    Dynamical response of the southwestern Laurentide Ice Sheet to rapid Bølling–Allerød warming

    The shift in climate that occurred between the Last Glacial Maximum (LGM) and the Early Holocene (ca. 18–12 kyr BP) displayed rates of temperature increase similar to present-day warming trends. The most rapid recorded changes in temperature occurred during the abrupt climate oscillations known as the Bølling–Allerød interstadial (14.7–12.9 kyr BP) and the Younger Dryas stadial (12.9–11.7 kyr BP). Reconstructing ice sheet dynamics during these climate oscillations provides the opportunity to assess long-term ice sheet evolution in reaction to a rapidly changing climate. Here, we use glacial geomorphological inversion methods (flowsets) to reconstruct the ice flow dynamics and the marginal retreat pattern of the southwestern sector of the Laurentide Ice Sheet (SWLIS). We combine our reconstruction with a recently compiled regional deglaciation chronology to depict ice flow dynamics that encompass the time period from pre-LGM to the Early Holocene. Our reconstruction portrays three macroscale reorganizations in the orientation and dynamics of ice streaming followed by regional deglaciation associated with rapid warming during the Bølling–Allerød interstadial. Initial westward flow is documented, likely associated with an early set of ice streams that formed during the advance to the LGM. During the LGM ice streaming displays a dominant north to south orientation. Ice sheet thinning at ∼15 ka is associated with a macroscale reorganization in ice stream flow, with a complex of ice streams recording south-eastward flow. A second macroscale reorganization in ice flow is then observed at ∼14 ka, in which southwestern ice flow is restricted to the Hay, Peace, Athabasca, and Churchill river lowlands. Rates of ice sheet retreat then slowed considerably during the Younger Dryas stadial; at this time, the ice margin was situated north of the Canadian Shield boundary and ice flow continued to be sourced from the northeast. Resulting from these changes in ice sheet dynamics, we recognize a three-part pattern of deglacial landform zonation within the SWLIS characterized by active ice margin recession, stagnation and downwasting punctuated by local surging (terrestrial ice sheet collapse): the outer deglacial zone contains large recessional moraines aligned with the direction of active ice margin retreat; the intermediate deglacial zone contains large regions of hummocky and stagnation terrain, in some areas crosscut by the signature of local surges, reflecting punctuated stagnation and downwasting; and the inner deglacial zone contains inset recessional moraines demarcating progressive regional ice margin retreat. We attribute these macroscale changes in ice flow geometry and associated deglacial behaviour to external climatic controls during the Bølling–Allerød and Younger Dryas but also recognize the role of internal (glaciological, lithological and topographic) controls in SWLIS dynamics

    Learning from community pharmacists' initial experiences of a workplace-based training program

    Introduction:  To prepare community pharmacists for the provision of clinical and patient-focused services, a novel postgraduate course for community pharmacists in the United Kingdom was developed. The program incorporated personal development planning against a personal development framework, workplace mentoring, employment of work-based assessment tools, activities that encouraged increased inter-professional working, reflection, and opportunities for peer support. Objectives were to identify course components that support development, describe the effect on practice, and explore the perceived advantages and disadvantages of this model of postgraduate education. Methods:  Interviews were conducted with a purposive sample of 15 community pharmacists after they had completed approximately one year of the three-year course. A topic guide covering approaches to learning, rationale for course selection, course experiences to date, and impact on practice was used. Interview recordings were thematically analyzed. Results:  Two themes were identified. ‘Support for learning’ describes the components of the course that provided support for learning, including opportunities to learn with and from others, workplace mentoring, and facilitated access to general practitioners. ‘Outcomes of learning’ encompasses how the course was a way of effecting change within existing roles and the increase in confidence and motivation to change practice. Conclusions:  The model has merit in supporting community pharmacists to develop the confidence and competence required for extended clinical and patient-focused roles. While this model of learning seems to provide educational value, further research is required to determine whether the additional resources required to provide workplace mentoring, use work-based assessment tools, and encourage inter-professional working are justified

    Patterns, levels and correlates of self-reported physical activity in urban black Soweto women

    BACKGROUND: Urban black South African women have a high prevalence of non-communicable diseases such as obesity and type 2 diabetes. The aim of this study was to assess the physical activity patterns of a cohort of middle-aged urban-dwelling black African women and to determine if physical activity is associated with anthropometric measures and metabolic outcomes in this population. METHODS: Physical activity and sitting time were assessed using the Global Physical Activity Questionnaire (GPAQ) in a cross-sectional study of 977 black African women (mean age 41.0 ± 7.84 years) from the Birth to Twenty study based in Soweto, Johannesburg. Anthropometric outcomes were measured and fasting blood glucose, insulin and lipid profile were analysed to determine metabolic disease risk and prevalence. RESULTS: Sixty-seven percent of the population were classified as active according to GPAQ criteria, and the domain that contributed most to overall weekly physical activity was walking for travel. Only 45.0% of women participated in leisure time activity. The prevalence of metabolic syndrome in this sample was 40.0%, and the prevalence of overweight and obesity was 29.2% and 48.0%, respectively. Women who reported owning a motor vehicle walked for travel less, and participated in more leisure-time activity (both p < 0.01), while women who owned a television reported significantly lower moderate-vigorous physical activity (MVPA), and walking for travel (both p < 0.01). Sitting time (mins/wk) was not different between the activity groups, but was associated with triglycerides and diastolic blood pressure. Total physical activity was inversely associated with fasting insulin, and physical activity in the work domain was associated with fat free soft tissue mass. CONCLUSIONS: The findings of this study show that the majority of urban dwelling black South African women are classified as physically active despite a high prevalence of obesity and metabolic disease risk factors. Sitting time had detrimental effects on both triglyceride levels and diastolic blood pressure whilst total physical activity attenuated fasting insulin levels. As walking for travel is a major contributor to physical activity, future research should attempt to determine whether the intensity of this activity plays a role in the prevention of cardiometabolic diseases.     Appetite regulation genes are associated with body mass index in black South African adolescents: a genetic association study

Genome-wide association studies have identified several variants that are robustly associated with obesity and body mass index (BMI), many of which are found within genes involved in appetite regulation. Currently, genetic association data for obesity are lacking in Africans-a single genome-wide association study and a few replication studies have been published in West Africa, but none have been performed in a South African population. OBJECTIVE: To assess the association of candidate loci with BMI in black South Africans. The authors focused on single nucleotide polymorphisms (SNPs) in the FTO, LEP, LEPR, MC4R, NPY2R and POMC genes. DESIGN: A genetic association study. PARTICIPANTS: 990 randomly selected individuals from the larger Birth to Twenty cohort (a longitudinal birth cohort study of health and development in Africans). MEASURES: The authors genotyped 44 SNPs within the six candidate genes that included known BMI-associated SNPs and tagSNPs based on linkage disequilibrium in an African population for FTO, LEP and NPY2R. To assess population substructure, the authors included 18 ancestry informative markers. Weight, height, sex, sex-specific pubertal stage and exact age collected during adolescence (13 years) were used to identify loci that predispose to obesity early in life. RESULTS: Sex, sex-specific pubertal stage and exact age together explain 14.3% of the variation in log(BMI) at age 13. After adjustment for these factors, four SNPs were individually significantly associated with BMI: FTO rs17817449 (p=0.022), LEP rs10954174 (p=0.0004), LEP rs6966536 (p=0.012) and MC4R rs17782313 (p=0.045). Together the four SNPs account for 2.1% of the variation in log(BMI). Each risk allele was associated with an estimated average increase of 2.5% in BMI. CONCLUSIONS: The study highlighted SNPs in FTO and MC4R as potential genetic markers of obesity risk in South Africans.     Analysis of the asymmetrically expressed Ablim1 locus reveals existence of a lateral plate Nodal-independent left sided signal and an early, left-right independent role for nodal flow

    BACKGROUND: Vertebrates show clear asymmetry in left-right (L-R) patterning of their organs and associated vasculature. During mammalian development a cilia driven leftwards flow of liquid leads to the left-sided expression of Nodal, which in turn activates asymmetric expression of the transcription factor Pitx2. While Pitx2 asymmetry drives many aspects of asymmetric morphogenesis, it is clear from published data that additional asymmetrically expressed loci must exist. RESULTS: A L-R expression screen identified the cytoskeletally-associated gene, actin binding lim protein 1 (Ablim1), as asymmetrically expressed in both the node and left lateral plate mesoderm (LPM). LPM expression closely mirrors that of Nodal. Significantly, Ablim1 LPM asymmetry was detected in the absence of detectable Nodal. In the node, Ablim1 was initially expressed symmetrically across the entire structure, resolving to give a peri-nodal ring at the headfold stage in a flow and Pkd2-dependent manner. The peri-nodal ring of Ablim1 expression became asymmetric by the mid-headfold stage, showing stronger right than left-sided expression. Node asymmetry became more apparent as development proceeded; expression retreated in an anticlockwise direction, disappearing first from the left anterior node. Indeed, at early somite stages Ablim1 shows a unique asymmetric expression pattern, in the left lateral plate and to the right side of the node. CONCLUSION: Left LPM Ablim1 is expressed in the absence of detectable LPM Nodal, clearly revealing existence of a Pitx2 and Nodal-independent left-sided signal in mammals. At the node, a previously unrecognised action of early nodal flow and Pkd2 activity, within the pit of the node, influences gene expression in a symmetric manner. Subsequent Ablim1 expression in the peri-nodal ring reveals a very early indication of L-R asymmetry. Ablim1 expression analysis at the node acts as an indicator of nodal flow. Together these results make Ablim1 a candidate for controlling aspects of L-R identity and patterning

    Glucocorticoids associate with cardiometabolic risk factors in black South Africans

    Circulating glucocorticoids are associated with metabolic syndrome and related cardiometabolic risk factors in non-Africans. This study investigated these associations in Africans, whose metabolic phenotype reportedly differs from Europeans. Adiposity, blood pressure, glycaemia, insulin resistance, and lipid profile, were measured in 316 African men and 788 African women living in Soweto, Johannesburg. The 2009 harmonized criteria were used to define metabolic syndrome. Serum glucocorticoids were measured using liquid chromatography-mass spectrometry. Cortisol was associated with greater odds presenting with metabolic syndrome (odds ratio (95% CI) =1.50 (1.04, 2.17) and higher systolic (beta coefficient, β (95% CI) =0.04 (0.01, 0.08)) and diastolic (0.05 (0.02, 0.09)) blood pressure, but higher HDL (0.10 (0.02, 0.19)) and lower LDL (−0.14 (−0.24, −0.03)) cholesterol concentrations, in the combined sample of men and women. In contrast, corticosterone was only associated with higher insulin sensitivity (Matsuda index; 0.22 (0.03, 0.41)), but this was not independent of BMI. Sex-specific associations were observed, such that both cortisol and corticosterone were associated with higher fasting glucose (standardized β (95% CI): 0.24 (0.12, 0.36) for cortisol and 0.12 (0.01, 0.23) for corticosterone) and HbA1c (0.13 (0.01, 0.25) for cortisol and 0.12 (0.01, 0.24) for corticosterone) in men only, but lower HbA1c (0.10 (−0.20, −0.01) for cortisol and −0.09 (−0.18, −0.03) for corticosterone) in women only. Our study reports for the first time that associations between circulating glucocorticoid concentrations and key cardiometabolic risk factors exhibit both glucocorticoid- and sex-specificity in Africans

    Microblogging, constituency service and impression management – UK MPs and the use of Twitter

    Twitter, a microblogging site which allows users to deliver statements, thoughts and links in 140 characters to followers as well as a wider Internet audience, is the latest online communications technology adopted by MPs. Assessing the use of early adopters, this article considers which MPs are most likely to use Twitter (e.g. tweeting), and how. Content analysis of tweeting MPs was conducted, and identified personal and political characteristics which may influence use. The data suggested that of the six characteristics tested, gender, party and seniority had most impact on adoption. Applying Jones and Pittman’s (1982) typology there is clear evidence that MPs use Twitter as a tool of impression management. Constituency service is a secondary function of the use of Twitter by MPs. Where MPs use Twitter as part of their constituency role it is to promote their local activity. We note that a small group of MPs use Twitter as a regular communication channel, but most are only occasionally dipping their toe into the microbloggersphere
