Genistein-induced proteome changes in the human endometrial carcinoma cell line, ishikawa
Epidemiological studies have shown that Asian populations display a lower incidence of hormone-dependant cancers, cardiovascular disease, osteoporosis, and menopausal ailments compared to Western societies. Available data support the proposal that lower incidence is associated with the high dietary consumption of isoflavones, such as genistein. This study used two-dimensional electrophoresis to characterize the effect of genistein on the proteome of an endometrial tumor cell model, namely the Ishikawa cell line. Proteome maps displaying approx 1800 proteins were obtained from cells treated with vehicle or genistein at physiologically attainable concentrations of 0.5, 5, or 50 μM or supra-physiological concentration, 500 μM. The effects of genistein on protein expression were characterized using image analysis software. A total 65 protein spots displayed a significant decrease in expression and 32 proteins displayed a significant increase in expression. Of these protein spots, 29 were randomly selected for characterization by matrix assisted laser desorption/ionization tandem mass spectrometry, yielding 18 different proteins. This type of analysis enabled the characterization of a wide range of cellular proteins and allowed for the identification of functional and biochemical pathways that may be regulated or affected by genistein, including cellular transcription, cell proliferation, stress response, or modulation of oncogenic pathways.15 page(s
Observing and Promoting Normative Developmental Outcomes: Reciprocity is Key
Background: Occupational therapists focus on caregiver-infant reciprocity, which is influenced by a host of biopsychosocial factors and is predictive of developmental outcomes across domains. It is important for early intervention professions to understand how different forms of reciprocity may predict infant development in salient domains (i.e., language, mobility, and co-occupation). Therefore, the purpose of this study was to investigate associations among related measures of development in and across age, while also exploring how reciprocity influences the acquisition of developmental milestones.
Method: We examined these important areas of development in relation to novel caregiver-infant co-occupational constructs in addition to well-established domains of reciprocity (i.e., language, touch, and emotional sensitivity). In a cohort of 16 caregiver-infant dyads, we investigated infant language, motor, and affective development at 8, 12, and 16 months of age in relation to caregiver-infant reciprocity in the same domains.
Results: Findings identify relations among domains, as well as novel, bidirectional associations among these domains, and caregiver-infant reciprocity. In particular, infant utterances, standing, and positive affect were related to caregiver sensitivity and responsivity to infant affect, touch, and/or physicality.
Conclusion: These findings suggest that aspects of caregiver-infant reciprocity may predict development in several important domains
Targeting small molecule drugs to T cells with antibody-directed cell-penetrating gold nanoparticles
We sought to develop a nanoparticle vehicle that could efficiently deliver small molecule drugs to target lymphocyte populations. The synthesized amphiphilic organic ligand-protected gold nanoparticles (amph-NPs) were capable of sequestering large payloads of small molecule drugs within hydrophobic pockets of their ligand shells. These particles exhibit membrane-penetrating activity in mammalian cells, and thus enhanced uptake of a small molecule TGF-β inhibitor in T cells in cell culture. By conjugating amph-NPs with targeting antibodies or camelid-derived nanobodies, the particles' cell-penetrating properties could be temporarily suppressed, allowing targeted uptake in specific lymphocyte subpopulations. Degradation of the protein targeting moieties following particle endocytosis allowed the NPs to recover their cell-penetrating activity in situ to enter the cytoplasm of T cells. In vivo, targeted amph-NPs showed 40-fold enhanced uptake in CD8+ T cells relative to untargeted particles, and delivery of TGF-β inhibitor-loaded particles to T cells enhanced their cytokine polyfunctionality in a cancer vaccine model. Thus, this system provides a facile approach to concentrate small molecule compounds in target lymphocyte populations of interest for immunotherapy in cancer and other diseases.Massachusetts Institute of Technology. Association between active genes occurs at nuclear speckles and is modulated by chromatin environment
Genes on different chromosomes can be spatially associated in the nucleus in several transcriptional and regulatory situations; however, the functional significance of such associations remains unclear. Using human erythropoiesis as a model, we show that five cotranscribed genes, which are found on four different chromosomes, associate with each other at significant but variable frequencies. Those genes most frequently in association lie in decondensed stretches of chromatin. By replacing the mouse α-globin gene cluster in situ with its human counterpart, we demonstrate a direct effect of the regional chromatin environment on the frequency of association, whereas nascent transcription from the human α-globin gene appears unaffected. We see no evidence that cotranscribed erythroid genes associate at shared transcription foci, but we do see stochastic clustering of active genes around common nuclear SC35-enriched speckles (hence the apparent nonrandom association between genes). Thus, association between active genes may result from their location on decondensed chromatin that enables clustering around common nuclear speckles
Military deployment, masculinity and trauma : reviewing the connections
This article reviews the literature on deployment trauma and examines the limitations
of conventional understandings of trauma as they relate to veterans’ experiences.
It suggests that the failure to take into account social influences and social
relationships limits the usefulness of conventional approaches to trauma. The article
considers the role that masculinity plays in male veterans’ experience of and
sense making about trauma. It is suggested that while formal recognition of posttraumatic
stress disorder in the DSM has provided a helpful language for veterans,
it is an incomplete response. A new model of masculinity that better enables the male
veteran to speak about trauma and to reconnect with others has implications for
counselling practice with veterans
Blood pressure gradients and cardiovascular risk factors in urban and rural populations in Abia State South Eastern Nigeria using the WHO STEPwise approach
Background: Developing countries of sub-Saharan Africa (SSA) face a double burden of non-communicable diseases (NCDs)
and communicable diseases. As high blood pressure (BP) is a common global cardiovascular (CV) disorder associated with
high morbidity and mortality, the relationship between gradients of BP and other CV risk factors was assessed in Abia State,
Nigeria.
Methods: Using the WHO STEPwise approach to surveillance of chronic disease risk factors, we conducted a populationbased
cross-sectional survey in Abia state, Nigeria from August 2011 to March 2012. Data collected at various steps
included: demographic and behavioral risk factors (Step 1); BP and anthropometric measurements (Step 2), and fasting
blood cholesterol and glucose (Step 3).
Results: Of the 2983 subjects with complete data for analysis, 52.1% were females and 53.2% were rural dwellers. Overall,
the distribution of selected CV disease risk factors was diabetes (3.6%), hypertension (31.4%), cigarette smoking (13.3%), use
of smokeless tobacco (4.8%), physical inactivity (64.2%) and being overweight or obese (33.7%). Presence of hypertension,
excessive intake of alcohol, smoking (cigarette and smokeless tobacco) and physical inactivity occurred more frequently in
males than in females (p,0.05); while low income, lack of any formal education and use of smokeless tobacco were seen
more frequently in rural dwellers than in those living in urban areas (p,0.05). The frequency of selected CV risk factors
increased as BP was graded from optimal, normal to hypertension; and high BP correlated with age, gender, smokeless
tobacco, overweight or obesity, annual income and level of education.
Conclusion: Given the high prevalence of hypertension in this part of Nigeria, there is an urgent need to focus on the
reduction of preventable CV risk factors we have observed to be associated with hypertension, in order to effectively reduce

Treatment for Mild Chronic Hypertension during Pregnancy.
BACKGROUND: The benefits and safety of the treatment of mild chronic hypertension (blood pressure, \u3c160/100 mm Hg) during pregnancy are uncertain. Data are needed on whether a strategy of targeting a blood pressure of less than 140/90 mm Hg reduces the incidence of adverse pregnancy outcomes without compromising fetal growth.
METHODS: In this open-label, multicenter, randomized trial, we assigned pregnant women with mild chronic hypertension and singleton fetuses at a gestational age of less than 23 weeks to receive antihypertensive medications recommended for use in pregnancy (active-treatment group) or to receive no such treatment unless severe hypertension (systolic pressure, ≥160 mm Hg; or diastolic pressure, ≥105 mm Hg) developed (control group). The primary outcome was a composite of preeclampsia with severe features, medically indicated preterm birth at less than 35 weeks\u27 gestation, placental abruption, or fetal or neonatal death. The safety outcome was small-for-gestational-age birth weight below the 10th percentile for gestational age. Secondary outcomes included composites of serious neonatal or maternal complications, preeclampsia, and preterm birth.
RESULTS: A total of 2408 women were enrolled in the trial. The incidence of a primary-outcome event was lower in the active-treatment group than in the control group (30.2% vs. 37.0%), for an adjusted risk ratio of 0.82 (95% confidence interval [CI], 0.74 to 0.92; P
CONCLUSIONS: In pregnant women with mild chronic hypertension, a strategy of targeting a blood pressure of less than 140/90 mm Hg was associated with better pregnancy outcomes than a strategy of reserving treatment only for severe hypertension, with no increase in the risk of small-for-gestational-age birth weight. Stellar dynamics in young clusters: the formation of massive runaways and very massive runaway mergers
In the present paper we combine an N-body code that simulates the dynamics of
young dense stellar systems with a massive star evolution handler that accounts
in a realistic way for the effects of stellar wind mass loss. We discuss two
topics:
1. The formation and the evolution of very massive stars (with a mass >120
Mo) is followed in detail. These very massive stars are formed in the cluster
core as a consequence of the successive (physical) collison of 10-20 most
massive stars of the cluster (the process is known as runaway merging). The
further evolution is governed by stellar wind mass loss during core hydrogen
burning and during core helium burning (the WR phase of very massive stars).
Our simulations reveal that as a consequence of runaway merging in clusters
with solar and supersolar values, massive black holes can be formed but with a
maximum mass of 70 Mo. In small metallicity clusters however, it cannot be
excluded that the runaway merging process is responsible for pair instability
supernovae or for the formation of intermediate mass black holes with a mass of
several 100 Mo.
2. Massive runaways can be formed via the supernova explosion of one of the
components in a binary (the Blaauw scenario) or via dynamical interaction of a
single star and a binary or between two binaries in a star cluster. We explore
the possibility that the most massive runaways (e.g., zeta Pup, lambda Cep,
