    The Highest-Copy Repeats are Methylated in the Small Genome of the Early Divergent Vascular Plant Selaginella moellendorffii

    Background The lycophyte Selaginella moellendorffii is a vascular plant that diverged from the fern/seed plant lineage at least 400 million years ago. Although genomic information for S. moellendorffii is starting to be produced, little is known about basic aspects of its molecular biology. In order to provide the first glimpse to the epigenetic landscape of this early divergent vascular plant, we used the methylation filtration technique. Methylation filtration genomic libraries select unmethylated DNA clones due to the presence of the methylation-dependent restriction endonuclease McrBC in the bacterial host. Results We conducted a characterization of the DNA methylation patterns of the S. moellendorffii genome by sequencing a set of S. moellendorffii shotgun genomic clones, along with a set of methylation filtered clones. Chloroplast DNA, which is typically unmethylated, was enriched in the filtered library relative to the shotgun library, showing that there is DNA methylation in the extremely small S. moellendorffii genome. The filtered library also showed enrichment in expressed and gene-like sequences, while the highest-copy repeats were largely under-represented in this library. These results show that genes and repeats are differentially methylated in the S.moellendorffii genome, as occurs in other plants studied. Conclusion Our results shed light on the genome methylation pattern in a member of a relatively unexplored plant lineage. The DNA methylation data reported here will help understanding the involvement of this epigenetic mark in fundamental biological processes, as well as the evolutionary aspects of epigenetics in land plants

    The Lunar Polar Hydrogen Mapper (LunaH-Map) Mission

    The Lunar Polar Hydrogen Mapper (LunaH-Map) mission will map hydrogen enrichments within permanently shadowed regions at the lunar south pole using a miniature neutron spectrometer. While hydrogen enrichments have been identified regionally from previous orbital missions, the spatial extent of these regions are often below the resolution of the neutron instruments that have flown on lunar missions. LunaH-Map will enter into an elliptical, low altitude perseline orbit which will enable the mission to spatially isolate and constrain the hydrogen enrichments within permanently shadowed regions. LunaH-Map will use a solid iodine ion propulsion system, X-Band radio communications through the NASA Deep Space Network, star tracker, C&DH and EPS systems from Blue Canyon Technologies, solar arrays from MMA Designs, LLC, mission design and navigation by KinetX.     LunaH-Map: Revealing Lunar Water with a New Radiation Sensor Array

    A new type of neutron and gamma-ray spectrometer called the Miniature Neutron Spectrometer (Mini-NS) has been developed, assembled, qualified and delivered as part of the Lunar Polar Hydrogen Mapper (LunaH-Map) cubesat mission. The LunaH-Map spacecraft is currently manifested as a secondary payload on the Space Launch System (SLS) Artemis-1 rocket. LunaH-Map will deploy from Artemis-1 and enter a low altitude perilune elliptical orbit around the Moon. The Mini-NS will measure the lunar epithermal neutron albedo, and measurements around perilune will be used to produce maps of hydrogen enrichments and depletions across the lunar South Pole region including both within and outside of permanently shadowed regions (PSRs). The Min-NS was designed to achieve twice the epithermal neutron count rate of the Lunar Prospector Neutron Spectrometer (LP-NS). The instrument response was characterized through the collection of pre-flight neutron counting data with a Cf-252 neutron source at Arizona State University across hundreds of power cycles, as well as across the expected temperature range. The instrument spatial response was characterized at the Los Alamos National Laboratories (LANL) Neutron Free In-Air Facility. The LunaH-Map Mini-NS was designed to fit within the cubesat form-factor and uses two detectors with eight sensor heads that can be operated independently. For future missions with different science goals that can be achieved with epithermal neutron detection, the number of Mini-NS sensor heads can easily be modified without requiring a complete re-design and re-qualification

    Efficacy and Safety of a Single Dose of Casirivimab and Imdevimab for the Prevention of COVID-19 Over an 8-Month Period: A Randomised, Double-Blind, Placebo-Controlled Trial

    BACKGROUND: There is an unmet need for COVID-19 prevention in patient populations who have not mounted or are not expected to mount an adequate immune response to complete COVID-19 vaccination. We previously reported that a single subcutaneous 1200 mg dose of the monoclonal antibody combination casirivimab and imdevimab (CAS + IMD) prevented symptomatic SARS-CoV-2 infections by 81·4% in generally healthy household contacts of SARS-CoV-2-infected individuals over a 1-month efficacy assessment period. Here we present additional results, including the 7-month follow-up period (months 2-8), providing additional insights about the potential for efficacy in pre-exposure prophylaxis settings. METHODS: This was a randomised, double-blind, placebo-controlled trial done in the USA, Romania, and Moldova in 2020-2021, before the emergence of omicron (B.1.1.529) and omicron-lineage variants. Uninfected and unvaccinated household contacts of infected individuals, judged by the investigator to be in good health, were randomly assigned (1:1) to receive 1200 mg CAS + IMD or placebo by subcutaneous injection according to a central randomisation scheme provided by an interactive web response system; randomisation was stratified per site by the test results of a local diagnostic assay for SARS-CoV-2 and age group at baseline. COVID-19 vaccines were prohibited before randomisation, but participants were allowed to receive COVID-19 vaccination during the follow-up period. Participants who developed COVID-19 symptoms during the follow-up period underwent RT-PCR testing. Prespecified endpoints included the proportion of previously uninfected and baseline-seronegative participants (seronegative-modified full analysis set) who had RT-PCR-confirmed COVID-19 in the follow-up period (post-hoc for the timepoints of months 2-5 and 6-8 only) and underwent seroconversion (ie, became seropositive, considered a proxy for any SARS-CoV-2 infections [symptomatic and asymptomatic]; prespecified up to day 57, post-hoc for all timepoints thereafter). We also assessed the incidence of treatment-emergent adverse events. This study is registered with ClinicalTrials.gov, NCT04452318. FINDINGS: From July 13, 2020, to Oct 4, 2021, 2317 participants who were RT-PCR-negative for SARS-CoV-2 were randomly assigned, of whom 1683 (841 assigned to CAS + IMD and 842 assigned to placebo) were seronegative at baseline. During the entirety of the 8-month study, CAS + IMD reduced the risk of COVID-19 by 81·2% (nominal p INTERPRETATION: CAS + IMD is not authorised in any US region as of Jan 24, 2022, because data show that CAS + IMD is not active against omicron-lineage variants. In this study, done before the emergence of omicron-lineage variants, a single subcutaneous 1200 mg dose of CAS + IMD protected against COVID-19 for up to 5 months of community exposure to susceptible strains of SARS-CoV-2 in the pre-exposure prophylaxis setting, in addition to the post-exposure prophylaxis setting that was previously shown.
