The purpose of this article is to provide an overview of nursing research in Latin America during the decade from 1988 to 1998. Data from the bi-annual Pan American Colloquia in Nursing Research from 1988 to 1998 were subject to secondary analysis. Findings indicate that most of the research emanated from Brazil, the only country with a doctoral program in nursing in the 1990's. Research topics included: public health issues, clinical studies (usually of adults), nursing care studies of process, therapeutic communication, and administrative issues such as standards of care and quality. The most common design was descriptive quantitative, although there were several qualitative studies. The analysis provides directions for future research and indicates areas of concern, especially the need for theory based nursing research. There is a possible accelerated biological aging in patients with substance use disorders (SUD). The evaluation of epigenetic clocks, which are accurate estimators of biological aging based on DNA methylation changes, has been limited to blood tissue in patients with SUD. Consequently, the impact of biological aging in the brain of individuals with SUD remains unknown. In this study, we evaluated multiple epigenetic clocks (DNAmAge, DNAmAgeHannum, DNAmAgeSkinBlood, DNAmPhenoAge, DNAmGrimAge, and DNAmTL) in individuals with SUD (n = 42), including alcohol (n = 10), opioid (n = 19), and stimulant use disorder (n = 13), and controls (n = 10) in postmortem brain (prefrontal cortex) and blood tissue obtained from the same individuals. We found a higher DNAmPhenoAge (β = 0.191, p-value = 0.0104) and a nominally lower DNAmTL (β = -0.149, p-value = 0.0603) in blood from individuals with SUD compared to controls. SUD subgroup analysis showed a nominally lower brain DNAmTL in subjects with alcohol use disorder, compared to stimulant use disorder and controls (β = 0.0150, p-value = 0.087). Cross-tissue analyzes indicated a lower blood DNAmTL and a higher blood DNAmAge compared to their respective brain values in the SUD group. This study highlights the relevance of tissue specificity in biological aging studies and suggests that peripheral measures of epigenetic clocks in SUD may depend on the specific type of drug used

The pyruvate kinase (PK) and ATPase activities taking part in nitrogen (N) assimilation is essential for the growth and development of plants. Studies on the kinetics of these enzymes reveal that its activities are dependent of the cofactors K+, Ca2+, and Mg2+. Therefore, the objective of the present work was to determine the effect of different doses of N on enzymatic activities of ATPase and PK as potentials biochemical indicators of the levels of K+, Mg2+, and Ca2+ in the roots and leaves of green bean plants. The N was applied to the nutrient solution as NH4NO3 at the following rates: 1.5, 3.0, 6.0, 12.0, 18.0, and 24.0 mM of N. These results indicate that deficient conditions of N (N1 and N2) were characterized by the lowest accumulation of K+, Mg2+ and Ca2+ in both total and soluble forms, and also minimum activities of PK and ATPase induced by K+, Mg2+ and Ca2+, with respect to the activity of basal PK and ATPase; this could mean near optimum conditions for these cations. On the contrary, high-N treatments (N4, N5 and N6) were characterized by presenting decreasing concentrations of total and soluble K+, Mg2+ and Ca2+ in roots and leaves of green bean plants; however, the activities of PK and ATPase induced with K+, Mg2+ and Ca2+ were increased reaching their maximum activity with respect to basal PK and ATPase, both enzymes reflecting the level of cations in roots and leaves, hence being considered as good physiological bioindicators of these cations.

A Human Stem Cell-Derived Neuronal Model of Morphine Exposure Reflects Brain Dysregulation in Opioid Use Disorder: Transcriptomic and Epigenetic Characterization of Postmortem-Derived iPSC Neurons

    Get PDF
INTRODUCTION: Human-derived induced pluripotent stem cell (iPSC) models of brain promise to advance our understanding of neurotoxic consequences of drug use. However, how well these models recapitulate the actual genomic landscape and cell function, as well as the drug-induced alterations, remains to be established. New METHODS: We engineered a novel induced pluripotent stem cell-derived model of neural progenitor cells and neurons from cultured postmortem human skin fibroblasts, and directly compared these to isogenic brain tissue from the donor source. We assessed the maturity of the cell models across differentiation from stem cells to neurons using RNA cell type and maturity deconvolution analyses as well as DNA methylation epigenetic clocks trained on adult and fetal human tissue. As proof-of-concept of this model\u27s utility for substance use disorder studies, we compared morphine- and cocaine-treated neurons to gene expression signatures in postmortem Opioid Use Disorder (OUD) and Cocaine Use Disorder (CUD) brains, respectively. RESULTS: Within each human subject (N = 2, 2 clones each), brain frontal cortex epigenetic age parallels that of skin fibroblasts and closely approximates the donor\u27s chronological age; stem cell induction from fibroblast cells effectively sets the epigenetic clock to an embryonic age; and differentiation of stem cells to neural progenitor cells and then to neurons progressively matures the cells DISCUSSION: In summary, we introduce an iPSC model generated from human postmortem fibroblasts that can be directly compared to corresponding isogenic brain tissue and can be used to model perturbagen exposure such as that seen in opioid use disorder. Future studies with this and other postmortem-derived brain cellular models, including cerebral organoids, can be an invaluable tool for understanding mechanisms of drug-induced brain alterations

Evaluation of a Brief Marriage Intervention for Internal Behavioral Health Consultants in Military Primary Care

    Get PDF
Military couples face significant challenges to their relationships including demanding schedules, multiple deployments, and frequent moves. Despite the high costs of chronic marital distress, very few military (or civilian) couples seek marriage therapy. The military services and the VA system have implemented collaborative care models in primary care where internal behavioral health consultants are integrated into primary care. Integrated primary care can reduce the stigma of behavioral health services and may increase the odds that couples would seek help earlier. There are no established couple interventions designed for use in primary care. The purpose of this presentation is to describe a program of research focused on adapting and validating The Marriage Checkup (MC) for use in an integrated primary care clinic.https://corescholar.libraries.wright.edu/urop_celebration/1019/thumbnail.jp
