    Embedded contact homology of prequantization bundles

    The 2011 PhD thesis of Farris demonstrated that the ECH of a prequantization bundle over a Riemann surface is isomorphic as a Z/2Z-graded group to the exterior algebra of the homology of its base. We extend this result by computing the Z-grading on the chain complex, permitting a finer understanding of this isomorphism. We fill in some technical details, including the Morse-Bott direct limit argument and some writhe bounds. The former requires the isomorphism between filtered Seiberg-Witten Floer cohomology and filtered ECH as established by Hutchings--Taubes. The latter requires the work on higher asymptotics of pseudoholomorphic curves by Cristofaro-Gardiner--Hutchings--Zhang. In a subsequent paper, we plan to extend these methods for Seifert fiber spaces and relate the ECH U-map to orbifold Gromov-Witten invariants of the base.Comment: 85 page

    Torus knot filtered embedded contact homology of the tight contact 3-sphere

    Knot filtered embedded contact homology was first introduced by Hutchings in 2015; it has been computed for the standard transverse unknot in irrational ellipsoids by Hutchings and for the Hopf link in lens spaces L(n,n-1) via a quotient by Weiler. While straightforward toric constructions can be used to understand the ECH chain complexes of open books along the unknot and Hopf link, they do not readily adapt to general torus knots and links. In this paper, we generalize the definition and invariance of knot filtered embedded contact homology to allow for degenerate knots with rational rotation numbers. We then develop new methods for understanding the embedded contact homology chain complex of positive torus knotted fibrations of the standard tight contact 3-sphere in terms of their presentation as open books and as Seifert fibered spaces. We provide Morse-Bott methods, using a doubly filtered complex and the energy filtered perturbed Seiberg-Witten theory developed by Hutchings and Taubes, and use them to compute the T(2,q) knot filtered embedded contact homology, for q odd and positive. In the sequel we complete the computation for positive T(p,q) knots (where there is a nonvanishing differential) and use our results to deduce quantitative existence results for torus knotted Reeb dynamics on the tight 3-sphere and the mean action of area preserving diffeomorphisms of once punctured surfaces of arbitrary genus arising as Seifert surfaces of positive torus knots.Comment: 85 pages, arXiv insists the primary is GT rather than S

The program was designed to address the needs of women with high-risk pregnancy and focused on mental health, physical health, energy conservation, emotional regulation, and social participation.     Size matters: An empirical study of neural network training for large vocabulary continuous speech recognition

    We have trained and tested a number of large neural networks for the purpose of emission probability estimation in large vocabulary continuous speech recognition. In particular, the problem under test is the DARPA Broadcast News task. Our goal here was to determine the relationship between training time, word error rate, size of the training set, and size of the neural network. In all cases, the network architecture was quite simple, comprising a single large hidden layer with an input window consisting of feature vectors from 9 frames around the current time, with a single output for each of 54 phonetic categories. Thus far, simultaneous increases to the size of the training set and the neural network improve performance; in other words, more data helps, as does the training of more parameters. We continue to be surprised that such a simple system works as well as it does for complex tasks. Given a limitation in training time, however, there appears to be an optimal ratio of training patterns to parameters of around 25:1 in these circumstances. Additionally, doubling the training data and system size appears to provide diminishing returns of error rate reduction for the largest systems

    Neuronal ceroid lipofuscinoses (NCLs; also known collectively as Batten Disease) are a family of autosomal recessive lysosomal storage disorders. Mutations in as many as 13 genes give rise to ∌10 variants of NCL, all with overlapping clinical symptomatology including visual impairment, motor and cognitive dysfunction, seizures, and premature death. Mutations in CLN6 result in both a variant late infantile onset neuronal ceroid lipofuscinosis (vLINCL) as well as an adult-onset form of the disease called Type A Kufs. CLN6 is a non-glycosylated membrane protein of unknown function localized to the endoplasmic reticulum (ER). In this study, we perform a detailed characterization of a naturally occurring Cln6 mutant (Cln6(nclf)) mouse line to validate its utility for translational research. We demonstrate that this Cln6(nclf) mutation leads to deficits in motor coordination, vision, memory, and learning. Pathologically, we demonstrate loss of neurons within specific subregions and lamina of the cortex that correlate to behavioral phenotypes. As in other NCL models, this model displays selective loss of GABAergic interneuron sub-populations in the cortex and the hippocampus with profound, early-onset glial activation. Finally, we demonstrate a novel deficit in memory and learning, including a dramatic reduction in dendritic spine density in the cerebral cortex, which suggests a reduction in synaptic strength following disruption in CLN6. Together, these findings highlight the behavioral and pathological similarities between the Cln6(nclf) mouse model and human NCL patients, validating this model as a reliable format for screening potential therapeutics

    Crystallization and preliminary X-ray analysis of the Plasmodium falciparum apicoplast DNA polymerase

    Infection by the parasite Plasmodium falciparum is the leading cause of malaria in humans. The parasite has a unique and essential plastid-like organelle called the apicoplast. The apicoplast contains a genome that undergoes replication and repair through the action of a replicative polymerase (apPOL). apPOL has no direct orthologs in mammalian polymerases and is therefore an attractive antimalarial drug target. No structural information exists for apPOL, and the Klenow fragment of Escherichia coli DNA polymerase I, which is its closest structural homolog, shares only 28% sequence identity. Here, conditions for the crystallization of and preliminary X-ray diffraction data from crystals of P. falciparum apPOL are reported. Data complete to 3.5 Å resolution were collected from a single crystal (2 × 2 × 5 ”m) using a 5 ”m beam. The space group P6522 (unit-cell parameters a = b = 141.8, c = 149.7 Å, α = ÎČ = 90, Îł = 120°) was confirmed by molecular replacement. Refinement is in progress

    “It still haunts me whether we did the right thing”: a qualitative analysis of free text survey data on the bereavement experiences and support needs of family caregivers

    Background Research suggests that there may be bereavement experiences and support needs which are specific to family caregivers providing end of life care (EoLC), although this remains an under-researched area. This paper focuses on themes relating to bereavement which were derived from an analysis of free text survey responses collected in a research priority setting exercise for palliative and EoLC. Methods The priority setting exercise involved a public survey, designed to generate research priorities. Rather than identify research topics, many people instead described their experiences and raised more general questions relating to palliative and end of life care. To explore these experiences and perspectives a supplementary thematic analysis was conducted on the survey responses. 1403 respondents took part, including patients, current and bereaved carers, health and social care professionals, volunteers and members of the public. Results Several grief issues were identified, which seem specific to the experiences of family caregivers. Responses demonstrated a relationship between death experiences, feelings of guilt and bereavement outcomes for some family caregivers, as well as caregiver experiences of a “void” created by the withdrawal of professional support after death. Communication and support needs were also identified by participants. Conclusion This analysis provides further evidence of some of the specific effects that caring for a loved one at the end of life can have on bereavement experiences. Finding ways of improving communication around the time of death and effective follow up approaches post death could help to address some of these issues
