Development of 50-kW-Class High-Temperature Superconducting Induction/Synchronous Motor With Continuous Drive Characteristics from Room Temperature
We investigate the rotational characteristics of a 50-kW-class high-temperature superconducting induction/synchro- nous motor in superconducting and non-superconducting states. The prototype motor was first placed in liquid nitrogen, and the efficiency contour was obtained. The liquid nitrogen was then gradually removed from the motor under the partial load (10 kW) condition. This showed that continuous operation is possible even at temperatures exceeding 130 K. Furthermore, we succeeded in driving at 11% of the rated output, even at room temperature. As a result, a practical high-temperature superconducting motor that can continue to operate with reduced output even when the cooling system fails could be realized
Intracellular cAMP contents regulate NAMPT expression via induction of C/EBPβ in adipocytes
A decline in intracellular nicotinamide adenine mononucleotide (NAD+) causes adipose tissue dysfunction. Nicotinamide phosphoribosyltransferase (NAMPT) catalyzes the rate-limiting step in the NAD+ biosynthesis pathway. However, the molecular mechanism that mediates regulation of NAMPT expression in adipocytes is yet to be elucidated. This study found that intracellular cAMP regulates NAMPT expression and promoter activity in 3T3-L1 adipocytes. cAMP-mediated Nampt promoter activity was suppressed by protein kinase A inhibitor H89, whereas AMP-activated protein kinase inhibitor compound C did not affect cAMP-mediated Nampt promoter activity. Intracellular cAMP induced CCAAT/enhancer-binding protein β (C/EBPβ) expression. Knockdown of C/EBPβ suppressed NAMPT expression and promoter activity. Furthermore, the Nampt promoter was activated by C/EBPβ, while LIP activated the dominant-negative form of C/EBPβ. Promoter sequence analysis revealed that the region from -96 to -76 on Nampt was required for C/EBPβ-mediated promoter activity. Additionally, chromatin immunoprecipitation assay demonstrated that C/EBPβ was bound to the promoter sequences of Nampt. Finally, NAMPT inhibitor FK866 suppressed adipogenesis in 3T3-L1 cells, and this suppressive effect was restored by nicotinamide mononucleotide treatment. These findings showed that intracellular cAMP increased NAMPT levels by induction of C/EBPβ expression and indicated that the induction of NAMPT expression was important for adipogenesis.ArticleBiochemical and Biophysical Research Communications. 522(3): 770-775. Three-dimensional architecture and assembly mechanism of the egg-shaped shell in testate amoeba Paulinella micropora
Unicellular euglyphid testate amoeba Paulinella micropora with filose pseudopodia secrete approximately 50 siliceous scales into the extracellular template-free space to construct a shell isomorphic to that of its mother cell. This shell-constructing behavior is analogous to building a house with bricks, and a complex mechanism is expected to be involved for a single-celled amoeba to achieve such a phenomenon; however, the three-dimensional (3D) structure of the shell and its assembly in P. micropora are still unknown. In this study, we aimed to clarify the positional relationship between the cytoplasmic and extracellular scales and the structure of the egg-shaped shell in P. micropora during shell construction using focused ion beam scanning electron microscopy (FIB-SEM). 3D reconstruction revealed an extensive invasion of the electron-dense cytoplasm between the long sides of the positioned and stacked scales, which was predicted to be mediated by actin filament extension. To investigate the architecture of the shell of P. micropora, each scale was individually segmented, and the position of its centroid was plotted. The scales were arranged in a left-handed, single-circular ellipse in a twisted arrangement. In addition, we 3D printed individual scales and assembled them, revealing new features of the shell assembly mechanism of P. micropora. Our results indicate that the shell of P. micropora forms an egg shape by the regular stacking of precisely designed scales, and that the cytoskeleton is involved in the construction process

High molecular weight amyloid β1-42 oligomers induce neurotoxicity via plasma membrane damage.
Amyloid β-protein (Aβ) molecules tend to aggregate and subsequently form low MW (LMW) oligomers, high MW (HMW) aggregates such as protofibrils, and ultimately fibrils. These Aβ species can generally form amyloid plaques implicated in the neurodegeneration of Alzheimer disease (AD), but therapies designed to reduce plaque load have not demonstrated clinical efficacy. Recent evidence implicates amyloid oligomers in AD neuropathology, but the precise mechanisms are uncertain. We examined the mechanisms of neuronal dysfunction from HMW-Aβ1-42 exposure by measuring membrane integrity, reactive oxygen species (ROS) generation, membrane lipid peroxidation, membrane fluidity, intracellular calcium regulation, passive membrane electrophysiological properties, and long-term potentiation (LTP). HMW-Aβ1-42 disturbed membrane integrity by inducing ROS generation and lipid peroxidation, resulting in decreased membrane fluidity, intracellular calcium dysregulation, depolarization, and impaired LTP. The damaging effects of HMW-Aβ1-42 were significantly greater than those of LMW-Aβ1-42. Advanced Biopolymer-Based Nanocomposites and Hybrid Materials
The exploitation of naturally occurring polymers to engineer advanced nanocomposites and hybrid materials is the focus of increasing scientific activity, explained by growing environmental concerns and interest in the peculiar features and multiple functionalities of these macromolecules. Natural polymers, such as polysaccharides and proteins, present a remarkable potential for the design of all kinds of materials for application in a multitude of domains. This Special Issue collected the work of scientists on the current developments in the field of multifunctional biopolymer-based nanocomposites and hybrid materials with a particular emphasis on their production methodologies, properties, and prominent applications. Sorotipos, "mating type" e ploidia de amostras de C. neoformans isoladas de pacientes no Brasil
Serotype, mating type and ploidy of 84 strains of Cryptococcus neoformans isolated from 61 AIDS and 23 non-AIDS patients admitted in a tertiary teaching hospital in São Paulo, Brazil were examined. Among 61 strains isolated from AIDS patients, 60 strains were var. grubii (serotype A). Only one strain was var. gattii (serotype B). No var. neoformans (serotype D) was found. Among 23 strains isolated from non-AIDS patients, 15 were var. grubii (serotype A) and the remaining 8 were var. gattii, all of which were serotype B. Seventy-three of the 75 serotype A strains were the heterothallic alpha type (MATalpha) and the remaining 2 were untypable (asexual). Most of the MATalpha strains (69/73) were haploid and the remaining 4 strains were diploid. Similarly, both of the 2 asexual strains among the 75 serotype A strains were haploid. There were no alpha-mating type (MATalpha) strains among the 84 isolates. All of the 8 var. gattii strains were serotype B and haploid. Among a total of 84 strains tested, neither serotype AD nor serotype D were found. Neither triploid nor tetraploid were found. These results suggest that the serological, sexual and ploidy characteristics in C. neoformans strains isolated from AIDS patients in São Paulo were rather simple, whereas strains isolated from non-AIDS patients presented serotype A and B with predominance of serotype A.Foram estudados os sorotipos, "mating type" e ploidia de 84 amostras de C. neoformans isoladas de 61 pacientes com AIDS e 23 não-AIDS em São Paulo. Das amostras isoladas de pacientes com AIDS, 60 foram identificadas como var. grubii (sorotipo A) e 1 como var. gattii (sorotipo B). Não houve isolamento do sorotipo D. Entre as amostras isoladas, de pacientes não-AIDS, 15 foram de var. grubii (sorotipo A) e as 8 restantes de var. gattii, todos do sorotipo B. Setenta e três dos 75 sorotipos A foram identificadas como cepas heterotálicas do fenótipo alfa (MATalfa) e as 2 remanescentes não-tipáveis (assexuada), eram haplóides. A maioria das cepas MATalfa (69/73) era haplóide sendo 4 diplóide. Não houve o isolamento de fenótipo a (MATa) entre as 84 cepas analisadas. Todas as 11 amostras de var. gattii eram do sorotipo B e haplóides. Não foram observados os sorotipos AD e C, nem células triplóides ou tetraplóides entre as 84 amostras estudadas. Os resultados sugerem, que as características sorológicas, sexuais e de ploidia de C. neoformans, isoladas de pacientes com AIDS em São Paulo, são particularmente simples, a maioria do sorotipo A, enquanto que nos pacientes não-AIDS foram observados tanto os sorotipos A quanto o B
