    Gram-negative pathogens often use conserved type three secretion systems (T3SS) for virulence. The Shigella type three secretion apparatus (T3SA) penetrates the host cell membrane and provides a unidirectional conduit for injection of effectors into host cells. The protein Spa47 localizes to the base of the apparatus and is speculated to be an ATPase that provides the energy for T3SA formation and secretion. Here, we developed an expression and purification protocol, producing active Spa47 and providing the first direct evidence that Spa47 is a bona fide ATPase. Additionally, size exclusion chromatography and analytical ultracentrifugation identified multiple oligomeric species of Spa47 with the largest greater than 8 fold more active for ATP hydrolysis than the monomer. An ATPase inactive Spa47 point mutant was then engineered by targeting a conserved Lysine within the predicted Walker A motif of Spa47. Interestingly, the mutant maintained a similar oligomerization pattern as active Spa47, but was unable to restore invasion phenotype when used to complement a spa47 null S. flexneri strain. Together, these results identify Spa47 as a Shigella T3SS ATPase and suggest that its activity is linked to oligomerization, perhaps as a regulatory mechanism as seen in some related pathogens. Additionally, Spa47 catalyzed ATP hydrolysis appears to be essential for host cell invasion, providing a strong platform for additional studies dissecting its role in virulence and providing an attractive target for anti-infective agents

    Plural policing in Europe:relationships and governance in contemporary security system

    References to ‘plural policing’, ‘policing beyond the police’ and the ‘extended policing family’ are now commonplace in many discussions of policing in late modern societies. There is a danger that claims about the dynamic and changing nature of plural policing themselves become a new orthodoxy and begin to lose a sense of local nuance and recognition of the importance of place-based specificity and context in understanding the particularities of policing. It is this need to unpack the complex ways in which contemporary plural policing is now configured at a local level within different national political environments that provides the underpinning rationale for this Special Issue. Focussing on aspects of relationships and governance in six jurisdictions across northern and western Europe, it provides important insights into how the policies, practices and narratives around plural policing reflect the influence of particular histories and geographies. The first three articles are focused primarily on the relationships which have emerged in the public sector through its own processes of pluralisation, in particular, through the introduction of policing auxiliaries or municipal policing in Scotland, England and The Netherlands. The fourth article considers both relationships and governance in pluralised policing in Paris, France. A detailed analysis of the governance of safety and security is taken up in the final two articles, examining the cases of Austria and Belgium. These articles clearly demonstrate that experiences of pluralised policing vary widely within Europe and call into question the assumed dominance of neo-liberal forces in this area

    Using coupled micropillar compression and micro-Laue diffraction to investigate deformation mechanisms in a complex metallic alloy Al13Co4

    In this investigation, we have used in-situ micro-Laue diffraction combined with micropillar compression of focused ion beam milled Al13Co4 complex metallic alloy to study the evolution of deformation in Al13Co4. Streaking of the Laue spots showed that the onset of plastic flow occured at stresses as low as 0.8 GPa, although macroscopic yield only becomes apparent at 2 GPa. The measured misorientations, obtained from peak splitting, enabled the geometrically necessary dislocation density to be estimated as 1.1 x 1013 m-2

    Adding a Brane to the Brane-Anti-Brane Action in BSFT

    We attempt to generalize the effective action for the D-brane-anti-D-brane system obtained from boundary superstring field theory (BSFT) by adding an extra D-brane to it to obtain a covariantized action for 2 D-branes and 1 anti-D-brane. We discuss the approximations made to obtain the effective action in closed form. Among other properties, this effective action admits solitonic solutions of codimension 2 (vortices) when one of the D-brane is far separated from the brane-anti-brane pair.Comment: 23 pages, 2 figures, minor revision

    Tipping in a Low-Dimensional Model of a Tropical Cyclone

    A presumed impact of global climate change is the increase in frequency and intensity of tropical cyclones. Due to the possible destruction that occurs when tropical cyclones make landfall, understanding their formation should be of mass interest. In 2017, Kerry Emanuel modeled tropical cyclone formation by developing a low-dimensional dynamical system which couples tangential wind speed of the eye-wall with the inner-core moisture. For physically relevant parameters, this dynamical system always contains three fixed points: a stable fixed point at the origin corresponding to a non-storm state, an additional asymptotically stable fixed point corresponding to a stable storm state, and a saddle corresponding to an unstable storm state. The goal of this work is to provide insight into the underlying mechanisms that govern the formation and suppression of tropical cyclones through both analytical arguments and numerical experiments. We present a case study of both rate and noise-induced tipping between the stable states, relating to the destabilization or formation of a tropical cyclone. While the stochastic system exhibits transitions both to and from the non-storm state, noise-induced tipping is more likely to form a storm, whereas rate-induced tipping is more likely to be the way a storm is destabilized, and in fact, rate-induced tipping can never lead to the formation of a storm when acting alone. For rate-induced tipping acting as a destabilizer of the storm, a striking result is that both wind shear and maximal potential velocity have to increase, at a substantial rate, in order to effect tipping away from the active hurricane state. For storm formation through noise-induced tipping, we identify a specific direction along which the non-storm state is most likely to get activated

    Collisions of Cosmic F- and D-strings

    Recent work suggests that fundamental and Dirichlet strings, and their (p,q) bound states, may be observed as cosmic strings. The evolution of cosmic string networks, and therefore their observational signals, depends on what happens when two strings collide. We study this in string perturbation theory for collisions between all possible pairs of strings; different cases involve sphere, disk, and annulus amplitudes. The result also depends on the details of compactification; the dependence on ratios of scales is only logarithmic, but this is still numerically important. We study a range of models and parameters, and find that in most cases these strings can be distinguished from cosmic strings that arise as gauge theory solitons.Comment: 42 pages, 7 figures; v.2: added references, expanded discussion of reconnection in field theor

    Fat-Free Mass Index in a Large Sample of Collegiate American Football Athletes

    International Journal of Exercise Science 17(4): 129-139, 2024. High levels of fat-free mass (FFM) are favorable for athletes and are related to sport performance. However, fat-free mass index (FFMI), which includes adjustments for height, may offer a better way to characterize FFM beyond raw values. As FFMI is understudied relative to sport, the purpose of the current study was to assess position and age group differences in FFMI among collegiate American football players. National Collegiate Athletic Association DIII (n=111) football players underwent body composition assessment via bioelectrical impedance analysis. FFMI was calculated by dividing FFM by height squared. One-way analyses of variance with Bonferroni post-hoc tests were conducted to evaluate differences in FFMI by position and age groups (α\u3c0.05). The overall mean FFMI was 23.50 ± 2.04 kg· m−2, with values ranging from 18.1–27.7 kg· m−2. FFMI was highest in linemen (24.8 ± 1.5 kg· m−2) and lowest in specialty players (20.6 ± 1.4 kg· m−2) (p\u3c0.05). No differences in FFMI were apparent across age groups (p\u3e0.05). Current findings demonstrate that an athlete’s upper limit for FFMI may exceed 25 kg· m−2, and differences exist across positions, likely due to position-specific demands. These measurements serve as a foundation for tailoring nutritional and exercise plans, forecasting athletic performance, and supplying coaches with standardized data about the potential for additional FFM accretion in collegiate American football players

    Satellite and ground-based measurements of XCO2 in a remote semiarid region of Australia

    In this study, we present ground-based measurements of column-averaged dry-air mole fractions (DMFs) of CO2 (or XCO2) taken in a semiarid region of Australia with an EM27/SUN portable spectrometer equipped with an automated clamshell cover. We compared these measurements to space-based XCO2 retrievals from the Greenhouse Gases Observing Satellite (GOSAT). Side-by-side measurements of EM27/SUN with the Total Carbon Column Observing Network (TCCON) instrument at the University of Wollongong were conducted in 2015-2016 to derive an XCO2 scaling factor of 0.9954 relative to TCCON. Although we found a slight drift of 0.13 % over 3 months in the calibration curve of the EM27/SUN vs. TCCON XCO2, the alignment of the EM27/SUN proved stable enough for a 2-week campaign, keeping the retrieved Xair values, another measure of stability, to within 0.5 % and the modulation efficiency to within 2 %. From the measurements in Alice Springs, we confirm a small bias of around 2 ppm in the GOSAT M-gain to H-gain XCO2 retrievals, as reported by the NIES GOSAT validation team. Based on the reported random errors from GOSAT, we estimate the required duration of a future campaign in order to better understand the estimated bias between the EM27/SUN and GOSAT.     An Improved Brane Anti-Brane Action from Boundary Superstring Field Theory and Multi-Vortex Solutions

    We present an improved effective action for the D-brane-anti-D-brane system obtained from boundary superstring field theory. Although the action looks highly non-trivial, it has simple explicit multi-vortex (i.e. codimension-2 multi-BPS D-brane) multi-anti-vortex solutions. The solutions have a curious degeneracy corresponding to different ``magnetic'' fluxes at the core of each vortex. We also generalize the brane anti-brane effective action that is suitable for the study of the inflationary scenario and the production of defects in the early universe.
