Stable maintenance of de novo assembled human artificial chromosomes in embryonic stem cells and their differentiated progeny in mice
De novo assembled alphoid(tetO)-type human artificial chromosomes (HACs) represent a novel promising generation of high capacity episomal vectors. Their function and persistence, and any adverse effects, in various cell types in live animals, have not, however, been explored. In this study we transferred the alphoid(tetO)-HAC into mouse ES cells and assessed whether the presence of this extra chromosome affects their pluripotent properties. Alphoid(tetO)-HAC-bearing ES cells were indistinguishable from their wild-type counterparts: they retained self-renewal potential and full capacity for multilineage differentiation during mouse development, whereas the HAC itself was mitotically and transcriptionally stable during this process. Our data provide the first example of fully synthetic DNA behaving like a normal chromosome in cells of living animals. It also opens a new perspective into functional genetic studies in laboratory animals as well as stem cell-based regenerative medicine
Simultaneous monitoring of the photometric and polarimetric activity of the young star PV Cep in the optical/near-infrared bands
We present the results of a simultaneous monitoring, lasting more than 2
years, of the optical and near-infrared photometric and polarimetric activity
of the variable protostar PV Cep. During the monitoring period, an outburst has
occurred in all the photometric bands, whose declining phase (J
3 mag) lasted about 120 days. A time lag of 30 days between
optical and infrared light curves has been measured and interpreted in the
framework of an accretion event. This latter is directly recognizable in the
significant variations of the near-infrared colors, that appear bluer in the
outburst phase, when the star dominates the emission, and redder in declining
phase, when the disk emission prevails. All the observational data have been
combined to derive a coherent picture of the complex morphology of the whole PV
Cep system, that, in addition to the star and the accretion disk, is composed
also by a variable biconical nebula. In particular, the mutual interaction
between all these components is the cause of the high value of the polarization
( 20%) and of its fluctuations. The observational data concur to
indicate that PV Cep is not a genuine EXor star, but rather a more complex
object; moreover the case of PV Cep leads to argue about the classification of
other recently discovered young sources in outburst, that have been considered,
maybe over-simplifying, as EXor.Comment: Accepted for publication on Ap
Spectral Dependence of Coherent Backscattering of Light in a Narrow-Resonance Atomic System
We report a combined theoretical and experimental study of the spectral and
polarization dependence of near resonant radiation coherently backscattered
from an ultracold gas of 85Rb atoms. Measurements in an approximately 6 MHz
range about the 5s^{2}S_{1/2}- 5p^{2}P_{3/2}, F=3 - F'=4 hyperfine transition
are compared with simulations based on a realistic model of the experimental
atomic density distribution. In the simulations, the influence of heating of
the atoms in the vapor, magnetization of the vapor, finite spectral bandwidth,
and other nonresonant hyperfine transitions are considered. Good agreement is
found between the simulations and measurements.Comment: 10 pages, 12 figur
