    Causalities of war: The connection between type VI secretion system and microbiota

Different animal and environmental niches contain defined resident microbiota that tend to be stable over time and offer protection against undesired intruders. Yet fluxes can occur, which alter the composition of a bacterial population. In humans, the microbiota are now considered a key contributor to maintenance of health and homeostasis, and its alteration leads to dysbiosis. The bacterial type VI secretion system (T6SS) transports proteins into the environment, directly into host cells or can function as an antibacterial weapon by killing surrounding competitors. Upon contact with neighbouring cells, the T6SS fires, delivering a payload of effector proteins. In the absence of an immunity protein, this results in growth inhibition or death of prey leading to a competitive advantage for the attacker. It is becoming apparent that the T6SS has a role in modulating and shaping the microbiota at multiple levels, which is the focus of this review.     Trends in Child Poverty by Race/Ethnicity: New Evidence Using an Anchored Historical Supplemental Poverty Measure

    Official poverty statistics have been criticized, however, for being based on an outdated measure of poverty (Blank, 2008; Citro and Michael, 1995). First put into use in the 1960s, the official poverty measure’s (OPM) concept of needs has been updated for inflation but still reflects the living standards, family budgets, and family structures of that time. Moreover, when tallying family resources, the OPM misses key government programs such as Food Stamps and tax credits that have expanded since the 1960s. For these reasons, the Census Bureau and the Bureau of Labor Statistics (BLS) implemented an improved “supplemental poverty measure” (SPM) in 2011 (Short 2011) for calendar years 2009 and 2010. This SPM is now released annually alongside the OPM (see Short 2015 for the latest data as of this writing), but the Census Bureau has no plans to produce the measure historically. However, historical data on levels and trends in poverty are essential for better understanding the progress the country has made since Lyndon B. Johnson’s famous declaration of the War on Poverty in the 1960s. Understanding what has been successful in the past is important for assessing what might be successful in the future for amelioration of economic disadvantage. What’s more, success and its sources may vary by race/ethnicity. We use a historical version of the SPM to provide the first estimates of racial/ethnic differences in child poverty for the period 1970 to the present using this improved measure. We begin our analysis in 1970 because that is the first year we can reliably distinguish non-Hispanic whites, African-Americans, and Latinos (unfortunately, data limitations prevent us from examining other groups over the long term). We detail below our data and methods, then describe our main results, and conclude briefly. Data and Methods We use data from multiple years of the Current Population Survey’s Annual Social and Economic Supplement (also known as the March CPS) combined with data from the Consumer Expenditure Survey (CEX) to produce SPM estimates for the period 1970 to 2014. We use a methodology similar to that used by the Census Bureau and the Bureau of Labor Statistics in producing their SPM estimates, but with adjustments for differences in available data over time. Our methodology differs from the SPM in only one respect. Instead of using a poverty threshold that is re-calculated over time, we use today’s threshold and carry it back historically by adjusting it for inflation using the CPI-U-RS. Because this alternative measure is anchored with today’s SPM threshold, we refer to it as an anchored supplemental poverty measure, or anchored SPM for short. An advantage of an anchored SPM is that poverty trends resulting from such a measure can be explained only by changes in income and net transfer payments (cash or in kind). Trends in poverty based on a relative measure (e.g. SPM poverty), on the other hand, could be due to over time changes in thresholds. Thus, an anchored SPM arguably provides a cleaner measure of how changes in income and net transfer payments have affected poverty historically (Wimer et al., 2013).[1] [1] All analyses in this paper are also available using quasi-relative poverty thresholds; results are available upon request

    Lived Clinical Education Experiences of Professional Master’s Athletic Training Program Graduates

    Purpose: Clinical education has been identified as a key aspect in the professional preparation of athletic trainers. The literature, however, has focused on clinical education at the undergraduate level rather than the graduate level, which is the future population of all CAATE-accredited athletic training programs. This study sought to describe the experiences of those who have completed athletic training clinical education at the master’s level, specifically focusing on the type of learning experiences they were granted and what influence the preceptor and the setting played in the overall experience. Method: This was a phenomenology study. Twelve graduates of CAATE-accredited professional master’s athletic training programs served as participants. All participants graduated after Spring 2014 and completed clinical education as a degree requirement. Three methods of data collection were utilized, including semi-structured interviews, prompted journaling and asynchronous online focus group discussion boards. Data was analyzed using a phenomenological reduction approach. Credibility of the data was confirmed with member checking, peer review and data triangulation. Results: Clinical education was determined to be a significant contributor to professional preparation of professional master’s athletic training students. Hands on-experiences, active preceptors and variety in clinical setting and opportunity were all cited as contributors to the success of clinical education. Participants described their experience as a significant commitment but valuable. Conclusions: Our findings suggest that clinical education should continue to be a requirement at the graduate level and that the clinical education curriculum should be designed to encourage hands-on learning and variety

    Space, politics and community: the case of Kinning Park Complex

    This thesis is about space, politics and the community. It examines how spatial politics constitutes a community through time. It explores the way that urban governance interacts with community politics, and more importantly, how people can rework politics through spatial practices. The thesis scales down to focus on a case study of Kinning Park Complex (KPC), an independently run community centre that was saved from closure by building users in 1996 following a 55 day sit-in. I track the trajectory of this space since 1996, to investigate the resourcefulness of the community to withstand multiple crises at local and national levels. KPC is a valuable social and political space that continues to exist in, against, and beyond neoliberalism. I focus on the paradoxical nature of KPC, as the space appears in-line with the current government plans to expand third sector projects in a context of austerity, whilst simultaneously striving to function as a non-hierarchical and not-for profit space. It is both an important site of social of reproduction and a symbolic community space. Through participatory methods and ethnographic observations, I have explored the social practices at KPC to investigate what they reveal about social relations and the structural problems that independent spaces face in the context of austerity. I draw upon the theories of Pierre Bourdieu and Jacques Rancière to elucidate the contradictions in their theoretical disagreement by relating their ideas to the rich empirical material that I gathered at KPC. Finally, I draw upon Doreen Massey to bridge this theoretical divide and to provide an essential spatial context to my work. The thesis brings to light the complexities, contradictions and tangible forms of labour involved in simultaneously struggling against, and providing services autonomously from, the state during austere times

    Post-transcriptional gene silencing (PTGS) involving small interfering RNA (siRNA)-directed degradation of RNA transcripts and transcriptional silencing via DNA methylation have each been proposed as mechanisms of genome defence against invading nucleic acids, such as transposons and viruses. Furthermore, recent data from plants indicates that many transposons are silenced via a combination of the two mechanisms, and siRNAs can direct methylation of transposon sequences. We investigated the contribution of DNA methylation and the PTGS pathway to transposon control in the filamentous fungus Neurospora crassa. We found that repression of the LINE1-like transposon, Tad, requires the Argonaute protein QDE2 and Dicer, each of which are required for transgene-induced PTGS (quelling) in N.crassa. Interestingly, unlike quelling, the RNA-dependent RNA polymerase QDE1 and the RecQ DNA helicase QDE3 were not required for Tad control, suggesting the existence of specialized silencing pathways for diverse kinds of repetitive elements. In contrast, Tad elements were not significantly methylated and the DIM2 DNA methyltransferase, responsible for all known DNA methylation in Neurospora, had no effect on Tad control. Thus, an RNAi-related transposon silencing mechanism operates during the vegetative phase of N.crassa that is independent of DNA methylation, highlighting a major difference between this organism and other methylation-proficient species

    Field Assessment of a Model Tuberculosis Outbreak Response Plan for Low-Incidence Areas

    Background: For a regional project in four low-incidence states, we designed a customizable tuberculosis outbreak response plan. Prior to dissemination of the plan, a tuberculosis outbreak occurred, presenting an opportunity to perform a field assessment of the plan. The purpose of the assessment was to ensure that the plan included essential elements to help public health professionals recognize and respond to outbreaks. Methods: We designed a semi-structured questionnaire and interviewed all key stakeholders involved in the response. We used common themes to assess validity of and identify gaps in the plan. A subset of participants provided structured feedback on the plan. Results: We interviewed 11 public health and six community stakeholders. The assessment demonstrated that (1) almost all of the main response activities were reflected in the plan; (2) the plan added value by providing a definition of a tuberculosis outbreak and guidelines for communication and evaluation. These were areas that lacked written protocols during the actual outbreak response; and (3) basic education about tuberculosis and the interpretation and use of genotyping data were important needs. Stakeholders also suggested adding to the plan questions for evaluation and a section for specific steps to take when an outbreak is suspected. Conclusion: An interactive field assessment of a programmatic tool revealed the value of a systematic outbreak response plan with a standard definition of a tuberculosis outbreak, guidelines for communication and evaluation, and response steps. The assessment highlighted the importance of education and training for tuberculosis in low-incidence areas

    Climate change impacts on wind energy generation in Ireland

    An ensemble of high-resolution regional climate model simulation data is used to examine the impacts of climate change on offshore and onshore wind energy genera- tion in Ireland. Two Representative Concentration Pathway (RCP) scenarios (RCP 4.5 and 8.5) are analysed for the mid-term (2041–2060) and the long-term (2081–2100) future. Wind energy is projected to decrease (≤2%) overall in future climate scenarios. Changes are evident by mid-century and are more pronounced by late 21st century, particularly for RCP 8.5 offshore. Seasonally, wind energy is projected to decrease by less than 6% in summer and to increase slightly in winter (up to 1.1%). The distinct changes in different parts of the power curve, presented here for the first time, show a reversed pattern of duration at certain levels of the power curve. In summer, there is an increase of low-power and a decrease of high-power generation, whereas during winter, there is a projected increase in the time spent at high power. This could lead to diverse consequences for system operators depending on the season. The impacts of climate change on the duration and frequency of long periods (longer than 24 h) of low-/high-power wind energy events in Ireland are also presented. The frequency of low-power events is projected to increase slightly, especially during summer. Onshore and offshore events are considered separately, demonstrating the complementarity of developing both onshore and offshore wind farms for future energy systems. Regional analysis highlights the benefit of developing a geographically dispersed wind farm network incorporating different local wind conditions
