic fluids we can have energy transfer from two fluids to a third one, or energy transfer from one cosmic fluid to the other two. It is shown that by requiring the positivity of energy densities there always is a matter component which violates the dominant energy condition in this kind of anisotropic cosmological scenarios.Comment: Accepted for publication in Astrophysics &Space Science, 8 page
