    Towards Healthy Diets for parents: efectiveness of a counselling intervention Eveline J.C. Hooft van Huysduynen Abstract Introduction and Objective: As parents’ modelling of dietary behaviour is one of the factors influencing children’s diets, improving parents’ diets is expected to result in improved dietary intake of their children. This thesis describes research that was conducted to develop and evaluate a counselling intervention to improve parental adherence to the Dutch dietary guidelines. Methods: A counselling intervention was developed, which was underpinned with the theory of planned behaviour and the transtheoretical model. In 20 weeks, five face-to-face counselling sessions were provided by a registered dietician who used motivational interviewing to improve parental adherence to the Dutch dietary guidelines. In addition, parents received three individually tailored email messages. During the counselling, the dietary guidelines and additional eating behaviours, that were hypothesized to affect diet quality, were addressed. The intervention was evaluated in a randomised controlled trial with 92 parents receiving the counselling and 94 parents as controls. Effects on dietary intake, biomarkers, intermediate markers of health and children’s dietary intake were evaluated. With mediation analyses, it was investigated if changes in dietary intake were established via changes in behavioural determinants. Thereby, it was also examined if spot urine samples could be used to replace 24 h urine samples for evaluating changes in sodium and potassium intake. Results: The intervention group increased their adherence to the dietary guidelines, as assessed with the Dutch Healthy Diet-index (ranging from 0 to 100 points), by 6.7 points more than the control group did. This improvement was achieved by small increases in the scores of seven out of ten index components. The most substantial changes were shown in fruit and fish intakes of which increases in fish intake were reflected in changes in fatty acid profiles derived from blood plasma. Also a small decrease in waist circumference was observed. Based on parental reports, the children in the intervention group increased their intakes of fruit, vegetables and fish more than the children in the control group. Improvements in parental fruit intake were mediated by changes in the behavioural determinants attitude and habit strength. Decreases in snack intake were mediated by changes in self-identity as a healthy eater. Although the results of a study in young Caucasian women showed that spot urine can be used to rank individuals for their ratios of sodium to potassium, no intervention effects on these ratios were observed. Conclusion: This thesis provides empirical knowledge on potential effective elements for counselling interventions aiming at improving the dietary pattern as a whole of parents and provides knowledge on methods to evaluate changes in dietary intake.</p

    Genomic analysis reveals selection for Asian genes in European pigs following human-mediated introgression

    The independent domestication of local wild boar populations in Asia and Europe about 10,000 years ago led to distinct European and Asian pig breeds, each with very different phenotypic characteristics. During the Industrial Revolution, Chinese breeds were imported to Europe to improve commercial traits in European breeds. Here we demonstrate the presence of introgressed Asian haplotypes in European domestic pigs and selection signatures on some loci in these regions, using whole genome sequence data. The introgression signatures are widespread and the Asian haplotypes are rarely fixed. The Asian introgressed haplotypes are associated with regions harbouring genes involved in meat quality, development and fertility. We identify Asian-derived non-synonymous mutations in the AHR gene that associate with increased litter size in multiple European commercial lines. These findings demonstrate that increased fertility was an important breeding goal for early nineteenth century pig farmers, and that Asian variants of genes related to this trait were preferentially selected during the development of modern European pig breeds

    New investigations around CYP11A1 and its possible involvement in an androstenone QTL characterised in Large White pigs

    <p>Abstract</p> <p>Background</p> <p>Previously, in boars with extreme androstenone levels, differential expression of the <it>CYP11A1 </it>gene in the testes has been characterised. <it>CYP11A1 </it>is located in a region where a QTL influencing boar fat androstenone levels has been detected in a Large White pig population. Clarifying the role of CYP11A1 in boar taint is important because it catalyses the initial step of androstenone synthesis and also of steroid synthesis.</p> <p>Results</p> <p>A genome-wide association study located <it>CYP11A1 </it>at approximately 1300 kb upstream from SNP H3GA0021967, defining the centre of the region containing the QTL for androstenone variation. In this study, we partially sequenced the <it>CYP11A1 </it>gene and identified several new single nucleotide polymorphisms (SNP) within it. Characterisation of one animal, heterozygous for <it>CYP11A1 </it>testicular expression but homozygous for a haplotype of a large region containing <it>CYP11A1</it>, revealed that variation of <it>CYP11A1 </it>expression is probably regulated by a mutation located downstream from the SNP H3GA0021967. We analysed <it>CYP11A1 </it>expression in LW families according to haplotypes of the QTL region's centre. Effects of haplotypes on <it>CYP11A1 </it>expression and on androstenone accumulation were not concordant.</p> <p>Conclusion</p> <p>This study shows that testicular expression of <it>CYP11A1 </it>is not solely responsible for the QTL influencing boar fat androstenone levels. As a conclusion, we propose to refute the hypothesis that a single mutation located near the centre of the QTL region could control androstenone accumulation in fat by regulating the <it>CYP11A1 </it>expression.</p

    Genome-wide association study of meat quality traits using whole-genome sequence data in a multi-breed sheep population

    This is the first study showing GWAS results from whole-genome sequence (WGS) data in sheep. The objective of this study was to fine map previously identified QTL and detect new QTL for meat quality traits in a multi-breed sheep population using WGS data. The traits measured were carcass fat depth, intramuscular fat, post-weaning eye muscle depth, post-weaning body weight and SF5. Corrected phenotypes were available on 13,363 to 26,769 Australian sheep depending on the trait. The GWAS for all five traits for the 50K, HD, and WGS identified 18, 52 and 250 QTL regions, respectively based on a 5% FDR threshold. Together, the largest QTLs explained a substantial part of the additive genetic variance (between 10.6 - 14.3%). Previously identified QTL on chromosomes 6 and 18 were confirmed. Single breed analysis for Merino sheep only, showed a reduced significance of the large QTL, but in similar regions. We were able to identify candidate genes involved in meat quality using functional annotation. We conclude that the WGS data provided more and clearer evidence of QTL regions, potentially resulting in more predictive SNPs and more detailed functional analysis of genes underlying the QTL regions

    Large scale genome-wide association and LDLA mapping study identifies QTLs for boar taint and related sex steroids

    <p>Abstract</p> <p>Background</p> <p>Boar taint is observed in a high proportion of uncastrated male pigs and is characterized by an unpleasant odor/flavor in cooked meat, primarily caused by elevated levels of androstenone and skatole. Androstenone is a steroid produced in the testis in parallel with biosynthesis of other sex steroids like testosterone and estrogens. This represents a challenge when performing selection against androstenone in breeding programs, without simultaneously decreasing levels of other steroids. The aim of this study was to use high-density genome wide association (GWA) in combination with linkage disequilibrium-linkage analysis (LDLA) to identify quantitative trait loci (QTL) associated with boar taint compounds and related sex steroids in commercial Landrace (n = 1,251) and Duroc (n = 918) breeds.</p> <p>Results</p> <p>Altogether, 14 genome wide significant (GWS) QTL regions for androstenone in subcutaneous fat were obtained from the LDLA study in Landrace and 14 GWS QTL regions in Duroc. LDLA analysis revealed that 7 of these QTL regions, located on SSC 1, 2, 3, 7 and 15, were obtained in both breeds. All 14 GWS androstenone QTLs in Landrace are also affecting the estrogens at chromosome wise significance (CWS) or GWS levels, while in Duroc, 3 of the 14 QTLs affect androstenone without affecting any of the estrogens. For skatole, 10 and 4 QTLs were GWS in the LDLA analysis for Landrace and Duroc respectively, with 4 of these detected in both breeds. The GWS QTLs for skatole obtained by LDLA are located at SSC 1, 5, 6, 7, 10, 11, 13 and 14.</p> <p>Conclusion</p> <p>This is the first report applying the Porcine 60 K SNP array for simultaneous analysis of boar taint compounds and related sex hormones, using both GWA and LDLA approaches. Several QTLs are involved in regulation of androstenone and skatole, and most of the QTLs for androstenone are also affecting the levels of estrogens. Seven QTLs for androstenone were detected in one breed and confirmed in the other, i.e. in an independent sample, although the majority of QTLs are breed specific. Most QTLs for skatole do not negatively affect other sex hormones and should be easier to implement into the breeding scheme.</p

    Entrepreneurs’ achieved success: developing a multi-faceted measure

    Firm performance is typically measured via objective financial indicators. However, researchers increasingly acknowledge that entrepreneurs do not measure their success solely in financial terms but that a range of often subjective indicators matter to them. This article contributes to the debate on entrepreneurial performance by studying how entrepreneurs assess their achieved success. ‘Entrepreneurs’ achieved success’ was conceptualized as a multi-faceted construct that includes entrepreneurs’ self-reported achievement of firm performance, workplace relationships, personal fulfilment, community impact, and personal financial rewards. It was measured via the Subjective Entrepreneurial Success–Achievement Scale (SES-AS). Over the course of three studies (N = 390) the factorial structure of ‘entrepreneurs’ achieved success’ was established and largely replicated in two cultures. Based on a nomological network, we documented relationships among ‘entrepreneurs’ achieved success’, quasi-objective indicators of firm performance, and entrepreneurs’ financial satisfaction, creativity, and health. Based on our research, we propose a new conceptual framework to study performance in the context of entrepreneurship. This framework acknowledges both the success criteria that entrepreneurs wish to achieve and those that they actually achieve, and extends our understanding of firm performance

    Pig farmers’ willingness to pay for management strategies to reduce aggression between pigs

    When deciding whether to invest in an improvement to animal welfare, farmers must trade-off the relative costs and benefits. Despite the existence of effective solutions to many animal welfare issues, farmers’ willingness to pay for them is largely unknown. This study modelled pig farmers’ decisions to improve animal welfare using a discrete choice experiment focused on alleviating aggression between growing/finishing pigs at regrouping. Eighty-two UK and Irish pig farm owners and managers were asked to choose between hypothetical aggression control strategies described in terms of four attributes; installation cost, on-going cost, impact on skin lesions from aggression and impact on growth rate. If they did not like any of the strategies they could opt to keep their current farm practice. Systematic variations in product attributes allowed farmers’ preferences and willingness to pay to be estimated and latent class modelling accounted for heterogeneity in responses. The overall willingness to pay to reduce lesions was low at £0.06 per pig place (installation cost) and £0.01 per pig produced (running cost) for each 1% reduction in lesions. Results revealed three independent classes of farmers. Farmers in Class 1 were unlikely to regroup unfamiliar growing/finishing pigs, and thus were unwilling to adopt measures to reduce aggression at regrouping. Farmers in Classes 2 and 3 were willing to adopt measures providing certain pre-conditions were met. Farmers in Class 2 were motivated mainly by business goals, whilst farmers in Class 3 were motivated by both business and animal welfare goals, and were willing to pay the most to reduce aggression; £0.11 per pig place and £0.03 per pig produced for each 1% reduction in lesions. Farmers should not be considered a homogeneous group regarding the adoption of animal welfare innovations. Instead, campaigns should be targeted at subgroups according to their independent preferences and willingness to pay

    Fine mapping of a QTL affecting levels of skatole on pig chromosome 7

    Abstract Background Previous studies in the Norwegian pig breeds Landrace and Duroc have revealed a QTL for levels of skatole located in the region 74.7–80.5 Mb on SSC7. Skatole is one of the main components causing boar taint, which gives an undesirable smell and taste to the pig meat when heated. Surgical castration of boars is a common practice to reduce the risk of boar taint, however, a selection for boars genetically predisposed for low levels of taint would help eliminating the need for castration and be advantageous for both economic and welfare reasons. In order to identify the causal mutation(s) for the QTL and/or identify genetic markers for selection purposes we performed a fine mapping of the SSC7 skatole QTL region. Results A dense set of markers on SSC7 was obtained by whole genome re-sequencing of 24 Norwegian Landrace and 23 Duroc boars. Subsets of 126 and 157 SNPs were used for association analyses in Landrace and Duroc, respectively. Significant single markers associated with skatole spanned a large 4.4 Mb region from 75.9–80.3 Mb in Landrace, with the highest test scores found in a region between the genes NOVA1 and TGM1 (p < 0.001). The same QTL was obtained in Duroc and, although less significant, with associated SNPs spanning a 1.2 Mb region from 78.9–80.1 Mb (p < 0.01). The highest test scores in Duroc were found in genes of the granzyme family (GZMB and GZMH-like) and STXBP6. Haplotypes associated with levels of skatole were identified in Landrace but not in Duroc, and a haplotype block was found to explain 2.3% of the phenotypic variation for skatole. The SNPs in this region were not associated with levels of sex steroids. Conclusions Fine mapping of a QTL for skatole on SSC7 confirmed associations of this region with skatole levels in pigs. The QTL region was narrowed down to 4.4 Mb in Landrace and haplotypes explaining 2.3% of the phenotypic variance for skatole levels were identified. Results confirmed that sex steroids are not affected by this QTL region, making these markers attractive for selection against boar taint
