    The Energetics of Halogenated Ethylenes (Ethynes) and 1,3-Butadienes (Butadiynes): A Computational and Conceptual Study of Substituent Effects and “Dimerization”

    The energetics of ethylenes and 1,3-butadienes may be interrelated by the reaction: RHC=CH2 + H2C=CHR\u27 → RHC=CH−CH=CHR\u27 + H2. Shown earlier to be nearly enthalpically thermoneutral for a variety of hydrocarbon cases, we are now interested in the related energetics of halogenated alkenes and alkynes. Using quantum chemical calculations, we have studied this as recast as the isodesmic reactions: 2(H2C=CHX) + H2C=CH−CH=CH2 → p,q-di-X-1,3-butadiene + 2H2C=CH2 2(HC≡CX) + HC≡C−C≡CH → di-X-butadiyne + 2HC≡CH. Here p,q- = 1,3-; 1,4- and 2,3- with X = F, Cl, Br, and I. The halogen and location-dependent deviations from near enthalpic thermoneutrality are discussed

    Stryd 25 vs. Stryd 27: Comparing Running Metrics Between a Predecessor and “The Next Gen Stryd”

    Wearable technology has claimed the top spot in the Worldwide Survey of Fitness Trends in all but two years since 2016. A popular wearable among runners is the Stryd power meter. The company markets its latest model, the Stryd 27, as 5x more responsive in measuring running power. Yet, it is unclear whether the new model performs differently than its predecessor. PURPOSE: This study aimed to compare running metrics of the Stryd 25 and Stryd 27 in self-paced and interval runs. METHODS: Participants consented (N = 16; 50% female; height = 174.1 ± 8.1 centimeters [cm]; mass = 73.0 ± 12.4 kilograms) and were equipped with the Stryd 25 and Stryd 27, attached randomly to the left and right shoelaces. Each Stryd was paired with a separate mobile device using the Stryd app. Researchers started and stopped recording on each Stryd simultaneously. Participants ran for 10 minutes at a self-selected pace counterclockwise around an indoor track (10 laps/mile) before resting for five minutes. Then participants ran 10 more minutes, alternating between fast and slow intervals: 120 seconds (s) × 2, 60 s × 2, 30 s × 4, and 15 s × 8. Fast and slow intervals were 20% faster and 20% slower, respectively, than the participant’s mean pace of the first run. The Stryd app recorded power in watts (W), cadence in steps per minute (spm), vertical oscillation (VO) in cm, and stride length in meters (m). Four independent t-tests were run to compare these measurements between the two Stryd models for the self-paced and interval runs. The alpha level was .05, and the effect size was Cohen’s d (0.2 small, 0.5 medium, 0.8 large). RESULTS: See Table 1. CONCLUSION: Four running metrics were statistically similar between the Stryd 25 and Stryd 27 during two indoor runs. Runners using the predecessor indoors can be confident it returns similar data to the newest model

    The Effectiveness of Running Power as a Metric of Exercise Intensity During Running Interval Training

    Wearable power meters are increasingly popular among runners with Coros and Stryd offering running power as a real-time, trackable of a metric. PURPOSE: This study compared running power (RP) to physiological measures, heart rate (HR) and oxygen consumption (VO2), across high and low intensity running intervals. METHODS: Thirteen adult participants (n = 6 male; height = 174.9 ± 6.9 cm; mass = 72.5 ± 12.0 kg) were equipped with a Stryd 27 RP meter, a Polar H10 HR monitor, and a Cosmed K5 portable metabolic unit. Participants’ self-selected RP was obtained during a 10-min run on an indoor track (10 laps/mile). After resting for five minutes, participants ran another 10 min, alternating between equal intervals of RP 20% higher and 20% lower than self-selected RP: 120 s × 2, 60 s × 2, 30 s × 4, and 15 s × 8. All devices were started simultaneously before each run. RP (W/kg) was sampled at 1 Hz. HR (bpm) and VO2 (mL/kg/min) were sampled at 0.1 Hz throughout the interval run. Data were analyzed from the 60 s mark through the end of the run. HR and VO2 data were interpolated to 1 Hz, and cross correlations (max lag = 60 s) were used to compare RP, HR, and VO2 (mean values in Table 1). RESULTS: There were weak to moderate correlations between RP and VO2 (r = 0.351; lag = -29.1 s), RP and HR (r = 0.475; lag = 9.38 s), and HR and VO2 (r = 0.572; lag = -29.1 s; Table 2). CONCLUSION: HR showed the strongest correlation and smallest time delay with RP. This may be practically useful because HR data is more readily available to runners than VO2. However, the correlation is only moderate. While related, the three metrics of running intensity are fundamentally different. When exercising at a moderate intensity, changes in HR or VO2, which take seconds to minutes to stabilize, may be less evident than changes in mechanical power, which are immediate. Thus, it is possible that HR and VO2 would show a stronger relationship with RP across intervals longer than the 120 s maximum observed here. While RP can be a useful metric, it may not be informative about physiological responses to running especially over short intervals or when running at high intensity

As observers over-attend to visual information presented in the left visual field, we investigated whether the spatial arrangement of the faces in a group would influence the magnitude of the cheerleader effect. In Experiment 1, target faces were presented twice in the centre of the display: once alone, and once in a group. Group images featured two distractor faces, which were presented in either the left or the right visual field, or on either side of the target. The location of the distractor faces did not modulate the size of the cheerleader effect, which was observed in each group configuration. In Experiment 2, we manipulated the location of the target faces, which were presented at the far left, far right, or centre of the group. Faces were again significantly more attractive in each group configuration, and the spatial location of the target face did not influence the size of the cheerleader effect. Together, our results show that the cheerleader effect is a robust phenomenon, which is not influenced by the spatial arrangement of the faces in the group

    Assessing the Reliability of Stryd 27 for Variable Speed Running

    Wearable technology is beneficial when it comes to tracking and optimizing performance. The Stryd 27 is a wearable footpod marketed as being more responsive in measuring power during running than the previous version (Stryd 25). However, the reliability of this newer device to return consistent values has not been determined. PURPOSE: This study aimed to observe whether Stryd 27 gives reliable metrics during variable speed running. METHODS: Sixteen participants (N = 16; 50% female; height = 174.1 ± 8.1 cm; mass = 73.0 ± 12.4 kg) were recruited, each equipped with two Stryd 27 footpods (updated to the same software version) affixed to the shoelaces of their running shoes. The researchers recorded data using the Stryd app on a mobile device that was connected to the Stryd 27 via Bluetooth. Recording on both devices were started and stopped at the same time. Each participant completed two, 10-minute runs on an indoor track. The initial run was used to establish a baseline. Following a 5-minute rest period, participants proceeded with the second run, during which they alternated between faster and slower intervals. The pace for these intervals was set to be 20% faster and 20% slower than what each participant’s average pace was during the first run. Reliability of power, cadence, form power, ground contact time (GCT), vertical oscillation (VO), leg spring stiffness (LSS), and stride length during the interval run was determined using coefficient of variation (CV) and intraclass correlation coefficient (ICC), with CV0.70 (p \u3c 0.05) being considered evidence of reliability. RESULTS: Reliability data are shown in Table 1. The following measures were found to be reliable: power, cadence, form power, GCC, and VO. The measures of LSS and stride length were not found to be reliable. CONCLUSION: Runners using the new Stryd 27 can have confidence that most measures return reliable values (power, cadence, form power, GCT, and VO). Unfortunately, two measures were observed to not meet the threshold for reliability (LSS and stride length). Athletes interested in these measures should be cautious when interpreting their data

    Incidence and prognostic significance of hypoxemia in fibrotic interstitial lung disease: an international cohort study.

    BACKGROUND Hypoxemia is a cardinal feature of fibrotic interstitial lung disease (ILD). The incidence, progression, and prognostic significance of hypoxemia in patients with fibrotic ILD is currently unknown. RESEARCH QUESTION What are the epidemiology of hypoxemia and its additive prognostic value in current risk prediction model in fibrotic ILD? METHODS We identified 848 patients with fibrotic ILD (258 with idiopathic pulmonary fibrosis (IPF)) in five prospective ILD registries from Australia, Canada, and Switzerland. Cumulative incidence of exertional and resting hypoxemia from the time of diagnosis was estimated at 1-year intervals in patients with baseline 6-minute walk tests, adjusted for competing risks of death and lung transplantation. Likelihood ratio tests were used to determine the prognostic significance of exertional and resting hypoxemia for 1-year mortality/transplantation when added to the ILD-GAP model. The cohort was divided into derivation and validation subsets to evaluate performance characteristics of the extended model (the "ILD-GAP-O2" model), which included oxygenation status as a predictor. RESULTS The 1-, 2-, and 5-year overall cumulative incidence was 6.1%, 17.3%, and 40.1% for exertional hypoxemia, and 2.4%, 5.6%, and 16.5% for resting hypoxemia, which were significantly higher in IPF patients compared to non-IPF patients (p<0.001 for both). Addition of exertional or resting hypoxemia to the ILD-GAP model improved 1-year mortality/transplantation prediction (p<0.001 for both). The ILD-GAP-O2 model had improved discrimination (C-index of 0.80 vs 0.75) and model fit (Akaike information criteria of 400 vs 422) in the validation cohort, with comparable calibration. INTERPRETATION IPF patients have higher cumulative incidence of exertional and resting hypoxemia than non-IPF patients. The extended ILD-GAP-O2 model provides additional risk stratification for 1-year prognosis in fibrotic ILD

    Part I. SARS-CoV-2 triggered \u27PANIC\u27 attack in severe COVID-19

    The coronavirus disease 2019 (COVID-19) pandemic has produced a world-wide collapse of social and economic infrastructure, as well as constrained our freedom of movement. This respiratory tract infection is nefarious in how it targets the most distal and highly vulnerable aspect of the human bronchopulmonary tree, specifically, the delicate yet irreplaceable alveoli that are responsible for the loading of oxygen upon red cell hemoglobin for use by all of the body\u27s tissues. In most symptomatic individuals, the disease is a mild immune-mediated syndrome, with limited damage to the lung tissues. About 20% of those affected experience a disease course characterized by a cataclysmic set of immune activation responses that can culminate in the diffuse and irreversible obliteration of the distal alveoli, leading to a virtual collapse of the gas-exchange apparatus. Here, in Part I of a duology on the characterization and potential treatment for COVID-19, we define severe COVID-19 as a consequence of the ability of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) to trigger what we now designate for the first time as a ‘Prolific Activation of a Network-Immune-Inflammatory Crisis’, or ‘PANIC’ Attack, in the alveolar tree. In Part II we describe an immunotherapeutic hypothesis worthy of the organization of a randomized clinical trial in order to ascertain whether a repurposed, generic, inexpensive, and widely available agent is capable of abolishing ‘PANIC’; thereby preventing or mitigating severe COVID-19, with monumental ramifications for world health, and the global pandemic that continues to threaten it

    Part II. high-dose methotrexate with leucovorin rescue for severe COVID-19: An immune stabilization strategy for SARS-CoV-2 induced \u27PANIC\u27 attack

    Here, in Part II of a duology on the characterization and potential treatment for COVID-19, we characterize the application of an innovative treatment regimen for the prevention of the transition from mild to severe COVID-19, as well as detail an intensive immunotherapy intervention hypothesis. We propose as a putative randomized controlled trial that high-dose methotrexate with leucovorin (HDMTX-LR) rescue can abolish \u27PANIC\u27, thereby \u27left-shifting\u27 severe COVID-19 patients to the group majority of those infected with SARS-CoV-2, who are designated as having mild, even asymptomatic, disease. HDMTX-LR is endowed with broadly pleiotropic properties and is a repurposed, generic, inexpensive, and widely available agent which can be administered early in the course of severe COVID-19 thus rescuing the critical and irreplaceable gas-exchange alveoli. Further, we describe a preventative treatment intervention regimen for those designated as having mild to moderate COVID-19 disease, but who exhibit features which herald the transition to the severe variant of this disease. Both of our proposed hypothesis-driven questions should be urgently subjected to rigorous assessment in the context of randomized controlled trials, in order to confirm or refute the contention that the approaches characterized herein, are in fact capable of exerting mitigating, if not abolishing, effects upon SARS-CoV-2 triggered \u27PANIC Attack\u27. Confirmation of our immunotherapy hypothesis would have far-reaching ramifications for the current pandemic, along with yielding invaluable lessons which could be leveraged to more effectively prepare for the next challenge to global health

    Leukocyte telomere length is unrelated to cognitive performance among non-demented and demented persons: An examination of long life family study participants

    OBJECTIVE: Leukocyte telomere length (LTL) is a widely hypothesized biomarker of biological aging. Persons with shorter LTL may have a greater likelihood of developing dementia. We investigate whether LTL is associated with cognitive function, differently for individuals without cognitive impairment versus individuals with dementia or incipient dementia. METHOD: Enrolled subjects belong to the Long Life Family Study (LLFS), a multi-generational cohort study, where enrollment was predicated upon exceptional family longevity. Included subjects had valid cognitive and telomere data at baseline. Exclusion criteria were age ≤ 60 years, outlying LTL, and missing sociodemographic/clinical information. Analyses were performed using linear regression with generalized estimating equations, adjusting for sex, age, education, country, generation, and lymphocyte percentage. RESULTS: Older age and male gender were associated with shorter LTL, and LTL was significantly longer in family members than spouse controls (p \u3c 0.005). LTL was not associated with working or episodic memory, semantic processing, and information processing speed for 1613 cognitively unimpaired individuals as well as 597 individuals with dementia or incipient dementia (p \u3c 0.005), who scored significantly lower on all cognitive domains (p \u3c 0.005). CONCLUSIONS: Within this unique LLFS cohort, a group of families assembled on the basis of exceptional survival, LTL is unrelated to cognitive ability for individuals with and without cognitive impairment. LTL does not change in the context of degenerative disease for these individuals who are biologically younger than the general population
