    Enraizamento de estacas de Luehea divaricata sob diferentes concentrações de ácido indolbutírico.

    Apesar da disponibilidade de sementes em plantas matrizes de Luehea divaricata, sua coleta é uma tarefa trabalhosa e onerosa, visto a maturação ser irregular ao longo do tempo, gerando dificuldades para produção de mudas por meio de sementes. Com base nisto, objetivou-se avaliar a concentração mais adequada de ácido indolbutírico (AIB) para a produção de mudas via estaquia de Luehea divaricata. As estacas foram obtidas de ramos de árvores adultas e regenerações naturais na primavera de 2003, preparadas com 10 cm a 15 cm, um par de folhas cortadas ao meio, tratadas com AIB (0, 2000, 4000, 6000 e 8000 mg L-1) e enraizadas em casa de vegetação por um período de 85 dias. A sobrevivência média na saída da casa de sombra foi de 29,2 %, com superioridade das estacas tratadas com 4.000 mg L-1 de AIB (42,5 %) e, a pleno sol, média de 19,8 % de sobrevivência, com superioridade das estacas tratadas com 2.000 mg L-1 de AIB (26,5 %).     Embryonic dormancy in seeds of Bactris gasipaes Kunth (peach-palm)

    Bactris gasipaes is a domesticated palm whose fruits are of great importance for the Amazonian people and whose heart of palm is also receiving economic interest in other brazilian and Latin America regions. The aim of this study was verify embryonic dormancy and its correlation with first cataphyll emergence in B. gasipaes seeds collected from four plants at Manaus city and four others at Coari city, both in the Amazonas state, Brazil. After extraction and cleaning, some of the seeds (4 replications of 25 per plant) were sown in a seedbed with a sawdust and sand mixture as substrate, and embryos (4 replications of 10 per plant), after extraction, were inoculated into half strength Murashige and Skoog cultures. Were used 100 seeds and 40 embryo per treatment. Whole seed and embryo germination varied between the different source plants and locations, with the greatest difference observed for the emergence of first cataphyll from seeds in the seedbed. For the most part of variables, results of seed and embryo were positively associated, namely, as one went up the other also, and vice versa. These results suggesting that, at least in part, seed dormancy in Bactris gasipaes is associated with embryonic dormancy. © 2017, Associacao Brasileira de Tecnologia de Sementes. All rights reserved

    Cathepsin K induces platelet dysfunction and affects cell signaling in breast cancer - molecularly distinct behavior of cathepsin K in breast cancer

    BACKGROUND: Breast cancer comprises clinically and molecularly distinct tumor subgroups that differ in cell histology and biology and show divergent clinical phenotypes that impede phase III trials, such as those utilizing cathepsin K inhibitors. Here we correlate the epithelial-mesenchymal-like transition breast cancer cells and cathepsin K secretion with activation and aggregation of platelets. Cathepsin K is up-regulated in cancer cells that proteolyze extracellular matrix and contributes to invasiveness. Although proteolytically activated receptors (PARs) are activated by proteases, the direct interaction of cysteine cathepsins with PARs is poorly understood. In human platelets, PAR-1 and −4 are highly expressed, but PAR-3 shows low expression and unclear functions. METHODS: Platelet aggregation was monitored by measuring changes in turbidity. Platelets were immunoblotted with anti-phospho and total p38, Src-Tyr-416, FAK-Tyr-397, and TGFβ monoclonal antibody. Activation was measured in a flow cytometer and calcium mobilization in a confocal microscope. Mammary epithelial cells were prepared from the primary breast cancer samples of 15 women with Luminal-B subtype to produce primary cells. RESULTS: We demonstrate that platelets are aggregated by cathepsin K in a dose-dependent manner, but not by other cysteine cathepsins. PARs-3 and −4 were confirmed as the cathepsin K target by immunodetection and specific antagonists using a fibroblast cell line derived from PARs deficient mice. Moreover, through co-culture experiments, we show that platelets activated by cathepsin K mediated the up-regulation of SHH, PTHrP, OPN, and TGFβ in epithelial-mesenchymal-like cells from patients with Luminal B breast cancer. CONCLUSIONS: Cathepsin K induces platelet dysfunction and affects signaling in breast cancer cells.     Characterization of buriti (Mauritia flexuosa) pulp oil and the effect of its supplementation in an In vivo experimental model.

    Mauritia flexuosa (buriti) pulp oil contains bioactive substances and lipids that are protective against cardiovascular and inflammatory diseases. We performed physical and chemical analyses to verify its quality and stability. buriti oil was stable according to the Rancimat test, presenting an induction period of 6.6 h. We evaluated the effect of supplementation with crude buriti oil and olive oil on metabolic parameters in 108 Swiss mice for 90 days. We investigated six groups: extra virgin olive oil (EVOO) 1 and 2 (1000 and 2000 mg/kg), buriti oil (BO) 1 and 2 (1000 and 2000 mg/kg), synergic (S) (BO1 + EVOO1), and control (water dose 1000 mg/kg). The animals were euthanized to examine their blood, livers, and fats. The supplementation did not interfere with food consumption, weight gain, and histological alterations in the liver. Group S showed the strongest relationship with the fractions HDL-c and non-HDL-c, indicating a possible cardioprotective effect. Moreover, we observed significantly higher IL-6 levels in the control, EVOO2, and BO1 groups than in the EVOO1 group. Resistin was also significantly higher for the synergic treatment than for the control. We conclude that BO combined with EVOO could be an excellent food supplement for human consumption
