    Synergistic interactions between kainate and mGlu receptors regulate bouton Ca2+ signalling and mossy fibre LTP

    It is currently unknown why glutamatergic presynaptic terminals express multiple types of glutamate receptors. We have addressed this question by studying both acute and long-term regulation of mossy fibre function in the hippocampus. We find that inhibition of both mGlu1 and mGlu5 receptors together can block the induction of mossy fibre LTP. Furthermore, mossy fibre LTP can be induced by the pharmacological activation of either mGlu1 or mGlu5 receptors, provided that kainate receptors are also stimulated. Like conventional mossy fibre LTP, chemically-induced mossy fibre LTP (chem-LTPm) depends on Ca2+ release from intracellular stores and the activation of PKA. Similar synergistic interactions between mGlu receptors and kainate receptors were observed at the level of Ca2+ signalling in individual giant mossy fibre boutons. Thus three distinct glutamate receptors interact, in both an AND and OR gate fashion, to regulate both immediate and long-term presynaptic function in the brain

    Bell inequalities as constraints on unmeasurable correlations

    The interpretation of the violation of Bell-Clauser-Horne inequalities is revisited, in relation with the notion of extension of QM predictions to unmeasurable correlations. Such extensions are compatible with QM predictions in many cases, in particular for observables with compatibility relations described by tree graphs. This implies classical representability of any set of correlations , , , and the equivalence of the Bell-Clauser-Horne inequalities to a non void intersection between the ranges of values for the unmeasurable correlation associated to different choices for B. The same analysis applies to the Hardy model and to the "perfect correlations" discussed by Greenberger, Horne, Shimony and Zeilinger. In all the cases, the dependence of an unmeasurable correlation on a set of variables allowing for a classical representation is the only basis for arguments about violations of locality and causality.Comment: Some modifications have been done in order to improve clarity of presentation and comparison with other approache

    Dissolved organic carbon retention by coprecipitation during the oxidation of ferrous iron (EGU2018-3906)

    Although the importance of Fe (hydr)oxides for soil organic matter (OM) stabilization and C retention by surface adsorption is well known, only recently has coprecipitation been recognized as an important process responsible for C storage in hydromorphic soils, such as rice paddy soils. Under periodic fluctuations in redox conditions the interaction between dissolved organic carbon (DOC) and Fe (hydr)oxides may not only involve organic coatings on mineral surfaces, but also Fe-DOC coprecipitates that form during the oxidation of soil solutions containing important amounts of DOC and Fe2+. The aim of this work was to provide new insights into the mechanisms involved, and the amount and selectivity of C retained during the coprecipitation process. A series of Fe-OM associations with increasing C loading was synthesized at pH 6 by surface adsorption or coprecipitation (oxidation of ferrous iron) utilizing rice-straw derived dissolved organic matter. The kinetics of Fe2+ oxidation and complexation, and the total and selective retention of DOC during the coprecipitation process were evaluated. Moreover, synthesized associations, as well as a field coprecipitate collected in situ from a paddy soil, were studied by X-ray diffraction, N2 gas adsorption-desorption isotherms, electrophoretic mobility measurements and thermogravimetric analyses. Coprecipitation resulted in higher organic C contents (49-213 mg g-1) with respect to adsorbed systems (18-47 mg g-1), and favoured the inclusion of OM within highly aggregated associations having particularly low BET specific surface areas. Coprecipitates sampled in situ also showed a similar retention of organic C confirming that this process may contribute significantly to the OM stabilization in paddy topsoils. The mechanisms involved in the retention of straw-derived DOM during coprecipitation were shown to be strongly dependent on C/Fe ratio of the solution. Although the overall coprecipitation process was highly selective for aromatic constituents, initial complexation of Fe2+ and precipitation as C-rich metal salts involved the selective interaction with aliphatic carboxylic constituents. The contribution of the latter mechanism to total C retention during coprecipitation was shown to increase with increasing solution C/Fe ratios. These aliphatic complexes formed during coprecipitation may play an important, though often underestimated, role in C stabilization in hydromorphic. The process-related selectivity of specific constituents of DOM during coprecipitation highlights the involvement of specific mechanisms, i.e. complexation, adsorption, salt precipitation, while ruling out non-selective, physical occlusion of OM within the forming coprecipitat

    Observations of a rotating macrospicule associated with an X-ray jet

    We attempt to understand the driving mechanism of a macrospicule and its relationship with a coronal jet. We study the dynamics of a macrospicule and an associated coronal jet captured by multi-spacecraft observations. Doppler velocities both in the macrospicule and the coronal jet are determined by EIS and SUMER spectra. Their temporal evolution is studied using X-ray and He II 304 images. A blueshift of -120+/-15 km/s is detected on one side of the macrospicule, while a redshift of 50+/-6 km/s is found at the base of the other side. The inclination angle of the macrospicule inferred from a stereoscopic analysis with STEREO suggests that the measured Doppler velocities can be attributed to a rotating motion of the macrospicule rather than a radial flow or an expansion. The macrospicule is driven by the unfolding motion of a twisted magnetic flux rope, while the associated X-ray jet is a radial outflow.Comment: 4 pages, 3 figures, accepted for publication in A&

    The anticipation, perception of affective touch in women with, recovered from Anorexia Nervosa

    Disruptions in reward processing and anhedonia have long been observed in Anorexia Nervosa (AN). Interoceptive deficits have also been observed in AN, including reduced tactile pleasure. However, the extent to which this tactile anhedonia is specifically liked to an impairment in a specialized, interoceptive C-tactile system originating at the periphery, or a more top-down mechanism in the processing of tactile pleasantness remains debated. Here, we investigated differences between patients with and recovered from AN and healthy controls in the perception of pleasantness of touch delivered in a CT-optimal versus a CT-non-optimal manner, and in their top-down, anticipatory beliefs about the perceived pleasantness of touch. To this end, we measured the anticipated pleasantness of various materials touching the skin and the perceived pleasantness of light, dynamic touch applied to theforearmof 27 women with AN, 24 women who have recovered and 30 healthy controls using C Tactile (CT) afferents-optimal (slow) and non-optimal (fast) velocities. Our results showed that both clinical groups anticipated tactile experiences and rated delivered tactile stimuli as less pleasant than healthy controls, but the latter difference was not related to the CT optimality of the stimulation. Instead, differences in the perception of CT-optimal touch were predicted by differences in top-down beliefs, alexithymia and interoceptive sensibility. Thus, tactile anhedonia in AN might persist as a trait even after otherwise successful recovery of AN and it is not linked to a bottom-up interoceptive deficit in the CT system, but rather to a learned, defective top-down anticipation of tactile pleasantness

    Updating Prospective Self-Efficacy Beliefs About Cardiac Interoception in Anorexia Nervosa: An Experimental and Computational Study.

    Patients with anorexia nervosa (AN) typically hold altered beliefs about their body that they struggle to update, including global, prospective beliefs about their ability to know and regulate their body and particularly their interoceptive states. While clinical questionnaire studies have provided ample evidence on the role of such beliefs in the onset, maintenance, and treatment of AN, psychophysical studies have typically focused on perceptual and 'local' beliefs. Across two experiments, we examined how women at the acute AN (N = 86) and post-acute AN state (N = 87), compared to matched healthy controls (N = 180) formed and updated their self-efficacy beliefs retrospectively (Experiment 1) and prospectively (Experiment 2) about their heartbeat counting abilities in an adapted heartbeat counting task. As preregistered, while AN patients did not differ from controls in interoceptive accuracy per se, they hold and maintain 'pessimistic' interoceptive, metacognitive self-efficacy beliefs after performance. Modelling using a simplified computational Bayesian learning framework showed that neither local evidence from performance, nor retrospective beliefs following that performance (that themselves were suboptimally updated) seem to be sufficient to counter and update pessimistic, self-efficacy beliefs in AN. AN patients showed lower learning rates than controls, revealing a tendency to base their posterior beliefs more on prior beliefs rather than prediction errors in both retrospective and prospective belief updating. Further explorations showed that while these differences in both explicit beliefs, and the latent mechanisms of belief updating, were not explained by general cognitive flexibility differences, they were explained by negative mood comorbidity, even after the acute stage of illness

    Obsessive-Compulsive Disorder and Decision Making under Ambiguity: A Systematic Review with Meta-Analysis.

    In the last decade, decision-making has been proposed to have a central role in obsessive-compulsive disorder (OCD) aetiology, since patients show pathological doubt and an apparent inability to make decisions. Here, we aimed to comprehensively review decision making under ambiguity, as measured by the Iowa Gambling Task (IGT), in OCD, using a meta-analytic approach. According to PRISMA Guidelines, we selected 26 studies for a systematic review and, amongst them, 16 studies were included in a meta-analysis, comprising a total of 846 OCD patients and 763 healthy controls (HC). Our results show that OCD patients perform significantly lower than HC at the IGT, pointing towards the direction of a decision making impairment. In particular, this deficit seems to emerge mainly in the last three blocks of the IGT. IGT scores in OCD patients under the age of 18 were still significantly lower than in HC. Finally, no difference emerged between medicated and unmedicated patients, since they both scored significantly lower at the IGT compared to HC. In conclusion, our results are in line with the hypothesis according to which decision making impairment might represent a potential endophenotype lying between the clinical manifestation of OCD and its neurobiological aetiology

    Clinical significance of PTEN and p-Akt co-expression in HER2-positive metastatic breast cancer patients treated with trastuzumab-based therapies

    Objective: The phosphatase and tensine homologue gene (PTEN) plays a crucial role in proliferation and survival of cancer cells by antagonizing the function of phosphatidylinositol 3'-kinase (PI3K), which, in turn, results in decreased Akt activity. We investigated the clinical impact of the expression of PTEN, p-Akt and PI3K in HER2-positive metastatic breast cancer (MBC) patients treated with trastuzumab-based therapies. Methods: Seventy-three patients treated with trastuzumab-based therapies were included and followed prospectively. PTEN, p-Akt and PI3K expression was determined by immunohistochemistry. Results: PTEN, p-Akt and PI3K resulted positive in 48%, 71% and 46.5% of patients, respectively. A significant correlation between PTEN and p-Akt (kappa 0.22, p = 0.03) and p-Akt and PI3K (kappa 0.20, p = 0.05) was observed. PTEN-positive patients had a progression-free survival (PFS) longer than PTEN-negative ones (p = 0.06). When grouped together, patients co-expressing PTEN and p-Akt had a statistically significant longer PFS as compared to the rest of patients (p = 0.01). At the multivariate analysis, PTEN and p-Akt co-expression was an independent predictor of lower risk of progression (hazard ratio 0.53, p = 0.05). Conclusion: In HER2-positive MBC, basal co-expression of PTEN and p-Akt might identify those patients who are more likely to benefit from trastuzumab-based therapies

    Explosive events - swirling transition region jets

    In this paper, we extend our earlier work to provide additional evidence for an alternative scenario to explain the nature of so-called `explosive events'. The bi-directed, fast Doppler motion of explosive events observed spectroscopically in the transition region emission is classically interpreted as a pair of bidirectional jets moving upward and downward from a reconnection site. We discuss the problems of such a model. In our previous work, we focused basically on the discrepancy of fast Doppler motion without detectable motion in the image plane. We now suggest an alternative scenario for the explosive events, based on our observations of spectral line tilts and bifurcated structure in some events. Both features are indicative of rotational motion in narrow structures. We explain the bifurcation as the result of rotation of hollow cylindrical structures and demonstrate that such a sheath model can also be applied to explain the nature of the puzzling `explosive events'. We find that the spectral tilt, the lack of apparent motion, the bifurcation, and a rapidly growing number of direct observations support an alternative scenario of linear, spicular-sized jets with a strong spinning motion.Comment: 9 pages, 3 figures, accepted for publication in Solar Physic
