    AChBP-targeted α-conotoxin correlates distinct binding orientations with nAChR subtype selectivity

    Neuronal nAChRs are a diverse family of pentameric ion channels with wide distribution throughout cells of the nervous and immune systems. However, the role of specific subtypes in normal and pathological states remains poorly understood due to the lack of selective probes. Here, we used a binding assay based on acetylcholine-binding protein (AChBP), a homolog of the nicotinic acetylcholine ligand-binding domain, to discover a novel α-conotoxin (α-TxIA) in the venom of Conus textile. α-TxIA bound with high affinity to AChBPs from different species and selectively targeted the α3ÎČ2 nAChR subtype. A co-crystal structure of Ac-AChBP with the enhanced potency analog TxIA(A10L), revealed a 20° backbone tilt compared to other AChBP–conotoxin complexes. This reorientation was coordinated by a key salt bridge formed between Arg5 (TxIA) and Asp195 (Ac-AChBP). Mutagenesis studies, biochemical assays and electrophysiological recordings directly correlated the interactions observed in the co-crystal structure to binding affinity at AChBP and different nAChR subtypes. Together, these results establish a new pharmacophore for the design of novel subtype-selective ligands with therapeutic potential in nAChR-related diseases

    MATHVIS is a researched and developed software application that is capable of uplifting the educational background of visually impaired children. Education which is a knowledge gaining activity that all individuals cherish throughout their lives and enables access to a career path is not confined to any age, gender or culture. This research focuses on providing access to education for all, including the visually impaired. MATHVIS has been developed using game based learning properties, mainly targeting the visually impaired. Several resources that facilitate learning for the visually impaired were identified. However, it was noted that resources were limited to learn mathematics using local languages for visually impaired children in Sri Lanka. Hence, the aim of the research was to develop the mathematical knowledge of visually impaired Sri Lankan students. Since the target user group for this game is visually impaired children, the requirements were based on their capabilities and requirements. Sound was a key feature used to facilitate communicating with visually impaired students. Therefore, user input, selection of options and other facilities were provided along with sound tracks and a menu to enable selecting native languages such as Sinhala and English. The game consists of three levels made up of simple maths concepts such as addition, subtraction, number series and equations as well as scenario based questions. Level one consists of simple addition and subtraction questions. Supporting the requirements of the user, the questions for this level are mainly one or two digit number calculations. Level two consists of two sections; the first includes questions from the number series and the second includes questions from equations. In section one, the user is provided with a blank space to input the answer where as in the second, the user calculates the value of a given unknown letter. Level three consists of scenario based questions where the user has to listen and understand the scenario to answer the relevant question correctly. The game was developed mainly using Java Language and applications such as Netbeans 6.0. Microsoft Access was used to store data. The only way in which a visually impaired child can communicate with this application is through sound. Therefore, this game uses voice instructions with the help of software such as Text Aloud and Jet Audio. The evaluators for the game were from different categories, namely; technical evaluators, the users of the system, visually impaired students and instructors for visually impaired. The students requested incorporating changes that would enhance the language selection process. The instructors recommended limiting the number range to two digits. All evaluators provided positive comments about functionalities and ease of use of the tool. This paper discusses features of the application, evaluation carried out and the technology and learning pedagogy used to develop the application. The paper also looks at how this application can be used to provide education for all, including visually impaired children

    Respiratory syncytial virus infection, TLR3 ligands, and proinflammatory cytokines induce CD161 ligand LLT1 expression on the respiratory epithelium

    D. Lyles (Editeur)During respiratory-virus infection, excessive lymphocyte activation can cause pathology both in acute infection and in exacerbations of chronic respiratory diseases. The costimulatory molecule CD161 is expressed on lymphocyte subsets implicated in promoting respiratory inflammation, including Th2, Th17, mucosally associated invariant T (MAIT) cells, and type 2 innate lymphoid cells. We asked whether the CD161 ligand LLT1 could be expressed on respiratory epithelial cells following respiratory-virus infection as a mechanism by which respiratory-virus infection could promote activation of proinflammatory lymphocytes. In response to respiratory syncytial virus (RSV) infection, expression of LLT1 was upregulated in the BEAS-2B respiratory epithelial cell line and primary human bronchial epithelial cells. Imaging studies revealed that LLT1 expression increased in both RSV-infected and cocultured uninfected cells, suggesting that soluble factors produced during infection stimulate LLT1 expression. TLR3 and TLR2/6 ligands led to a rapid increase in LLT1 mRNA in respiratory epithelial cells, as did the proinflammatory cytokines type I interferons, interleukin 1 beta (IL-1 beta), and tumor necrosis factor alpha (TNF-alpha), which are produced early in respiratory-virus infection. Immunohistochemistry confirmed the increase in LLT1 protein on the epithelial cell surface, and live-cell confocal microscopy demonstrated accumulation of epithelial LLT1 at synapses formed with CD161(+) T lymphocytes. LLT1 expression by the respiratory epithelium in response to respiratory-virus infection and inflammatory cytokines represents a novel link between innate immunity and lymphocyte activation. As a regulator of CD161(+) proinflammatory lymphocytes, LLT1 could be a novel therapeutic target in inflammation caused by respiratory-virus infection

    Molluscs of the genus Conus (cone shells) are carnivorous, feeding on marine worms, small fish and other marine molluscs. They capture their prey by injecting venom containing hundreds of neurally active peptide components. These peptides are classed as conotoxins and consist of small disulfide-bonded peptides exhibiting a high degree of post-translational modifications (PTMs). The functional roles of these modifications remain largely unknown. Two of the most frequently observed modifications are Îł-carboxylation of glutamate and hydroxylation of proline (Buczek et al. Cell Mol Life Sci 62:3067, 2005). Vc1.1 is an α-conotoxin from Conus victoriae (Sandall et al. Biochemistry 42(22):6904–6911, 2003) and the only form of this peptide which has been detected in the venom is the Îł-glutamate and hydroxyproline (Vc1.1.P6O:E14- Gla) version of the molecule (Jakubowski et al. Toxicon 47(6):688–699, 2006). In order to investigate the role of PTMs, we did mass spectral profiling of the venom duct of C. victoriae looking at changes in mass and the number of peptides detected. We synthesised a number of predicted Vc1.1-PTM peptides together with the three possible disulfide isoforms of Vc1.1 and assessed the possible functional role of the PTM conopeptides by measuring the in vitro activity at the cognate neuronal nicotinic acetylcholine receptors (nAChRs). In addition we looked for their presence Vc1.1 venom by mass spectrometry and by this approach we were able to detect unmodified Vc1.1 in C. victoriae venom for the first time.A. Townsend, B.G.
