    The Leishmania donovani Lipophosphoglycan Excludes the Vesicular Proton-ATPase from Phagosomes by Impairing the Recruitment of Synaptotagmin V

    We recently showed that the exocytosis regulator Synaptotagmin (Syt) V is recruited to the nascent phagosome and remains associated throughout the maturation process. In this study, we investigated the possibility that Syt V plays a role in regulating interactions between the phagosome and the endocytic organelles. Silencing of Syt V by RNA interference revealed that Syt V contributes to phagolysosome biogenesis by regulating the acquisition of cathepsin D and the vesicular proton-ATPase. In contrast, recruitment of cathepsin B, the early endosomal marker EEA1 and the lysosomal marker LAMP1 to phagosomes was normal in the absence of Syt V. As Leishmania donovani promastigotes inhibit phagosome maturation, we investigated their potential impact on the phagosomal association of Syt V. This inhibition of phagolysosome biogenesis is mediated by the virulence glycolipid lipophosphoglycan, a polymer of the repeating Galβ1,4Manα1-PO4 units attached to the promastigote surface via an unusual glycosylphosphatidylinositol anchor. Our results showed that insertion of lipophosphoglycan into ganglioside GM1-containing microdomains excluded or caused dissociation of Syt V from phagosome membranes. As a consequence, L. donovani promatigotes established infection in a phagosome from which the vesicular proton-ATPase was excluded and which failed to acidify. Collectively, these results reveal a novel function for Syt V in phagolysosome biogenesis and provide novel insight into the mechanism of vesicular proton-ATPase recruitment to maturing phagosomes. We also provide novel findings into the mechanism of Leishmania pathogenesis, whereby targeting of Syt V is part of the strategy used by L. donovani promastigotes to prevent phagosome acidification

    LGR5 Is a Negative Regulator of Tumourigenicity, Antagonizes Wnt Signalling and Regulates Cell Adhesion in Colorectal Cancer Cell Lines

    BACKGROUND: LGR5 (Leucine-rich repeat-containing G-protein coupled receptor 5) is the most established marker for intestinal stem cells. Mouse models show that LGR5+ cells are the cells of origin of intestinal cancer, and LGR5 expression is elevated in human colorectal cancers, however very little is known about LGR5 function or its contribution to the stem cell phenotype and to colorectal cancer. PRINCIPAL FINDINGS: We have modulated the expression of LGR5 by RNAi (inhibitory RNAs) or overexpression in colorectal cancer cell lines. Paradoxically, ablation of LGR5 induces increased invasion and anchorage-independent growth, and enhances tumourigenicity in xenografts experiments. Conversely, overexpression of LGR5 augments cell adhesion, reduces clonogenicity and attenuates tumourigenicity. Expression profiling revealed enhanced wnt signalling and upregulation of EMT genes upon knockdown of LGR5, with opposite changes in LGR5 overexpressing cells. These findings suggest that LGR5 is important in restricting stem cells to their niche, and that loss of LGR5 concomitant with activated wnt signalling may contribute to the invasive phenotype of colorectal carcinomas

    Diel and seasonal patterns in activity and home range size of green turtles on their foraging grounds revealed by extended Fastloc-GPS tracking

    An animal’s home range is driven by a range of factors including top-down (predation risk) and bottom-up (habitat quality) processes, which often vary in both space and time. We assessed the role of these processes in driving spatiotemporal patterns in the home range of the green turtle (Chelonia mydas), an important marine megaherbivore. We satellite tracked adult green turtles using Fastloc-GPS telemetry in the Chagos Archipelago and tracked their fine-scale movement in different foraging areas in the Indian Ocean. Using this extensive data set (5,081 locations over 1,675 tracking days for 8 individuals) we showed that green turtles exhibit both diel and seasonal patterns in activity and home range size. At night, turtles had smaller home ranges and lower activity levels, suggesting they were resting. In the daytime, home ranges were larger and activity levels higher, indicating that turtles were actively feeding. The transit distance between diurnal and nocturnal sites varied considerably between individuals. Further, some turtles changed resting and foraging sites seasonally. These structured movements indicate that turtles had a good understanding of their foraging grounds in regards to suitable areas for foraging and sheltered areas for resting. The clear diel patterns and the restricted size of nocturnal sites could be caused by spatiotemporal variations in predation risk, although other factors (e.g. depth, tides and currents) could also be important. The diurnal and seasonal pattern in home range sizes could similarly be driven by spatiotemporal variations in habitat (e.g. seagrass or algae) quality, although this could not be confirmed

    The wavelength dependence of the two-photon ionisation of metastable D(2s) by a laser beam has been measured near the 2s - 3p resonant frequency. A sharp structure is observed in the broad resonance profile at quite moderate intensity. Its origin is shown to be the spontaneous decay of the 3p state that competes with photoionisation

    We describe a new experiment dealing with the multiphoton ionization of metastable atomic hydrogen H(2s). The ion yield has been measured in a relative way but it provides absolute photoionization rates. In the present contribution, the wavelength is tuned in the vicinity of the 2s-3p transition and the laser intensity is in the 10(7)-5 x 10(10) W cm(-2) range. Experimental photoionization rates and resonance width are found to be in agreement with theoretical predictions

    ABSTRACT Clariidae is a group that includes many species that have great economic importance in both fisheries and fish culture. Monogenean parasites of fishes assigned to this family have been studied in Cameroon, but there have been no studies on Clarias submarginatus Peter, 1882, a fish that is traditionally consumed by the local people. The examination of 43 specimens identified as C. submarginatus from Lake Ossa (first record of this fish species in the Sanaga basin), revealed that some of them belong to Quadriacanthus and are new. Their identification was made based on the morphology and the size of sclerotized parts of the haptor and the male and female copulatory complexes. Quadriacanthus macruncus Bahanak, Nack & Pariselle sp. nov. and Quadriacanthus submarginati Bahanak, Nack & Pariselle sp. nov. are characterized by the morphology of their accessory piece, pointed, slightly curved and wider at medium level, equipped with two subterminal, symmetrical and similar spines for Q. macruncus sp. nov. and made up of one long tip flanked by a short spine and a bulb for Q . submarginati sp. nov., while Quadriacanthus ossaensis Bahanak, Nack & Pariselle sp. nov. is distinguished by the unique morphology of the penis, ending in a fork. The new species of Quadriacanthus are herein described and their host specificity is discussed

    International audienceTwelve specimens of Heterobranchus longifilis sampled from the River Boumba were examined for their monogenean gill parasites. Quadriacanthus longifilisi N'Douba,Lambert & Euzet 1999 and Q. thysi N'Douba, Lambert & Euzet 1999 are newly recorded in the studied locality. Three new species have also been described: Q. tricorniculai n. sp., Q. triunguisin. sp., and Q. ndoubai n. sp. Quadriacanthus tricorniculai and Q. triunguisi are characterized by the unique morphology of their accessory pieces, which end in a three-head hydrashapedstructure, with tentacles being longer in Q. tricorniculai; proximal extremity of accessorypiece is stronger for Q. triunguisi, while Q. ndoubai is distinguished by the size of its male copulatory organ (larger than 80 mu m) and the fork shaped distal extremity of the accessory piece of the male copulatory complex. The variability of monogenean species richness of Hetero-branchus longifilis between Ivory Coast and Cameroon is discussed
