    Silent synapses generate sparse and orthogonal action potential firing in adult-born hippocampal granule cells.

    In adult neurogenesis young neurons connect to the existing network via formation of thousands of new synapses. At early developmental stages, glutamatergic synapses are sparse, immature and functionally 'silent', expressing mainly NMDA receptors. Here we show in 2- to 3-week-old young neurons of adult mice, that brief-burst activity in glutamatergic fibers is sufficient to induce postsynaptic AP firing in the absence of AMPA receptors. The enhanced excitability of the young neurons lead to efficient temporal summation of small NMDA currents, dynamic unblocking of silent synapses and NMDA-receptor-dependent AP firing. Therefore, early synaptic inputs are powerfully converted into reliable spiking output. Furthermore, due to high synaptic gain, small dendritic trees and sparse connectivity, neighboring young neurons are activated by different distinct subsets of afferent fibers with minimal overlap. Taken together, synaptic recruitment of young neurons generates sparse and orthogonal AP firing, which may support sparse coding during hippocampal information processing

    Synaptic Integration of Adult-Born Hippocampal Neurons Is Locally Controlled by Astrocytes.

    Adult neurogenesis is regulated by the neurogenic niche, through mechanisms that remain poorly defined. Here, we investigated whether niche-constituting astrocytes influence the maturation of adult-born hippocampal neurons using two independent transgenic approaches to block vesicular release from astrocytes. In these models, adult-born neurons but not mature neurons showed reduced glutamatergic synaptic input and dendritic spine density that was accompanied with lower functional integration and cell survival. By taking advantage of the mosaic expression of transgenes in astrocytes, we found that spine density was reduced exclusively in segments intersecting blocked astrocytes, revealing an extrinsic, local control of spine formation. Defects in NMDA receptor (NMDAR)-mediated synaptic transmission and dendrite maturation were partially restored by exogenous D-serine, whose extracellular level was decreased in transgenic models. Together, these results reveal a critical role for adult astrocytes in local dendritic spine maturation, which is necessary for the NMDAR-dependent functional integration of newborn neurons

    The Physics of Heavy Flavours at SuperB

    This is a review of the SuperB project, covering the accelerator, detector, and highlights of the broad physics programme. SuperB is a flavour factory capable of performing precision measurements and searches for rare and forbidden decays of Bu,d,sB_{u,d,s}, DD, τ\tau and Υ(nS)\Upsilon({\mathrm{nS}}) particles. These results can be used to test fundamental symmetries and expectations of the Standard Model, and to constrain many different hypothesised types of new physics. In some cases these measurements can be used to place constraints on the existence of light dark matter and light Higgs particles with masses below 10GeV/c210GeV/c^2. The potential impact of the measurements that will be made by SuperB on the field of high energy physics is also discussed in the context of data taken at both high energy in the region around the \Upsilon({\mathrm{4S}})$, and near charm threshold.Comment: 49 pages, topical review submitted to J.     Trypan Blue Dye Enters Viable Cells Incubated with the Pore-Forming Toxin HlyII of Bacillus cereus

    Trypan blue is a dye that has been widely used for selective staining of dead tissues or cells. Here, we show that the pore-forming toxin HlyII of Bacillus cereus allows trypan blue staining of macrophage cells, despite the cells remaining viable and metabolically active. These findings suggest that the dye enters viable cells through the pores. To our knowledge, this is the first demonstration that trypan blue may enter viable cells. Consequently, the use of trypan blue staining as a marker of vital status should be interpreted with caution. The blue coloration does not necessarily indicate cell lysis, but may rather indicate pore formation in the cell membranes and more generally increased membrane permeability

    Measurements of Charmless Hadronic b->s Penguin Decays in the pi+pi-K+pi- Final State and First Observation of B0 -> rho0K+pi-

    We report measurements of charmless hadronic B^0 decays into the pi+pi-K+pi+ final state. The analysis uses a sample of 657x10^6 BBbar pairs collected with the Belle detector at the KEKB asymmetric-energy e+e- collider at the Y(4S) resonance. The decay B^0 -> rho0 Kpi is observed for the first time; the significance is 5.0sigma and the corresponding partial branching fraction for M_Kpi in (0.75,1.20) GeV/c^2 is [2.8 +- 0.5(stat) +-0.5(syst)] x 10^{-6}. We also obtain the first evidence for B^0 -> f0Kpi with 3.5sigma significance and for B^0 -> pi+pi-K*0 with 4.5sigma significance. For the two-body decays B^0 -> rho0K*0 and B^0 -> f0K*0, the significances are 2.7sigma and 2.5sigma, respectively, and the upper limits on the branching fractions are 3.4x10^{-6} and 2.2x10^{-6} at 90% confidence level.Comment: 6 pages, 3 figures. accepted by PRD(RC

    Search for CP violation in the decays D(s)+KS0π+D^+_{(s)} \to K_S^0\pi^+ and D(s)+KS0K+D^+_{(s)} \to K_S^0K^+

    We have searched for CP violation in the charmed meson decays D(s)+KS0π+D^{+}_{(s)}\to K^0_S\pi^+ and D(s)+KS0K+D^{+}_{(s)}\to K^0_S K^+ using 673 fb1^{-1} of data collected with the Belle detector at the KEKB asymmetric-energy e+ee^+e^- collider. No evidence for CP violation is observed. We report the most sensitive CP asymmetry measurements to date for these decays: ACPD+KS0π+=(0.71±0.19±0.20)A_{CP}^{D^+\to K^0_S\pi^+}=(-0.71\pm0.19\pm0.20)%, ACPDs+KS0π+=(+5.45±2.50±0.33)A_{CP}^{D^+_s\to K^0_S\pi^+}=(+5.45\pm2.50\pm0.33)%, ACPD+KS0K+=(0.16±0.58±0.25)A_{CP}^{D^+\to K^0_S K^+}=(-0.16\pm0.58\pm0.25)%, and ACPDs+KS0K+=(+0.12±0.36±0.22)A_{CP}^{D^+_s\to K^0_S K^+}=(+0.12\pm0.36\pm0.22)%, where the first uncertainties are statistical and the second are systematic

    We report an improved measurement of D+KS0K+D^+ \to K_S^0 K^+ and Ds+KS0π+D^+_s\to K_S^0 \pi^+ branching ratios using 605 fb1^{-1} of data collected with the Belle detector at the KEKB asymmetric-energy e+ee^+ e^- collider. The measured branching ratios with respect to the Cabibbo-favored modes are B(D+KS0K+)/B(D+KS0π+)\mathcal{B}(D^+ \to K_S^0 K^+)/\mathcal{B}(D^+ \to K_S^0 \pi^+) = 0.1899±0.0011±\pm0.0011\pm0.0022 and B(Ds+KS0π+)/B(Ds+KS0K+)\mathcal{B}(D^+_s \to K_S^0 \pi^+)/\mathcal{B}(D^+_s \to K_S^0 K^+) = 0.0803±0.0024±\pm0.0024\pm0.0019 where the first uncertainties are statistical and the second are systematic

    Evidence for the Suppressed Decay B- -> DK-, D -> K+pi-

    The suppressed decay chain B- -> DK-, D -> K+pi-, where D indicates a anti-D0 or D0 state, provides important information on the CP-violating angle phi_3. We measure the ratio R_{DK} of the decay rates to the favored mode B- -> DK-, D -> K-pi+ to be R_{DK} = [1.63^{+0.44}_{-0.41}(stat)^{+0.07}_{-0.13}(syst)] x 10^{-2}, which indicates the first evidence of the signal with a significance of 4.1sigma. We also measure the asymmetry A_{DK} between the charge-conjugate decays to be A_{DK} = -0.39^{+0.26}_{-0.28}(stat)^{+0.04}_{-0.03}(syst). The results are based on the full 772 x 10^6 B anti-B pair data sample collected at the Upsilon(4S) resonance with the Belle detector.Comment: 6 pages, 2 figures, 2 tables, accepted by Physical Review Letter

    Measurement Of |V_ub| From Inclusive Charmless Semileptonic B Decays

    We present the partial branching fraction for inclusive charmless semileptonic B decays and the corresponding value of the CKM matrix element |Vub|, using a multivariate analysis method to access ~90% of the B -> Xu l nu phase space. This approach dramatically reduces the theoretical uncertainties from the b-quark mass and non-perturbative QCD compared to all previous inclusive measurements. The results are based on a sample of 657 million B -Bbar pairs collected with the Belle detector. We find that Delta BR(B -> Xu l nu; p^*B_l>1.0 GeV/c=1.963 x (1 +/- 0.088(stat.) +/- 0.081(sys.)) x 10^-3. Corresponding values of |Vub| are extracted using several theoretical calculations.Comment: 9 pages, 1 figure, 2 tables. Published in PR
