    Microbial and heavy metal contaminant of antidiabetic herbal preparations formulated in Bangladesh

    The aim of the current study was to evaluate microbial contamination in terms of microbial load (total aerobic count and total coliform count) and specific pathogenic bacteria (Salmonella spp., Escherichia coli, particularly Escherichia coli 0157) in thirteen antidiabetic herbal preparations (ADHPs) from Dhaka City. All the thirteen ADHPs had been found contaminated with fungi and different pathogenic bacteria. From the data, it is found that only two of these preparations (ADHP-1 and ADHP-12) complied with the safety limit (as stated in different Pharmacopoeias and WHO guidelines) evaluated by all different microbial counts. None of these herbal preparations could assure the safety as all of them were contaminated by fungi. The overall safety regarding heavy metal content (Zn, Cu, Mn, Cr, Cd, and Pb) was assured as none of them exceeded the safety limit of the daily intake. Microbial contaminants in these herbal preparations pose a potential risk for human health and care should be taken in every step involved in the preparation of these herbal preparations to assure safety.Rausan Zamir, Anowar Hosen, M.     Is gastro-oesophageal reflux a factor in exercise-induced asthma?

    AbstractExercise-induced bronchoconstriction (EIB) occurs in the majority of patients with asthma. The relationship between asthma and gastro-oesophageal reflux (GER) is well defined, and the reports of exertional gastro-oesophageal acid reflux in healthy subjects, prompted us to study the relationship between EIB and GER.Following an overnight fast and medication withholding, 15 asthmatics and 15 normal subjects were placed on continuous monitoring of oesophageal pH and ECG. After baseline monitoring of oesophageal pH, at rest, for 30 min, spirometry was performed. Thereafter, the subjects underwent rigorous treadmill exercise for 8 min followed by spirometry, 10 min after running.Twelve out of 15 asthmatics and none in the control group demonstrated significant fall in FEV1 in response to exercise. However, only six out of 15 normal subjects and three in the asthmatic group had evidence of GER during or following exercise.We concluded that there is no significant correlation between EIB and GER in patients with asthma

    A New Advanced Backcross Tomato Population Enables High Resolution Leaf QTL Mapping and Gene Identification.

    Quantitative Trait Loci (QTL) mapping is a powerful technique for dissecting the genetic basis of traits and species differences. Established tomato mapping populations between domesticated tomato (Solanum lycopersicum) and its more distant interfertile relatives typically follow a near isogenic line (NIL) design, such as the S. pennellii Introgression Line (IL) population, with a single wild introgression per line in an otherwise domesticated genetic background. Here, we report on a new advanced backcross QTL mapping resource for tomato, derived from a cross between the M82 tomato cultivar and S. pennellii This so-called Backcrossed Inbred Line (BIL) population is comprised of a mix of BC2 and BC3 lines, with domesticated tomato as the recurrent parent. The BIL population is complementary to the existing S. pennellii IL population, with which it shares parents. Using the BILs, we mapped traits for leaf complexity, leaflet shape, and flowering time. We demonstrate the utility of the BILs for fine-mapping QTL, particularly QTL initially mapped in the ILs, by fine-mapping several QTL to single or few candidate genes. Moreover, we confirm the value of a backcrossed population with multiple introgressions per line, such as the BILs, for epistatic QTL mapping. Our work was further enabled by the development of our own statistical inference and visualization tools, namely a heterogeneous hidden Markov model for genotyping the lines, and by using state-of-the-art sparse regression techniques for QTL mapping

    X-ray phase contrast tomography; proof of principle for post-mortem imaging

    Objectives: To demonstrate the feasibility of using X-ray phase contrast tomography to assess internal organs in a post-mortem piglet model, as a possible non-invasive imaging autopsy technique. Methods: Tomographic images of a new-born piglet were obtained using a Free Space Propagation (FSP) X-ray phase contrast imaging setup at a synchrotron (European Synchrotron Radiation Facility, Grenoble, France). A monochromatic X-ray beam (52 keV) was used in combination with a detector pixel size of 46x46 μm2. A phase-retrieval algorithm was applied to all projections, which were then reconstructed into tomograms using the filtered-back projection algorithm. Images were assessed for diagnostic quality. Results: Images obtained with the FSP setup presented high soft tissue contrast and sufficient resolution for resolving organ structure. All of the main body organs (heart, lungs, kidneys, liver and intestines) were easily identified and adequately visualised. In addition, grey/white matter differentiation in the cerebellum while still contained within the skull was shown. Conclusions: The feasibility of using X-ray phase contrast tomography as a post-mortem imaging technique in an animal model has been demonstrated. Future studies will focus on translating this experiment to a laboratory-based setup

    The effect of methylphenidate on three forms of response inhibition in boys with AD/HD

    Item does not contain fulltextThe current study was aimed at (a) investigating the effect of three doses methylphenidate (MPH) and placebo on inhibition of a prepotent response, inhibition of an ongoing response, and interference control in Attention Deficit/Hyperactivity Disorder (AD/HD), and (b) studying dose-response relations for the three forms of response inhibition. To meet these aims, the following tasks were selected: two versions of the Stop Paradigm for inhibition of a prepotent response, a Circle Tracing Task and a recently developed Follow Task for inhibition of an ongoing response, and the Stroop Color-Word Test and an Eriksen Flanker Task for interference control. These tasks were administered to 23 boys with AD/HD during four treatment conditions: 5 mg MPH, 10 mg MPH, 20 mg MPH, and placebo. A pseudorandomized, multiple-blind, placebo-controlled, within-subject design was used. As hypothesized, inhibitory control in children with AD/HD improved under MPH compared to placebo. However, this effect was only significant for inhibition of a prepotent response and inhibition of an ongoing response (as measured by the Follow Task), but not for interference control. The relation between treatment condition and response was linear. However, this linear relation was due to improved inhibitory control under MPH compared to placebo, because no effects of MPH dose were observed for any of the response inhibition measures

The study used a novel paradigm to examine both thought suppression and response inhibition in OCD, where the modulatory effects of stimuli relevant to OCD could also be assessed. Additionally, the study compared inhibitory impairments in OCD patients with and without co-morbid depression, as depression is the major co-morbidity of OCD. Method: Volitional response suppression and unintentional thought suppression to emotive and neutral stimuli were examined using a novel thought stop-signal task. The thought stop-signal task was administered to non-depressed OCD patients, depressed OCD patients and healthy controls (n=20 per group). Results: Motor inhibition impairments were evident in OCD patients, while motor response performance did not differ between patients and controls. Switching to a new response but not motor inhibition was affected by stimulus relevance in OCD patients. Additionally, unintentional thought suppression as measured by repetition priming was intact. OCD patients with and without depression did not differ on any task performance measures, though there were significant differences in all self-reported measures. Conclusions: Results support motor inhibition deficits in OCD that remain stable regardless of stimulus meaning or co-morbid depression. Only switching to a new response was influenced by stimulus meaning. When response inhibition was successful in OCD patients, so was the unintentional suppression of the accompanying thought.Peer reviewe

    Dynamic Analysis of Vascular Morphogenesis Using Transgenic Quail Embryos

    Background: One of the least understood and most central questions confronting biologists is how initially simple clusters or sheet-like cell collectives can assemble into highly complex three-dimensional functional tissues and organs. Due to the limits of oxygen diffusion, blood vessels are an essential and ubiquitous presence in all amniote tissues and organs. Vasculogenesis, the de novo self-assembly of endothelial cell (EC) precursors into endothelial tubes, is the first step in blood vessel formation [1]. Static imaging and in vitro models are wholly inadequate to capture many aspects of vascular pattern formation in vivo, because vasculogenesis involves dynamic changes of the endothelial cells and of the forming blood vessels, in an embryo that is changing size and shape. Methodology/Principal Findings: We have generated Tie1 transgenic quail lines Tg(tie1:H2B-eYFP) that express H2B-eYFP in all of their endothelial cells which permit investigations into early embryonic vascular morphogenesis with unprecedented clarity and insight. By combining the power of molecular genetics with the elegance of dynamic imaging, we follow the precise patterning of endothelial cells in space and time. We show that during vasculogenesis within the vascular plexus, ECs move independently to form the rudiments of blood vessels, all while collectively moving with gastrulating tissues that flow toward the embryo midline. The aortae are a composite of somatic derived ECs forming its dorsal regions and the splanchnic derived ECs forming its ventral region. The ECs in the dorsal regions of the forming aortae exhibit variable mediolateral motions as they move rostrally; those in more ventral regions show significant lateral-to-medial movement as they course rostrally. Conclusions/Significance: The present results offer a powerful approach to the major challenge of studying the relative role(s) of the mechanical, molecular, and cellular mechanisms of vascular development. In past studies, the advantages of the molecular genetic tools available in mouse were counterbalanced by the limited experimental accessibility needed for imaging and perturbation studies. Avian embryos provide the needed accessibility, but few genetic resources. The creation of transgenic quail with labeled endothelia builds upon the important roles that avian embryos have played in previous studies of vascular development

    Nitrogen management studies in maize (Zea mays L.) hybrids

    A field experiment was conducted to study the effect of different nitrogen management methods on yield, yield components and quality attributes of maize hybrids (single cross-6142 and double cross-4444) under irrigated conditions. Nitrogen dose is met either by PM (poultry manure) or urea according to each treatment. PM was incorporated at the time of presowing irrigation whereas fertigation method at knee height stage and foliar spray at flowering were use for the application of urea. Results showed that plant height, cob diameter, number of grains per cob, grain yield and biological yield were significantly affected by the hybrids. Significantly, higher plant height, cob diameter, number of grains per cob, grain yield and biological yield were produced by single cross-6142. There was no significant difference occur between both hybrids on seed oil and protein contents. N management by the application of T3 (60% N from PM + 38.5% N from urea through fertigation + 1.5% N from urea through foliar application) produced significantly more plant height, cob diameter, number of grains per cob, grain yield, biological yield, seed protein and seed oil contents. The interaction of single cross-6142 and T3 (60% N from PM + 38.5% N from urea through fertigation + 1.5% N from urea through foliar application) was found superior in production of more plant height, cob diameter and number of grains per cob. However, interaction between maize hybrids and N application methods for grain yield, biological yield, seed protein and oil contents was reported nonsignificant. It can be concluded that single cross hybrid-6142) and T3 (60% N from PM, 38.5% N from urea through fertigation and 1.5% N from urea through foliar application) could be used successfully for improving maize yield under the irrigated conditions

    Direct Image to Point Cloud Descriptors Matching for 6-DOF Camera Localization in Dense 3D Point Cloud

    We propose a novel concept to directly match feature descriptors extracted from RGB images, with feature descriptors extracted from 3D point clouds. We use this concept to localize the position and orientation (pose) of the camera of a query image in dense point clouds. We generate a dataset of matching 2D and 3D descriptors, and use it to train a proposed Descriptor-Matcher algorithm. To localize a query image in a point cloud, we extract 2D keypoints and descriptors from the query image. Then the Descriptor-Matcher is used to find the corresponding pairs 2D and 3D keypoints by matching the 2D descriptors with the pre-extracted 3D descriptors of the point cloud. This information is used in a robust pose estimation algorithm to localize the query image in the 3D point cloud. Experiments demonstrate that directly matching 2D and 3D descriptors is not only a viable idea but also achieves competitive accuracy compared to other state-of-the-art approaches for camera pose localization
