    High-voltage pulse discharge as factor of the methanogenesis initiation

    Results of scientific research on influence of the high-voltage pulse discharge on process of bioorganic waste methane sludge fermentation in agricultural production are given in the article. Powerful infra- and ultrasonic fluctuations leading to emergence of shock waves exert strong impact on processes: disinfecting, cleanings and deflocculation of bioorganic mix, as well as on activity of various physical and chemical changes of synthesis products. Selecting the modes of high-voltage pulse processing, it is possible to provide highly productive anaerobic bacteria. At the same time "artificial selection" as a result of which weak microorganisms perish is observed, and the strong group remains. Viable species of microorganisms, having received at the order nutrient medium, as a result of destruction and death of weak microorganisms, begin to breed quickly, increasing growth of a biofilm responsible for a biogas exit. For the analysis of experimental data on development of technological process for anaerobic sludge fermentation the method of trans-resonant functional topography which provided informational content of the active environment concerning ions and free radicals arising in the course of modifying the organic substratum was used.Keywords: biogas unit, anaerobic sludge fermentation, electro technology, methanogenesis,pulse discharge, bioorganic waste, water substratum, organic fertilizers, processing, ecology

    Visual Analytics for Epidemiologists: Understanding the Interactions Between Age, Time, and Disease with Multi-Panel Graphs

    Visual analytics, a technique aiding data analysis and decision making, is a novel tool that allows for a better understanding of the context of complex systems. Public health professionals can greatly benefit from this technique since context is integral in disease monitoring and biosurveillance. We propose a graphical tool that can reveal the distribution of an outcome by time and age simultaneously.We introduce and demonstrate multi-panel (MP) graphs applied in four different settings: U.S. national influenza-associated and salmonellosis-associated hospitalizations among the older adult population (≥65 years old), 1991-2004; confirmed salmonellosis cases reported to the Massachusetts Department of Public Health for the general population, 2004-2005; and asthma-associated hospital visits for children aged 0-18 at Milwaukee Children's Hospital of Wisconsin, 1997-2006. We illustrate trends and anomalies that otherwise would be obscured by traditional visualization techniques such as case pyramids and time-series plots.MP graphs can weave together two vital dynamics--temporality and demographics--that play important roles in the distribution and spread of diseases, making these graphs a powerful tool for public health and disease biosurveillance efforts

    Nonlinear relativistic optics in the single cycle, single wavelength regime and kilohertz repetition rate

    Pulses of few optical cycles, focused on one wavelength with relativistic intensities can be produced at a kilohertz repetition rate.     Since the term “thoracic outlet syndrome” (TOS) has been introduced, there have been disputes about the accuracy of the diagnosis, definition, diagnostic workup and treatment of this condition. Existing  clinical variants include involvement of blood vessels and nerve fibers. Neurogenic TOS, consisting of the brachial plexus compression, is the most common variant of TOS, which is also the most hardly proven  by physical findings, imaging or electrodiagnostic studies. A long history of searching the “gold standard” for TOS diagnosis has led to a  large number of publications. The current paper reviews the symptoms and diagnostic approaches to the neurogenic TOS.С тех пор как был введен термин «синдром верхней апертуры грудной клетки» (thoracic outlet syndrome, TOS), не прекращаются споры о правомерности такого диагноза, его дефинициях,  возможной диагностике и лечении. Заинтересованность разных анатомических структур  обусловливает многообразие клинических вариантов. Факт поражения плечевого сплетения  при наиболее частом варианте болезни – нейрогенном – редко удается подтвердить  инструментально. За 60 лет поисков «золотого стандарта» диагностики TOS накопилось  большое количество публикаций. В обзоре уделено внимание клиническим проявлениям  нейрогенного варианта, а также рассмотрены методы инструментальной диагностики этой патологии

    Modulation of enhancer looping and differential gene targeting by Epstein-Barr virus transcription factors directs cellular reprogramming

    Epstein-Barr virus (EBV) epigenetically reprogrammes B-lymphocytes to drive immortalization and facilitate viral persistence. Host-cell transcription is perturbed principally through the actions of EBV EBNA 2, 3A, 3B and 3C, with cellular genes deregulated by specific combinations of these EBNAs through unknown mechanisms. Comparing human genome binding by these viral transcription factors, we discovered that 25% of binding sites were shared by EBNA 2 and the EBNA 3s and were located predominantly in enhancers. Moreover, 80% of potential EBNA 3A, 3B or 3C target genes were also targeted by EBNA 2, implicating extensive interplay between EBNA 2 and 3 proteins in cellular reprogramming. Investigating shared enhancer sites neighbouring two new targets (WEE1 and CTBP2) we discovered that EBNA 3 proteins repress transcription by modulating enhancer-promoter loop formation to establish repressive chromatin hubs or prevent assembly of active hubs. Re-ChIP analysis revealed that EBNA 2 and 3 proteins do not bind simultaneously at shared sites but compete for binding thereby modulating enhancer-promoter interactions. At an EBNA 3-only intergenic enhancer site between ADAM28 and ADAMDEC1 EBNA 3C was also able to independently direct epigenetic repression of both genes through enhancer-promoter looping. Significantly, studying shared or unique EBNA 3 binding sites at WEE1, CTBP2, ITGAL (LFA-1 alpha chain), BCL2L11 (Bim) and the ADAMs, we also discovered that different sets of EBNA 3 proteins bind regulatory elements in a gene and cell-type specific manner. Binding profiles correlated with the effects of individual EBNA 3 proteins on the expression of these genes, providing a molecular basis for the targeting of different sets of cellular genes by the EBNA 3s. Our results therefore highlight the influence of the genomic and cellular context in determining the specificity of gene deregulation by EBV and provide a paradigm for host-cell reprogramming through modulation of enhancer-promoter interactions by viral transcription factors

    A protocol for a systematic literature review: comparing the impact of seasonal and meteorological parameters on acute respiratory infections in Indigenous and non-Indigenous peoples

    Background: Acute respiratory infections (ARI) are a leading cause of morbidity and mortality globally, and are often linked to seasonal and/or meteorological conditions. Globally, Indigenous peoples may experience a different burden of ARI compared to non-Indigenous peoples. This protocol outlines our process for conducting a systematic review to investigate whether associations between ARI and seasonal or meteorological parameters differ between Indigenous and non-Indigenous groups residing in the same geographical region. Methodology: A search string will be used to search PubMed®, CAB Abstracts/CAB Direct©, and Science Citation Index® aggregator databases. Articles will be screened using inclusion/exclusion criteria applied first at the title and abstract level, and then at the full article level by two independent reviewers. Articles maintained after full article screening will undergo risk of bias assessment and data will be extracted. Heterogeneity tests, meta-analysis, and forest and funnel plots will be used to synthesize the results of eligible studies. Discussion and registration: This protocol paper describes our systematic review methods to identify and analyze relevant ARI, season, and meteorological literature with robust reporting. The results are intended to improve our understanding of potential associations between seasonal and meteorological parameters and ARI and, if identified, whether this association varies by place, population, or other characteristics. The protocol is registered in the PROSPERO database (#38051)

    Principles of meiotic chromosome assembly revealed in S. cerevisiae

    During meiotic prophase, chromosomes organise into a series of chromatin loops emanating from a proteinaceous axis, but the mechanisms of assembly remain unclear. Here we use Saccharomyces cerevisiae to explore how this elaborate three-dimensional chromosome organisation is linked to genomic sequence. As cells enter meiosis, we observe that strong cohesin-dependent grid-like Hi-C interaction patterns emerge, reminiscent of mammalian interphase organisation, but with distinct regulation. Meiotic patterns agree with simulations of loop extrusion with growth limited by barriers, in which a heterogeneous population of expanding loops develop along the chromosome. Importantly, CTCF, the factor that imposes similar features in mammalian interphase, is absent in S. cerevisiae, suggesting alternative mechanisms of barrier formation. While grid-like interactions emerge independently of meiotic chromosome synapsis, synapsis itself generates additional compaction that matures differentially according to telomere proximity and chromosome size. Collectively, our results elucidate fundamental principles of chromosome assembly and demonstrate the essential role of cohesin within this evolutionarily conserved process
