    Palladium-catalyzed synthesis of pyrrole-, indole- and phenanthridine-fused polycyclic aromatic compounds from ortho-dihaloarenes

    Convenient palladium-catalyzed syntheses of polycyclic nitrogen heterocycles from ortho-dihaloarenes were developed. First, syntheses of pyrrole-fused derivatives via domino C-C/C-N coupling of ortho-dihaloarenes with imines were accomplished. Employment of ortho-dihaloarenes for sequential regioselective Suzuki reaction/double C-N coupling afforded corresponding indole-fused structures. Finally, syntheses of previously unexplored phenanthridine-fused architectures by regioselective Sonogashira reaction followed by domino or one-pot C-N coupling/hydroamination/C-H arylation were described

    The antimicrobial resistance patterns and associated determinants in Streptococcus suis isolated from humans in southern Vietnam, 1997-2008

    <p>Abstract</p> <p>Background</p> <p><it>Streptococcus suis </it>is an emerging zoonotic pathogen and is the leading cause of bacterial meningitis in adults in Vietnam. Systematic data on the antimicrobial susceptibility profiles of <it>S. suis </it>strains isolated from human cases are lacking. We studied antimicrobial resistance and associated resistance determinants in <it>S. suis </it>isolated from patients with meningitis in southern Vietnam.</p> <p>Methods</p> <p><it>S. suis </it>strains isolated between 1997 and 2008 were investigated for their susceptibility to six antimicrobial agents. Strains were screened for the presence and expression of tetracycline and erythromycin resistance determinants and the association of <it>tet</it>(M) genes with <it>Tn</it>916- like transposons. The localization of tetracycline resistance gene <it>tet</it>(L) was determined by pulse field gel electrophoresis and Southern blotting.</p> <p>Results</p> <p>We observed a significant increase in resistance to tetracycline and chloramphenicol, which was concurrent with an increase in multi-drug resistance. In tetracycline resistance strains, we identified <it>tet</it>(M), <it>tet</it>(O), <it>tet</it>(W) and <it>tet</it>(L) and confirmed their expression. All <it>tet</it>(M) genes were associated with a <it>Tn</it>916-like transposon. The co-expression of <it>tet</it>(L) and other tetracycline resistance gene(s) encoding for ribosomal protection protein(s) was only detected in strains with a minimum inhibitory concentration (MIC) of tetracycline of ≥ 64 mg/L</p> <p>Conclusions</p> <p>We demonstrated that multi-drug resistance in <it>S. suis </it>causing disease in humans in southern Vietnam has increased over the 11-year period studied. We report the presence and expression of <it>tet</it>(L) in <it>S. suis </it>strains and our data suggest that co-expression of multiple genes encoding distinct mechanism is required for an MIC ≥ 64 mg/L to tetracycline.</p

    Carotenoid producing Bacillus aquimaris found in chicken gastrointestinal tracts

    Pigmented spore-forming bacterial strains were isolated from the gastrointestinal tracts of chickens for screening for heat-stable carotenoid-producing strains that could be applied as feed supplements. Of the seven heat-stable pigmented isolates screened, only two, yellow Sporosarcina saromensis CH1 and red-orange Bacillus aquimaris CH9, produced pigments with typical carotenoid absorbance peaks (400–500 nm). The CH9 carotenoids exhibited higher scavenging activity (73.2%) of DPPH free radicals than the CH1 carotenoids (35.9%) and carotenoids of the reference B. indicus HU36 strain (78.4%), in comparison to 100% activity of acid ascorbic at 18.75 M as the standard. The CH9 strain produced high levels of carotenoids (439 g [g DW]-1) and formed nearly 100% spores, whereas the CH1 strain produced low levels of carotenoids (92 g [g DW]-1) and only achieved 30% sporulation. Chromatographic and spectral profiles of the carotenoids found in CH9 indicated the presence of as many as 11 different carotenoid types closely related to 1-HO-demethylspheroidene and keto/hydroxyl derivatives of carotene. We successfully produced concentrated orange CH9 spore powder at a high concentration of 6.1 × 1011 CFU g-1; these spores were much more heat-stable (66% survival at 80°C for 20 min) than the reference B. indicus HU36 spores (9% survival at 50°C for 20 min). In conclusion, B. aquimaris CH9 is a promising probiotic carotenoid-producing strain, with heat-stable spores that should withstand the heat-treatment processing required for feed and food supplement production

    The Vietnam Initiative on Zoonotic Infections (VIZIONS): A Strategic Approach to Studying Emerging Zoonotic Infectious Diseases

    The effect of newly emerging or re-emerging infectious diseases of zoonotic origin in human populations can be potentially catastrophic, and large-scale investigations of such diseases are highly challenging. The monitoring of emergence events is subject to ascertainment bias, whether at the level of species discovery, emerging disease events, or disease outbreaks in human populations. Disease surveillance is generally performed post hoc, driven by a response to recent events and by the availability of detection and identification technologies. Additionally, the inventory of pathogens that exist in mammalian and other reservoirs is incomplete, and identifying those with the potential to cause disease in humans is rarely possible in advance. A major step in understanding the burden and diversity of zoonotic infections, the local behavioral and demographic risks of infection, and the risk of emergence of these pathogens in human populations is to establish surveillance networks in populations that maintain regular contact with diverse animal populations, and to simultaneously characterize pathogen diversity in human and animal populations. Vietnam has been an epicenter of disease emergence over the last decade, and practices at the human/animal interface may facilitate the likelihood of spillover of zoonotic pathogens into humans. To tackle the scientific issues surrounding the origins and emergence of zoonotic infections in Vietnam, we have established The Vietnam Initiative on Zoonotic Infections (VIZIONS). This countrywide project, in which several international institutions collaborate with Vietnamese organizations, is combining clinical data, epidemiology, high-throughput sequencing, and social sciences to address relevant one-health questions. Here, we describe the primary aims of the project, the infrastructure established to address our scientific questions, and the current status of the project. Our principal objective is to develop an integrated approach to the surveillance of pathogens circulating in both human and animal populations and assess how frequently they are exchanged. This infrastructure will facilitate systematic investigations of pathogen ecology and evolution, enhance understanding of viral cross-species transmission events, and identify relevant risk factors and drivers of zoonotic disease emergence

    Preparation and Desalination Performance of PA/UiO-66/PES Composite Membranes

    UiO-66 nanoparticles are considered highly potential fillers for the application in desalination membranes. In this study, UiO-66 nanoparticles were anchored to PES membrane substrates, which were subsequently subjected to the interfacial polymerization reaction to coat a layer of polyamide (PA) on their surface. For comparison, a blank membrane incorporating no UiO-66 and a reference membrane incorporating ZrO2 (instead of UiO-66) were prepared. All prepared membranes were tested for their desalination performance. The membranes containing UiO-66 were found to outperform the blank and the reference counterparts. The reason for this outperformance is possibly attributed to the hydrophilicity of UiO-66 nanoparticles and the presence of nanochannels in their structure

    Spatiotemporal analysis and assessment of risk factors in transmission of African swine fever along the major pig value chain in Lao Cai province, Vietnam

    African swine fever (ASF) is a contagious and lethal hemorrhagic disease with a case fatality rate approaching 100% in domestic pigs. The main objectives of this study were to describe the spatiotemporal analysis as well as to assess the potential risk factors along the pig value chain in Lao Cai province, Vietnam. A total of 925 outbreaks were reported from 2019 to 2020. The three clusters (primary, secondary and 5th) were observed near the Chinese border. The most temporal clusters were detected between May and August during the study period. In addition, we evaluated the association between ASF outbreak locations to the nearest main roads and elevation. For ASF outbreak locations to the nearest main roads, compared with the reference (1,500 m), the farm locations with <500 m (55.31 times) showed a significantly increased risk of ASF outbreaks. Farmers perceived that the highest risk of ASF transmission may come from collectors and slaughterers, intermediaries inside and outside the commune, feed agents and maize agents in the commune, and pig retailers. Both commercial and household pig producers considered minimizing the number of people going in and out of pig stables and improving healthcare and husbandry procedures to be both very important and feasible. There is a need for compliance by all pig producers and other actors in the pig value chain to adopt biosecurity practices. Therefore, awareness, knowledge and understanding of infection and risks of ASF need to be improved. Veterinary officials at the provincial and district levels need to improve capacity and resources to perform laboratory analysis for ASF and need to coordinate with local actors on the control and prevention of ASF in the community

    A New Modeling Approach for Spatial Prediction of Flash Flood with Biogeography Optimized CHAID Tree Ensemble and Remote Sensing Data

    Flash floods induced by torrential rainfalls are considered one of the most dangerous natural hazards, due to their sudden occurrence and high magnitudes, which may cause huge damage to people and properties. This study proposed a novel modeling approach for spatial prediction of flash floods based on the tree intelligence-based CHAID (Chi-square Automatic Interaction Detector)random subspace, optimized by biogeography-based optimization (the CHAID-RS-BBO model), using remote sensing and geospatial data. In this proposed approach, a forest of tree intelligence was constructed through the random subspace ensemble, and, then, the swarm intelligence was employed to train and optimize the model. The Luc Yen district, located in the northwest mountainous area of Vietnam, was selected as a case study. For this circumstance, a flood inventory map with 1866 polygons for the district was prepared based on Sentinel-1 synthetic aperture radar (SAR) imagery and field surveys with handheld GPS. Then, a geospatial database with ten influencing variables (land use/land cover, soil type, lithology, river density, rainfall, topographic wetness index, elevation, slope, curvature, and aspect) was prepared. Using the inventory map and the ten explanatory variables, the CHAID-RS-BBO model was trained and verified. Various statistical metrics were used to assess the prediction capability of the proposed model. The results show that the proposed CHAID-RS-BBO model yielded the highest predictive performance, with an overall accuracy of 90% in predicting flash floods, and outperformed benchmarks (i.e., the CHAID, the J48-DT, the logistic regression, and the multilayer perception neural network (MLP-NN) models). We conclude that the proposed method can accurately estimate the spatial prediction of flash floods in tropical storm areas

    Markhacanasin C, cycloartane triterpenoid from the leaves of <i>Markhamia stipulata</i> var. <i>canaense</i> V.S. Dang

    <p>One new cycloartane triterpenoid, named markhacanasin C (<b>1</b>), together with three known triterpenoids, oleanolic acid (<b>2</b>), ursolic acid (<b>3</b>) and 6<i>β</i>,19<i>α</i>-dihydroxyursolic acid (<b>4</b>) were isolated by various chromatographic methods from the most cytotoxic fraction of the ethyl acetate extract of <i>Markhamia stipulata</i> var. <i>canaense</i> V.S. Dang leaves. Among them, <b>4</b> was reported for the first time from the genus <i>Markhamia</i>, while <b>2</b> and <b>3</b> were found for the first time from this species. Their structures were elucidated by IR, UV, HR-ESI-MS and NMR experiments. The cytotoxicity of isolated compounds (<b>3</b> and <b>4)</b> against three human cancer cell lines (HeLa, HepG2, and MCF-7) were evaluated. At the concentration of 100 μg/mL, <b>3</b> exhibited significant cytotoxic activity (86.36 ± 3.69%).</p
