    Contributions from the reaction e+e- --> pi+pi- e+e- to the pion form factor measurement via radiative return method are discussed basing on the results of a Monte Carlo generator (EKHARA). The generator contains contributions from the initial and final state emission of a e+e- pair from e+e- --> pi+pi- production diagrams and the pi+pi- pair production from space-like and time-like Bhabha diagrams. A detailed study is performed for the Phi- factory energy. Tests of the generation procedure are also presented

    Electrophysiological Indicators of the Age-Related Deterioration in the Sensitivity to Auditory Duration Deviance

    The present study investigates age-related changes in duration discrimination in millisecond time domain. We tested young (N = 20, mean age = 24.5, SD = 2.97) and elderly (N = 20, mean age = 65.2, SD = 2.94) subjects using the mismatch negativity (MMN) paradigm. White-noise bursts of two different durations (50 and 10 ms) were presented in two oddball blocks. In one block (Increment Condition), the repetitive sequence of 10 ms standards was interspersed by occasional 50 ms deviants. In the Decrement Condition, the roles of the two stimuli were reversed. We analyzed the P1-N1 complex, MMN and P3a and found the effect of age for all these components. Moreover, the impact of stimulus presentation condition (increment/decrement) was observed for MMN and P3a. Our results confirmed the previous evidence for deteriorated duration discrimination in elderly people. Additionally, we found that this effect may be influenced by procedural factors.Peer reviewe

    Altered event-related potentials and theta oscillations index auditory working memory deficits in healthy aging

    Speech comprehension deficits constitute a major issue for an increasingly aged population, as they may lead older individuals to social isolation. Since conversation requires constant monitoring, updating and selecting information, auditory working memory decline, rather than impoverished hearing acuity, has been suggested a core factor. However, in stark contrast to the visual domain, the neurophysiological mechanisms underlying auditory working memory deficits in healthy aging remain poorly understood, especially those related to on-the-fly information processing under increasing load. Therefore, we investigated the behavioral costs and electrophysiological differences associated with healthy aging and working memory load during continuous auditory processing. We recorded EEG activity from 27 younger (∼25 years) and 29 older (∼70 years) participants during their performance on an auditory version of the n-back task with speech syllables and 2 workload levels (1-back; 2-back). Behavioral measures were analyzed as indices of function; event-related potentials as proxies for sensory and cognitive processes; and theta oscillatory power as a reflection of memory and central executive function. Our results show age-related differences in auditory information processing within a latency range that is consistent with a series of impaired functions, from sensory gating to cognitive resource allocation during constant information updating, especially under high load

    mRNA expression and immunohistochemical localization of inducible nitric oxide synthase (NOS-2) in the muscular niche of Trichinella spiralis.

    The aim of this study was to demonstrate iNOS mRNA expression in muscular phase of experimental trichinellosis and to localize iNOS protein in T. spiralis-infected muscles using specific anti-iNOS monoclonal antibodies. The expression of iNOS mRNA in skeletal muscles from Trichinella spiralis-infected mice was examined using the reverse transcription PCR assay. Fragments of skeletal muscles were also subjected to the immunohistochemical reaction using specific anti-iNOS monoclonal antibodies followed by Dako-Ark test. mRNA for iNOS measured on day 21 after infection was expressed in the muscular phase of trichinellosis. Positive immunostaining for iNOS occurred in infiltrating mononuclear cells around the encapsulated larvae. iNOS-positive cells could be traced from the 21st day post infection (dpi); on 42 dpi and 90 dpi most cells expressed iNOS. By assessing expression of protein and its mRNA it can be concluded that iNOS is active in the pathology of skeletal muscle tissue in experimental trichinellosis

    Structural bases of peptidoglycan recognition by lysostaphin SH3b domain.

    Staphylococcus simulans lysostaphin cleaves pentaglycine cross-bridges between stem peptides in the peptidoglycan of susceptible staphylococci, including S. aureus. This enzyme consists of an N-terminal catalytic domain and a cell wall binding domain (SH3b), which anchors the protein to peptidoglycan. Although structures of SH3bs from lysostaphin are available, the binding modes of peptidoglycan to these domains are still unclear. We have solved the crystal structure of the lysostaphin SH3b domain in complex with a pentaglycine peptide representing the peptidoglycan cross-bridge. The structure identifies a groove between β1 and β2 strands as the pentaglycine binding site. The structure suggests that pentaglycine specificity of the SH3b arises partially directly by steric exclusion of Cβ atoms in the ligand and partially indirectly due to the selection of main chain conformations that are easily accessible for glycine, but not other amino acid residues. We have revealed further interactions of SH3b with the stem peptides with the support of bioinformatics tools. Based on the structural data we have attempted engineering of the domain specificity and have investigated the relevance of the introduced substitutions on the domain binding and specificity, also in the contexts of the mature lysostaphin and of its bacteriolytic activity

    Inhibin-α, E-cadherin, calretinin and Ki-67 antigen in the immunohistochemical evaluation of canine and human testicular neoplasms

    Introduction. The steady increase of dogs with diagnosed testicular neoplasms observed in recent years prompted us to carry out immunohistochemical (IHC) studies for their better characterization. The aim of the study was to analyze most common canine testicular neoplasms (seminomas, Leydig cell and Sertoli cell tumors) with selected IHC markers and to compare the expressions of these proteins in corresponding canine and human testicular tumors. Material and methods. Studies were carried out on testicular canine tumors: 40 cases of seminoma, 40 cases of Leydig cell tumor and 40 cases of Sertoli cell tumor. Moreover, 15 cases of human seminomas and 5 cases of human Leydig cell tumors were also analyzed. Immunohistochemistry was performed on paraffin sections by standard technique using monoclonal anti-human antibodies against E-cadherin, inhibin-α, calretinin and Ki-67. The slides were subjected to computer-aided image analysis and the intensity of the immunoreactivity was assessed by a semi-quantitative scoring system. Results. Due to the very low prevalence of the Sertoli cell-derived tumors in the human population, we were able to examine the markers’ expression only in the canine gonadal tumors. We revealed that, apart from E-cadherin in Leydig cell tumors and calretinin in seminomas, the expression of all the analyzed markers in canine and human testicular tumors was similar. E.g. there was no immunoexpression of inhibin-α in 75% of canine and 100% of human cases of seminoma. The immunoreactivity of Ki-67 was intense in 40% of canine and 60% of human seminomas. Immunoexpression of inhibin-α in Leydig cell tumor was intense in 70% of canine and 100% of human cases, respectively. Also the immunoreactivity of calretinin was intense in 75% of cases of canine and 100% of human Leydig cell tumors. In 50% of canine and 40% of human Leydig cell tumors, the immunoexpression of Ki-67 was weak. Conclusions. The applied anti-human monoclonal antibodies against common antigens and markers of human testicular neoplasms could be routinely used for the immunohistochemical evaluation of canine testicular tumors.

    NON-DESTRUCTIVE TESTING OF THE HISTORIC TIMBER ROOF STRUCTURES OF THE NATIONAL MUSEUM IN STOCKHOLM, SWEDEN

    The National Museum in Stockholm is Sweden’s leading museum of art and design. Behind the now closed doors of the National Museum and its construction coverings a renovation project is taking place, which began in 2014 and will be completed in 2018. The renovation project will create a modern museum, a brighter atmosphere for the arts and for its visitors. Part of the renovation project was to de-sign and upgrade the timber roof structure for new loads according to Eurocode 5. As the roof structure was originally built in 1860, it was important to evaluate the condition and mechanical properties of the original load-bearing timber members. The roof structure, which is of main interest, comprises roof trusses and lantern structures.The main aim of the project regarding the timber roof structure in the National Museum was a safety verification in both the ultimate state (ULS) and the serviceability limit state (SLS). The preliminary capacity calculations for the roof structure showed that the roof members were under the new load conditions (with increased loads due to security and environmental requirements) utilized above 100%. Therefore, an investigation into the properties of the old timber members and connections was of vital importance. The need for non-destructive testing (NDT) of structural timber is well known under the name of strength grading and the concept has been used for many years to classify timber with respect to mechanical performance. The stress wave technique used in the project was based on commercial instruments such as FAKOPP\uae. The timber quality investigation for the attic roof structures showed that the material was generally in very good condition. As a result, according to SS-EN 338-2009, the timber strength could be assigned strength class C27–C30.
