    Pattern-based multi-cloud architecture migration

    Many organizations migrate on-premise software applications to the cloud. However, current coarse-grained cloud migration solutions have made such migrations a non transparent task, an endeavor based on trial-anderror. This paper presents Variability-based, Pattern-driven Architecture Migration .V-PAM), a migration method based on (i) a catalogue of fine-grained service-based cloud architecture migration patterns that target multi-cloud, (ii) a situational migration process framework to guide pattern selection and composition, and (iii) a variability model to structure system migration into a coherent framework. The proposed migration patterns are based on empirical evidence from several migration projects, best practice for cloud architectures and a systematic literature review of existing research. Variability-based, Pattern-driven Architecture Migration allows an organization to (i) select appropriate migration patterns, (ii) compose them to define a migration plan, and (iii) extend them based on the identification of new patterns in new contexts. The patterns are at the core of our solution, embedded into a process model, with their selection governed by a variability model

    An approach to improve accuracy in probabilistic models using state refinement

    Probabilistic models are useful in the analysis of system be- haviour and non-functional properties. Reliable estimates and measurements of probabilities are needed to annotate behaviour models in order to generate accurate predictions. However, this may not be su cient, and may still lead to inaccurate results when the system model does not properly re ect the probabilistic choices made by the environment. Thus, not only should the probabilities be accurate in prop- erly re ecting reality, but also the model that is being used. In this paper we propose state re nement as a technique to mitigate this problem, showing that it is guaranteed to preserve or increase the accuracy of the initial model. We present a framework for iteratively improving the accuracy of a probabilistically annotated behaviour model with re- spect to a set of benchmark properties through iterative state re nements

    A grateful heart: parents’ reflections on gratitude and its development in their children.

    Too much focus has been placed on positive psychology’s view of gratitude, which is broad in scope but operationalized often merely as a positive emotion. I argue that gratitude should be defined as a virtue, as it has been conceptualized by a multitude of philosophers, psychologists, and other scholars who have studied it (Annas, 2011; Baumgarten-Tramer, 1938; Emmons, 2009; Tudge, Freitas, & O’Brien, 2015). As a virtue, gratitude is experienced when (a) a benefactor purposefully and willingly gives a benefit to the beneficiary, and the beneficiary (b) recognizes the intentionality of the benefactor and feels good about it, (d) freely reciprocates based on the benefactor’s needs and wants, if and when the opportunity presents itself, and (e) is consistent in this type of behavioral response (Tudge et al., 2015). The goals of the present research were, in a diverse sample: (1) to investigate whether parents place importance on gratitude for their children; (2) to examine whether parents’ experiences of gratitude affect their thoughts and actions regarding their children’s gratitude; and (3) and to investigate parenting strategies to foster and encourage gratitude, including exploring potential differences based on social class, society, and racial/ethnic group membership. Drawing from semi-structured interviews conducted in the United States and China with parents of children aged 7 to 14, representing seven distinct cultural groups, 100 interviews were coded using directed content analysis based on a triadic coding scheme. A priori codes were established based on literature and new codes were determined based on parent responses. Through parents’ own words, it became apparent that gratitude is significant in all of the cultures represented. The close ties that gratitude helped foster and solidify among family, friends and the community were clear based on the examples given by parents of being helped by benefactors. The need to encourage gratitude in children was evident when parents spoke of children who were not able to truly understand gratitude and who did not demonstrate it enough. Children’s young age was a key factor limiting their understanding of gratitude, regardless of whether the children were in middle childhood or adolescence. Parents also spoke of children who were shy as being less inclined to express gratitude and those who were out-going as being more likely to express gratitude. Strategies parents used to promote gratitude were quite similar such as speaking to children about gratitude or reminding them to express it appropriately Cultural differences were found as well. Gratitude was conceptualized differently depending on the language and cultural group. In this study when examining transcripts from Brazilian or Mexican immigrants in Portuguese or Spanish or the Chinese parents’ transcripts translated into English from Mandarin, special care was taken in interpretation of the interviews. Further, parents varied in why they felt that their children should feel gratitude depending on whether parents were middle-class or working-class. Overall, the present study showed both similarities and differences among the various cultural groups in parents’ thoughts about and experiences of gratitude and how they relate to their opinions of and encouraging gratitude in their children

    Is an understanding of moral obligation associated with the moral virtue gratitude?

    The purpose of the current study was to examine gratitude as a virtue, defined by a benefactor freely giving some kind of benefit to a beneficiary who recognizes the boon, and then freely and intentionally chooses to reciprocate based on the wishes of the benefactor, and its relation to moral obligation, specifically heteronomous versus autonomous obligation in seven to fourteen year olds. Vignette interviews, an open-ended questionnaire determining expressions of gratitude, and the GAQ showed that there was no significant link between expressions of gratitude and children’s and adolescents’ justifications for helping a benefactor based on the type of moral obligation expressed. Nor were differences found based on age or gender. However, an unexpected type of obligation, consequences to the benefactor, was related to age such that younger children were more likely to be concerned about the consequences to their benefactors if they did not help them than were older children. Additionally, those children and adolescents who expressed concrete or connective gratitude scored higher on the GAC, as did girls versus boys

    Uncovering the heterogeneity and temporal complexity of neurodegenerative diseases with Subtype and Stage Inference

    The heterogeneity of neurodegenerative diseases is a key confound to disease understanding and treatment development, as study cohorts typically include multiple phenotypes on distinct disease trajectories. Here we introduce a machine-learning technique—Subtype and Stage Inference (SuStaIn)—able to uncover data-driven disease phenotypes with distinct temporal progression patterns, from widely available cross-sectional patient studies. Results from imaging studies in two neurodegenerative diseases reveal subgroups and their distinct trajectories of regional neurodegeneration. In genetic frontotemporal dementia, SuStaIn identifies genotypes from imaging alone, validating its ability to identify subtypes; further the technique reveals within-genotype heterogeneity. In Alzheimer’s disease, SuStaIn uncovers three subtypes, uniquely characterising their temporal complexity. SuStaIn provides fine-grained patient stratification, which substantially enhances the ability to predict conversion between diagnostic categories over standard models that ignore subtype (p = 7.18 × 10−4) or temporal stage (p = 3.96 × 10−5). SuStaIn offers new promise for enabling disease subtype discovery and precision medicine

    BALB/c Mice Infected with Antimony Treatment Refractory Isolate of Leishmania braziliensis Present Severe Lesions due to IL-4 Production

    Leishmaniasis is a neglected disease that affects more than 12 million people worldwide. In Brazil, the cutaneous disease is more prevalent with about 28,000 new cases reported each year, and L. braziliensis is the main causative agent. The interesting data about the infection with this parasite is the wide variety of clinical manifestations that ranges from single ulcerated lesions to mucocutaneous and disseminated disease. However, experimental models to study the infection with this parasite are difficult to develop due to high resistance of most mouse strains to the infection, and the mechanisms underlying the distinct manifestations remain poorly understood. Here, the authors use a mouse experimental model of infection with different L. braziliensis isolates, known to induce diseases with distinct severity in the human hosts, to elucidate immune mechanisms that may be involved in the different manifestations. They showed that distinct parasite isolates may modulate host response, and increased IL-4 production and Arg I expression was related to more severe disease, resulting in longer length of disease with larger lesions and reduced parasite clearance. These findings may be useful in the identification of immunological targets to control L. braziliensis infection and potential clinical markers of disease progression

    HemaMax™, a Recombinant Human Interleukin-12, Is a Potent Mitigator of Acute Radiation Injury in Mice and Non-Human Primates

    HemaMax, a recombinant human interleukin-12 (IL-12), is under development to address an unmet medical need for effective treatments against acute radiation syndrome due to radiological terrorism or accident when administered at least 24 hours after radiation exposure. This study investigated pharmacokinetics, pharmacodynamics, and efficacy of m-HemaMax (recombinant murine IL-12), and HemaMax to increase survival after total body irradiation (TBI) in mice and rhesus monkeys, respectively, with no supportive care. In mice, m-HemaMax at an optimal 20 ng/mouse dose significantly increased percent survival and survival time when administered 24 hours after TBI between 8–9 Gy (p<0.05 Pearson's chi-square test). This survival benefit was accompanied by increases in plasma interferon-γ (IFN-γ) and erythropoietin levels, recovery of femoral bone hematopoiesis characterized with the presence of IL-12 receptor β2 subunit–expressing myeloid progenitors, megakaryocytes, and osteoblasts. Mitigation of jejunal radiation damage was also examined. At allometrically equivalent doses, HemaMax showed similar pharmacokinetics in rhesus monkeys compared to m-HemaMax in mice, but more robustly increased plasma IFN-γ levels. HemaMax also increased plasma erythropoietin, IL-15, IL-18, and neopterin levels. At non-human primate doses pharmacologically equivalent to murine doses, HemaMax (100 ng/Kg and 250 ng/Kg) administered at 24 hours after TBI (6.7 Gy/LD50/30) significantly increased percent survival of HemaMax groups compared to vehicle (p<0.05 Pearson's chi-square test). This survival benefit was accompanied by a significantly higher leukocyte (neutrophils and lymphocytes), thrombocyte, and reticulocyte counts during nadir (days 12–14) and significantly less weight loss at day 12 compared to vehicle. These findings indicate successful interspecies dose conversion and provide proof of concept that HemaMax increases survival in irradiated rhesus monkeys by promoting hematopoiesis and recovery of immune functions and possibly gastrointestinal functions, likely through a network of interactions involving dendritic cells, osteoblasts, and soluble factors such as IL-12, IFN-γ, and cytoprotectant erythropoietin

    A modified Camel and Cactus Test detects presymptomatic semantic impairment in genetic frontotemporal dementia within the GENFI cohort

    Impaired semantic knowledge is a characteristic feature of some forms of frontotemporal dementia (FTD), particularly the sporadic disorder semantic dementia. Less is known about semantic cognition in the genetic forms of FTD caused by mutations in the genes MAPT, C9orf72, and GRN. We developed a modified version of the Camel and Cactus Test (mCCT) to investigate the presence of semantic difficulties in a large genetic FTD cohort from the Genetic FTD Initiative (GENFI) study. Six-hundred-forty-four participants were tested with the mCCT including 67 MAPT mutation carriers (15 symptomatic, and 52 in the presymptomatic period), 165 GRN mutation carriers (33 symptomatic, 132 presymptomatic), and 164 C9orf72 mutation carriers (56 symptomatic, 108 presymptomatic) and 248 mutation-negative members of FTD families who acted as a control group. The presymptomatic mutation carriers were further split into those early and late in the presymptomatic period (more than vs. within 10 years of expected symptom onset). Groups were compared using a linear regression model, adjusting for age and education, with bootstrapping. Performance on the mCCT had a weak negative correlation with age (rho = −0.20) and a weak positive correlation with education (rho = 0.13), with an overall abnormal score (below the 5th percentile of the control population) being below 27 out of a total of 32. All three of the symptomatic mutation groups scored significantly lower than controls: MAPT mean 22.3 (standard deviation 8.0), GRN 24.4 (7.2), C9orf72 23.6 (6.5) and controls 30.2 (1.6). However, in the presymptomatic groups, only the late MAPT and late C9orf72 mutation groups scored lower than controls (28.8 (2.2) and 28.9 (2.5) respectively). Performance on the mCCT correlated strongly with temporal lobe volume in the symptomatic MAPT mutation group (rho > 0.80). In the C9orf72 group, mCCT score correlated with both bilateral temporal lobe volume (rho > 0.31) and bilateral frontal lobe volume (rho > 0.29), whilst in the GRN group mCCT score correlated only with left frontal lobe volume (rho = 0.48). This study provides evidence for presymptomatic impaired semantic knowledge in genetic FTD. The different neuroanatomical associations of the mCCT score may represent distinct cognitive processes causing deficits in different groups: loss of core semantic knowledge associated with temporal lobe atrophy (particularly in the MAPT group), and impaired executive control of semantic information associated with frontal lobe atrophy. Further studies will be helpful to address the longitudinal change in mCCT performance and the exact time at which presymptomatic impairment occurs

    Anxiety and Depression in Adults with Autism Spectrum Disorder: A Systematic Review and Meta-analysis

    Adults with autism spectrum disorder (ASD) are thought to be at disproportionate risk of developing mental health comorbidities, with anxiety and depression being considered most prominent amongst these. Yet, no systematic review has been carried out to date to examine rates of both anxiety and depression focusing specifically on adults with ASD. This systematic review and meta-analysis examined the rates of anxiety and depression in adults with ASD and the impact of factors such as assessment methods and presence of comorbid intellectual disability (ID) diagnosis on estimated prevalence rates. Electronic database searches for studies published between January 2000 and September 2017 identified a total of 35 studies, including 30 studies measuring anxiety (n = 26 070; mean age = 30.9, s.d. = 6.2 years) and 29 studies measuring depression (n = 26 117; mean age = 31.1, s.d. = 6.8 years). The pooled estimation of current and lifetime prevalence for adults with ASD were 27% and 42% for any anxiety disorder, and 23% and 37% for depressive disorder. Further analyses revealed that the use of questionnaire measures and the presence of ID may significantly influence estimates of prevalence. The current literature suffers from a high degree of heterogeneity in study method and an overreliance on clinical samples. These results highlight the importance of community-based studies and the identification and inclusion of well-characterized samples to reduce heterogeneity and bias in estimates of prevalence for comorbidity in adults with ASD and other populations with complex psychiatric presentations
