Genesis at work: Advancing inclusive innovation through manufacturing extension
US manufacturing is struggling with both a productivity and job quality challenge. These challenges are interconnected, reinforcing the need for increased coordination of economic and workforce development efforts. This article outlines the evaluation findings of a novel business-facing initiative called the Genesis Movement, to understand its role in reshaping the workforce experience within small- and medium-sized manufacturing businesses in Chicago, Illinois. Spearheaded in 2014 by the Illinois Manufacturing Excellence Center (IMEC), Genesis starts with the premise that workforce practices are central to business operations, productivity, and competitivenessâand therefore, manufacturing extension services need to promote improvements to job quality in support of long-term business success. This integrated approach represents a form of âinclusive innovation,â expanding the capacities of firms and workers to adapt in a competitive environment where pressures to maintain high quality while reducing costs are ever-present. Firms that participate in Genesis learn to adopt an inclusive organizational culture, using front-line worker engagement, skills training, and job quality improvements to drive performance and process innovation. As such, Genesis offers transferable lessons that could be leveraged by other manufacturing-supporting organizations to benefit their industry clients and the workers they employ
Syrian Refugees and the Digital Passage to Europe: Smartphone Infrastructures and Affordances
This research examines the role of smartphones in refugeesâ journeys. It traces the risks and possibilities afforded by smartphones for facilitating information, communication, and migration flows in the digital passage to Europe. For the Syrian and Iraqi refugee respondents in this France-based qualitative study, smartphones are lifelines, as important as water and food. They afford the planning, navigation, and documentation of journeys, enabling regular contact with family, friends, smugglers, and those who help them. However, refugees are simultaneously exposed to new forms of exploitation and surveillance with smartphones as migrations are financialised by smugglers and criminalized by European policies, and the digital passage is dependent on a contingent range of sociotechnical and material assemblages. Through an infrastructural lens, we capture the dialectical dynamics of opportunity and vulnerability, and the forms of resilience and solidarity, that arise as forced migration and digital connectivity coincide
A Twist Code Determines the Onset of Osteoblast Differentiation
AbstractRunx2 is necessary and sufficient for osteoblast differentiation, yet its expression precedes the appearance of osteoblasts by 4 days. Here we show that Twist proteins transiently inhibit Runx2 function during skeletogenesis. Twist-1 and -2 are expressed in Runx2-expressing cells throughout the skeleton early during development, and osteoblast-specific gene expression occurs only after their expression decreases. Double heterozygotes for Twist-1 and Runx2 deletion have none of the skull abnormalities observed in Runx2+/â mice, a Twist-2 null background rescues the clavicle phenotype of Runx2+/â mice, and Twist-1 or -2 deficiency leads to premature osteoblast differentiation. Furthermore, Twist-1 overexpression inhibits osteoblast differentiation without affecting Runx2 expression. Twist proteins' antiosteogenic function is mediated by a novel domain, the Twist box, which interacts with the Runx2 DNA binding domain to inhibit its function. In vivo mutagenesis confirms the antiosteogenic function of the Twist box. Thus, relief of inhibition by Twist proteins is a mandatory event precluding osteoblast differentiation
CRISPR-UnLOCK: Multipurpose Cas9-Based Strategies for Conversion of Yeast Libraries and Strains

The development of genome-wide collections of individually manipulated strains (libraries) has allowed for high-throughput genetic screens and an emerging global view of this single-celled Eukaryote. The success of strain construction has relied on the innate ability of budding yeast to accept foreign DNA and perform homologous recombination, allowing for efficient plasmid construction (in vivo) and integration of desired sequences into the genome. The development of molecular toolkits and âintegration cassettesâ have provided fungal systems with a collection of strategies for tagging, deleting, or over-expressing target genes; typically, these consist of a C-terminal tag (epitope or fluorescent protein), a universal terminator sequence, and a selectable marker cassette to allow for convenient screening. However, there are logistical and technical obstacles to using these traditional genetic modules for complex strain construction (manipulation of many genomic targets in a single cell) or for the generation of entire genome-wide libraries. The recent introduction of the CRISPR/Cas gene editing technology has provided a powerful methodology for multiplexed editing in many biological systems including yeast. We have developed four distinct uses of the CRISPR biotechnology to generate yeast strains that utilizes the conversion of existing, commonly-used yeast libraries or strains. We present Cas9-based, marker-less methodologies for (i) N-terminal tagging, (ii) C-terminally tagging yeast genes with 18 unique fusions, (iii) conversion of fluorescently-tagged strains into newly engineered (or codon optimized) variants, and finally, (iv) use of a Cas9 âgene driveâ system to rapidly achieve a homozygous state for a hypomorphic query allele in a diploid strain. Previously, it was time-consuming to hand-edit data and then set up simulation runs to find the effect and impact of the input data on a spacecraft. MPS Editor provides the user the capability to create/edit/update models and sequences, and immediately try them out using what appears to the user as one piece of software. MPS Editor provides an integrated sequencing environment for users. It provides them with software that can be utilized during development as well as actual operations. In addition, it provides them with a single, consistent, user friendly interface. MPS Editor uses the Eclipse Rich Client Platform to provide an environment that can be tailored to specific missions. It provides the capability to create and edit, and includes an Activity Dictionary to build the simulation spacecraft models, build and edit sequences of commands, and model the effects of those commands on the spacecraft. MPS Editor is written in Java using the Eclipse Rich Client Platform. It is currently built with four perspectives: the Activity Dictionary Perspective, the Project Adaptation Perspective, the Sequence Building Perspective, and the Sequence Modeling Perspective. Each perspective performs a given task. If a mission doesn't require that task, the unneeded perspective is not added to that project's delivery. In the Activity Dictionary Perspective, the user builds the project-specific activities, observations, calibrations, etc. Typically, this is used during the development phases of the mission, although it can be used later to make changes and updates to the Project Activity Dictionary. In the Adaptation Perspective, the user creates the spacecraft models such as power, data store, etc. Again, this is typically used during development, but will be used to update or add models of the spacecraft. The Sequence Building Perspective allows the user to create a sequence of activities or commands that go to the spacecraft. It provides a simulation of the activities and commands that have been created
Field-induced Ordering in Critical Antiferromagnets
Transfer-matrix scaling methods have been used to study critical properties
of field-induced phase transitions of two distinct two-dimensional
antiferromagnets with discrete-symmetry order parameters: triangular-lattice
Ising systems (TIAF) and the square-lattice three-state Potts model (SPAF-3).
Our main findings are summarised as follows. For TIAF, we have shown that the
critical line leaves the zero-temperature, zero -field fixed point at a finite
angle. Our best estimate of the slope at the origin is . For SPAF-3 we provided evidence that the zero-field correlation
length diverges as , with , through analysis of the critical curve at plus crossover
arguments. For SPAF-3 we have also ascertained that the conformal anomaly and

Catalytic living ring-opening metathesis polymerization
In living ring-opening metathesis polymerization (ROMP), a transition-metalâcarbene complex polymerizes ring-strained olefins with very good control of the molecular weight of the resulting polymers. Because one molecule of the initiator is required for each polymer chain, however, this type of polymerization is expensive for widespread use. We have now designed a chain-transfer agent (CTA) capable of reducing the required amount of metal complex while still maintaining full control over the living polymerization process. This new method introduces a degenerative transfer process to ROMP. We demonstrate that substituted cyclohexene rings are good CTAs, and thereby preserve the âlivingâ character of the polymerization using catalytic quantities of the metal complex. The resulting polymers show characteristics of a living polymerization, namely narrow molecular-weight distribution, controlled molecular weights and block copolymer formation. This new technique provides access to well- defined polymers for industrial, biomedical and academic use at a fraction of the current costs and significantly reduced levels of residual ruthenium catalyst
Tracing controversies in hacker communities: ethical considerations for internet research
This paper reflects on the ethics of internet research on community controversies. Specifically, it focuses on controversies concerning gendered, social interaction in hacking communities. It addresses the question how internet researchers should treat and represent content that individuals controversially discussed online. While many internet sources are likewise technically public, they may yet suggest distinct privacy expectations on the part of involved individuals. In internet research, ethical decision-making regarding which online primary sources may be, e.g., referenced and quoted or require anonymisation is still ambiguous and contested. Instead of generalisable rules, the context dependence of internet research ethics has been frequently stressed. Given this ambiguity, the paper elaborates on ethical decisions and their implications by exploring the case of a controversial hackerspaces.org mailing list debate. Performing masculinity, influencing health: A qualitative mixed-methods study of young Spanish men
Background: The literature shows how gender mandates contribute to differences in exposure and vulnerability to certain health risk factors. This paper presents the results of a study developed in the south of Spain, where research aimed at understanding men from a gender perspective is still limited.Objective: The aim of this paper is to explore the lay perceptions and meanings ascribed to the idea of
masculinity, identifying ways in which gender displays are related to health.Design: The study is based on a mixed-methods data collection strategy typical of qualitative research.
We performed a qualitative content analysis focused on manifest and latent content.Results: Our analysis showed that the relationship between masculinity and health was mainly defined with regard to behavioural explanations with an evident performative meaning. With regard to issues such as driving, the use of recreational drugs, aggressive behaviour, sexuality, and body image, important connections were established between manhood acts and health outcomes. Different ways of understanding and performing the male identity also emerged from the results. The findings revealed the implications of these aspects in the processes of change in the identity codes of men and women.Conclusions: The study provides insights into how the category âmanâ is highly dependent on collective practices and performative acts. Consideration of how males perform manhood acts might be required in guidance on the development of programmes and policies aimed at addressing gender inequalities in health in a particular local context.This research received no specific grant from
