    The influence of metabolic profile of obese men on the severity of erectile dysfunction: Are metabolically healthy obese individuals protected?

    Objective: To determine the prevalence of erectile dysfunction (ED) in metabolically healthy obese (MHO) individuals, and to compare ED severity and hypogonadism prevalence in MHO, metabolically unhealthy obese (MUO) and metabolically healthy non-obese individuals.Material and methods: ED patients (n=460) were evaluated by standardized protocol, that included clinical evaluation, abridged 5-item version of the International Index of Erectile Function (IIEF-5) questionnaire survey, and Penile Duplex Doppler Ultrasound (PDDU) exam. Patients were classified as obese [body mass index (BMI) =30.0 kg/m2] and non-obese (BMI <30.0 kg/m2), and metabolic health status was defined by National Cholesterol Education Program Adult Treatment Panel III (NCEP ATPIII) criteria. Statistical analysis was performed and statistical significance was considered at p-level <0.05.Results: The mean age of the subjects was 56.2±10.5 years. MHO was present in 40% of obese individuals (n=37). MUO had lower mean peak systolic velocity (mPSV) compared to MHO (28.1 cm/s vs. 36.9 cm/s; p=0.005), and IIEF-5 scores were also lower in MUO compared to MHO patients (10.2 vs. 13.1; p=0.018). No statistical differences in IIEF-5 score, mPSV and hypogonadism prevalence between MHO and metabolically healthy non-obese (MHNO) patients were observed.Conclusion: Our results lead us to conclude that healthy metabolic profile protects obese individuals from severity of ED. The strong association between obesity and ED may be otherwise attributed to metabolic abnormalities present in the obese

    Cumulative Effect of Cardiovascular Risk Factors on Regulation of AMPK/SIRT1-PGC-1 alpha-SIRT3 Pathway in the Human Erectile Tissue

    Cardiovascular disease risk factors (CVDRF), especially diabetes mellitus (DM), disrupt oxidative stress response. This condition underlies endothelial dysfunction, early manifested in men as erectile dysfunction. The current study is aimed at elucidating the impact of CVDRF in the oxidation responsive AMPK/SIRT1-PGC-1 alpha-SIRT3 pathway and related miRNAs in the human corpus cavernosum. Human penile tissue fragments from individuals submitted to programmed urological surgeries (n=27), aged 43-63 years, were clustered depending on the presence of CVDRF; the control group included samples from patients without CVDRF, and groups A and B included samples from patients with DM and additional CVDRF, totalizing = 3 CVDRF (group B). Dual-immunolabelling of SIRT3, SOD2, or GPX1 with alpha-actin in tissue sections was carried out. The assessment of expression levels of NOX1, phospho-AMPK alpha, total AMPK alpha, SIRT1, PGC-1 alpha, SIRT3, SOD2, and GPX1 was performed by western blotting and of miR-200a, miR-34a, miR-421, and miR-206 by real-time PCR. Phospho-AMPK alpha and SIRT3 expression was found significantly increased in group B relative to other groups, suggesting a marked influence of CVDRF, additional to DM, in the regulation of these enzymes. NOX1 was also increased in group B relative to controls. Only an increasing tendency was observed in the phospho-AMPK alpha/total AMPK alpha ratio, SIRT1, and PGC-1 alpha expression in groups A and B when compared with controls. Concerning antioxidant enzymes, GPX1 expression was found incremented in group A, but SOD2 expression was decreased in groups A and B, comparative with controls. Group B presented significantly diminished levels of miR-421 and miR-200a, but only a decreasing trend on miR-34 and miR-206 expression was observed. Taken together, our findings demonstrated that besides DM, additional CVDRF presented a cumulative effect in the cellular response to oxidative unbalance, contributing to AMPK/SIRT1-PGC-1 alpha-SIRT3 pathway activation.     Thermal detection of single e-h pairs in a biased silicon crystal detector

    We demonstrate that individual electron-hole pairs are resolved in a 1 cm2^2 by 4 mm thick silicon crystal (0.93 g) operated at \sim35 mK. One side of the detector is patterned with two quasiparticle-trap-assisted electro-thermal-feedback transition edge sensor (QET) arrays held near ground potential. The other side contains a bias grid with 20\% coverage. Bias potentials up to ±\pm 160 V were used in the work reported here. A fiber optic provides 650~nm (1.9 eV) photons that each produce an electron-hole (eh+e^{-} h^{+}) pair in the crystal near the grid. The energy of the drifting charges is measured with a phonon sensor noise σ\sigma \sim0.09 eh+e^{-} h^{+} pair. The observed charge quantization is nearly identical for h+h^+'s or ee^-'s transported across the crystal.Comment: 4 journal pages, 5 figure

    We present results on phonon quasidiffusion and Transition Edge Sensor (TES) studies in a large, 3 inch diameter, 1 inch thick [100] high purity germanium crystal, cooled to 50 mK in the vacuum of a dilution refrigerator, and exposed with 59.5 keV gamma-rays from an Am-241 calibration source. We compare calibration data with results from a Monte Carlo which includes phonon quasidiffusion and the generation of phonons created by charge carriers as they are drifted across the detector by ionization readout channels. The phonon energy is then parsed into TES based phonon readout channels and input into a TES simulator

    Validation of Phonon Physics in the CDMS Detector Monte Carlo

    The SuperCDMS collaboration is a dark matter search effort aimed at detecting the scattering of WIMP dark matter from nuclei in cryogenic germanium targets. The CDMS Detector Monte Carlo (CDMS-DMC) is a simulation tool aimed at achieving a deeper understanding of the performance of the SuperCDMS detectors and aiding the dark matter search analysis.     Characterization of SuperCDMS 1-inch Ge Detectors

    The newly commissioned SuperCDMS Soudan experiment aims to search for WIMP dark matter with a sensitivity to cross sections of 5×10^(−45)cm^2 and larger (90% CL upper limit). This goal is facilitated by a new set of germanium detectors, 2.5 times more massive than the ones used in the CDMS-II experiment, and with a different athermal phonon sensor layout that eliminates radial degeneracy in position reconstruction of high radius events. We present characterization data on these detectors, as well as improved techniques for correcting position-dependent variations in pulse shape across the detector. These improvements provide surface-event discrimination sufficient for a reach of 5×10^(−45)cm^2

    A Search for WIMPs with the First Five-Tower Data from CDMS

    We report first results from the Cryogenic Dark Matter Search (CDMS II) experiment running with its full complement of 30 cryogenic particle detectors at the Soudan Underground Laboratory. This report is based on the analysis of data acquired between October 2006 and July 2007 from 15 Ge detectors (3.75 kg), giving an effective exposure of 121.3 kg-d (averaged over recoil energies 10--100 keV, weighted for a weakly interacting massive particle (WIMP) mass of 60 \gev). A blind analysis, incorporating improved techniques for event reconstruction and data quality monitoring, resulted in zero observed events. This analysis sets an upper limit on the WIMP-nucleon spin-independent cross section of 6.6×1044\times10^{-44} cm2^2 (4.6×1044\times10^{-44} cm2^2 when combined with previous CDMS Soudan data) at the 90% confidence level for a WIMP mass of 60 \gev. By providing the best sensitivity for dark matter WIMPs with masses above 42 GeV/c2^2, this work significantly restricts the parameter space for some of the favored supersymmetric models.Comment: 5 pages, 4 figures, submitted to PRL 28 March 200

    Using improved Ge and Si detectors, better neutron shielding, and increased counting time, the Cryogenic Dark Matter Search (CDMS) experiment has obtained stricter limits on the cross section of weakly interacting massive particles (WIMPs) elastically scattering from nuclei. Increased discrimination against electromagnetic backgrounds and reduction of neutron flux confirm WIMP-candidate events previously detected by CDMS were consistent with neutrons and give limits on spin-independent WIMP interactions which are >2X lower than previous CDMS results for high WIMP mass, and which exclude new parameter space for WIMPs with mass between 8-20 GeV/c^2.Comment: 4 pages, 4 figure

    Bioformulation of microbial biocontrol agents for a sustainable agriculture

    The application of microbial based biopesticides has become a sustainable alternative to the use of chemicals to prevent yield losses due to plant pathogens. However, microbial based biopesticides are often unsuccessfully formulated and do not meet the demanding regulatory standards required by the agencies, which hinders their commercialization. Hence, an outline on the approaches to attain more effective formulations might be useful for the development of future more effective products. With this aim, this chapter reports the current state of biocontrol strategies and describes the principles of microbial biocontrol formulations. Emphasis is placed on techniques and tools available for the development and characterisation of microbial products. To provide glimpses on the possible formulations, the different existing additives, carriers, inoculation techniques and formulation types are exhaustively reviewed.
