    Materials Contrast in Piezoresponse Force Microscopy

    Piezoresponse Force Microscopy contrast in transversally isotropic material corresponding to the case of c+ - c- domains in tetragonal ferroelectrics is analyzed using Green's function theory by Felten et al. [J. Appl. Phys. 96, 563 (2004)]. A simplified expression for PFM signal as a linear combination of relevant piezoelectric constant are obtained. This analysis is extended to piezoelectric material of arbitrary symmetry with weak elastic and dielectric anisotropies. This result provides a framework for interpretation of PFM signals for systems with unknown or poorly known local elastic and dielectric properties, including nanocrystalline materials, ferroelectric polymers, and biopolymers.Comment: 20 pages, 3 figures, 1 table, accepted to Appl.     The relationship between workplace innovation and organizational culture: a case study of a Victorian public sector organization.

    To improve public sector innovation more understanding is needed about the impact of organizational culture on workplace innovation. This has been neglected in literature despite the public sector representing up to 30 per cent of local economies and Workplace Innovation contributing well over 30 per cent of Sector innovation. This thesis investigated the relationship between Workplace Innovation and Organizational Culture within the context of a large Victorian Public Sector Organization. Culture in this study is treated at three levels; public sector culture, organizational culture, and group (department) level culture. The thesis utilized a case study approach within a Victorian State Government Department and an explanatory sequential mixed methods approach was undertaken. A survey of 479 employees was analyzed using correlation, ANOVA, T-Tests, regression and structural equation modelling. Public Sector Culture was shown as a significant antecedent of Workplace Innovation predicting 24.6 per cent of variation and identifying significant variation in Individual Innovation, Organizational Innovation, Team Innovation and Workplace Innovation Climate. Analysis of Demographic and Employment Characteristics found considerable variation between groups. Group level culture was particularly influential in the relationships identified. Triangulation with qualitative data corroborated the findings, and a final mixed methods integration including a second triangulation of findings identified additional factors explaining the relationships including sustained organizational change that reinforced the strength of subcultures. This thesis accessed as its subject organization a Department of State, an entity which is rarely available for research. For the first time to date the results empirically identified a significant relationship between Public Sector Culture and Workplace Innovation. As an unusual and critical case the findings could be generalized to public sector theory. The findings identified significant correlations where culture was an antecedent to workplace innovation which impacted on employee’s capacity to innovate. The research extended the theory of public sector innovation by proving a relationship between Public Sector Culture and Workplace Innovation. In addition, the thesis extended the theory of Public Sector Culture highlighting its importance to Workplace Innovation and the negative impact on innovation of constant organizational change. The thesis built on the theory of public sector management identifying culture as important when managing public sector innovation. The research has implications for management of innovative public sector workplaces and cultures

    Magnetodielectric coupling in Mn3O4

    We have investigated the dielectric anomalies associated with spin ordering transitions in the tetragonal spinel Mn3_3O4_4, using thermodynamic, magnetic, and dielectric measurements. We find that two of the three magnetic ordering transitions in Mn3_3O4_4 lead to decreases in the temperature dependent dielectric constant at zero applied field. Applying a magnetic field to the polycrystalline sample leaves these two dielectric anomalies practically unchanged, but leads to an increase in the dielectric constant at the intermediate spin-ordering transition. We discuss possible origins for this magnetodielectric behavior in terms of spin-phonon coupling. Band structure calculations suggest that in its ferrimagnetic state, Mn3_3O4_4 corresponds to a semiconductor with no orbital degeneracy due to strong Jahn-Teller distortion.Comment: 6 pages, 7 figure

    Ipl1/Aurora B kinase coordinates synaptonemal complex disassembly with cell cycle progression and crossover formation in budding yeast meiosis

    Several protein kinases collaborate to orchestrate and integrate cellular and chromosomal events at the G2/M transition in both mitotic and meiotic cells. During the G2/M transition in meiosis, this includes the completion of crossover recombination, spindle formation, and synaptonemal complex (SC) breakdown. We identified Ipl1/Aurora B kinase as the main regulator of SC disassembly. Mutants lacking Ipl1 or its kinase activity assemble SCs with normal timing, but fail to dissociate the central element component Zip1, as well as its binding partner, Smt3/SUMO, from chromosomes in a timely fashion. Moreover, lack of Ipl1 activity causes delayed SC disassembly in a cdc5 as well as a CDC5-inducible ndt80 mutant. Crossover levels in the ipl1 mutant are similar to those observed in wild type, indicating that full SC disassembly is not a prerequisite for joint molecule resolution and subsequent crossover formation. Moreover, expression of meiosis I and meiosis II-specific B-type cyclins occur normally in ipl1 mutants, despite delayed formation of anaphase I spindles. These observations suggest that Ipl1 coordinates changes to meiotic chromosome structure with resolution of crossovers and cell cycle progression at the end of meiotic prophase

    Optimizing DNA Extraction Methods for Nanopore Sequencing of Neisseria gonorrhoeae Directly from Urine Samples

    Empirical gonorrhea treatment at initial diagnosis reduces onward transmission. However, increasing resistance to multiple antibiotics may necessitate waiting for culture-based diagnostics to select an effective treatment. There is a need for same-day culture-free diagnostics that identify infection and detect antimicrobial resistance. We investigated if Nanopore sequencing can detect sufficient Neisseria gonorrhoeae DNA to reconstruct whole genomes directly from urine samples. We used N. gonorrhoeae-spiked urine samples and samples from gonorrhea infections to determine optimal DNA extraction methods that maximize the amount of N. gonorrhoeae DNA sequenced while minimizing contaminating host DNA. In simulated infections, the Qiagen UCP pathogen mini kit provided the highest ratio of N. gonorrhoeae to human DNA and the most consistent results. Depletion of human DNA with saponin increased N. gonorrhoeae yields in simulated infections but decreased yields in clinical samples. In 10 urine samples from men with symptomatic urethral gonorrhea, ≥92.8% coverage of an N. gonorrhoeae reference genome was achieved in all samples, with ≥93.8% coverage breath at ≥10-fold depth in 7 (70%) samples. In simulated infections, if ≥104 CFU/ml of N. gonorrhoeae was present, sequencing of the large majority of the genome was frequently achieved. N. gonorrhoeae could also be detected from urine in cobas PCR medium tubes and from urethral swabs and in the presence of simulated Chlamydia coinfection. Using Nanopore sequencing of urine samples from men with urethral gonorrhea, sufficient data can be obtained to reconstruct whole genomes in the majority of samples without the need for culture

    Solution structure of a bacterial microcompartment targeting peptide and its application in the construction of an ethanol bioreactor

    Targeting of proteins to bacterial microcompartments (BMCs) is mediated by an 18-amino-acid peptide sequence. Herein, we report the solution structure of the N-terminal targeting peptide (P18) of PduP, the aldehyde dehydrogenase associated with the 1,2-propanediol utilization metabolosome from Citrobacter freundii. The solution structure reveals the peptide to have a well-defined helical conformation along its whole length. Saturation transfer difference and transferred NOE NMR has highlighted the observed interaction surface on the peptide with its main interacting shell protein, PduK. By tagging both a pyruvate decarboxylase and an alcohol dehydrogenase with targeting peptides, it has been possible to direct these enzymes to empty BMCs in vivo and to generate an ethanol bioreactor. Not only are the purified, redesigned BMCs able to transform pyruvate into ethanol efficiently, but the strains containing the modified BMCs produce elevated levels of alcohol

    Antarctica on foot: the energy expended to walk, ski and man-haul

    Polar exploration often involves travelling on foot and thus is physically intensive, with long-term excursions typically resulting in weight loss. Few studies have investigated the energy expended under such circumstances. Here, we present a range of prediction equations for estimating metabolic rate from heart rate or accelerometry data for specific activities including skiing and man-hauling which can be applied to either short- or long-term excursions. We also use some of these equations to estimate the energy expended undertaking various activities by a team of explorers while attempting to traverse the Antarctic continent during the austral winter of 2013 (as part of the White Mars Project during The Coldest Journey). Calibration equations based on either accelerometry data (from which overall dynamic body acceleration, ODBA, is derived) or heart rate showed good relationships with rate of oxygen consumption, particularly when person height was included. Periods of skiing and man- hauling on The Coldest Journey were estimated to be more energetically demanding (30.0 and 31.1 kJ min−1, respectively) than walking (24.9 kJ min−1), or other outdoor work (21.9 kJ min−1). Estimates of energy expenditure during The Coldest Journey were similar to measures obtained in previous, comparative scenarios. We hope that future expeditions to Antarctica will use these prediction equations to further our understanding of the energy costs of exploring Antarctica and the nutritional requirements needed to guard against emaciation

    Late Holocene palynology and palaeovegetation of tephra-bearing mires at Papamoa and Waihi Beach, western Bay of Plenty, North Island, New Zealand.

    The vegetation history of two mires associated with Holocene dunes near the western Bay of Plenty coast, North Island, New Zealand, is deduced from pollen analysis of two cores. Correlation of airfall tephra layers in the peats, and radiocarbon dates, indicate that the mires at Papamoa and Waihi Beach are c. 4600 and c. 2900 conventional radiocarbon years old, respectively. Tephras used to constrain the chronology of the pollen record include Rotomahana (1886 AD), Kaharoa (700 yr B.P.), Taupo (Unit Y; 1850 yr B.P.), Whakaipo (Unit V; 2700 yr B.P.), Stent (Unit Q; 4000 yr B.P.), Hinemaiaia (Unit K; 4600 yr B.P.), and reworked Whakatane (c. 4800 yr B.P.) at Papamoa, and Kaharoa and Taupo at Waihi Beach. Peat accumulation rates at Papamoa from 4600 - 1850 yr B.P. range from 0.94 to 2.64 mm/yr (mean 1.37 mm/yr). At Waihi Beach, from 2900 yr B.P. - present day, they range from 0.11 to 0.21 mm/yr (mean 0.20 mm/yr). Peat accumulation at both sites was slowest from 1850 to 700 yr B.P., suggesting a drier overall climate during this interval. At both sites, the earliest organic sediments, which are underlain by marine or estuarine sands, yield pollen spectra indicating salt marsh or estuarine environments. Coastal vegetation communities declined at both sites, as sea level gradually fell or the coast prograded, and were eventually superseded by a low moor bog at Papamoa, and a mesotrophic swamp forest at Waihi Beach. These differences, and the marked variation in peat accumulation rates, probably reflect local hydrology and are unlikely to have been climatically controlled. The main regional vegetation during this period was mixed northern conifer-angiosperm forest. Kauri (Agathis australis) formed a minor component of these forests, but populations of this tree have apparently not expanded during the late Holocene at these sites, which are near its present southern limit. Occasional shortlived forest disturbances are detectable in these records, in particular immediately following the deposition of Taupo Tephra. However, evidence for forest clearance during the human era is blurred by the downward dislocation of modern adventi ve pollen at these sites, preventing the clear differentiation of the Polynesian and European eras
