Detection of shrimp viral pathogens with a simple dot-blot enzyme immunoassay protocol
Dot-blot nitrocellulose enzyme immunoassays (DB-NC-EIA) were developed for the detection of two shrimp viral pathogens, white-spot virus (WSV), and yellow-head virus (YHV). The assays utilized HRP-conjugated virus-specific polyclonal antibodies to detect virus antigen present in gill homogenates of infected shrimp spotted onto nitrocellulose membrane. The DB-NC-EIA was highly specific and sensitive enough to detect the presence of viral proteins before the appearance of overt symptoms. The assays are one of the simplest and most rapid detection methods available for WSV and YHV
Results of sunflower-seed oil and rapeseed oil alcoholysis with isopropyl alcohol are represented. The effect of catalyst (cationite with immobilized metal ions) nature on the vegetable oil conversion has been investigated. The possibility of side reactions when using above mentioned catalysts was assessed based on chromatography results and the reaction mixture optical density values. The conclusion about the influence of nature of metal ion immobilized on cationite surface, and the oil type on the reaction of vegetable oils alcoholysis with isopropyl alcohol has been made. The results obtained shows that above mentioned Lewis acids are expedient to be used as vegetable oils alcoholysis catalysts.
Detection of white spot syndrome virus (WSSV) from hemolymph of Penaeid shrimps Penaeus japonicus by reverse passive latex agglutination assay using high-density latex particles
A truncated version of the white spot syndrome virus (WSSV) 27.5 kDa envelope protein was expressed as a histidine tag fusion protein in Escherichia coli. The bacterial expression system allowed the production of up to 10 mg of purified recombinant protein per liter of bacterial culture. Antiserum from a rabbit immunized with the recombinant protein recognized the 27.5 kDa viral envelope protein of WSSV isolated from different geographic regions. The antiserum did not recognize any of the other known WSSV structural proteins. A sensitive immunodot assay for WSSV was developed using the specific rabbit polyclonal antiseru
Haemagglutination as a low-cost detection method for gill-associated virus and by inference, yellowhead virus in Penaeus monodon Fabricius, 1798
A quantitative, low-cost test based on haemagglutination (HA) using chicken erythrocytes was developed to indicate the viral load of Australian yellowhead-like virus, gill-associated virus (GAV), in Penaeus monodon. The study tested the haemolymph, gill, lymphoid organ, heart, sub-cutaneous tissue, eye stalk, pleopods, uropods and the central nerve cord for agglutination activity in 100 prawns, with the haemolymph and gill tissue giving the highest end-point titres of 1:1370 and 1:361 respectively. The sensitivity of the test was demonstrated by testing two different populations of P. monodon, which showed a highly significant difference (P<0.001) in HA activity, indicating a difference in viral load. By testing three other penaeid prawn species (n=20), Penaeus esculentus, Penaeus merguiensis and Penaeus longistylis, and the crayfish Cherax quadricarinatus, it was demonstrated that natural agglutinins were not causing the high agglutination in the population of P. monodon being tested. It was also demonstrated that there was no effect of freezing and thawing of samples on HA activity. The speed and low cost of this test makes it a very useful tool, particularly in the developing world, for on-farm testing of penaeid prawns to indicate yellowhead virus and GAV loads which can contribute to management practices with respect to harvesting of ponds.James Munro and Leigh Owen
