    Flight test of ARINC 741 configuration low gain SATCOM system on Boeing 747-400 aircraft

    The Boeing company conducted a flight test of a SATCOM system similar to the ARINC 741 configuration on a production model 747-400. A flight plan was specifically designed to test the system over a wide variety of satellite elevations and aircraft attitudes as well as over land and sea. Interface bit errors, signal quality and aircraft position and navigational inputs were all recorded as a function of time. Special aircraft maneuvers were performed to demonstrate the potential for shadowing by aircraft structures. Both a compass rose test and the flight test indicated that shadowing from the tail is insignificant for the 747-400. However, satellite elevation angles below the aircraft horizon during banking maneuvers were shown to have a significant deleterious effect on SATCOM communications

    Dual Method Headphone Amplifier

    Many high impedance headphones underperform their full potential when directly connected to the audio source. Amplifiers boost the audio signal and provide the headphones with sufficient power to ensure their maximum performance. The invention of transistors caused vacuum tube implementation to decline, leaving many audiophiles unsatisfied with the transistor’s sound signature. Vacuum tubes and transistors both amplify signals, however the distinct “tube sound” has vanished. We have designed and created a product where the user selectively switches between solid-state transistor and tube amplification to compare the sound signatures of each amplification method. The ability to switch between the solid-state and tube amplifiers creates the ability to achieve a more customized sound for individual songs and improve the user’s listening experience. This requires the design of two separate amplifiers and circuitry to switch back and forth between amplification methods without pausing the music or unplugging any device

    End of Project ReportObjectives: (i) to develop rapid screening procedures for the determination of milk protein polymorphism (genetic variants) (ii) to determine the frequency distribution of milk protein genetic variants in a large population of Irish Holstein-Friesians and to determine if there was an association between κ-casein variant and milk yield and composition in this group of animals, and (iii) to make Cheddar and low-moisture part-skim Mozzarella cheese from different κ-casein genetic variant milks and to assess any effect on cheese yield, composition and functional characteristics. Conclusions:Analysis of 6,007 individual Irish Holstein-Friesian milks showed that the phenotype distribution of the κ-casein BB variant was very low at 1.98% compared to 53.07% for κ-casein AA and 44.95% for κ-casein AB. While no statistically significant associations were observed between κ-casein variant and milk yield and composition, κ-casein BB variant milks had superior rennet coagulation properties to that of the AA or AB variants. Generally, κ-casein variant had little effect on compositional attributes of cheese apart from FDM (fat in dry matter) which was significantly higher in cheeses from κ-casein BB milk than in those from κ-casein AA milk. Generally, κ-casein variant had no significant effects on either primary or secondary proteolysis, or on the sensory and/or textural characteristics of Cheddar or Mozzarella cheese throughout ripening; or on the functional characteristics (e.g. flow and stretch) of baked Mozzarella on storage for 90 days at 4°C. However, κ-casein BB variant milk gave significantly higher actual, and moisture adjusted yields of Cheddar and Mozzarella cheese than either κ-casein AB or AA variant milks. For example, the moisture adjusted Cheddar yield from κ-casein BB milk was 8.2% higher than from κ-casein AA milk. In the case of Mozzarella, the moisture adjusted yield was 12% higher. Based on the results, it is estimated that the actual yield of cheese in a plant producing 20,000 tonnes per year from κ-casein AA milk would increase to approximately 21,180 tonnes of Cheddar, or 21,780 tonnes of Mozzarella if made from κ-casein BB milk. Where κ-casein AB milk is used instead of κ-casein BB milk, the estimated yield of Mozzarella would increase to 21,580 tonnes.Department of Agriculture, Food and the Marin

    Profiles of Urine Samples Taken from Ecstasy Users at Rave Parties: Analysis by Immunoassays, HPLC, and GC-MS

    The abuse of the designer amphetamines such as 3,4-methylenedioxymethamphetamine (MDMA, Ecstasy) is increasing throughout the world. They have become popular drugs, especially at all-night techno dance parties (Raves), and their detection is becoming an important issue. Presently, there are no MDMA- or MDA-specific immunoassays on the market, and detection of the designer amphetamines is dependent upon the use of commercially available amphetamine assays. The success of this approach has been difficult to assess because of the general unavailability of significant numbers of samples from known drug users. The objectives of the present study are to characterize the drug content of urine samples from admitted Ecstasy users by chromatographic methods and to assess the ability of the available amphetamine/methamphetamine immunoassays to detect methylenedioxyamphetamines. We found that, when analyzed by high-performance liquid chromatography with diode-array detection (HPLC-DAD), 64% of 70 urine samples (by gas chromatography-mass spectrometry [GC-MS]: 88% of 64 urine samples) obtained from Rave attendees contained MDMA and/or 3,4-methylenedioxyamphetamine (MDA) alone or in combination with amphetamine, methamphetamine, or other designer amphetamines such as 3,4-methylenedioxyethylamphetamine (MDEA). This suggests that the majority of the Ravers are multi-drug users. At the manufacturer's suggested cutoffs, the Abbott TDx Amphetamine/Methamphetamine II and the new Roche HS Amphetamine/MDMA assays demonstrated greater detection sensitivity for MDMA than the other amphetamine immunoassays tested (Abuscreen OnLine Hitachi AMPS, Abuscreen OnLine Integra AMPS, Abuscreen OnLine Integra AMPSX, CEDIA AMPS, and EMIT II AMPS). There is 100% agreement between each of the two immunoassays with the reference chromatographic methods, HPLC-DAD and GC-MS, for the detection of methylenedioxyamphetamine

    Pharmacokinetics and Excretion of Gamma-Hydroxybutyrate (GHB) in Healthy Subjects

    In Europe and the United States, the recreational use of gammahydroxy butyric acid (GHB) at dance clubs and "rave” parties has increased substantially. In addition, GHB is used to assist in the commission of sexual assaults. The aim of this controlled clinical study was to acquire pharmacokinetic profiles, detection times, and excretion rates in human subjects. Eight GHB-naïve volunteers were administered a single 25-mg/kg body weight oral dose of GHB, and plasma, urine, and oral fluid specimens were analyzed by using gas chromatography-mass spectrometry (GC-MS). Liquid-liquid extraction was performed after acid conversion of GHB to gamma-butyrolactone. Limits of quantitation of 0.1 (oral fluid), 0.2 (urine), and 0.5 µg/mL (plasma) could be achieved in the selected ion monitoring mode. GHB plasma peaks of 39.4 ± 25.2 µg/mL (mean ± SEM) occurred 20-45 min after administration. The terminal plasma elimination half-life was 30.4 ± 2.45 min, the distribution volume 52.7 ± 15.0 L, and the total clearance 1228 ± 233 µL/min. In oral fluid, GHB could be detected up to 360 min, with peak concentrations of 203 ± 92.4 µg/mL in the 10-min samples. In urine, 200 ± 71.8 and 230 ± 86.3 µg/mL, were the highest GHB levels measured at 30 and 60 min, respectively. Only 1.2 ± 0.2% of the dose was excreted, resulting in a detection window of 720 min. Common side-effects were confusion, sleepiness, and dizziness; euphoria and change of vital functions were not observed. GHB is extensively metabolized and rapidly eliminated in urine and oral fluid.
