Electrophysiological correlates of idiom comprehension: semantic composition does not follow lexical retrieval
We investigated the extent to which the literal meanings of the words forming literally-plausible idioms (e.g., break the ice) are retrieved from memory and semantically composed during sentence comprehension. Idiom strings were embedded in highly predictable, literal and idiomatic contexts. EEG data showed that the integration of the idiom\u2019s conventional meaning occurred at the end of the expression and affected the Post-N400 Positivity. Time-Frequency Representation of the EEG clarified that prior to integrating the idiom\u2019s meaning, an increase in power of higher gamma frequency band was associated only with literal processing. We argue that idioms comprehension proceeds in discrete steps: idiom recognition hampers semantic composition processes so that readers wait until the end of the expression to integrate the idiom meaning with the representation of the sentence
Identifying SARS-CoV-2 antiviral compounds by screening for small molecule inhibitors of Nsp3 papain-like protease
The COVID-19 pandemic has emerged as the biggest life-threatening disease of this century. Whilst vaccination should provide a long-term solution, this is pitted against the constant threat of mutations in the virus rendering the current vaccines less effective. Consequently, small molecule antiviral agents would be extremely useful to complement the vaccination program. The causative agent of COVID-19 is a novel coronavirus, SARS-CoV-2, which encodes at least nine enzymatic activities that all have drug targeting potential. The papain-like protease (PLpro) contained in the nsp3 protein generates viral non-structural proteins from a polyprotein precursor, and cleaves ubiquitin and ISG protein conjugates. Here we describe the expression and purification of PLpro. We developed a protease assay that was used to screen a custom compound library from which we identified dihydrotanshinone I and Ro 08-2750 as compounds that inhibit PLpro in protease and isopeptidase assays and also inhibit viral replication in cell culture-based assays
Obstructive sleep apnea and Fuhrman grade in patients with clear cell renal cell carcinoma treated surgically
PURPOSE: To assess the association between obstructive sleep apnea (OSA) and Fuhrman grade in patients with clear cell renal cell carcinoma (ccRCC). As secondary endpoints, we studied its association with tumor size, metastasis-free survival (MFS) and cancer-specific survival (CSS). METHODS: We reviewed the databases of two tertiary care centers, identifying 2579 patients who underwent partial or radical nephrectomy for ccRCC between 1991 and 2014. Descriptive statistics were used to compare pathologic variables between patients with and without OSA. Linear and logistic regression models were used to assess the association of OSA with Fuhrman grade and tumor size. A Cox proportional hazards model was used to determine OSA association with MFS and CSS. A pathway analysis was performed on a cohort with available gene expression data. RESULTS: In total, 172 patients (7 %) had self-reported OSA at diagnosis. More patients with OSA had high Fuhrman grade compared to those without OSA [51 vs. 38 %; 13 % risk difference; 95 % confidence interval (CI), 5-20 %; p = 0.003]. On multivariable analysis, the association remained significant (OR 1.41; 95 % CI 1.00-1.99; p = 0.048). OSA was not associated with tumor size (p > 0.5), MFS (p = 0.5) or CSS (p = 0.4). A trend toward vascular endothelial growth factor pathway enrichment was seen in OSA patients (p = 0.08). CONCLUSIONS: OSA is associated with high Fuhrman grade in patients undergoing surgery for ccRCC. Pending validation of this novel finding in further prospective studies, it could help shape future research to better understand etiological mechanisms associated
Identifying SARS-CoV-2 antiviral compounds by screening for small molecule inhibitors of Nsp5 main protease
The coronavirus 2019 (COVID-19) pandemic, caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), spread around the world with unprecedented health and socio-economic effects for the global population. While different vaccines are now being made available, very few antiviral drugs have been approved. The main viral protease (nsp5) of SARS-CoV-2 provides an excellent target for antivirals, due to its essential and conserved function in the viral replication cycle. We have expressed, purified and developed assays for nsp5 protease activity. We screened the nsp5 protease against a custom chemical library of over 5000 characterised pharmaceuticals. We identified calpain inhibitor I and three different peptidyl fluoromethylketones (FMK) as inhibitors of nsp5 activity in vitro, with IC(50) values in the low micromolar range. By altering the sequence of our peptidomimetic FMK inhibitors to better mimic the substrate sequence of nsp5, we generated an inhibitor with a subnanomolar IC(50). Calpain inhibitor I inhibited viral infection in monkey-derived Vero E6 cells, with an EC(50) in the low micromolar range. The most potent and commercially available peptidyl-FMK compound inhibited viral growth in Vero E6 cells to some extent, while our custom peptidyl FMK inhibitor offered a marked antiviral improvement
Basic composition and enriched integration in idiom processing: An EEG study.
We investigated the extent to which the literal meanings of the words forming literally-plausible idioms (e.g., break the ice) are semantically composed, and how the idiomatic meaning is integrated in the unfolding sentence representation. Participants read ambiguous idiom strings embedded in highly predictable, literal and idiomatic contexts, while their EEG was recorded. Control sentences only contained the idiom-final word whose cloze values were as high as in literal and idiomatic contexts. Event Related Potentials data showed that differences in the amplitude of a Frontal positivity (PNP) emerged at the beginning and at the end of the idiom strings, with the Idiomatic context condition associated with more positive voltages. The Time-Frequency analysis of the EEG showed an increase in power of the middle gamma frequency band only in the Literal context condition. These findings suggest that sentence revision mechanisms, associated with the frontal PNP, are involved in idiom meaning integration, and that the literal semantic composition of the idiomatic constituents, associated with changes in gamma frequency, is not carried out after idiom recognition
