Physiological demands of officiating in the game of Rugby Union at the senior level
The aim of this study was to evaluate the physiological demands of officiating in
the code of Rugby Union at an elite level and to propose a relevant programme
of fitness testing and training arising from the results obtained. A group of ten
RFU National Panel referees were selected as subjects and all were analysed
performing match duties as both a referee and a touch-judge during a number of
level 1 and 2 league matches. Each subject performed a range of laboratory tests
to establish various physiological parameters, which were then compared with the
match analysis findings of heart rate and total match distances. [Continues.
Culturomics of Anaerobic Sludge
Ever increasing concerns surrounding climate change are making alternative energy sources a focus of global discussion. Anaerobic digestion holds promise as one method of generating renewable energy with a low carbon footprint. The simple principle that underlies anaerobic digestion is the degradation of organic material by microorganisms to produce digestate and biogas. While this process occurs without human intervention in a variety of environments, there is still large scope for refinement of the process in order to increase its efficiency in industry. A key problem that limits technology advancement is the understanding of the microbiomes within anaerobic digesters. This can be attributed to the fastidious growth requirements of many of the organisms involved and as such, novel methods for culturing must be adopted if we are to advance our understanding. Work undertaken used a number of different growth backgrounds and isolation chips as a novel method for the enrichment of methanogens and anaerobic fungi from anaerobic sludge samples. 16S rRNA gene sequencing technology was used to assign taxonomy to 1,152 samples. A potential novel bacterial species was further investigated using fluorescent in-situ hybridisation and Sanger sequencing with a view to demonstrating the efficacy of this culturing method for the isolation and culturing of novel microbes. This alternative approach for the cultivation of multiple microbes in isolation has shown promise in terms of the number of novel species grown. Given that 16 samples were investigated at an individual level and one of these has been identified as a novel species, it is likely that novel species have also been cultured within the other 1,136 samples that have not yet been studied individually. This experiment provides a starting point from which a more streamlined method can be developed or altered to produce different microbial populations

Human exercise-induced circulating progenitor cell mobilization is nitric oxide-dependent and is blunted in South Asian men

Copyright @ 2010 American Heart Foundation.Objectiveâ Circulating progenitor cells (CPC) have emerged as potential mediators of vascular repair. In experimental models, CPC mobilization is critically dependent on nitric oxide (NO). South Asian ethnicity is associated with reduced CPC. We assessed CPC mobilization in response to exercise in Asian men and examined the role of NO in CPC mobilization per se.
Methods and Resultsâ In 15 healthy, white European men and 15 matched South Asian men, CPC mobilization was assessed during moderate-intensity exercise. Brachial artery flow-mediated vasodilatation was used to assess NO bioavailability. To determine the role of NO in CPC mobilization, identical exercise studies were performed during intravenous separate infusions of saline, the NO synthase inhibitor l-NMMA, and norepinephrine. âFlow-mediated vasodilatation (5.8%±0.4% vs 7.9%±0.5%; P=0.002) and CPC mobilization (CD34+/KDR+ 53.2% vs 85.4%; P=0.001; CD133+/CD34+/KDR+ 48.4% vs 73.9%; P=0.05; and CD34+/CD45â 49.3% vs 78.4; P=0.006) was blunted in the South Asian group. CPC mobilization correlated with flow-mediated vasodilatation and l-NMMA significantly reduced exercise-induced CPC mobilization (CD34+/KDR+ â3.3% vs 68.4%; CD133+/CD34+/KDR+ 0.7% vs 71.4%; and CD34+/CD45â â30.5% vs 77.8%; all P<0.001).
Conclusionâ In humans, NO is critical for CPC mobilization in response to exercise. Reduced NO bioavailability may contribute to imbalance between vascular damage and repair mechanisms in South Asian men.British Heart Foundatio

Effects of prenatal and postnatal depression, and maternal stroking, at the glucocorticoid receptor gene
In animal models, prenatal and postnatal stress is associated with elevated hypothalamicâpituitary axis (HPA) reactivity mediated via altered glucocorticoid receptor (GR) gene expression. Postnatal tactile stimulation is associated with reduced HPA reactivity mediated via increased GR gene expression. In this first study in humans to examine the joint effects of prenatal and postnatal environmental exposures, we report that GR gene (NR3C1) 1-F promoter methylation in infants is elevated in the presence of increased maternal postnatal depression following low prenatal depression, and that this effect is reversed by self-reported stroking of the infants by their mothers over the first weeks of life
The influence of emotional reaction on help seeking by victims of school bullying
Research has started to focus on how victims of school bullying cope, but there is still very little understanding if why pupils cope in one way and not another. This paper aimed to examine the effects of gender, school-stage, frequency of victimization and different emotions (anger, vengeance, self-pity, indifference, and helplessness) upon the choice of social support that children report using. Questionnaires were completed by 6282 Maltese schoolchildren aged between 9 and 14 years old. Analyses revealed that specific patterns of emotion and victimization predict whether pupils report using certain sources of social support. Results are discussed in relation to their relevance for possible intervention, future research needs and implications for the theoretical framework used
HUMAN CALORIMETER FOR DIRECT AND INDIRECT MEASUREMENT OF 24H ENERGY-EXPENDITURE
1. A calorimeter for the continuous measurement of heat production and heat loss in the human subject, for at least 24 h, is described. The calorimeter operated on the heat-sink principle for direct calorimetry and an open-circuit system for indirect calorimetry.
2. Sensible heat loss was measured using a water-cooled heat exchanger, and the temperature of water entering the heat exchanger was controlled to maintain a mean temperature gradient of zero across the chamber walls.
3. Evaporative heat loss was determined from ingoing and outgoing wet-and-dry bulb temperatures and air flow-rates.
4. Problems associated with the calculation of evaporative heat loss and the estimation of the volume of incoming air in open-circuit systems are considered.
5. The calibration, limits of accuracy, sources of error and experiments with subjects are discussed
Patterns of satellite tagged hen harrier disappearances suggest widespread illegal killing on British grouse moors
Natural England commenced a Hen Harrier Recovery Project in 2002. This tracking study was funded exclusively by Natural England and is part of their on-going work on hen harrier conservation. We thank Hamish Smith and staff at the Hawk and Owl Trust for contributing data from four hen harriers they have tracked. Does bariatric surgery prior to total hip or knee arthroplasty reduce post-operative complications and improve clinical outcomes for obese patients? Systematic review and meta-analysis.
AIMS: Our aim was to determine whether, based on the current literature, bariatric surgery prior to total hip (THA) or total knee arthroplasty (TKA) reduces the complication rates and improves the outcome following arthroplasty in obese patients. METHODS: A systematic literature search was undertaken of published and unpublished databases on the 5 November 2015. All papers reporting studies comparing obese patients who had undergone bariatric surgery prior to arthroplasty, or not, were included. Each study was assessed using the Downs and Black appraisal tool. A meta-analysis of risk ratios (RR) and 95% confidence intervals (CI) was performed to determine the incidence of complications including wound infection, deep vein thrombosis (DVT), pulmonary embolism (PE), revision surgery and mortality. RESULTS: From 156 potential studies, five were considered to be eligible for inclusion in the study. A total of 23 348 patients (657 who had undergone bariatric surgery, 22 691 who had not) were analysed. The evidence-base was moderate in quality. There was no statistically significant difference in outcomes such as superficial wound infection (relative risk (RR) 1.88; 95% confidence interval (CI) 0.95 to 0.37), deep wound infection (RR 1.04; 95% CI 0.65 to 1.66), DVT (RR 0.57; 95% CI 0.13 to 2.44), PE (RR 0.51; 95% CI 0.03 to 8.26), revision surgery (RR 1.24; 95% CI 0.75 to 2.05) or mortality (RR 1.25; 95% CI 0.16 to 9.89) between the two groups. CONCLUSION: For most peri-operative outcomes, bariatric surgery prior to THA or TKA does not significantly reduce the complication rates or improve the clinical outcome. This study questions the previous belief that bariatric surgery prior to arthroplasty may improve the clinical outcomes for patients who are obese or morbidly obese. Mild cold effects on hunger, food intake, satiety and skin temperature in humans.
BACKGROUND: Mild cold exposure increases energy expenditure and can influence energy balance, but at the same time it does not increase appetite and energy intake. OBJECTIVE: To quantify dermal insulative cold response, we assessed thermal comfort and skin temperatures changes by infrared thermography. METHODS: We exposed healthy volunteers to either a single episode of environmental mild cold or thermoneutrality. We measured hunger sensation and actual free food intake. After a thermoneutral overnight stay, five males and five females were exposed to either 18°C (mild cold) or 24°C (thermoneutrality) for 2.5 h. Metabolic rate, vital signs, skin temperature, blood biochemistry, cold and hunger scores were measured at baseline and for every 30 min during the temperature intervention. This was followed by an ad libitum meal to obtain the actual desired energy intake after cold exposure. RESULTS: We could replicate the cold-induced increase in REE. But no differences were detected in hunger, food intake, or satiety after mild cold exposure compared with thermoneutrality. After long-term cold exposure, high cold sensation scores were reported, which were negatively correlated with thermogenesis. Skin temperature in the sternal area was tightly correlated with the increase in energy expenditure. CONCLUSIONS: It is concluded that short-term mild cold exposure increases energy expenditure without changes in food intake. Mild cold exposure resulted in significant thermal discomfort, which was negatively correlated with the increase in energy expenditure. Moreover, there is a great between-subject variability in cold response.
