The influence of post-fixation on visualising vimentin in the retina using immunofluorescence method

    Get PDF
Background: Post-fixation of sections is especially required for cryostat sections of fresh frozen tissues. Vimentin is an intermediate filament in both fibrillary and non-fibrillary form, expressed in Müller’s cells and astrocytes of the retina. Our aim was to determine the best post-fixation method for visualising vimentin in archival mouse eyes. Materials and methods: We used an archival mouse eye, slightly pre-fixed with paraformaldehyde and stored at –80°C for 4 years. We tried three fixatives (paraformaldehyde [PFA], alcohol/acetic acid [AAA] and methanol) for post-fixation of eye sections. Results: We showed that post-fixation alters the labelling properties of vimentin expressed in the retina. In the sections with no post-fixation, vimentin positivity was observed in and around the nuclei in non-fibrillary form. In PFA post-fixed sections, the vimentin in the retina was not observed as fibrils. Positivity was observed in the nuclei and in perinuclear regions of the cells. In AAA post-fixed sections, positive labelling was observed around the nuclei as fibrils. In methanol post-fixed sections, labelling was observed around the nuclei as fibrils. Conclusions: We conclude that post-fixation with AAA is more convenient for immunofluorescent labelling of vimentin in the retina for slightly PFA pre-fixed and long-term stored retina. A Real Time Image Processing Subsystem: GEZGIN

    Get PDF
In this study, a real-time image processing subsystem, GEZGIN, which is currently being developed for BILSAT-1, a 100kg class micro-satellite, is presented. BILSAT-1 is being constructed in accordance with a technology transfer agreement between TÜBITAK-BILTEN (Turkey) and SSTL (UK) and planned to be placed into a 650 km sunsynchronous orbit in Summer 2003. GEZGIN is one of the two Turkish R&D payloads to be hosted on BILSAT-1. One of the missions of BILSAT-1 is constructing a Digital Elevation Model of Turkey using both multi-spectral and panchromatic imagers. Due to limited down-link bandwidth and on-board storage capacity, employment of a realtime image compression scheme is highly advantageous for the mission. GEZGIN has evolved as an implementation to achieve image compression tasks that would lead to an efficient utilization of both the down-link and on-board storage. The image processing on GEZGIN includes capturing of 4-band multi-spectral images of size 2048x2048 8- bit pixels, compressing them simultaneously with the new industry standard JPEG2000 algorithm and forwarding the compressed multi-spectral image to Solid State Data Recorders (SSDR) of BILSAT-1 for storage and down-link transmission. The mission definition together with orbital parameters impose a 6.5 seconds constraint on real-time image compression. GEZGIN meets this constraint by exploiting the parallelism among image processing units and assigning compute intensive tasks to dedicated hardware. The proposed hardware also allows for full reconfigurability of all processing units

The present study was conducted to explore the role of different types of vermicomposts (VCs) prepared from different substrates to improve soil health (physical and chemical properties) and wheat plant growth under field conditions. Different combinations of vermicompost prepared from different substrates (cow dung, paper waste, and rice straw) and inorganic fertilizers were applied in soil using wheat as a test plant. The impact of three different VCs on physico-chemical characteristics and nutrient availability in soil was evaluated to examine their efficacy in combination with chemical fertilizers. Temporal trends in vermicomposting treatments at various stages showed significant improvement in physico-chemical attributes of the VCs substrates. All the plant physiological attributes showed significant response where N: P: K 100:50:50 kg ha?1 + 10 t ha?1 cow dung vermicompost was applied. In addition, post-harvest analysis of soil not only revealed that different combinations of the vermicomposting treatments improved the soil health by improving the physico-chemical attributes of the soil. Conclusively, application of cow dung vermicompost along with recommended NPK not only improved crop yield, soil health, reduced insect (aphid) infestation but also fortified grains with Zn and Fe. © 2019 by the authors.7527, 9017; Provozn? Quasi-free Compton Scattering from the Deuteron and Nucleon Polarizabilities

Cross sections for quasi-free Compton scattering from the deuteron were measured for incident energies of 236--260 MeV at the laboratory angle -135 degrees. The recoil nucleons were detected in a liquid-scintillator array situated at 20 degrees. The measured differential cross sections were used, with the calculations of Levchuk et al., to determine the polarizabilities of the bound nucleons. For the bound proton, the extracted values were consistent with the accepted value for the free proton. Combining our results for the bound neutron with those from Rose et al., we obtain one-sigma constraints of alpha_n = 7.6-14.0 and beta_n = 1.2-7.6.Comment: 4 pages, 3 figures, accepted in PR

In this study, the cyanido complexes given by the formula [Ni(Abut)Ni(CN)4]·8H2O (C1), [Cu(Abut)2Ni(CN)4]·7H2O (C2), [Zn(Abut)Ni(CN)4]·8H2O (C3) and [Cd(Abut)Ni(CN)4]·7H2O (C4) were obtained by microwave synthesis method. The powder forms of the complexes were characterized by elemental, FT-IR spectroscopy, and thermal analysis. And also antibacterial, antibiofilm and anticancer activities were investigated. The splitting stretching bands of cyanido groups in the FT-IR spectra of C1-C4 indicated the assets of terminal and end cyanido groups. The antibacterial activities of C1-C4 were tested with nine Gram negative and six Gram positive bacteria. The most efficient antibacterial activity of complexes was observed at 1000 µg/ml-1 concentration. Anticancer activity was tested using HeLa cell line and MTT test. The studied cyanide complexes have been shown to decrease the viability of HeLa cells with IC50 values 14.86, 6.5, 7.2 and 19.2 µg/ml for C1, C2, C3 and C4 complex, respectively

In this study, we practically reviewed the negative effects of social networks on today’s youth which became somewhat indispensable to them. The study was based on the university students attending public and private universities and the high-school students. The number of participants included in the study is 894 individuals in total. The study was completed in an eight month period. Within this period, specific student groups were asked to fill in a survey composed of eighty questions on their own free will and choices. While the survey had two sections, the 60 questions in the first section were prepared in a 5-point Likert scale. In the first section, questions towards the use of social networks were answered. The last 20 questions were demographic which were arranged as multiple-choice questions. The data from the survey were analyzed via the statistical program SPSS 15. The results of the analysis and findings were presented as tables and explanations at the end of the study. This study contributes to the understanding of social status and problems experienced by today’s youth related to the social networking web pages. At the end of the research, we came to the conclusion that social networks had a negative effect on the youth

Utilisation of Infrared Stabilised Immature Rice Grains in a Cereal Based Fermented Food: Tarhana

    Get PDF
Immature rice grain is one of the under-valued by-products of rice milling industry. In this study, it was aimed to investigate the effects of immature rice grain flour (IRGF) substitution to tarhana, which is a traditional cereal-based, fermented food product. IRGF was replaced by refined wheat flour at the levels of 0 (control), 30, 50, 70, and 100%, and the effect of this replacement on the contents of crude fat, ash, protein, dietary fibre, phytic acid, minerals, vitamins B1 and B2, tocopherols, γ-oryzanol contents and sensory properties were investigated. It was shown that stabilised IRGF remarkably improved nutritional properties of the end product without adversely affecting sensory quality up to 50%. In conclusion, it could be suggested that stabilised IRGF has a promising potential for many food applications due to being cheap, accessible, nutritious, and gluten-free
