    SPEXOR passive spinal exoskeleton decreases metabolic cost during symmetric repetitive lifting

    PURPOSE: Besides mechanical loading of the back, physiological strain is an important risk factor for low-back pain. Recently a passive exoskeleton (SPEXOR) has been developed to reduce loading on the low back. We aimed to assess the effect of this device on metabolic cost of repetitive lifting. To explain potential effects, we assessed kinematics, mechanical joint work, and back muscle activity. METHODS: We recruited ten male employees, working in the luggage handling department of an airline company and having ample experience with lifting tasks at work. Metabolic cost, kinematics, mechanical joint work and muscle activity were measured during a 5-min repetitive lifting task. Participants had to lift and lower a box of 10 kg from ankle height with and without the exoskeleton. RESULTS: Metabolic cost was significantly reduced by 18% when wearing the exoskeleton. Kinematics did not change significantly, while muscle activity decreased by up to 16%. The exoskeleton took over 18-25% of joint work at the hip and L5S1 joints. However, due to large variation in individual responses, we did not find a significant reduction of joint work around the individual joints. CONCLUSION: Wearing the SPEXOR exoskeleton decreased metabolic cost and might, therefore, reduce fatigue development and contribute to prevention of low-back pain during repetitive lifting tasks. Reduced metabolic cost can be explained by the exoskeleton substituting part of muscle work at the hip and L5S1 joints and consequently decreasing required back muscle activity

    Early neoplastic and metastatic mammary tumours of transgenic mice detected by 5-aminolevulinic acid-stimulated protoporphyrin IX accumulation

    A photodynamic technique for human breast cancer detection founded upon the ability of tumour cells to rapidly accumulate the fluorescent product protoporphyrin IX (PpIX) has been applied to transgenic mouse models of mammary tumorigenesis. A major goal of this investigation was to determine whether mouse mammary tumours are reliable models of human disease in terms of PpIX accumulation, for future mechanistic and therapeutic studies. The haeme substrate 5-aminolevulinic acid (5-ALA) (200 mg kg−1) was administered to mouse strains that develop mammary tumours of various histological subtypes upon expression of the transgenic oncogenes HRAS, Polyoma Virus middle T antigen, or Simian Virus 40 large T antigen in the mammary gland. Early neoplastic lesions, primary tumours and metastases showed consistent and rapid PpIX accumulation compared to the normal surrounding tissues, as evidenced by red fluorescence (635 nm) when the tumours were directly illuminated with blue light (380–440 nm). Detection of mouse mammary tumours at the stage of ductal carcinoma in situ by red fluorescence emissions suggests that enhanced PpIX synthesis is a good marker for early tumorigenic processes in the mammary gland. We propose the mouse models provide an ideal experimental system for further investigation of the early diagnostic and therapeutic potential of 5-ALA-stimulated PpIX accumulation in human breast cancer patients

    Characteristics and drivers of forest cover change in the post-socialist era in Croatia: evidence from a mixed-methods approach

    © 2016, Springer-Verlag Berlin Heidelberg.Extensive forests in Croatia represent an important biological and economic resource in Europe. They are characterised by heterogeneity in forest management practices dating back to the socialist planned economy of the pre-1991 era. In this study we investigated the difference in rates of deforestation and reforestation in private- and state-owned forests during the post-socialist period and the causal drivers of change. The selected region of Northern Croatia is characterised by a high percentage of privately owned forests with minimal national monitoring and control. We used a mixed-methods approach which combines remote sensing, statistical modelling and a household-based questionnaire survey to assess the rates of forest cover change and factors influencing those changes. The results show that predominantly privately owned forests in Northern Croatia have recorded a net forest loss of 1.8 % during the 1991–2011 period, while Croatia overall is characterised by a 10 % forest cover increase in predominantly state-owned forests. Main factors influencing forest cover changes in private forests are slope, altitude, education structure, population age and population density. The results also show that the deforestation in private forests is weakening overall, mostly due to the continuation of the de-agrarisation and de-ruralisation processes which began during socialism

    De novo characterization of the gametophyte transcriptome in bracken fern, Pteridium aquilinum

    <p>Abstract</p> <p>Background</p> <p>Because of their phylogenetic position and unique characteristics of their biology and life cycle, ferns represent an important lineage for studying the evolution of land plants. Large and complex genomes in ferns combined with the absence of economically important species have been a barrier to the development of genomic resources. However, high throughput sequencing technologies are now being widely applied to non-model species. We leveraged the Roche 454 GS-FLX Titanium pyrosequencing platform in sequencing the gametophyte transcriptome of bracken fern (<it>Pteridium aquilinum</it>) to develop genomic resources for evolutionary studies.</p> <p>Results</p> <p>681,722 quality and adapter trimmed reads totaling 254 Mbp were assembled <it>de novo </it>into 56,256 unique sequences (i.e. unigenes) with a mean length of 547.2 bp and a total assembly size of 30.8 Mbp with an average read-depth coverage of 7.0×. We estimate that 87% of the complete transcriptome has been sequenced and that all transcripts have been tagged. 61.8% of the unigenes had blastx hits in the NCBI nr protein database, representing 22,596 unique best hits. The longest open reading frame in 52.2% of the unigenes had positive domain matches in InterProScan searches. We assigned 46.2% of the unigenes with a GO functional annotation and 16.0% with an enzyme code annotation. Enzyme codes were used to retrieve and color KEGG pathway maps. A comparative genomics approach revealed a substantial proportion of genes expressed in bracken gametophytes to be shared across the genomes of <it>Arabidopsis</it>, <it>Selaginella </it>and <it>Physcomitrella</it>, and identified a substantial number of potentially novel fern genes. By comparing the list of <it>Arabidopsis </it>genes identified by blast with a list of gametophyte-specific <it>Arabidopsis </it>genes taken from the literature, we identified a set of potentially conserved gametophyte specific genes. We screened unigenes for repetitive sequences to identify 548 potentially-amplifiable simple sequence repeat loci and 689 expressed transposable elements.</p> <p>Conclusions</p> <p>This study is the first comprehensive transcriptome analysis for a fern and represents an important scientific resource for comparative evolutionary and functional genomics studies in land plants. We demonstrate the utility of high-throughput sequencing of a normalized cDNA library for <it>de novo </it>transcriptome characterization and gene discovery in a non-model plant.</p

    Threat memory reminder under matrix metalloproteinase 9 inhibitor doxycycline globally reduces subsequent memory plasticity

    Associative memory can be rendered malleable by a reminder. Blocking the ensuing re-consolidation process is suggested as a therapeutic target for unwanted aversive memories. Matrix metalloproteinase (MMP)-9 is required for structural synapse remodelling involved in memory consolidation. Inhibiting MMP-9 with doxycycline is suggested to attenuate human threat conditioning. Here, we investigate whether MMP-9 inhibition also interferes with threat memory re-consolidation. N=78 male and female human participants learned the association between two visual conditioned stimuli (CS+) and a 50% chance of an unconditioned nociceptive stimulus (US), and between CS- and the absence of US. On day 7, one CS+ was reminded without reinforcement 3.5 hours after ingesting either 200 mg doxycycline, or placebo. On day 14, retention of CS memory was assessed under extinction, by fear-potentiated startle. Contrary to our expectations, we observed a greater CS+/CS- difference in participants who were reminded under doxycycline, compared to placebo. Participants who were reminded under placebo showed extinction learning during the retention test, which was not observed in the doxycycline group. There was no difference between the reminded and the non-reminded CS+ in either group. In contrast, during re-learning after the retention test, CS+/CS- difference was more pronounced in the placebo than the doxycycline group. To summarize, a single dose of doxycycline appeared to have no specific impact on re-consolidation, but to globally impair extinction learning, and threat re-learning, after drug clearance.SIGNIFICANCE STATEMENTMMP-9 inhibition appears to attenuate memory consolidation. It could also be a target for blocking reconsolidation. Here, we test this hypothesis in human threat conditioning. We find that doxycycline has no specific impact on a reminded cue, but confers a global reduction in extinction learning and threat learning beyond the clearance of the drug. This may point towards a more long-lasting impact of doxycycline treatment on memory plasticity

    In order to better understand the mechanisms of learning disabilities it is important to evaluate the electroencephalogram parameters and their relation to the results of the Wechsler Intelligence Scale. Thirty-six children with complaints of learning disability were studied. Electroencephalograms were carried out while awake and resting, and the values for absolute and relative powers calculated. The results were compared with those of 36 healthy children paired with respect to age, gender and maternal scholastic level. In the group with learning disabilities, the absolute (in the delta, theta and alpha 1 bands) and relative (theta) power values were higher and the relative power alpha 2 value significantly lower at the majority of the electrodes in relation to the control group. There was a high positive correlation in the children with learning disabilities between the relative power alpha 2 and the verbal, performance and total IQ values. These quantitative electroencephalogram findings in children with learning disabilities have a clear relation with psychological measurements and could be due to brain immaturity.Para compreender melhor os mecanismos das dificuldades de aprendizagem Ă© importante avaliar a relação entre parĂąmetros do eletrencefalograma e resultados da Escala Weschler de InteligĂȘncia. Foram estudadas 36 crianças com queixas de dificuldades de aprendizagem. Foi realizado o eletrencefalograma durante vigĂ­lia, em repouso e calculados os valores de potĂȘncia absoluta e relativa. Os resultados foram comparados aos de 36 crianças sadias pareadas quanto a idade, gĂȘnero e escolaridade materna. As potĂȘncias absoluta (das faixas delta, teta e alfa 1) e relativa (teta) foram maiores e a potĂȘncia relativa alfa 2 foi significativamente menor, na maioria dos eletrodos, no grupo com dificuldade de aprendizagem em relação ao grupo controle. Nas crianças com dificuldade de aprendizagem houve correlação elevada e positiva entre a potĂȘncia relativa alfa 2 e o QI. Os achados do eletrencefalograma quantitativo das crianças com dificuldade aprendizagem tĂȘm nĂ­tida relação com medidas psicolĂłgicas e podem ser decorrentes de imaturidade cerebral
